Based on an ortho-lithiation protocol of 2,2′-dibromo-1,1′-binaphthyl four tetrasubstituted binaphthyls, 2,2′-dibromo-3,3′-diiodo-, 3,3′-dibromo-2,2′-diiodo-, 2,2′,3,3′-tetrabromo-, and 2,2′,3,3′-tetraiodo-1,1′-binaphthyls have been prepared in excellent yield which in turn proved to be versatile key intermediates in the synthesis of various 2,2′,3,3′-tetrasubstituted 1,1′-binaphthyl derivatives.  相似文献

This paper describes the commercially-viable synthesis and isolation of 2-ethoxy-3-pyridylboronic acid on a ca. 70 g scale via a directed ortho-metalation reaction on readily-available 2-ethoxypyridine. A range of efficient cross-coupling reactions of 2-ethoxy-3-pyridylboronic acid with selected aryl/heteroaryl halides under palladium-catalysed Suzuki-Miyaura conditions yield novel 2-ethoxy-3-aryl/heteroaryl-pyridines in high yield (heteroaryl=pyridyl, pyrimidyl, pyrazyl). The X-ray crystal structure of 2-ethoxy-3-pyridylboronic acid reveals that the boronic acid group takes part in an intramolecular O-H?O bond with the adjacent ethoxy substituent, and an intermolecular O-H?N bond.  相似文献

A new approach for the synthesis of fluorinated 1H-quinazolin-4-ones and 4-substituted quinazolines has been developed. 6-Fluoro-1H-quinazolin-4-ones were obtained by intramolecular cyclization of fluorine-containing S-ethyl N-benzoylisothioureas. Nucleophilic substitution reactions at positions 2 and 7, as well as alkylation at 1-position of quinazolinones were investigated. In addition, the synthesis of fluorine-containing 4-aminoquinazolines was carried out.  相似文献

2-(4-Chlorophenyl)-2-methyl-1,3-dioxolane 2a was lithiated ortho to the ketal group by treatment with butyllithium in THF at 0°C. Related 2-aryl-2-methyl-1,3-dioxolanes possessing a chlorine substituent at the meta position of the aryl group 2b,c were lithiated with butyllithium in THF at −78°C at the position between the two directing groups. The lithio species thus generated were treated with various electrophiles to give ortho-functionalized acetophenone derivatives.  相似文献

Chiral 3-N-arylsulfonyloxazolidin-2-ones 1a-f, prepared from (l)-amino acids, were reacted with lithium diisopropylamide in anhydrous THF and HMPA. A C3-symmetric macrocycle with alternating sugar amino acid and tyrosine residues was synthesized in seven steps from tyrosine tert-butyl ester and a sugar amino acid precursor derived from D-glucosamine. An Fmoc-protected D-glucosamine derivative was oxidized at C-6 to give the sugar amino acid, which was immediately coupled to tyrosine tert-butyl ester to produce an orthogonally protected building block. This building block was subsequently elongated to the trimer via the dimer, and finally cyclized to give the C3-symmetric macrocycle.  相似文献

Benzo[c]phenanthridine alkaloid, 7-hydroxynitidine, was synthesized from readily available 2-benzyloxy-6-bromo-3,4-dimethoxybenzaldehyde 5 and napthylamine 6 using reductive amination followed by radical cyclization in eight steps. LTMP (5 equiv) metalates randomly veratric acid (1). Under external quench conditions, the thermodynamically more stable lithium 2-lithio-3,4-dimethoxybenzoate (2) reacts with a variety of electrophiles to give versatile building units that are not easily accessible by conventional means. Under in situ quench conditions (LTMP/TMSCl), a reversal of regioselectivity is observed and 6-trimethylsilyl-3,4-dimethoxybenzoic acid (10) is formed predominantly.  相似文献

An efficient and straightforward procedure has been developed for the synthesis of highly substituted mono- and bis-N-aryl-3-aminodihydropyrrol-2-one-4-carboxylates via a one-pot, four-component domino reaction of amines, dialkyl acetylenedicarboxylates and formaldehyde in the presence of InCl3 (20 mol%) in MeOH at ambient temperature. The salient advantages of this method are mild reaction conditions, environmentally benign, high to excellent yields, shorter reaction times, easy operation and no column chromatographic separation.  相似文献

Reaction of N-aryl substituted maleimides with aliphatic ketazines leads to the formation of 4,5-dihydropyridazin-3(2H)-ones in moderate yields. The reaction proceeds through 1,4-addition of an azine to the maleimide double bond, as confirmed by separation of the corresponding adducts in some cases, which are then converted into 4,5-dihydropyridazin-3(2H)-ones. In addition, the solid-state synthesis of 4,5-dihydropyridazin-3(2H)-ones is demonstrated for the first time.  相似文献

It was found that the presence of DNA significantly enhanced the 4f luminescence of terbium(III) in its complexes with a number of 2-oxo-4-hydroxyquinoline-3-carboxylic acid amide derivatives (L1-L6). For the high-sensitivity determination of DNA, L1 and L4 were chosen from two proposed groups, in complexes with which terbium ions exhibited the most intense luminescence. Under optimum conditions, the luminescence intensity of terbium was proportional to the concentration of DNA over the ranges 0.025–1.2 μg/mL (detection limit of 10 ng/mL) and 0.01–1.2 λg/mL (detection limit of 3 ng/mL) for L1 and L4, respectively. The mechanisms of the interaction of Tb-L complexes with DNA molecules were hypothesized.  相似文献

The X-ray analysis of 1-oxo-3-thiophen-2-yl-isochroman-4-carboxylic acid methyl ester 1 confirmed its trans-configuration and a conformation with diaxial H-3 and H-4 atoms in solid state. NMR experiments indicated that trans-1 exists in solution in both expected conformers. In CDCl3 and especially in CD3OD or DMSO, the conformational equilibrium is shifted towards the conformer with diequatorial H-3 and H-4, which was also determined by 2D NOESY experiments. The shift is due to the greater polarity of that conformer deduced by ab initio calculations.  相似文献

The metalation of (quasi)alkoxy-substituted dibromobenzenes C6H3(OR)Br2 with lithium diisopropylamide (LDA) has been investigated. For 1-(quasi)alkoxy-3,5-dibromobenzenes (R = Me, TMS), different selectivities were observed depending on reaction conditions and the size of the alkoxy group. The methoxy group was an effective ortho-director whereas this was not the case for the bulky trimethylsilyloxy group. The metalation of related 2,5-dibromoanisole was also examined showing a significant meta-directing effect by the methoxy group. The thermal stability of aryllithium intermediates is significantly lower when lithium is flanked by a bromine and a methoxy group, whereas 4-(quasi)alkoxy-2,6-dibromoaryllithiums are less labile.  相似文献

We investigated the reductive elimination of several functionalized and non-functionalized vic-dibromides with 1,2-diphenyl-, 1,1,2,2-tetraphenyl- and 1-phenyl-2-(2-pyridyl)-1,2-disodioethane. The reaction, involving some of the less expensive organic and inorganic reagents, proceeds under mild conditions, and is tolerant of a variety of functional groups. Extension of this procedure to similar 1,2-disubstituted compounds was also investigated. Reductive eliminations run on stereochemical probe compounds strongly suggest that this reaction proceeds via a “single electron” reductive elimination reaction pathway.  相似文献
