A novel fluorescent probe for Cu2+ determination based on the fluorescence quenching of glyphosate (Glyp)-functionalized quantum dots (QDs) was firstly reported. Glyp had been used to modify the surface of QDs to form Glyp-functionalized QDs following the capping of thioglycolic acid on the core–shell CdTe/CdS QDs. Under the optimal conditions, the response was linearly proportional to the concentration of Cu2+ between 2.4 × 10−2 μg mL−1 and 28 μg mL−1, with a detection limit of 1.3 × 10−3 μg mL−1 (3δ). The Glyp-functionalized QDs fluorescent probe offers good sensitivity and selectivity for detecting Cu2+. The fluorescent probe was successfully used for the determination of Cu2+ in environmental samples. The mechanism of reaction was also discussed.  相似文献

Nanometer-sized fluorescent particles have been successfully synthesized. A synchronous fluorescence method, with high sensitivity and selectivity, has been developed for rapid determination of protein with functionalized CdS as a fluorescence probe. When Δλ=260 nm, maximum synchronous fluorescence is produced at 274 nm at pH 7.0. Under optimal conditions, the calibration graphs are linear over the range 0.1-3.0 μg ml−1 for bovine serum albumin (BSA), 0.1-11.0 μg ml−1 for γ-globulin (γ-G) and 0.1-1.4 μg ml−1 for human serum albumin (HSA), respectively. Limits of determination were 0.01 μg ml−1 for BSA, 0.019 μg ml−1 for γ-G and 0.021 μg ml−1 for HSA, respectively. The relative standard deviations of seven replicate measurements were 1.8% for 1.0 μg ml−1 BSA, 2.2% for 1.0 μg ml−1 γ-G and 2.3% for 1.0 μg ml−1 HSA.  相似文献

In answer to the ever-increasing need to perform the simultaneous analysis of environmental hazards, microcarrier-based multiplex technologies show great promise. Further integration with biofunctionalized quantum dots (QDs) creates new opportunities to extend the capabilities of multicolor flow cytometry with their unique fluorescence properties. Here, we have developed a competitive microbead-based flow cytometric immunoassay using QDs fluorescent labels for simultaneous detection of two analytes, bringing the benefits of sensitive, rapid and easy-of-manipulation analytical tool for environmental contaminants. As model target compounds, the cyanobacterial toxin microcystin-LR and the polycyclic aromatic hydrocarbon compound benzo[a]pyrene were selected. The assay was carried out in two steps: the competitive immunological reaction of multiple targets using their exclusive sensing elements of QD/antibody detection probes and antigen-coated microsphere, and the subsequent flow cytometric analysis. The fluorescence of the QD-encoded microsphere was thus found to be inversely proportional to target analyte concentration. Under optimized conditions, the proposed assay performed well within 30 min for the identification and quantitative analysis of the two environmental contaminants. For microcystin-LR and benzo[a]pyrene, dose–response curves with IC50 values of 5 μg L−1 and 1.1 μg L−1 and dynamic ranges of 0.52–30 μg L−1 and 0.13–10 μg L−1 were obtained, respectively. Recovery was 92.6–106.5% for 5 types of water samples like bottled water, tap water, surface water and seawater using only filtration as sample pretreatment.  相似文献

The new method presented in this article achieved the goal of capturing Salmonella typhimurium via immunoreaction and rapid in situ detection of the CdSe/ZnS quantum dots (QDs) labeled S. typhimurium by self-assembly light-emitting diode-induced fluorescence detection (LIF) microsystem on a specially designed multichannel microfluidic chip. CdSe/ZnS QDs were used as fluorescent markers improving detection sensitivity. The microfluidic chip developed in this study was composed of 12 sample channels, 3 mixing zones, and 6 immune reaction zones, which also acted as fluorescence detection zones. QDs–IgG–primary antibody complexes were generated by mixing CdSe/ZnS QDs conjugated secondary antibody (QDs–IgG) and S. typhimurium antibody (primary antibody) in mixing zones. Then, the complexes went into immune reaction zones to label previously captured S. typhimurium in the sandwich mode. The capture rate of S. typhimurium in each detection zone was up to 70%. The enriched QDs-labeled S. typhimurium was detected using a self-assembly LIF microsystem. A good linear relationship was obtained in the range from 3.7 × 10 to 3.7 × 105 cfu mL−1 using the equation I = 0.1739 log (C) − 0.1889 with R2 = 0.9907, and the detection limit was down to 37 cfu mL−1. The proposed method of online immunolabeling with QDs for in situ fluorescence detection on the designed multichannel microfluidic chip had been successfully used to detect S. typhimurium in pork sample, and it has shown potential advantages in practice.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

