Enantiomeric separations by cyclodextrin-modified capillary electrophoresis of chiral carbamates containing a 4-substituted imidazole heterocycle, pharmacologically acting as histamine H3-receptor antagonists, were carried out. Various cyclodextrins (alpha-, beta- and substituted beta-cyclodextrins) were investigated. Baseline resolution was achieved with six out of 11 compounds. Heptakis-2,3,6-trimethyl-beta-cyclodextrin modified buffers seemed to be most efficient in separating most of the investigated compounds. The optical purity of one pair of enantiomers was determined. 相似文献

Two new diaza-crown ether derivatives (R-1, RS-1) have been synthesized from 1,4,10,13-tetraoxa-7,16-diazacyclooctadecane and tested as potential chiral selectors in capillary electrophoresis (CE) for the chiral separation of five amino acid derivatives. The individual use of the selectors did not lead to chiral differentiation. However, they enhanced the enantioselective effect of different cyclodextrins in dual selector systems. A chemometric methodology was proposed to optimize the migration time, height equivalent to a theoretical plate and separation of a mixture of a series of imidazole compounds by capillary electrophoresis. The optimization process was based on a special polynomial from 9 or 18 preliminary experiments. This method connects a general simplex method to a computer. A simplex two or three optimization-capillary electrophoresis (STO-CE) method has been developed in our laboratory. The most efficient separation was achieved with acetonitrile-phosphate buffer, pH 4.70, (5.30+94.70 (v/v)) with a beta-cyclodextrin concentration in the background electrolyte equal to 5.80 mM and a capillary temperature of 35 degrees C. Similar results were obtained using simple step-wise scanning. The higher relative difference obtained for these values with these two methods (simplex and step-wise scanning) was 5% for the beta-cyclodextrin concentration factor. 相似文献

This paper reviews chiral separations of primary amines by capillary electrophoresis and crown ether as chiral selector. Two possible mechanisms of chiral recognition by host-guest complexation are discussed: (i) The substituents of the crown ether act as barriers for the guest compounds, and (ii) lateral electrostatic interactions between host and guest occur. Experimental conditions affecting the separation are discussed in detail. A literature overview of practical applications is presented as well. More than 80 different primary amines were analyzed, whereupon the majority could be resolved using a screening method. It is shown that a synergistic effect on the resolution of chiral amines is observed when the chiral crown ether and cyclodextrins are simultaneously used in the same buffer system. This approach opens interesting perspectives for further method optimization. 相似文献

Summary The enantiomeric resolution of (±)-econazole, (±)-miconazole and (±)-sulconazole was achieved on a Chiralpak WH column. The
mobile phase used was hexane-2-propanol-diethylamine (400:99:1,v/v/v). The flow rates of the mobile phase used were 0.50 and 1.00 mL min−1. The values of α of the resolved enantiomers of econazole, miconazole and sulconazole were in the range of 1.68 to 1.23 while
the values of Rs varied from 2.42 to 1.10. The resolution of these antifungal agents on Chiralpak WH column is governed by
ligand exchange mechanism. Hydrophobic interactions also play an important role for the enantiomeric resoltuion of antifungal

Uncharged cyclodextrins were tested as chiral selectors for the enantiomeric separation of 13 glycyl dipeptides with capillary electrophoresis. Initial experiments were performed on 10 mmol/L of a cyclodextrin in 0.1 mol/L phosphoric acid -0.088 mol/L triethanolamine. Some of the resolved dipeptides were nonaromatic, which is noteworthy since, to our knowledge, no examples of the separation of small, nonaromatic molecules have been published. Mobility difference plots for Gly-DL-Leu and Gly-DL-Phe with heptakis(2,6-di-O-methyl)-beta-cyclodextrin showed relatively flat profiles in a large concentration range, which is an advantage for the development of robust quantitative analytical methods. The use of a background electrolyte (BGE) solution with pH 3.0 gave irreproducible results for two of the dipeptides, the acidic Gly-DL-Asp and Gly-DL-Glu; this pH is not advisable for the development of robust methods for these two peptides. The need for purer chiral selectors was demonstrated by comparing different batches of heptakis(2,6-di-Omethyl)-beta-cyclodextrin from the same supplier. A BGE consisting of malonic acid and triethanolamine was introduced to give better buffer capacity than the original BGE at pH 3.0. 相似文献

