A flow injection set-up based on potentiometric detection and gas diffusion device for the determination of total nitrogen in food is described. The detection system consisted of two ammonium-sensitive electrodes placed sequentially and each alternately operating as reference electrode. Tubular electrodes without an inner reference solution were prepared with a PVC membrane composed of nonactin in Tris (2-ethylhexyl) phosphate and potassium tetrakis (4-chlorophenyl) borate to reduce the membrane resistance. The food sample digests were inserted into the system, and the ammonium present was converted into ammonia gas. The gas diffused through a gas-permeable membrane to a buffer acceptor stream with a pH that ensured transformation to the ammonium cation, which was potentiometrically detected. Good agreement between FIA results and those provided by the reference procedure was obtained, with relative deviation errors below 5%. Using the proposed system, low reagent consumption is possible, a sampling rate of about 30 samples/h was achieved, as well as a good reproducibility for consecutive injections of the same sample (variation coefficient < 2%).  相似文献

Dietary exposure of Thais to pesticides during 1989-1996   总被引:1,自引:0,他引:1  
To monitor the exposure of the Thai to pesticide residues in foods, the intake of pesticides based on the total diet approach has been studied in Thailand continually since 1989. Food items were chosen and their proportions of daily consumption were calculated from the national food consumption survey conducted by the Department of Health in 1986. Seventy-seven items of food including drinking water were classified into 12 groups according to their sources of contamination and the analytical methods used. Shopping lists and standard procedures for collecting samples and cooking were developed. Twelve food composites from each of the 4 regions were analyzed for nearly 100 pesticides. All analyses used multiresidue analytical methods, and the total daily dietary intakes of pesticide residues were estimated from the average amount of food consumed by Thais. Among 24 pesticides found in the 8 year study, DDT, dimethoate, methamidophos, and parathion methyl were found every year. However, dietary intakes of all pesticides were far below the established acceptable daily intake. In 1999, the study methodology was redesigned to accommodate the new national food consumption survey data.  相似文献

A one day representative mixed diet of an adult Korean was collectedfrom the data based on the food intake of 108 healthy subjects between theages 20 and 50. Sampling for the Korean total diet was carried out by usinga market basket study based on the Korean standard food consumption schemereported by the Korean Nutrition Society. Average consumption frequency ofdifferent food items for a one day representative mixed diet of an adult Koreanand the amount of each item to prepare a one day Korean representative totaldiet are surveyed. The analytical methods involve both instrumental and radiochemicalneutron activation techniques developed for the determination of the elementsCs, I, Sr, Th and U in various kinds of food samples. Concentrations of traceelements including 5 important elements for radiological protection, U, Th,Cs, Sr and I in the Korean total diet and the 4 most frequently consumed Koreanfoodstuffs have been analyzed by neutron activation analysis. Detection limitsfor U, Th, Sr and I were improved to ppb levels by radiochemical separationafter neutron irradiation. Five biological NIST reference materials were alsoanalyzed for quality control of the analysis. Seventeen trace elements inthe Korean total diet and four Korean representative foodstuffs were alsoanalyzed quantitatively by instrumental neutron activation analysis.  相似文献

Results are reported for an interlaboratory validation study of 2 commercially available Iateral flow devices (dipstick tests) designed to detect peanut residues in food matrixes. The test samples used in this study were cookies containing peanuts at 7 different concentrations in the range of 0-30 mg peanuts/kg food matrix. The test samples with sufficient and proven homogeneity were prepared in our laboratory. The analyses of the samples (5 times per level by each laboratory) were performed by 18 laboratories worldwide which submitted a total of 1260 analytical results. One laboratory was found to be an outlier for one of the test kits. In general, both test kits performed well. However, some false-negative results were reported for all matrixes containing < 21 mg peanuts/kg cookie. It must be stressed that the test kits were challenged beyond their cut-off limits (> or = 5 mg/kg, depending on the food matrix). One test kit showed fewer false-negative results, but it led to some false-positive results for the blank materials. The sensitivity of the dipstick tests approaches that achieved with enzyme-linked immunosorbent assays.  相似文献

