Rigorous numerical modeling of optical systems has attracted interest in diverse research areas ranging from biophotonics to photolithography. We report the full-vector electromagnetic numerical simulation of a broadband optical imaging system with partially coherent and unpolarized illumination. The scattering of light from the sample is calculated using the finite-difference time-domain (FDTD) numerical method. Geometrical optics principles are applied to the scattered light to obtain the intensity distribution at the image plane. Multilayered object spaces are also supported by our algorithm. For the first time, numerical FDTD calculations are directly compared to and shown to agree well with broadband experimental microscopy results.  相似文献

由于光在水中传输时的衰减和散射效应, 水下成像系统通常很难达到令人满意的成像效果, 而成像过程中由于光散射产生的背景光则是造成水下图像退化的主要原因. 本文对自然光照和人工光照两种不同光照条件下的水下成像背景光进行了建模和仿真分析. 结果显示: 自然光照下的无穷远处背景光与衰减系数成反比, 与散射系数成正比; 人工光照下的无穷远处背景光则与衰减系数、散射系数和相机-光源距离有关. 两种光照条件下的背景光都可以用无穷远处背景光的指数衰减表达式来表示. 水下图像背景光的强度主要与水体光学参数、相机-景物的距离、相机-光源的距离及相机成像角等因素有关. 本文的研究结论不仅可用于水下成像系统的设计与优化, 也可用来从水下图像背景光中估计水体光学参数、相机-景物距离等图像信息.  相似文献

光声成像技术是一种非常有前景的前列腺早期检测与成像的新技术. 在现有前列腺光声成像技术中, 通常采用的腺体外光源辐照方式由于受外围组织的影响, 容易减弱深部前列腺组织的光能量吸收, 导致辐照范围减小, 引起光声成像深度不足, 难以实现无损前列腺癌的检测. 本文基于前列腺组织的结构特征, 依据一种以柱状弥散光纤在尿道对前列腺实施光辐照、并利用外置于直肠内的长焦区聚焦式超声换能器检测光声信号的前列腺光声扫描成像技术, 构建了光声成像实验系统, 开展了仿体实验. 测试结果表明, 系统能够实现样品中吸收体的定位和成像, 结合柱弥散光源体内辐照可使成像深度提高, 同时侧向成像范围也较大. 初步结果表明, 借助柱弥散光源进行体内光辐照的光声信号新激发方式结合长焦区超声探头在前列腺癌早期无创诊断上具有潜在的应用前景.  相似文献

散射介质对光的散射是当前限制光学成像深度或距离的一个严重的问题.本文首先数值模拟比较了光透过随机散射介质成像研究中常用的基于光学记忆效应(memory effect, ME)和自相关(autocorrelation, AC)方法的HIOER算法和乒乓(Ping-Pang, PP)算法的优缺点.通过对HIOER算法和PP算法的恢复效果和迭代次数进行比较,发现PP算法在保持较高恢复效果的前提下拥有更快的运行速度.实验中,利用连续HeNe激光器和旋转毛玻璃产生赝热光源,通过物镜对随机散射介质后数毫米距离内的不同形状物体进行了单帧成像,并采用PP算法成功地恢复出微米量级物体的实际图像.这一研究结果将进一步促进ME和AC方法在深层生物组织医学成像研究上的应用.最后,实验研究了不同的物镜和散射介质的间距对成像恢复的放大率、分辨率和图像强度的影响特性,并进行了详细研究.  相似文献

A new microscope combines optical sectioning by fluorophore excitation using a single light sheet with structured illumination. Several images with laterally intensity-modulated light sheets are recorded from scattering fluorescent specimens. By applying a simple data processing scheme, the nonmodulated volumes are identified. The blurred features become dark, and the resultant images are improved in terms of contrast and resolution. Hence, the instrument is capable of discriminating against contributions to the image that are induced by the optical properties of the specimen. The new microscope's capabilities are demonstrated by imaging the internals of the head of an adult Drosophila melanogaster (fruit fly) expressing green fluorescent protein-labeled polycomb proteins.  相似文献

Dark-field illumination is known to enhance scattering contrast in optical microscopy. We combined this concept with Fourier domain optical coherence microscopy (OCM). The detection and illumination paths are decoupled, and only the scattered light originating from the sample generates the tomogram signal, whereas any specular reflection is highly suppressed. We analyze and discuss this dark-field OCM concept and present its superior imaging quality on live cell samples.  相似文献

The scattering of plane acoustic waves at area expansions in flow ducts is analysed using the vortex sheet model where the flow at the expansion is modelled as a jet, with a vortex sheet emanating from the edge. Of particular interest is the influence of the flow field on acoustic scattering and absorption.First, it is demonstrated that the stability properties of the shear layer can be simulated by modifying the edge condition within the vortex sheet model. To this end the accuracy for the region where the shear layer is changing from unstable to stable is improved by introducing a gradually relaxed Kutta edge condition with empirically derived coefficients. For low Strouhal numbers the vortex sheet model applies and for higher Strouhal numbers the two models converge.Second, it is demonstrated that the acoustic transmission through the jet expansion region can be determined by neglecting the scattering there. Incorporating this assumption, the vortex sheet model reproduces well the experimental results on transmission and absorption for an area expansion. This result supports the assumption that the main part of the scattering occurs at the area expansion at least for the low-frequency range. Furthermore, the influence of the flow field is particularly strong for small Strouhal numbers.  相似文献