A multi-residue method for the determination of organochlorine pesticides in fish feed samples was developed and optimized. The method is based on a cleanup step of the extracted fat, carried out by liquid–liquid extraction on diatomaceous earth cartridge with n-hexane/acetonitrile (80/20, v/v) followed by solid phase extraction (SPE) with silica gel–SCX cartridge, before the identification and quantification of the residues by gas chromatography–triple quadrupole tandem spectrometry (GC–MS/MS). Performance characteristics, such as accuracy, precision, linear range, limits of detection (LOD) and quantification (LOQ), for each pesticide were determined. Instrumental LODs ranged from 0.01 to 0.11 μg L−1, LOQs were in the range of 0.02–0.35 μg L−1, and calibration curves were linear (r2 > 0.999) in the whole range of explored concentrations (5–100 μg L−1). Repeatability values were in the range of 3–15%, evaluated from the relative standard deviation of six samples spiked at 100 μg kg−1 of fat, and in compliance with that derived by the Horwitz's equation. No matrix effects or interfering substances were observed in fish feed analyses. The proposed method allowed high recoveries (92–116%) of spiked extracted fat samples at 100 μg kg−1, and very low LODs (between 0.02 and 0.63 μg kg−1) and LOQs (between 0.05 and 2.09 μg kg−1) determined in fish feed samples.  相似文献

A fluorescence ratiometric sensor for OCl has been developed based on a novel dual fluorophore response approach. The sensor molecule contains a coumarin fluorophore and a rhodamine fluorophore, and the two fluorophores are directly linked to an OCl recognition group. The structure of the sensor was characterized by ESI-MS, NMR, and X-ray crystallographic analysis. Upon treatment with OCl, both fluorophores in the sensor responded simultaneously at two separate optical windows, with large enhancement of the fluorescence ratio (I578/I501) from 0.01 to 39.55. The fluorescence ratios for the sensor showed a good linearity with the concentration of OCl in the range of 0.2–40 μM and the detection limits is 0.024 μM (S N−1 = 3). Investigation of reaction products indicated that the sensor reaction with OCl produced two new fluorescent molecules, which were responsible for the fluorescence changes in two optical windows. In addition, the sensor showed high selectivity to OCl over other reactive oxygen species, reactive nitrogen species, cations, and anions. The sensor has also been successfully applied to detection of OCl in natural water samples with satisfactory recovery.  相似文献

Twelve commercially available edible marine algae from France, Japan and Spain and the certified reference material (CRM) NIES No. 9 Sargassum fulvellum were analyzed for total arsenic and arsenic species. Total arsenic concentrations were determined by inductively coupled plasma atomic emission spectrometry (ICP-AES) after microwave digestion and ranged from 23 to 126 μg g−1. Arsenic species in alga samples were extracted with deionized water by microwave-assisted extraction and showed extraction efficiencies from 49 to 98%, in terms of total arsenic. The presence of eleven arsenic species was studied by high performance liquid chromatography–ultraviolet photo-oxidation–hydride generation atomic–fluorescence spectrometry (HPLC–(UV)–HG–AFS) developed methods, using both anion and cation exchange chromatography. Glycerol and phosphate sugars were found in all alga samples analyzed, at concentrations between 0.11 and 22 μg g−1, whereas sulfonate and sulfate sugars were only detected in three of them (0.6-7.2 μg g−1). Regarding arsenic toxic species, low concentration levels of dimethylarsinic acid (DMA) (<0.9 μg g−1) and generally high arsenate (As(V)) concentrations (up to 77 μg g−1) were found in most of the algae studied. The results obtained are of interest to highlight the need to perform speciation analysis and to introduce appropriate legislation to limit toxic arsenic species content in these food products.  相似文献