A simple method for the simultaneous and trace analysis of four synthetic food azo dyes including carmoisine, ponceau 4R, sunset yellow, and allura red from some foodstuff samples was developed by combining dispersive μ‐solid‐phase extraction and high‐performance liquid chromatography with diode array detection. Zein‐modified magnetic Fe3O4 nanoparticles were prepared and used for μ‐solid‐phase extraction of trace amounts of mentioned food dyes. The prepared modified magnetic nanoparticles were characterized by scanning electron microscopy and FTIR spectroscopy. The factors affecting the extraction of the target analytes such as pH, amount of sorbent, extraction time, type and volume of the desorption eluent, and desorption time were investigated. Under the optimized conditions, the method provided good repeatability with relative standard deviations lower than 5.8% (n = 9). Limit of detection values ranged between 0.3 and 0.9 ng/mL with relatively high enrichment factors (224–441). Comparing the obtained results indicated that Fe3O4 nanoparticles modified by zein biopolymer show better analytical application than bare magnetic nanoparticles. The proposed method was also applied for the determination of target synthetic food dyes in foodstuff samples such as carbonated beverage, snack, and candy samples.  相似文献

A method based on solid-phase microextraction and gas chromatography flame photometric detector for the determination of organophosphorus pesticides (OPPs) in food samples was described. Three kinds of vinyl crown ether polar fibers were prepared with sol-gel process and used for the analytes. The new coatings showed higher extraction efficiency and sensitivity for organophosphorus pesticides compared with commercial fibers—85 μm PA and 65 μm PDMS-DVB. Specifically, the benzo-15-crown-5 coating was the most effective for the target analytes. Several factors affecting the performance of SPME such as extraction temperature and time, salt addition, and dilution ratios of samples were optimized. The apparent recoveries of spiked food samples (apple juice, apple and tomato) were determined to be over 55.3% and the limits of detection (LODs) were in the range of 0.003-0.09 ng/g for the OPP studied. The method was applied to determine the concentrations of OPP in real food samples.  相似文献

This paper reports on the application of a potentiometric sensor array to the food analysis field, in order to distinguish simple tastes and to classify food samples. This array is formed by a set of non-specific all-solid-state potentiometric sensors and has been used in combination with principal component analysis (PCA) for the classification of food samples in batch and in flow injection mode. First attempt was to classify synthetic samples prepared with controlled variability. Once this ability is proven, satisfactory classification results are presented for commercial waters, orange-based drinks and tea samples. An interesting correlation is achieved between the natural juice content and its first calculated component, which allows for a very simple tool for screening purposes.  相似文献

A procedure for the multiresidue determination of organochlorine pesticides and polychlorinated biphenyls in fish muscle samples has been developed. The method is based on the microwave-assisted extraction (MAE) of food samples from an acetonitrile-water (95 + 5, v/v) mixture followed by SPE cleanup of the extracts and analysis by GC with an electron capture detector. MAE operational parameters, such as the extraction solvent, temperature, and time, were optimized with respect to the extraction efficiency of the target compounds from food samples with 10-13% fat content. The chosen extraction technique allows reduction of the solvent consumption and extraction time when compared with methods already used. Acetonitrile is a good extraction solvent for low-fat matrixes (2-20% fat content), such as fish samples, because it does not significantly dissolve the highly polar proteins, salts, and sugars commonly found in food and gives high recoveries of a wide polarity range of analytes. For purification, SPE using LC-Florisil was shown to be sufficient for the removal of coextracted substances. Recoveries > 78% with RSD values < 15% were obtained for all compounds under the selected conditions. Method quantification limits were in the 5-10 microg/kg range. The method was applied to the analysis of samples of herring (Clupea harengus) purchased at the local fish market. The method is rapid and reliable for the determination of organochlorine analytes in fish muscle.  相似文献

The consumption of food and drugs adulterated with female sex hormones can have an extremely adverse effect on human health. Therefore, developing appropriate monitoring methods for the identification of various exogenous female sex hormones is crucial for minimizing and eliminating the related health risks. Herein, 17 female hormones categorized into two groups: estrogen and progestin, were analyzed using reversed-phase liquid chromatography coupled to Orbitrap or triple quadrupole mass spectrometry. The fragmentation patterns for all compounds were discovered, and fragmented structures were also derived from them through liquid chromatography–high-resolution mass spectrometry followed by qualitative sample analysis. In addition, a quantitative analysis of 67 samples of illicit drugs and dietary supplements was performed using the validated liquid chromatography-tandem mass spectrometry method. Female hormone components were detected in two samples of an unauthorized injectable solution and a tablet-type drug. Medroxyprogesterone was detected in the samples in the range of 96.4–206 ng/g. Notably, eight components similar in structure to steroids were simultaneously detected as male sex hormones by confirming their fragmentation ion patterns using liquid chromatography–high-resolution mass spectrometry. The developed methods thus offer a dependable and practically applicable approach for the screening and detection of exogenous female sex hormones in real food and drug samples to ensure public health.  相似文献

