trans-Cumylcyclohexanol (TCC) is used as a chiral auxiliary for the stereoselective addition of tributyltinlithium to N-acylpyrrolidinium/piperidinium ion with 70-80% diastereoselectivity at 0 °C. After removal of the minor diastereomer by radial chromatography, enantiopure N-methyl-2-(tributylstannyl)pyrrolidine and piperidine were produced by reductive removal of the auxiliary. 相似文献

A radical cascade involving 6-endo cyclization of aryl radicals generated from N-acryloyl-N-(1-methylethenyl)-9-bromophenanthren-10-ylmethylamines, followed by 5-endo-trig cyclization of the resulting α-amidoyl radicals afforded phenanthroindolizidines bearing a methyl substituent at the angular C13a position. 2,3,6-Trimethoxy derivative was synthesized by using this method, but its spectral data were not in accord with those of literature values reported for hypoestestatin 1. A concise and practical strategy to obtain C3 reverse-prenylated pyrrolidinoindoline scaffold has been executed in 28.8% overall yield. The key conjugative step involved a Booker-Milburn-Feudoloff reaction involving an NCS-mediated activation of indole, followed by coupling to C5 dimethylallylalcohol. This linchpin step proceeded in 74% yield. The overall sequence proceeded in five steps from commercially available N-methyltryptamine with a single protection-deprotection operation and a single redox manipulation. Mechanistic insights of NCS activation, and an ensuing rearrangement of the isoprene unit were gained by rationally varying the C3 substituent. 相似文献

Completion of the total synthesis of (±)-nominine (1) is described in detail. Based on the results of the preceding two papers, total synthesis of (±)-nominine was accomplished diverging from the intermediate 7. Thus, following pyrrolidine ring formation through transformation from 7 to 8, the C-ring was constructed by radical cyclization to form 10 from the enyne precursor 9. Subsequent elaboration of the C-ring, followed by formation of the azabicyclic ring system, completed a total synthesis of (±)-1. Single-crystal X-ray analysis of (±)-1 unambiguously confirmed its molecular structure and racemic crystal structure. 相似文献

A facile synthetic route to N-polyfluoro(trimethylsilyl)ethyl azole derivatives was developed starting from N-bromo(chloro)polyfluoroethyl-substituted azoles. A short and efficient stereoselective total synthesis of (±)-7-deoxy-trans-dihydronarciclasine, a highly potent antineoplastic agent and constituent of the Amaryllidaceae alkaloids, is described. Starting from a known arylcyclohexylamine-type precursor 6, the C-ring with the required stereochemistry is constructed using a chemo- and stereoselective enone reduction (NaBH4/CaCl2 system) and a Mitsunobu reaction. Reaction of Ln(NO3)3·6H2O with H2L [H2L=N,N′-bis(salicylidene)propane-1,2-diamine] gives rise to five new coordination polymers, viz. [Pr(H2L)(NO3)3(MeOH)]n (1) and [Ln(H2L)1.5(NO3)3]n [Ln=La (2), Eu (3), Sm (4) and Gd (5)]. Crystal structural analysis reveals that H2L effectively functions as a bridging ligand forming one-dimensional (1D) chain and two-dimensional (2D) open-framework polymers. Solid-state fluorescence spectra of 3 and 4 exhibit typical red fluorescence of Eu(III) and Sm(III) ions at room temperature while 2 emits blue fluorescence of ligand H2L. The lowest triplet level of ligand H2L was calculated on the basis of the phosphorescence spectrum of 5. The energy transfer mechanisms in the lanthanide polymers were described and discussed. 相似文献

A series of N-(1,2,4-triazole-4-yl)-N′-(fluorine-containing-phenyl)carbamimidothioates 5a-i were synthesized by reacting 4-amine-1,2,4-trizaole with corresponding aryl isothiocyanates in ethanol at room temperature and, in a subsequent step, with methyl iodide. The use of succinamic acid (H2sucm) in CuII/N,N′,N″-donor [2,2′:6′,2″-terpyridine (terpy), 2,6-bis(3,5-dimethylpyrazol-1-yl)pyridine (dmbppy)] reaction mixtures yielded compounds [Cu(Hsucm)(terpy)]n(ClO4)n (1), [Cu(Hsucm)(terpy)(MeOH)](ClO4) (2), [Cu2(Hsucm)2(terpy)2](ClO4)2 (3), [Cu(ClO4)2(terpy)(MeOH)] (4), [Cu(Hsucm)(dmbppy)]n(NO3)n·3nH2O (5.3nH2O), and [CuCl2(dmbppy)]·H2O (6·H2O). The succinamate(−1) ligand exists in four different coordination modes in the structures of 1–3 and 5, i.e., the μ2-κO:κO′:κO″ in 1 and 5 which involves asymmetric chelating coordination of the carboxylato group and ligation of the amide O-atom leading to 1D coordination polymers, the μ2-κ2O:κO′ in 3 which involves asymmetric chelating and bridging coordination of the carboxylato group, and the asymmetric chelating mode in 2. The primary amide group, either coordinated in 1 and 5, or uncoordinated in 2 and 3, participate in hydrogen bonding interactions, leading to interesting crystal structures. Characteristic IR bands of the complexes are discussed in terms of the known structures and the coordination modes of the Hsucm− ligands. The thermal decomposition of complex 5·3nH2O was monitored by TG/DTG and DTA measurements. 相似文献

A general method was developed to construct monocyclic α-methylene-γ-butyrolactone moiety. The key step is to introduce the α-methylene group by the ozonolysis of mono-substituted alkenes followed by reacting with a preheated mixture of CH2Br2-Et2NH. A high yielding cyclisation of the readily available N-(4,4-diethoxybutyl)-2-(3,4-dimethoxyphenyl)acetamide to 8,9-bis(methyloxy)-2,3,6,10b-tetrahydropyrrolo[2,1-a]isoquinolin-5(1H)-one is described. The first synthesis of the fungal natural product (−)-xylariamide A 1 is reported. N,O-Bis(trimethylsilyl)acetamide induced coupling of d-tyrosine with (E)-but-2-enedioic acid 2,5-dioxo-pyrrolidin-1-yl ester methyl ester 5 produced the dechloro natural product 6, which was subsequently monochlorinated using oxone and KCl to yield synthetic 1. (−)-Xylariamide A 1, (+)-xylariamide A 2 and (−)-dechloroxylariamide A 6 displayed no cytotoxic or antimicrobial activity. 相似文献

A (Z)-vinyl (N,N-diisopropyl)carbamate treated with Grignard reagents, under Wenkert Nickel-catalysed conditions, gave access to several substituted (Z)-alkenyl derivatives. A simple two-step procedure for the preparation of 2-trifluoromethyl-1(substituted aryl)-4(1H)-quinolones utilizing electrophilic trifluoroacetamidoyl chlorides as the starting materials is described.
