A rapid method for detection of Salmonella typhimurium contamination in packaged alfalfa sprouts using solid phase microextraction/gas chromatography/mass spectrometry (SPME/GC/MS) integrated with chemometrics was investigated. Alfalfa sprouts were inoculated with S. typhimurium, packed into commercial LDPE bags and stored at 10 + 2 °C for 0, 1, 2 and 3 days. Uninoculated sprouts were used as control samples. A SPME device was used to collect the volatiles from the headspace above the samples and the volatiles were identified using GC/MS. Chemometric techniques including linear discriminant analysis (LDA) and artificial neural network (ANN) were used as data processing tools. Numbers of Salmonella were followed using a colony counting method. From LDA, it was able to differentiate control samples from sprouts contaminated with S. typhimurium. The potential to predict the number of contaminated S. typhimurium from the SPME/GC/MS data was investigated using multilayer perceptron (MLP) neural network with back propagation training. The MLP comprised an input layer, one hidden layer, and an output layer, with a hyperbolic tangent sigmoidal transfer function in the hidden layer and a linear transfer function in the output layer. The MLP neural network with a back propagation algorithm could predict number of S. typhimurium in unknown samples using the volatile fingerprints. Good prediction was found as measured by a regression coefficient (R2 = 0.99) between actual and predicted data.  相似文献

High temperature gas chromatography and gas chromatography/mass spectrometry are used in the characterization of complex natural mixtures of permethylated oligosaccharides released from proteins or lipids. The high resolution allows separation of isomeric compounds and the mass range extends to oligosac-charides around molecular mass 2000 daltons or 10 sugar residues for isomalto-oligosaccharides. The mass spectra of permethylated oligosaccharide alditols from mucin glycopeptides are very informative and the approach allows a simple and rapid characterization of these complex components.  相似文献

The chemical composition of Schizonepeta tenuifolia Briq. (Sch.t.Briq.) is mainly composed of several volatile substances that affect multiple pharmacological targets and provide clinical efficacy. In this work, a headspace/solid-phase microextraction gas chromatography/mass spectrometry (HS-SPME-GC/MS) method was developed to evaluate the profiles of volatile compounds in Sch.t.Briq. The optimization of SPME conditions was carried out using four kinds of fiber, extraction time and temperature, desorption temperature and time, and sample amount. The GC/MS analysis allowed the tentative identification of 21 compounds, with similarities higher than 85%, in accordance with the NIST/Wiley mass spectral library. Major components such as (+)-menthone (14.32%), (−)-pulegone (47.73%), 2-hydroxy-2-isopropenyl-5-methylcyclohexane (5.97%), cis-pulegone oxide (4.12%), and schizonal (5.36%) were identified by comparison of retention time and mass spectral data of standards isolated from Sch.t.Briq. The contents of these compounds were about above 78% against total amounts of volatile compounds extracted from Sch.t.Briq. Based on optimized SPME method, 19 different Sch.t.Briq. samples collected from markets in Korea and China were analyzed to obtain the profiling data of volatile compounds. In addition, principal component analysis (PCA) was performed on the profiling data in order to classify the samples collected from the different regions. PCA could possibly visualize the grouping tendencies of the studied varieties of herbal samples, as well as the identification of the volatiles responsible for discriminating the groups.  相似文献

Analytical methods that are used for the detection and identification of chemicals related to the Chemical Weapons Convention in complex organic solutions were developed. A matrix modified by the addition of complex diesel oil background compounds and interferences was used for the development of a gas chromatography-mass spectrometry or gas chromatography-tandem mass spectrometry (GC-MS or GC-MS/MS) method for unambiguous identification of the scheduled chemicals. Co-elution of diethyl N,N-dimethylphosphoramidate and ethyl N,N-dimethylphosphoramidocyanidate, which are not separated by GC, was identified by GC-MS. Although GC-MS can identify the compounds, GC-MS/MS identification has greater accuracy and sensitivity in the case of complex matrixes. Four scheduled chemicals were accurately and selectively identified against numerous interfering substances by GC-MS/MS. The fragmentation chemistry of these compounds using MS/MS analysis was investigated.  相似文献

