Two simple and accurate spectrophotometric methods are presented for the determination of beta-lactam drugs, flucloxacillin (Fluclox) and dicloxacillin (Diclox), in pure and in different pharmaceutical preparations. The charge transfer (CT) reactions between Fluclox and Diclox as electron donors and 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) pi-acceptor and potassium iodate via oxidation reduction reaction where the highly coloured complex species or the liberated iodine have been spectrophotometrically studied. The optimum experimental conditions have been studied carefully. Beer's law is obeyed over the concentration range of 2-450 microg ml(-1) for Fluclox and 10-450 microg ml(-1) for Diclox using DDQ reagent and at 50-550 microg ml(-1) for Fluclox and 50-560 microg ml(-1) for Diclox using iodate method, respectively. For more accurate results, Ringbom optimum concentration range is calculated and found to be 6-450 and 15-450 microg ml(-1) for Fluclox and Diclox using DDQ, respectively, and 65-550 and 63-560 microg ml(-1) for Fluclox and Diclox using iodine, respectively. The Sandell sensitivity is found to be 0.018 and 0.011 microg cm(-2) for DDQ method and 0.013 and 0.011 microg cm(-2) for iodate method for Fluclox and Diclox, respectively, which indicates the high sensitivity of both methods. Standard deviation (S.D.=0.01-0.80 and 0.07-0.98) and relative standard deviation (R.S.D.=0.13-0.44 and 0.11-0.82%) (n=5) for DDQ and iodate methods, respectively, refer to the high accuracy and precision of the proposed methods. These results are also confirmed by between-day precision of percent recovery of 99.87-100.2 and 99.90-100% for Fluclox and Diclox by DDQ method and 99.88-100.1 and 99.30-100.2% for Fluclox and Diclox by iodate method, respectively. These data are comparable to those obtained by British and American pharmacopoeias assay for the determination of Fluclox and Diclox in raw materials and in pharmaceutical preparations.  相似文献

Five spectrophotometric methods and one fluorimetric method have been developed and validated for the analysis of clozapine. The spectrophotometric methods were based on the charge-transfer complexation reaction between clozapine as electron donor and each of iodine as sigma-acceptor or 7,7,8,8-tetracyanoquinondimethane (TCNQ), 2,3-dichloro-5,6-dicyano-1,4-benzo-quinone (DDQ), tetracyanoethane (TCNE), and p-chloranilic acid (pCA) as pi-acceptors. The obtained complexes were measured spectrophotometrically at 365, 843, 460, 414, and 520 nm for iodine, TCNQ, DDQ, TCNE, and pCA, respectively. The fluorimetric method was based on the oxidation of clozapine in the presence of perchloric acid by cerium (IV), and subsequent measuring the fluorescence of the produced cerium (III) fluorimetrically at lambda(excitation) 260 and lambda(emission) 355 nm. Under the optimum assay conditions, Beer's law was obeyed at concentrations ranged from 4-200 microg mL(-1) for the spectrophotometric methods and from 24-250 ng mL(-1) for the fluorimetric method. The limits of detection for the spectrophotometric methods were 1.12, 1.76, 2.22, 0.95, and 13.26 microg mL(-1) for iodine, TCNQ, DDQ, TCNE, and pCA, respectively. The limit of detection for the fluorimetric method was 6.69 ng mL(-1). The proposed methods were successfully applied to the analysis of clozapine in tablets with good recoveries. The fluorimetric method could also be applied to the analysis of clozapine in spiked urine samples. The molar ratios and the reaction mechanisms were investigated.  相似文献