Competitive binding of a Tris run buffer with chiral crown ether in chiral capillary electrophoresis
In capillary electrophoresis of primary amine racemates using (+)-(18-crown-6)-tetracarboxylic acid (18C6H4) as a chiral selector, chiral recognition emanates from the differences in the complex formation between 18C6H4 and the two protonated amine enantiomers. The presence of buffer constituents such as tris(hydroxymethyl)aminomethane (Tris) or Na+, capable of forming complexes with 18C6H4, is thus detrimental to the chiral separation of primary amines. Such a competitive binding of buffer constituents was studied by comparing the electrophoretic mobilities of racemic analytes obtained in Tris/citric acid and triethylamine/citric acid buffers. We developed a simple fitting method to determine the competitive binding constant and applied it to the Tris buffer system. The competitive binding constant of Tris with 18C6H4 obtained at pH 3.0 was 27 +/- 4. 相似文献

The enantiomeric separation of a series of acidic pharmaceuticals (mostly nonsteroidal anti-inflammatory drugs) has been investigated in NACE systems using single-isomer amino beta-CD derivatives. The first part of this study consisted of the selection of the basic experimental conditions to separate efficiently the enantiomers of acidic drugs. Several parameters, such as the nature of the ionic BGE components, were studied and a methanolic solution of ammonium acetate containing the cationic CD was selected as BGE. A D-optimal design with 20 experimental points was then applied and the nature and concentration of the CD were found to have a significant effect on the enantiomeric resolution for all studied compounds. Resolution (R(s)) values were always higher with 6-monodeoxy-6-mono(3-hydroxy)propylamino-beta-CD (PA-beta-CD) compared to those obtained with 6-monodeoxy-6-mono(2-hydroxy)propylamino-beta-CD (IPA-beta-CD). However, the latter led to shorter migration times. Generic NACE conditions were then selected by means of the multivariate approach in order to obtain the highest R(s) values in a minimum amount of time. Finally, dependence of separation selectivity, resolution, as well as mobility difference on chiral selector concentration was discussed and binding constants with PA-beta-CD were estimated for the two enantiomers of one of the model compounds, suprofen in these NACE systems. 相似文献

Despite the separation efficiency of capillary electrophoresis (CE) is much higher than other chromatographic methods, it is sometimes difficult to perfectly separate the complex ingredients in biological samples. One possible and simple way to develop the separation effect in CE is to add some modifiers in the running buffer. In this paper, the suitable running buffer modifiers were explored to simultaneously separate and detect six typical flavonoids (apigenin, luteolin, kaempferol, quercetin, (+)-catechin and (−)-epicatechin) which are the main active ingredients in chrysanthemum by capillary zone electrophoresis with amperometric detection (CZE-AD). It was found that when β-cyclodextrin (β-CD) and the mixture of methanol and ethanol were used as running buffer modifiers, a baseline separation of the six analytes could be accomplished in less than 20 min and the detection limits were as low as 10−7 or 10−8 g ml−1. Other factors affecting the CZE separation, such as working potential, pH value and ionic strength of running buffer, separation voltage and sample injection time were extensively investigated. Under the optimum conditions, a successful practical application on the determination of chrysanthemum samples confirmed the validity and practicability of this method. 相似文献

The chiral resolution of baclofen was achieved by capillary electrophoresis using a fused-silica capillary (60 cm x 75 microm ID). The background electrolyte (BGE) was phosphate buffer (pH 7.0, 50 mM)-acetonitrile (95:5 v/v) containing 10 mM beta-cyclodextrin. The applied voltage was 15 kV. The values of alpha and R(s) were 1.06 and 1.00, respectively. The electrophoretic conditions were optimized varying the pH and the ionic strength of the BGE, concentrations of beta-cyclodextrin and acetonitrile and the applied voltage. 相似文献

Direct chiral separation of chiral peptide nucleic acid (PNA) monomers has been achieved for the first time by capillary electrophoresis (CE) with charged cyclodextrins as chiral selectors added to the electrophoretic buffer. Selectively modified 6-deoxy-6-N-histamino-beta-cyclodextrin and sulfobutyl ether-beta-CD were successfully used as chiral selectors for the enantiomeric separation of chiral monomers based on different aminoethylamino acids bearing thymine or adenine as nucleobases. Chiral separations were obtained at low selector concentrations (1-3 mM) with good enantioselectivity and resolution factors. Separations were optimized as a function of pH in order to exploit the effect of the electrostatic interactions between the oppositely charged selector and selectand. The method has been applied to the analysis of the enantiomeric excess of chiral monomers used for the solid phase synthesis of chiral PNA oligomers. CE chiral analysis showed that a very high enantiomeric purity was generally achieved in the synthesis of all monomers, except for histidine and aspartic acid based monomers in which ca. 10% of the "wrong" enantiomer was always present. 相似文献