An on-line method, based on coupling dynamic ultrasonic extraction (DUE), continuously sampling the suspension of sample and solvent, high performance liquid chromatographic separation with diode array detection, has been developed for the determination of the flavonoids, including baicalin, baicalein and wogonin, from the root of Scutellaria baicalensis Georgi. Variables influencing the DUE were evaluated by orthogonal test. The extraction yields of baicalin, baicalein and wogonin in the roots of S. baicalensis Georgi obtained from five different cultivated areas are 73.8–131.5 μg mg−1 (RSD ≤ 6.24%), 6.8–15.9 μg mg−1 (RSD ≤ 5.36%) and 4.4–14.3 μg mg−1 (RSD ≤ 5.30%), respectively. The limits of detection for baicalin, baicalein and wogonin are 0.30, 0.37 and 0.41 μg mL−1, respectively. Linearity is from 0.55 to 109 μg mL−1 for baicalin, from 0.51 to 105 μg mL−1 for baicalein and from 0.53 to 102 μg mL−1 for wogonin. Compared with off-line continuous flow-DUE, the proposed method would be more convenient for the determination of the analytes and the rapid optimization of the extraction process. The extraction yields of flavonoids obtained by the proposed method are comparable with those obtained by dynamic microwave assisted extraction, static ultrasonic extraction and reflux extraction. The result indicated that the proposed method is suitable to determine the active components in Chinese herbal medicine.  相似文献

A novel method for selective determination of Cr(III) and Cr(VI) in environmental water samples was developed based on target-induced fluorescence quenching of glutathione-stabilized gold nanoclusters (GSH-Au NCs). Fluorescent GSH-Au NCs were synthesized by a one-step approach employing GSH as reducing/protecting reagent. It was found that Cr(III) and Cr(VI) showed pH-dependent fluorescence quenching capabilities for GSH-Au NCs, and thus selective determination of Cr(III) and Cr(VI) could be achieved at different pHs. Addition of EDTA was able to effectively eliminate the interferences from other metal ions, leading to a good selectivity for this method. Under optimized conditions, Cr(III) showed a linear range of 25–3800 μg L−1 and a limit of detection (LOD) of 2.5 μg L−1. The Cr(VI) ion demonstrated a linear range of 5–500 μg L−1 and LOD of 0.5 μg L−1. The run-to-run relative standard deviations (n = 5) for Cr(III) and Cr(VI) were 3.9% and 2.8%, respectively. The recoveries of Cr(III) and Cr(VI) in environmental water samples were also satisfactory (76.3–116%). This method, with its simplicity, low cost, high selectivity and sensitivity, could be used as a promising tool for chromium analysis in environmental water samples.  相似文献

Supramolecular solvents are here proposed firstly as extractants in solid sample microextractions. The approach was evaluated by extracting flumequine (FLU) and oxolinic acid (OXO), two widely used veterinary medicines, from fish and shellfish muscle using a supramolecular solvent made up of decanoic acid (DeA) reverse micelles. The antibiotics were extracted in a single step (∼15 min), at room temperature, using 400 μL of solvent. After centrifugation, an aliquot of the extract was directly analyzed by liquid chromatography and fluorescence, without the need of clean-up or solvent evaporation. Contrary to the previously reported methods, both OXO and FLU were quantitatively extracted from fish and shellfish, independently of sample composition. The high extraction efficiencies observed for these antibiotics were a consequence of their amphiphilic character which resulted in the formation of DeA-OXO and DeA-FLU mixed aggregates. The quality parameters of this quantitative method including sensitivity, linearity, selectivity, repeatability, trueness, ruggedness, stability, decision limit and detection capability were evaluated according to the 2002/657/EC Commission Decision. Quantitation limits in the different samples analyzed (salmon, sea trout, sea bass, gilt-head bream, megrim and prawns) ranged between 6.5 and 22 μg kg−1 for OXO and, 5 and 15 μg kg−1 for FLU. These limits were far below the current maximum residue limits (MRLs) set by the European Union (EU) (i.e. 100 and 600 μg kg−1, for OXO and FLU, respectively). The trueness of the method was determined by analyzing a Certified Reference Material (CMR, BCR®-725) consisting of a lyophilised salmon tissue material. Recoveries for fortified samples (50–100 μg kg−1 of OXO and 50–600 μg kg−1 of FLU) and their relative standard deviations were in the intervals 99–102% and 0.2–5%, respectively. The repeatability, expressed as relative standard deviation, was 3.6% for OXO and 2.3% for FLU ([OXO] = [FLU] = 200 μg kg−1 and n = 11).  相似文献