A new rapid method for the determination of actinides in food samples has been developed at the Savannah River Site Environmental Lab (Aiken, SC, USA) that can be used for emergency response or routine food samples. If a radiological dispersive device or improvised nuclear device event occurs, there will be a urgent need for rapid analyzes of many different environmental matrices, as well as food samples, to support dose mitigation and protect general populations from radioactivity that may enter the food chain. The recent accident at Fukushima nuclear power plant in March, 2011 reinforces the need to have rapid analyzes for radionuclides in environmental and food samples. The new method to determine actinides in food samples utilizes a furnace ashing step, a rapid sodium hydroxide fusion method, a lanthanum fluoride matrix removal step, and a column separation process with stacked TEVA, TRU, and DGA resin cartridges. The furnace ashing and rapid fusion steps are performed in relatively inexpensive, reusable zirconium crucibles. Alpha emitters are prepared using rare earth micro precipitation for counting by alpha spectrometry. The method showed high chemical recoveries and effective removal of interferences. The determination of actinides in food samples can be performed in less than 8 h for 10 g samples with excellent quality for emergency samples using short count times. Larger food samples (100 g) may be processed in 24 h or less. The rapid fusion technique is a rugged sample digestion method that ensures that any refractory actinide particles are effectively digested. This method can be used to meet the derived intervention level guidelines recommended by the U.S. Food and Drug Administrations.  相似文献

The establishment of an immunoaffinity chromatography (IAC) for simultaneously selective extraction of four illegal colorants Sudan dyes (Sudan I, II, II and IV) from food samples was described. The IAC column was constructed by covalently coupling monoclonal antibody (mAb) against Sudan I to CNBr-activated Sepharose 4B and packed into a common solid phase extraction (SPE) cartridge. It was observed that IAC column was able to separately capture Sudan I, II, III and IV with maximum capacity of 295, 156, 184 and 173ng, respectively. The extraction conditions including loading, washing and eluting solutions were carefully optimized. Under optimal conditions, the extraction recoveries of the IAC column for Sudan I-IV at two different spiked concentrations were within 95.3-106.9%. After 50 times repeated usage, 64% of the maximum capacity was still remained. Six food samples randomly collected from local supermarket without spiking Sudan dyes were extracted with IAC column and detected by high performance liquid chromatography (HPLC). It was found that there was no detectable Sudan II, III and IV in all six food samples, but Sudan I with the content of 2.7-134.5ngg(-1) was detected in three food samples. To further verify the extraction efficiency, other three negative samples were spiked with Sudan I-IV at the concentrations of 20ngg(-1) and 50ngg(-1), which were then extracted with IAC column. The extraction recoveries and relative standard deviation (RSD) were 68.6-96.0% and 4.8-15.2%, respectively, demonstrating the feasibility of the prepared IAC column for Sudan dyes extraction.  相似文献

This study aimed to develop polyelectrolyte‐structured antimicrobial food packaging materials that do not contain any antimicrobial agents. Cationic hydroxyethyl cellulose was synthesized and characterized by Fourier‐transform infrared, 1H NMR, and 13C NMR spectroscopy. Its nitrogen content was determined by Kjeldahl method. Polyelectrolyte‐structured antimicrobial food packaging materials were prepared using hydroxyethyl cellulose, cationic hydroxyethyl cellulose, and sodium alginate. Antimicrobial activity of materials was defined by inhibition zone method (disc diffusion method). Thermal stability of samples was evaluated by thermal gravimetric analysis and differential scanning calorimetry. Surface morphology of samples was investigated by SEM. The obtained results prove that produced food packaging materials have good thermal and antimicrobial properties, and they can be used as food packaging material in many industries.  相似文献