The charge transphere (CT) reactions between CQP and PYM as electron donors and 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) pi-acceptor and iodine sigma-acceptor reagents to give highly coloured complex species have been spectrophotometrically studied. The optimum experimental conditions have been studied carefully. Beer' law is obeyed over the concentration range of 1.0-15 microg ml(-1) for CQP and 1.0-40 microg ml(-1) for PYM using I(2) and at 5.0-53 microg ml(-1) for CQP and 1.0-46 microg ml(-1) for PYM using DDQ reagents, respectively. For more accurate results, Ringbom optimum concentration range is calculated and found to be 10-53 and 8-46 microg ml(-1) for CQP and PYM using DDQ, respectively and 5-15 and 8-40 microg ml(-1) for CQP and PYM using iodine, respectively. The Sandell sensitivity is found to be 0.038 and 0.046 g cm(-2) for DDQ method and 0.0078 and 0.056 g cm(-2) for I(2) method for CQP and PYM, respectively which indicates the high sensitivity of both methods. Standard deviation (S.D.=0.012-0.014 and 0.013-0.015) and relative standard deviation (R.S.D.=0.09-1.4 and 1.3-1.5%) (n=5) for DDQ and I(2) methods respectively, refer to the high accuracy and precision of the proposed methods. These results are also confirmed by between day precision of percent recovery of 99-100.6%, and 98-101% for CQP and PYM by DDQ method and 99-102% and 99.2-101.4% for CQP and PYM by I(2) method respectively. These data are comparable to those obtained by British and American pharmacopoeias assay for the determination of CQP and PYM in raw materials and in pharmaceutical preparations.  相似文献

A simple and rapid spectrophotometric method has been developed for the determination of tricyclic anti-depressant drugs such as trazodone (TZH), amineptine (APH) and amitriptyline (ATPH) hydrochlorides in pure form and in different pharmaceutical preparations. The charge transfer (CT) reaction between TZH, APH and ATPH as electron donors and TCNQ as electron acceptor was utilized for their spectrophotometric determination. The optimum experimental conditions, like time, temperature, stoichiometry, solvents, for the CT complex formation are established. The method permits the determination of TZH, APH and ATPH over a concentration range of 10-400, 10-440 and 10-300 microg ml(-1), respectively. The sensitivity (S) is found to be 0.09, 0.087 and 0.069 g cm(-2) for TZH, APH and ATPH, respectively. The SD values are found to be 0.146-0.293, 0.154-0.285 and 0.091-0.212 and RSD values are 0.142-1.92, 0.297-1.92 and 0.212-0.915 for TZH, APH and ATPH, respectively. The low values of the relative standard deviation indicate the high accuracy and precision of the method. The mean recovery values obtained together with a high correlation coefficient values, amount in the range 98-101.5, 98.7-102.9 and 93-101.9 for TZH, APH and ATPH, respectively. The method is applicable for the assay of the investigated drugs in different dosage forms and the results are in good agreement with those obtained by the official method.  相似文献

Two simple and accurate spectrophotometric methods for determination of Rifampicin (RIF) are described. The first method is based on charge transfer (CT) complex formation of the drug with three pi-electron acceptors either 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), 7,7,7,8-Tetracyanoquinodimethane (TCNQ) or 2,3,5,6-Tetrachloro-1,4-benzoquinone (p-chloranil) in acetonitrile. The method is followed spectrophotometrically by measuring the maximum absorbance at 584 nm, 761 nm (680 nm) or 560 nm for DDQ, TCNQ and p-chloranil, respectively. Under the optimized experimental conditions, the calibration curves showed a linear relationship over the concentration ranges of 5-140 microg/ml, 2-45 microg/ml (5-120 microg/ml) and 15-200 microg/ml, respectively. The second method is based on the reaction of RIF with iron(III) forming a water insoluble violet complex which is extracted into chloroform. The method determines RIF in concentration range of 10-240 microg/ml at 540 nm. The proposed methods applied to determination of RIF in capsule, human serum and urine samples with good accuracy and precision. The results were compared statistically with the official method and showed no significant different between the methods compared in terms of accuracy and precision.  相似文献