Summary The need to determine the migration of toxic unreacted compounds in bisphenol diglycidyl ether epoxy resins prompted us to investigate the HPLC properties of bisphenol F diglycidyl ether and its hydrolysis products in the water-based food simulants 3% (w/v) acetic acid, distilled water and 15% (v/v) ethanol. Peaks were identified by reversed-phase HPLC thermospray mass spectrometry and gas chromatography/mass spectrometry.  相似文献

Sample enrichment technology and instrumental sensitivity are no longer limiting factors in arson debris analysis. When pyrolyzed, petroleum based composite materials may produce artifacts. Urban air is another major source of interference since some of the components typically found in air, in particular alkylated aromatic hydrocarbons, are customarily used as indicators for the presence of gasoline. It is suggested to replace qualitative analysis with a quantitative approach, taking the sample matrix into consideration.  相似文献

建立了捕集阱顶空气相色谱/质谱测定水中二氯一溴甲烷的方法。采用正交实验设计对平衡温度、平衡时间、循环次数3个参数进行了优化,在平衡温度70 ℃、平衡时间20 min、循环次数2次的优化条件下,对水中的二氯一溴甲烷进行测定。结果显示,在0.1~10.0 μg/L范围内,二氯一溴甲烷的质量浓度和峰面积呈良好的线性关系,相关系数为0.9991。方法的检出限(S/N=3)为0.03 μg/L,定量限(S/N=10)为0.1 μg/L,回收率为83.1%~111.3%,相对标准偏差为1.7%~5.2%(n=6)。将该方法应用于水中二氯一溴甲烷的定性定量分析,效果良好。  相似文献

The separation of structurally related angiotensin-converting enzyme (ACE) inhibitors lisinopril, cilazapril, ramipril and quinapril and their corresponding active diacid forms (prilates) by conventional TLC silica gel 60 plates was contrasted with that afforded by monolithic ultra-thin-layer chromatographic (UTLC) plates. For the use of UTLC plates technical modifications of the commercially available equipments for the sample application, development and detection were made. Plates were developed in modified horizontal developing chamber using ethyl acetate-acetone-acetic acid-water (4:1:0.25:0.5, v/v). Detection of the separated compounds was performed densitometrically in absorption/reflectance mode at 220 nm and after exposure to iodine also by image analysis. The obtained results showed that monolithic layer is more efficient for the separation of structurally similar polar compounds, such as prilates than conventional silica layers. Identification of the compounds was confirmed by ESI-MS after their on-line extraction from the UTLC and TLC plates by means of Camag TLC-MS interface.  相似文献

The aim of the present study was to set‐up a Fast gas chromatography/mass spectrometry method for the analysis of cholesterol oxidation products (COPs). A silylated mixture of seven oxysterol standards was injected into a Fast GC/MS system. A capillary GC column (10 m×0.1 mm internal diameter×0.1 μm film thickness) coated with 95% dimethyl‐ and 5% diphenyl‐polysiloxane, was used. The method gave a fast (total analysis time=3.5 min) and satisfactory resolution (R>1.2) of the COPs standards, with a good repeatability and sensitivity, similar to those of conventional GC/MS; recoveries were tested on mice liver. Fast GC/MS method suitability for COPs analysis in food was also tested on an oxidized sardine fillet, which had been previously saponified and purified by NH2 solid‐phase extraction (SPE); a good repeatability and sensitivity was also obtained. The analytical performance of the Fast GC/MS method for the determination of COPs, together with the consequent significant reduction of the analysis time and consumables, demonstrates that Fast GC/MS represents a valid alternative to conventional GC/MS and evinces the great potential of such an analytical technique, which could be applied for both food and biological samples.  相似文献