A liquid chromatography/electrospray ionization time-of-flight mass spectrometry (LC/ESI-TOFMS) method has been developed for the determination of 12 pesticides (namely, carbendazim, thiabendazole, imazalil, tridemorph, triadimefon, bitertanol, prochloraz, flutriafol, myclobutanil, iprodione, diphenylamine and procymidone) in fruit-based baby food (multi-fruit jars and juices intended for infant consumption). The developed method consists of a sample treatment step based on liquid-liquid extraction using acetonitrile, followed by a clean-up step based on dispersive solid-phase extraction (SPE) with a primary-secondary amine (PSA). Multi-fruit and apple juices were processed by a SPE procedure using Oasis HLB cartridges. Subsequent identification and quantitation was accomplished by LC/ESI-TOFMS analysis: the confirmation of the target pesticides was based on accurate mass measurements of selected ions (protonated molecules ([M+H]+) and fragment ions). Confirmation studies were accomplished at low concentration levels (10 microg kg-1) and accuracy errors lower than 2 ppm were obtained in most cases. Baby food extracts spiked at 10 microg kg-1 fortification level yielded average recoveries in the range 78-105% with relative standard deviations less than 10% for most of the analytes. Limits of detection (LODs) were between 0.1 and 4 microg kg-1 depending on the pesticide studied. Finally, the proposed method was applied to a total of 33 baby food samples from Spain and the United Kingdom. Although imazalil, thiabendazole and carbendazim were detected in a high number--over 60%- of baby food samples, none of the samples tested were found to be above the 0.01 mg kg-1 EU standard.  相似文献

A new selective and fast procedure based on magnetic dispersive solid‐phase extraction and zeta potential analysis is proposed for the determination of Cd (II) in some food samples. In the developed method, novel magnetic nanoparticles modified with thioglycolic acid were synthesized/characterized and also applied for quantitative determination of trace amounts of Cd (II) in food samples. The prepared nanoparticles were characterized via infrared spectroscopy, electron microscopy and adsorption–desorption experiments. These magnetic nanocomposites carrying Cd (II) could be easily separated from real samples simply by applying an external magnetic field; no filtration or centrifugation was necessary. Several parameters affecting the analytical performance, such as sample pH, amounts of nanocomposite sample, desorption solution volumes and coexisting ions, were investigated in detail. The detection limit of the method was 0.1 μg l?1 while the relative standard deviation was 1.2% for a Cd (II) concentration of 0.5 mg l?1. The proposed method was successfully applied to food sample analysis and standard reference material samples with satisfactory results, and excellent recoveries were obtained in the range 95–99% even when the matrix, such as sea water, was complex.  相似文献

Meat samples are prepared by passing meat through a food chopper, bowl cutter, or food processor, subsampling the meat, and mixing the meat with granular diatomaceous earth. No drying step is necessary. Supercritical CO2 is then used to extract crude fat (which is defined as the components of meat that are extractable with petroleum ether, without digestion of the sample). Extracted material is deposited on glass wool contained in collection vials. After removal of any residual moisture from the extracts, percent crude fat is determined by weight gain of the collection vial. This method has been peer-verified by 3 laboratories, for a wide variety of raw and processed meat products containing 6-28% crude fat. Samples were prepared at the submitting laboratory. Ground samples were split into 4 portions, packed in Whirlpack bags, and immediately frozen. Frozen samples were sent by overnight room temperature, and percent fat was determined (in triplicate), without further processing of the samples. Analysis of the samples was completed within 1 week of sample prepara. tion. On the basis of this study, it can be estimated that all repeatability and reproducibility values are <3.0. Mean accuracy of the direct gravimetric supercritical fluid extraction method for meat samples ranged from +0.22 to -1.41 when the method was compared with AOAC Method 960.39. Interferences are unlikely but would include any nonfat substance that is added to (processed) meat, is soluble in nonpolar solvents, and is present in a quantity that would alter results. This method is expected to perform equally well for all meats with fat content within the stated range of applicability.  相似文献

A liquid chromatography-tandem mass spectrometry method has been developed for the determination of residues of alkylbenzyldimethylammonium, didecyldimethylammonium, didodecyldimethylammonium, and benzyldodecylhydroxyethylammonium compounds in various food matrixes. These quaternary ammonium compounds (QAs) are used in the food industry as disinfectants. According to the Dutch Food Law, the total mass (expressed as cetyltrimethylammonium chloride) of QAs in food products shall not exceed the legislative limit of 0.5 mg/kg. Samples were extracted by a simple salting-out procedure, using acetonitrile and sodium chloride; about 100 samples could be prepared and analyzed daily. Special care had to be taken to thoroughly homogenize samples and to avoid the use of contaminated labware. The method was validated by a procedure in compliance with EU Directive 2002/657. From the matrixes of ice cream and minced meat, recoveries of more than 95% with a relative standard deviation of about 3% were obtained by 3 different analysts (n = 54). Detection limits were in the low microg/kg range. The decision limit (CCalpha) was determined to be 0.55 mg/kg. Dairy and meat products, collected in The Netherlands, were analyzed (761 samples). In 1% of the meat samples, 2% of the ice cream and milkshake samples, and 24% of the whipped cream samples, the Dutch legislative limit was exceeded. Over 2000 injections could be performed on a single column without deterioration of the peak shapes or recoveries.  相似文献