A simple, accurate, and sensitive spectrophotometric method for analysis of selective serotonin reuptake inhibitors (SSRIs) has been developed and validated. The analysis was based on the formation of colored charge-transfer complexes between the intact molecule of SSRI drug as an n-electron donor and each of tetracyanoquinodimethane (TCNQ) or p-chloranilic acid (pCA) as electron acceptors. The formed complexes were measured spectrophotometrically at 842 and 520 nm for TCNQ and pCA, respectively. Different variables and parameters affecting the reactions were studied and optimized. Under the optimum reaction conditions, linear relationships with good correlation coefficients (0.9975-0.9996) were found between the absorbances and the concentrations of the investigated drugs in the concentration ranges of 4-50 and 20-400 microg/mL with TCNQ and pCA, respectively. With all the investigated drugs, TCNQ gave more sensitive assays than pCA; the limits of assay detection were 2.5-4.8 and 20-40 microg/mL with TCNQ and pCA, respectively. The intra- and interassay precisions were satisfactory; the relative standard deviations did not exceed 2%. The proposed procedures were successfully applied to the analysis of the studied drugs in pure form and pharmaceutical formulations with good accuracy; the recovery values were 98.4-102.8 +/- 1.24-1.81%. The results obtained from the proposed method were statistically comparable with those obtained from the previously reported methods.  相似文献

A polythiophene derivative substituted with electron‐rich alkynes as a side chain was synthesized using the Suzuki polycondensation reaction. The electron‐rich alkynes underwent the “click chemistry”‐type quantitative addition reaction with strong acceptor molecules, such as tetracyanoethylene (TCNE) and 7,7,8,8‐tetracyanoquinodimethane (TCNQ), resulting in the formation of donor–acceptor chromophores. All polymers showed excellent solubilities in the common organic solvents as well as good thermal stabilities with their 5% decomposition temperatures exceeding 230 °C. The TCNE‐/TCNQ‐adducted polymers displayed well‐defined charge‐transfer (CT) bands in the low energy region. The CT energy of the TCNE‐adducted polymer was 2.56 eV (484 nm), which was much greater than that of the TCNQ‐adducted polymer [1.65 eV (750 nm)]. This result was supported by the electrochemical measurements. The electrochemical band gaps of the TCNE‐adducted polymers were much greater than those of the corresponding TCNQ‐adducted polymers. Furthermore, the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) energy levels, determined from the first oxidation and first reduction peak potentials, respectively, decreased with the increasing acceptor addition amount. All these results suggested that the energy levels of the polythiophene derivative can be tuned by varying the species and amount of the acceptor molecules using this postfunctionalization method. © 2010 Wiley Periodicals, Inc. 用2,4-甲苯二异氰酸酯将蓖麻油及聚乙二醇(PEG)偶联成聚氨酯,同时通过自由基聚合使2-乙烯基吡啶(VP)与蓖麻油的双键共聚,生成互穿网络聚合物(IPN),其产率及凝胶含量大多超过93%,力学性能很好.当IPN与LiClO4络合后呈现离子导电现象;与TCNE或TCNQ络合后产生电子导电,与LiClO4及TCNE(TCNQ)络合后,呈现比上述两种导电率更大且具有协同效应的混合导电率,在25℃混合电导率可达到10-3~10-2S/cm.  相似文献

The interaction of the interesting polynitrogen cyclic base 1,4,7-trimethyl-1,4,7-triazacyclononane (TMTACN) with the sigma-acceptor iodine and pi-acceptors tetracyanoethylene (TCNE), 7,7,8,8-tetracyanoquinodimethane (TCNQ) and tetrachloro-p-benzoquinone (chloranil) have been studied spectrophotometrically and cyclic voltametrically in chloroform at 20 degrees C. Based on the obtained data, the formed charge-transfer complexes were formulated as [(TMTACN)I](+).I(3)(-), [(TMTACN)(TCNE)(5)], [(TMTACN)(TCNQ)(3)] and [(TMTACN)(chloranil)(3)] where the stoichiometry of the reactions, donor:acceptor molar ratios, were shown to equal 1:2 for iodine complex, 1:3 for chloranil and TCNQ complexes and 1:5 for TCNE complex.  相似文献