This paper describes a novel analytical method, flash gas chromatography (FGC), for the analysis of volatile compounds from Houttuynia cordata Thunb. This method does not demand time-consuming extraction process. The ground powder of the plant material can be directly applied for the analysis and only a few milligrams of sample are needed. The identification of the components was made by FGC-MS. The results between FGC and ordinary GC (using the extracted essential oil as sample) were compared and found that FGC offered a similar number and types of the components with GC. FGC is a novel and feasible method for the quality control of traditional Chinese medicines (TCMs).  相似文献

A matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS)-based method has been developed for rapid differentiation between Panax ginseng and Panax quinquefolius, two herbal medicines with similar chemical and physical properties but different therapeutic effects. This method required only a small quantity of samples, and the herbal medicines were analyzed by MALDI-MS either after a brief extraction step, or directly on the powder form or small pieces of raw samples. The acquired MALDI-MS spectra showed different patterns of ginsenosides and small chemical molecules between P. ginseng and P. quinquefolius, thus allowing unambiguous differentiation between the two Panax species based on the specific ions, intensity ratios of characteristic ions or principal component analysis. The approach could also be used to differentiate red ginseng or P. quinquefolius adulterated with P. ginseng from pure P. ginseng and pure Panax quinquefolium. The intensity ratios of characteristic ions in the MALDI-MS spectra showed high reproducibility and enabled quantitative determination of ginsenosides in the herbal samples and percentage of P. quinquefolius in the adulterated binary mixture. The method is simple, rapid, robust, and can be extended for analysis of other herbal medicines.  相似文献

C18固相萃取膜适宜处理大体积地下水样现场采样而且易于运输、贮存。利用C18固相膜萃取以及GC/MS联用的方法对地下水中痕量半挥发性有机污染物进行了萃取以及定性、定量分析。优化了固相膜萃取的地下水采样量和浓缩体积。有机氯农药和多环芳烃的平均回收率分别为85%~110.1%、90.3%~115.1%;方法检出限达到10^-9g/L;相对标准偏差均小于15%。本方法用于北京地区地下水中的有机污染物分析,并给出地下水样C18固相膜萃取的GC/MS测定结果。  相似文献

A facile method based on high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry (HPLC/(+)ESI-MSn) has been established for the analysis of polyoxypregnane glycosides in the stems of Marsdenia tenacissima. The data reveals the ability of MSn in the structural elucidation of polyoxypregnane glycosides including the nature of the polyoxypregnane core, the kinds of the substituents and the types of sugar residues. Offline Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) is also performed to assign accurate elemental compositions. In this study, eighteen polyoxypregnane glycosides have been investigated. Among these components, five compounds are unambiguously identified as Marsdenoside K, Tencissoside A, B, C and D; two compounds are established as novel compounds based on mass spectral data; and the other eleven compound's structures are tentatively proposed. Furthermore, breakdown curves are constructed to distinguish five pairs of isomers among these eighteen compounds. As far as our knowledge, this is the first report on identification of polyoxypregnane glycosides in the stems of M. tenacissima by HPLC/ESI-MSn directly, which could save time and material consuming efforts in traditional phytochemistry analyses.  相似文献

A sensitive derivatization and extraction method is proposed for the determination of hydroxy-PCBs in urine. Phenolic hydroxyl groups of PCBs were allowed to react with five different reagents such as iodomethane, iodoethane, iodopropane, BSTFA and MTBSTFA. Propylated products at 100 °C for 30 min showed the best sensitivity with mass selective detector. Extraction recoveries and relative standard deviations of hydroxy-PCBs by SPE using C2 column were in the range of 78.0-112.3% and 2.5-9.6%, respectively. Instrumental detection limits for derivatized hydroxy-PCBs were in the range of 1-2 pg and were 10-1000 times more sensitive than those of non-derivatized hydroxy-PCBs. The correlation coefficients of the linear regression curves exceed 0.99, and the intra- and inter-day precisions were evaluated by RSDs within 10% at the concentrations of 0.4 and 4.0 ng/mL.  相似文献