A novel on-line solid-phase microextraction–high-performance liquid chromatography(SPME–HPLC)system was developed for the determination of heterocyclic aromatic amines(HAAs) in food samples. A poly(vinylphenylboronic acid-co-ethylene glycol dimethacrylate) polymer monolith was prepared for on-line efficient extraction and large-volume injection was used to increase the sensitivity of detection.The polymermonolith, based on a ternary porogen, was prepared by in situ polymerization of vinylphenylboronic acid(VPBA) and ethylene glycol dimethacrylate(EGDMA) in a fused-silica capillary column. It showed good permeability, high extraction capacity, and high selectivity. The column-tocolumn reproducibility was satisfactory, and the enrichment factors for HAAs were 3746–7414.Conditions influencing the on-line extraction efficiency, including p H of sample solutions, flow rate of extraction and desorption, and desorption volume, were investigated. The proposed method had low limit of detection(0.10–0.15 ng/L) and good linearity. Trace HAAs in roast beef and lamb samples were determined, and the amounts of 2-amino-3-methylimidazo[4,5-f]quinoline, 2-amino-3,4-dimethylimidazo[4,5-f]quinoline, 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline, 2-amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline, and 2-amino-3,4,7,8-tetramethyl-3H-imidazo[4,5-f]quinoxaline in these samples were 0.235–2.08 ng/g. The recoveries for the five HAAs ranged from74.3% to 119%, and the relative standard deviation(RSDs) were less than 8.2%. The results showed that the proposed on-line method was highly sensitive for monitoring HAAs in different food samples.  相似文献

The metabolism of selenium (Se) in the human body has yet not completely been unravelled and hence, an efficient method for characterization and on-line monitoring of the main Se-compound in human urine after consumption of Se-rich food was developed. Total Se-concentration in human urine after consumption of several Se-rich products was measured with inductively coupled plasma mass spectrometry (ICP-MS). The highest Se concentration in urine was observed after 4-10 h. The urine samples were brought onto a reversed phase column and the Se was detected by ICP-MS. Parameters for liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS-MS) measurements were optimized by using commercially available sugars, because it is known that some of the urinary metabolites contain a sugar moiety. In order to characterize the predominant Se-metabolite, it was necessary to extensively clean-up the sample and preconcentrate the species. The main metabolite was measured on its precursor ion on three different m/z according to three isotopes of Se. Relative peak surfaces matched the relative abundances of the isotopes. The product ions could be measured in a human urine sample in accordance to the product ions of the commercially available sugars. Moreover, the evidence of a selenosugar was demonstrated by the use of the Se-isotopes when measuring the product ions. LC-ESI-MS-MS was proven to be very efficient for the characterization of the main urinary Se-metabolite and can be used for on-line monitoring of the compound in urine samples. The method can be extended for clinical screening after consumption of Se-(en)rich(ed) food by use of the Se-isotopic profile and/or of the typical product ions of (methyl)-N-acetyl-hexosamines.  相似文献

A method was developed for the detection of Listeria monocytogenes in food. The method is based on real-time PCR using hydrolysis probes (5' Nuclease). This advanced PCR method was designed to reduce the time necessary to achieve results from PCR reactions and to enable the user to monitor the amplification of the PCR product simultaneously, in real-time. After DNA isolation using the BIOTECON foodproof ShortPrep II Kit designed for the rapid preparation of L. monocytogenes DNA for direct use in PCR, the real-time detection of L. monocytogenes DNA is carried out using the foodproof Listeria monocytogenes Detection Kit. The kit provides primers and hydrolysis probes for sequence-specific detection, convenient premixed reagents, and controls for reliable interpretation of results. For the internal comparison study, three different foods (soft cheese, coalfish, and smoked ham) were analyzed, chosen from the 15 food groups recommended by the AOAC Research Institute for detection of L. monocytogenes. From each food, 20 samples were inoculated with a low level (1-10 CFU/25 g) and 20 samples with a high level (10-50 CFU/25 g) of L. monocytogenes. Additionally, five nonspiked samples were prepared from each food. Depending on the matrix, the food samples were examined with the test kits and compared with the cultural methods according to the U.S. Food and Drug Administration's Bacteriological Analytical Manual or the U.S. Department of Agriculture/Food Safety and Inspection Service Microbiology Laboratory Guidebook.  相似文献