An analytical method has been developed for the separation of glyoxal (Go), methylglyoxal (MGo), and dimethylglyoxal (DMGo) by MEKC using stilbenediamine (SD) as derivatizing reagent, separation time 6.5 min, SDS as micellar medium at pH 8, and sodium tetraborate (0.1 M) as buffer. Uncoated fused-silica capillary, effective length 50 cm x 75 microm id; applied voltage 20 kV and photodiode array detection, were used. Calibration was linear within 0.02-150 microg/mL with detection limits 3.5-5.8 ng/mL. Go and MGo, observed for diabetic and healthy volunteers, were within 0.098-0.193 microg/mL Go and 0.106-0.245 microg/mL MGo with RSD 1.6-3.5 and 1.7-3.4%, respectively, in diabetics against 0.016-0.046 microg/mL Go and 0.021-0.066 microg/mL MGo with RSDs 1.5-3.5 and 1.4-3.6%, respectively, in healthy volunteers. Go and MGo in diabetics were also measured by standard addition and DMGo as an internal standard. Additives do not contribute significantly to Go and MGo matrix.  相似文献

A simple reversed-phase high-performance liquid chromatography (HPLC) method with ultraviolet detection at 280 nm was developed for simultaneous quantitation of furosemide and hydrochlorothiazide along with phenol red as a nonabsorbable marker for in situ permeability studies in anaesthetized rats. A jejunal segment of approximately 10 cm was isolated and cannulated in both ends for inlet and outlet solution. The perfusate was collected every 10 min, and samples were analyzed using the developed method. The mobile phase was acetonitrile-water-triethylamine-glacial acetic acid (41.5 + 57.4 + 0.1 + 0.9, adjusted to pH 5.6) at a flow rate of 1 mL/min; the run time was 9 min. The calibration graphs were linear for all 3 compounds (r > 0.999) across the concentration range of 7.93-125 microg/mL for phenol red and 6.25-100 microg/mL for hydrochlorothiazide and furosemide. The limits of quantitation were 7.2, 8.9, and 6.8 microg/mL for furosemide, hydrochlorothiazide, and phenol red, respectively. The coefficients of variation for intraassay and interassay precision were less than or equal to 7.6%, and the accuracy was between 93.2-103.4%. Using the single pass intestinal perfusion technique and the suggested HPLC method for sample analysis, mean values of 0.25 x 10(-4) (+/-0.16) cm/s and 0.22 x 10(-4) (+/-0.13) cm/s were obtained for furosemide and hydrochlorothiazide, respectively.  相似文献

A simple and sensitive spectrophotometric method is suggested for analysis of 3 antihistaminic drugs, acrivastine (I), mequitazine (II), and dimethindene maleate (III). The method is based on reaction of the drugs with 7,7,8,8-tetracyanoquinodimethane (TCNQ) in acetonitrile to form highly stable colored products that are measured at 750, 766, and 844 nm for I and II, and 480 and 618 nm for III. Beer's law is obeyed in the ranges of 5-60 microg/mL for 1, 5-50 microg/mL for II, and 10-70 microg/mL for III. The optimum assay conditions and their applicability to the determination of the cited drugs in pharmaceutical formulations are described. The method is statistically analyzed as compared with the European Pharmacopoeia (2001) method for the analysis of dimethindene maleate and reference methods for acrivastine and mequitazine drugs revealing good accuracy and precision.  相似文献