Eight neutral cyclodextrins were tested for the enantiomeric separation of alanyl and leucyl dipeptides by capillary electrophoresis at pH 3, and seven out of the eight cyclodextrins proved suitable for the separation of one or more of the dipeptide enantiomer pairs. The best results were obtained with heptakis(2,6-di-O-methyl)-beta-cyclodextrin. The dipeptides that were separated were mainly the aromatic and the more lipophilic aliphatic dipeptides. Mobility difference plots at pH 3.0 with malonic acid-triethanolamine as background electrolyte showed that the aromatic dipeptides had higher affinities for the cyclodextrin than the nonpolar, aliphatic dipeptides. The results suggested that, under the conditions applied, the C-terminal amino acid rather than the N-terminal one is involved in the chiral discrimination. 相似文献

The chiral selectivity of a novel amphiphile, N-(2-hydroxydodecyl)-L-threonine (2-HDT), was evaluated for enantiomeric resolution of three binaphthyl derivatives (+/-)-1,1'-bi-2-naphthol, (+/-)-1,1'-binaphthyl-2,2'-diamine, and (+/-)-1,1'-binaphthyl-2,2'-diylhdrogen phosphate (BNP) by micellar EKC. The effects of three organic modifiers, methanol, isopropanol, and acetonitrile, on the separations of enantiomers of these compounds were investigated. Separation of enantiomers could be achieved in relatively dilute solutions of the pure surfactant. However, best separations of enantiomers were obtained only in the presence of 10% v/v acetonitrile. Enantiomeric impurity in nonracemic mixtures of R- and S-forms of BNP was determined. 相似文献

A series of aldo-bis-indole derivatives (aldo-BINs) was prepared by aromatic C-alkylation reactions of aldoses and indole in acetic acid solution. Common monosaccharides such as glucose, mannose, galactose, fucose, xylose, rhamnose, ribose, arabinose and N-acetylglucosamine were smoothly derivatized to form the UV absorbing aldo-BINs. The use of a capillary electrophoretic method to separate these novel aldo-BIN derivatives was established. The capillary electrophoresis conditions were set by using borate buffer (100 mM) at high pH (pH 9.0). The limit of determination was assessed to be 25 nM. The enantioseparation of D, L-pairs of aldo-BINs based on chiral ligand-exchange capillary electrophoresis technology was also achieved by using modified hydroxypropyl-β-cyclodextrin as the chiral selector in the presence of borate buffer. This aldose labeling method was applied successfully to the compositional and configurational analysis of saccharides, exemplified by a rapid and efficient method to simultaneously analyze the composition and configuration of saccharides from the medicinal herbs Cordyceps sinensis and Dendrobium huoshanense. 相似文献

Perfect control of electroosmotic flow (EOF) was achieved by dovetailing successive multiple ionic-polymer layer (SMIL) coated capillaries. The direction and magnitude of the EOF was perfectly controllable over the pH range 2-13. Zone diffusion was not observed, even if the inner wall of the dovetailed capillary was discontinuous, or if the sample zone passed through the connected part of the capillary because the RSDs of migration time, theoretical plates, symmetry factor and S/N of the marker were almost the same when seamless capillary and dovetailed capillary were compared. The dovetailed capillary was applied to cyclodextrin modified capillary zone electrophoresis. The control of the EOF enabled us to control both the resolution and the migration order of the enantiomers. The migration time was also controllable and, therefore, the best condition between separation and migration time could be determined by controlling the EOF. Partial filling affinity electrokinetic chromatography with a protein used as a chiral selector was also studied. The migration of the pseudostationary phase was controllable by EOF, and detection of the solute at 214 nm was possible. Therefore, the EOF-controlled dovetailed capillary has great potential to expand the application of the separation technique. 相似文献

The enantiomeric separation of gemfibrozil chiral analogues was performed by capillary zone electrophoresis (CZE). Resolution of the enantiomers was achieved using heptakis(2,3,6-tri-O-methyl)-beta-cyclodextrin (TM-beta-CD) as chiral selector dissolved into a buffer solution. In order to optimize the separation conditions, type, pH and concentration of running buffer and chiral selector concentration were varied. For each pH value, the optimum chiral selector concentration that produced the resolution of the isomers was found. The migration order of labile diastereoisomers formed was valued at the optimum experimental conditions by adding a pure optical isomer to the racemic mixture. Data from 1H NMR studies confirmed host-guest interaction between TM-beta-CD and 5-(2,5-dimethylphenoxy)-2-ethylpentanoic acid sodium salt. The hypothesized stoichiometry host:guest was 1:1. An apparent equilibrium constant (Ka) was estimated monitoring the chemical shift variation as a function of TM-beta-CD concentration. Salt effect on complexation equilibrium constant was also investigated. 相似文献