光声成像是采用“光激发-超声波成像”的新型成像技术,它是检测强散射介质内部光吸收分布的一种有效医学影像技术.用短脉冲激光照射强散射介质(如生物组织),强散射介质由于光声效应产生超声信号,使用具有成像能力的声透镜把声压分布成像于像面上,然后利用具有空间分辨能力的阵列光声传感器,把同一像面上的光声信号强度记录下来,最后根据光声信号强度的空间分布进行图像重组.根据成像系统物像共轭原理,同一物平面的光声信号到达像面的时延相等,而不同物面的光声信号到达同一个探测器平面的时延各不相同,因此,利用BOXCAR的门控积分 关键词: 光声层析成像 声透镜 光声信号  相似文献

We study theoretically the focusing properties of a two‐dimensional periodic array of silver‐coated silica nanospheres. The array contains an adsorbed nanosphere of the same type which serves as point source when the array is illuminated by a plane wave. By using a rigorous multiple‐scattering theory we show that under plane wave illumination, the array focuses the incident light in a tight area with size of about 1/70 of the wavelength. At the same time, the near‐field is amplified by three orders of magnitude. The frequency corresponding to the focusing and amplification effects depends on the thickness of the silver nanoshell offering a tunable response of the structure. We present measurements of the scalar-field light scattering of individual dimer, trimer, and tetrahedron shapes among colloidal clusters. By measuring the electric field with quantitative phase imaging at the sample plane and then numerically propagating to the far-field scattering plane, the two-dimensional light-scattering patterns from individual colloidal clusters are effectively and precisely retrieved. The measured scattering patterns are consistent with simulated patterns calculated from the generalized multiparticle Mie solution.  相似文献

水下偏振成像技术是目前水下成像研究的热点,由于自然光在水中衰减大,水下成像系统多采用主动照明方式。针对分焦平面偏振成像系统中偏振照明光源与偏振探测像元偏振方向不匹配引起采集图像偏振信息存在的偏差,进而影响目标图像增强质量的问题,提出了一种分焦平面偏振成像系统光源标定方法。阐述了偏振光源的标定原理,然后给出偏振光源标定的实施步骤,最后采用偏振去雾算法和图像质量评价方法对标定前后的水下目标图像进行了图像增强和图像质量评价。评价结果表明,标定后的增强图像质量优于未标定的增强图像质量,平均梯度最大提升了2.48倍。该标定方法简单有效,实用性强,适用于分焦平面偏振成像系统偏振光源标定。  相似文献

 中子数字成像过程中,散射中子可降低像质致使提取样品的定量信息变得困难。针对该情况,分析了中子微光成像系统图像散射降质的原理,采用点扩展函数的叠加来表征散射中子引起图像降质的过程,借助于蒙特卡罗方法(MC)对点扩展函数进行模拟和建模,将点扩展函数描述为样品厚度以及样品到探测器距离为参量的解析函数,研究了点扩展函数的计算方法。研究结果表明,借助于MC方法构建系统的点扩展函数之解析函数,并利用该函数对中子图像进行散射校正,是一种行之有效的定量中子数字成像散射校正方法。  相似文献

非均匀发光光源、大视场角等因素会造成光学成像检测系统像面照度分布不均匀,进而导致检测效率下降。研究非均匀发光光源和大视场角对像面照度均匀性的影响程度,首先建立光源间距与受光面接收照度间的关系模型,仿真分析不同LED间距对像面照度均匀性的影响,然后建立光学耦合系统的物面张角和像面照度间的关系模型,仿真分析物面张角对像面照度均匀性的影响。实验结果表明:在非均匀发光光源和大视场角的作用下,检测系统会造成像面照度严重不均匀现象。研究结果为后续像面照度校正算法设计提供了理论依据。  相似文献

The optical design of a two‐dimensional imaging soft X‐ray spectrometer is described. A monochromator will produce a dispersed spectrum in a narrow vertical illuminated stripe (~2 µm wide by ~2 mm tall) on a sample. The spectrometer will use inelastically scattered X‐rays to image the extended field on the sample in the incident photon energy direction (vertical), resolving the incident photon energy. At the same time it will image and disperse the scattered photons in the orthogonal (horizontal) direction, resolving the scattered photon energy. The principal challenge is to design a system that images from the flat‐field illumination of the sample to the flat field of the detector and to achieve sufficiently high spectral resolution. This spectrometer provides a completely parallel resonant inelastic X‐ray scattering measurement at high spectral resolution (~30000) over the energy bandwidth (~5 eV) of a soft X‐ray absorption resonance.  相似文献

The fluorescence from the out-of-focus region excited by the sidelobes of a Bessel beam is the major concern for light-sheet fluorescence microscopy(LSFM) with Bessel beam plane illumination. Here, we propose a method of applying the subtractive imaging to overcome the limitation of the conventional LSFM with Bessel beam plane illumination. In the proposed method, the sample is imaged twice by line scanning using the extended solid Bessel beam and the ring-like Bessel beam. By subtracting between the two images with similar out-of-focus blur, the improved image quality with the suppression of the Bessel beam sidelobes and enhanced sectioning ability with improved contrast are demonstrated.  相似文献

Recently, a light sheet-based technique called objective-coupled planar illumination (OCPI) microscopy [Holekamp et al., Neuron 57, 661 (2008)] was shown to permit low-phototoxicity, high-speed, three-dimensional fluorescence imaging of extended tissue samples. Here, we introduce two major improvements in OCPI microscopy. First, we miniaturize the objective coupler by using a uniaxial gradient-index lens to produce the light sheet. Second, we demonstrate theoretically and experimentally that refractive index mismatch at the fluid/tissue interface causes a significant defocus aberration. By introducing the ability to tune the angle of the light sheet, we show that defocus correction in a miniaturized OCPI microscope leads to a significant improvement in image sharpness deeper into tissue.  相似文献