The reactions of [fac-Re(CO)(3)(bpy)(MeOH)](PF(6)), bpy = 2.2'-bipyridine, with the TCNX ligands (TCNE = tetracyanoethene, TCNQ = 7,7,8,8-tetracyano-p-quinodimethane, and TCNB = 1,2,4,5-tetracyanobenzene) in CH(2)Cl(2) gave very different results. No reaction was observed with TCNB whereas TCNE produced very labile intermediates which converted under mild conditions to structurally characterized [(mu-CN)[fac-Re(CO)(3)(bpy)](2)](PF(6)) with an eclipsed conformation relative to the almost linear Re-CN-Re axis (Re-N(NC) 2.134(8) A, Re-C(CN) 2.098(8) A). With TCNQ, a stable tetranuclear complex [(mu(4)-TCNQ)[Re(CO)(3)(bpy)](4)](BF(4))(4) was obtained. Its structural, electrochemical, and spectroscopic analysis indicates only negligible charge transfer from the rhenium(I) centers to the extremely strong pi acceptor TCNQ. Evidence includes a calculated charge of only -0.09 for coordinated TCNQ according to the empirical structure/charge correlation of Kistenmacher, a high-energy nitrile stretching band nu(CN) = 2235 cm(-1), and unprecedented large anodic shifts >0.7 V of the reduction potentials. DFT calculations were used to confirm and explain the absence of electron delocalization from the electron-rich metals to the TCNQ acceptor bridge. Correspondingly, the X-band and high-frequency (285 GHz) EPR data (g = 2.007) as well as the IR and UV-vis-NIR spectroelectrochemical results (marginal nu(CO) shifts, TCNQ(*-) chromophore bands) support the almost exclusive confinement of the added electron in [(mu(4)-TCNQ)[Re(CO)(3)(bpy)](4)](3+) to the TCNQ bridge.  相似文献

Two simple and sensitive extractive spectrophotometric methods have been developed for determination of zolmitriptan (ZTP) in tablets. These methods are based on the formation of yellow ion-pair complexes between ZTP and tropaeolin OO (TPOO) and bromothymol blue (BTB) in citrate-phosphate buffer of pH 4.0 and 6.0, respectively. The formed complexes were extracted with dichloromethane and measured at 411.5 and 410 nm for TPOO and BTB, respectively. The best conditions of the reactions were studied and optimized. Beer's law was obeyed in the concentration ranges of 2-20 and 1.5-17 microg/mL with molar absorptivities of 1.42 x 10(4) and 1.60 x 10(4) L/mol/cm for the TPOO and BTB methods, respectively. Correlation coefficients were 0.9998 and 0.9999 for TPOO and BTB methods, respectively. Limits of detection of the TPOO and BTB methods were 0.341 and 0.344 microg/mL, respectively, and the limits of quantitation were 1.034 and 1.051 microg/mL, respectively. Sandell's sensitivity and stability constant were also calculated. The proposed methods have been applied successfully for the analysis of the drug in its dosage forms. No interference was observed from excipients present in tablets. Statistical comparison of the results with those obtained by a high-performance liquid chromatography method showed excellent agreement and indicated no significant differences in accuracy and precision.  相似文献

The reactions of the electron donor 1-methylpiperidine (1MP) with the π-acceptors 7,7,8,8-tetracyanoquinodimethane (TCNQ), tetracyanoethylene (TCNE), 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ), 2,3,5,6-tetrachloro-1,4-benzoquinone (chloranil = CHL) and iodine (I2) were studied spectrophotometrically in chloroform at room temperature. The electronic and infrared spectra of the formed molecular charge-transfer (CT) complexes were recorded. The obtained results showed that the stoichiometries of the reactions are not fixed and depend on the nature of the acceptor. Based on the obtained data, the formed charge-transfer complexes were formulated as [(1MP)(TCNE)2], [(1MP)(DDQ)]·H2O, [(1MP)(CHL)] and [(1MP)I]I3, while in the case of 1MP–TCNQ reaction, a short-lived CT complex is formed followed by rapid N-substitution by TCNQ forming the final reaction products 7,7,8-tricyano-8-piperidinylquinodimethane (TCPQDM). The five solids products were isolated and have been characterized by electronic spectra, infrared spectra, elemental analysis and thermal analysis.  相似文献

A simple and sensitive fluorometric method for the determination of nalidixic acid was established by using 0.75M sodium borohydride and 7.5% hydrogen peroxide solution as fluorogenic reagents. Analyte concentrations of 0.0232-11.6 pg/mL could be determined with high precision and accuracy by the method. A relative standard deviation of 1.75% was obtained for a nalidixic acid concentration of 0.232 pg/mL. The method was satisfactorily applied to the determination of nalidixic acid in human serum, fish muscle, and chicken muscle, and the calibration curves were linear from 0.23 to 58.00 pg/mL, from 9.28 to 32.48 mg/kg, and from 4.64 to 23.20 mg/kg, respectively. The specificity of the reaction is also discussed.  相似文献