The complex nature of botanicals and essential oils makes it difficult to identify all of the constituents by gas chromatography/mass spectrometry (GC/MS) alone. In this paper, automated sequential, multidimensional gas chromatography/mass spectrometry (GC-GC/MS) was used to obtain a matrix-specific, retention time/mass spectrometry library of 190 juniper berry oil compounds. GC/MS analysis on stationary phases with different polarities confirmed the identities of each compound when spectral deconvolution software was used to analyze the oil. Also analyzed were distillates of juniper berry and its oil as well as gin from four different manufacturers. Findings showed the chemical content of juniper berry can be traced from starting material to final product and can be used to authenticate and differentiate brands.  相似文献

Four flavonoids (rutin, hyperoside, quercitrin and quercetin) in Houttuynia cordata Thunb. and Saururus chinensis (Lour.) Bail. were determined by capillary electrophoresis with wall-jet amperometric detection. The working electrode was a 500 μm diameter carbon disc electrode and the detection potential was +0.95 V (versus Ag/AgCl). Effects of several important factors, such as the running buffer and its corresponding pH and concentration, separation voltage, injection time were investigated to acquire the optimum conditions for separation of these four flavonoids. Baseline separation for the four flavonoids was obtained within 21 min in a 60 cm length capillary at a separation voltage of 15 kV with a 60 mmoL/L Na2B4O7-120 mmoL/L NaH2PO4 buffer (pH 8.8) as running buffer. The relationship between peak currents and analyte concentrations was linear over about two orders of magnitude with detection limits (defined as S/N = 3) ranging from 0.02 to 0.05 μg/mL for all analytes. This method was applied for the determination of the above four flavonoids in H. cordata Thunb. and S. chinensis (Lour.) Bail. with simple extraction procedures, and the assay results were satisfactory.  相似文献

A gas chromatography/mass spectrometry (GC/MS) method was developed to study the pharmacokinetics of ligustilide following oral administration to rats. The method was used for the analysis of samples taken from rats. Biological samples were prepared by liquid-liquid extraction (LLE) using an n-hexane-ether (2:1) solvent mixture for a sample clean-up step and analyzed by GC/MS with a quadrupole MS detector in selected ion monitoring mode (m/z 190). The calibration curves were linear over the concentration range 0.172-8.60 microg/mL (r > 0.99) for blood samples and a different range (r > 0.99) for different tissue samples. The limit of detection (LOD) was 1.0 ng/mL or 1.0 ng/g (three times the signal-noise ratio). Within- and between-day precision expressed as the relative standard deviation (RSD) for the method was 1.58-3.88 and 2.99-4.91%, respectively. The recovery for all samples was >80%, except for liver samples (>70%). The main pharmacokinetic parameters obtained were: T(max) = 0.65 +/- 0.07 h, C(max) = 1.5 +/- 0.2 microg/mL, AUC = 34 +/- 6 h microg/mL and K(a) = 3.5 +/- 0.6/h. The experimental results showed that ligustilide was easily absorbed, but its elimination was slow, from 3 to 12 h after oral administration. The concentrations of ligustilide in rat cerebellum, cerebrum, spleen and kidney were higher than those in other organs.  相似文献

Retention indices of standard organic compounds of environmental interest were determined by gas chromatography/mass spectrometry, using a DB-5 fused-silica capillary column. Retention indices are useful references for tentative compound identification by gas chromatography, or confirmation by gas chromatography/mass spectrometry. They provide elution order for isomers that might be indistinguishable based on mass spectra. Modified Kovats and Lee retention indices are given for polycyclic aromatic hydrocarbons; sulfur heterocycles; nitrogen heterocycles; aromatic amines; oxygen heterocycles; phenols; alcohols; ketones; alkanes; nitriles; and methylesters of fatty, dicarboxylic, and aromatic acids for comparison and reference. Retention index values for heterocycles by gas chromatography/mass spectrometry are comparable with gas chromatography values previously reported.  相似文献