A gas chromatography–tandem mass spectrometric (GC–MS/MS) method has been established for the determination of cyanide in surface water. This method is based on the derivatization of cyanide with 2-(dimethylamino)ethanethiol in surface water. The following optimum reaction conditions were established: reagent dosage, 0.7 g L−1 of 2-(dimethylamino)ethanethiol; pH 6; reaction carried out for 20 min at 60 °C. The organic derivative was extracted with 3 mL of ethyl acetate, and then measured by using GC–MS/MS. Under the established conditions, the detection and quantification limits were 0.02 μg L−1 and 0.07 μg L−1 in 10-mL of surface water, respectively. The calibration curve had a linear relationship relationship with y = 0.7140x + 0.1997 and r2 = 0.9963 (for a working range of 0.07–10 μg L−1) and the accuracy was in a range of 98–102%; the precision of the assay was less than 7% in surface water. The common ions Cl, F, Br, NO3, SO42−, PO43−, K+, Na+, NH4+, Ca2+, Mg2+, Ba2+, Mn4+, Mn2+, Fe3+, Fe2+ and sea water did not interfere in cyanide detection, even when present in 1000-fold excess over the species. Cyanide was detected in a concentration range of 0.07–0.11 μg L−1 in 6 of 10 surface water samples.  相似文献

We have demonstrated the design of a new type fluorescence assay based on the inner filter effect (IFE) of gold nanoparticles (AuNPs) on the fluorescence of quantum dots (QDs). With a high extinction coefficient, AuNPs are expected to be capable of functioning as powerful absorbers. QDs with tunable emission wavelength are ideal fluorophores because the emission spectra of the rationally synthesized QDs can perfectly overlap with the absorption band of the absorber. Aminothiols are chosen as the model analytes, and the IFE-based fluorescent method for detection of aminothiols was suggested. Under the optimum conditions, the response is linearly proportional to the concentration of cysteine in the range of 0.05-0.9 μg mL−1. The present IFE-based fluorescent strategy could be also used to detect glutathione and homocysteine. The linear concentration ranges were 0.05-1.0 μg mL−1 for glutathione and 0.01-1.0 μg mL−1 for homocysteine.  相似文献

Competitive electrochemical enzyme-linked immunosorbent assays based on disposable screen-printed electrodes have been developed for quantitative determination of ochratoxin A (OTA). The assays were carried out using monoclonal antibodies in the direct and indirect format. OTA working range, I50 and detection limits were 0.05-2.5 and 0.1-7.5 μg L−1, 0.35 (±0.04) μg L−1 and 0.9 (±0.1) μg L−1, 60 and 100 μg L−1 in the direct and indirect assay format, respectively. The immunosensor in the direct format was selected for the determination of OTA in wheat. Samples were extracted with aqueous acetonitrile and the extract analyzed directly by the assay without clean-up. The I50 in real samples was 0.2 μg L−1 corresponding to 1.6 μg/kg in the wheat sample with a detection limit of 0.4 μg/kg (calculated as blank signal −3σ). Within- and between-assay variability were less than 5 and 10%, respectively. A good correlation (r = 0.9992) was found by comparative analysis of naturally contaminated wheat samples using this assay and an HPLC/immunoaffinity clean-up method based on the AOAC Official Method 2000.03 for the determination of OTA in barley.  相似文献