A very simple, ultra-sensitive and highly selective non-extractive spectrophotometric method for the determination of trace amount of molybdenum(VI) using 5,7-dibromo-8-hydroxyquinoline (DBHQ) has been developed. 5,7-Dibromo-8-hydroxyquinoline reacts in a slightly acidic solution (0.05 - 1.0 M H2SO4) with molybdenum(VI) to give a deep greenish-yellow chelate which has an absorption maximum at 401 nm. The reaction is instantaneous and the absorbance remains stable for over 24 h. The average molar absorption coefficient and Sandell's sensitivity were found to be 4.13 x 10(3) L mol(-1) cm(-1) and 7 ng cm(-2) of molybdenum(VI), respectively. Linear calibration graphs were obtained for 0.1 - 50 microg mL(-1) of molybdenum(VI). The stoichiometric composition of the chelate is 1:3 (Mo:DBHQ). A large excess of over 50 cations, anions and some common complexing agents (e.g. EDTA, oxalate, citrate, phosphate, thiourea, SCN-) do not interfere with the determination. The method was successfully used in the determination of molybdenum in several Standard Reference Materials (alloys, steels and waters) as well as in some environmental waters (inland and surface), biological samples (human blood and urine), soil samples, solution containing both molybdenum(V) and molybdenum(VI) and complex synthetic mixtures. The method has high precision and accuracy (S = +/-0.01 for 0.5 microg mL(-1)).  相似文献

A sensitive and selective method for determining the residual monoethanolamine in a developmental drug substance is developed and validated. Marfey's reagent, which is commonly used for the chiral analysis of amino acids, is reacted with the primary amine group of monoethanolamine and then analyzed by high-performance liquid chromatography-UV at 340 nm. Quantitation is performed by a standard addition method by preparing drug substance samples with added monoethanolamine ranging from 0.25-1.0 microg/mL (equivalent to 12.5-50 ppm with respect to the drug substance). The method performance is evaluated for linearity, specificity, detection and quantitation limits, accuracy, precision, and sample stability. The method is linear from 0.25-1.0 microg/mL with a coefficient of determination (r(2)) > 0.95. The accuracy and precision obtained is 105.5 +/- 4.8% (n = 3). The limits of detection and quantitation are 0.03 and 0.10 microg/mL, respectively. Instrument precision (% relative standard deviation of six injections of a derivatized 0.5 microg/mL monoethanolamine solution on two separate days) is >/= 2.0%. This method is suitable for the determination of monoethanolamine at the 25 ppm level in drug substance.  相似文献

A simple and rapid extraction spectrophotometric procedure has been developed for the determination of tricyclic anti-depressant drugs such as trazodone (TZH), amineptine (APH) and amitriptyline (ATPH) hydrochlorides in pure form and in different dosage forms. The method involves the formation of intense yellow ion-pairs between these drugs under investigation and methyl orange (MO) and bromocresol green (BCG) reagents followed by their extraction with 1,2-dichloroethane and quantitative microdetermination at 420 and 410 nm using MO or BCG, respectively. The optimum experimental conditions for the ion-pairs formation are established. The method permits the determination of TZH, APH and ATPH over a concentration range of 2-50, 2-50 and 1-25 microg ml(-1) for TZH, APH and ATPH, using MO and 1-25 microg ml(-1) for TZH, APH and ATPH, using BCG, respectively. The Sandell sensitivity (S) is found to be 0.106, 0.1071 and 0.0907 g cm(-2) for TZH, APH and ATPH, respectively, using MO reagent and 0.0788, 0.0661 and 0.0494 g cm(-2) for TZH, APH and ATPH, respectively, using BCG. The method is applicable for the assay of the investigated drugs in different dosage forms and the results are in good agreement with those obtained by the official method.  相似文献