In this study we propose a chromogenic platform for rapid analysis of organophosphate (OP) and carbamate (CM) insecticide residues, based on recombinant Drosophila melanogaster acetylcholinesterase (R-DmAChE) as enzyme and indoxyl acetate as substrate. The visible chromogenic strip had the advantages identical to those of commonly used lateral flow assays (LFAs) with utmost simplicity in sample loading and result observation. After optimization, depending on the color intensity (CI) values, the well-established assay has the capabilities of both qualitative measurement via naked eyes and quantitative analysis by colorimetric reader with the desirable IC50 values against the tested six insecticides (0.06 μg mL−1 of carbofuran, 0.28 μg mL−1 of methomyl, 0.03 μg mL−1 of dichlorvos, 31.6 μg mL−1 of methamidophos, 2.0 μg mL−1 of monocrotophos, 6.3 μg mL−1 of omethoate). Acceptable matrix effects and satisfactory detection performance were confirmed by in-parallel LC–MS/MS analysis in different vegetable varieties at various spiked levels of 10−3 to 101 μg g−1. Overall, the testified suitability and applicability of this novel platform meet the requirements for practical use in food safety management and environmental monitoring, especially in the developing world.  相似文献

An efficient analytical method for simultaneous determination of 12 SFEs in serum is described. The method involves solid-phase extraction to isolate of SFEs from interfering species, especially cholesteryl esters, conversion to trimethylsilyl (TMS) ether derivatives for the direct analysis by gas chromatography–mass spectrometry (GC–MS) using a high temperature MXT-1 (Silcosteel-treated stainless steel) capillary column. All SFEs as their TMS derivatives were well separated with excellent peak shapes within 12 min. Overall recoveries ranged from 88% to 119%, with a detection limits for SFEs ranged from 2 to 30 μg L−1. The linearity as correlation coefficient was higher than 0.99 except for pregnenolone-3-arachidate (r2 = 0.98) in the concentration range of 5–3000 μg L−1. Ten serum samples obtained from volunteers were also analyzed and quantitatively determined of DHEA-3-palmitate and pregnenolone-3-stearate in 1.8–1195.8 μg L−1 concentration. The devised high temperature GC–MS method could be useful for identification of SFEs in biological specimens including serum.  相似文献

Two non-electroactive biomarkers methylglyoxal (MGo) and glyoxal (Go) in urine and environmental water samples were determined for the first time by capillary electrophoresis with amperometric detection (CE-AD) after derivatizing with an electroactive compound 2-thiobarbituric acid. Experimental conditions of derivatization and CE-AD detection were optimized. Highly linear response was obtained for these two biomarkers over three orders of magnitude with good correlation (r2 > 0.999). The limits of detection (LODs) and limits of quantitation (LOQs) of MGo and Go were 0.2 μg L−1 and 1.0 μg L−1, 0.5 μg L−1 and 2.0 μg L−1, respectively. The average recovery and relative standard deviation (RSD) were within the range of 90.9–101.3% and 0.7–2.2%, respectively. The proposed CE-AD method provides a reliable and sensitive quantitative evaluation of MGo and Go in real sample matrices by employing relatively simple and inexpensive instrument.  相似文献

Phthalates (PAEs) are ubiquitous toxic chemical compounds. During the last few years, some phthalate metabolites (MPAEs) have been proposed as appropriate biomarkers in human urine samples to determine PAE human intake and exposure. So, it is necessary to have fast, easy, robust and validated analytical methods to determine selected MPAEs in urine human samples. Two different instrumental methods based on gas (GC) and ultra-high performance liquid (UHPLC) chromatography coupled to mass spectrometry (MS) have been optimized, characterized and validated for the simultaneous determination of nine primary and secondary phthalate metabolites in urine samples. Both instrumental methods have similar sensitivity (detection limits ranged from 0.03 to 8.89 pg μL−1 and from 0.06 to 0.49 pg μL−1 in GC–MS and UHPLC–MS2, respectively), precision (repeatability, expressed as relative standard deviation, which was lower than 8.4% in both systems, except for 5OH-MEHP in the case of GC–MS) and accuracy. But some advantages of the UHPLC–MS2 method, such as more selectivity and lower time in the chromatographic runs (6.8 min vs. 28.5 min), have caused the UHPLC–MS2 method to be chosen to analyze the twenty one human urine samples from the general Spanish population. Regarding these samples, MEP showed the highest median concentration (68.6 μg L−1), followed by MiBP (23.3 μg L−1), 5cx-MEPP (22.5 μg L−1) and MBP (19.3 μg L−1). MMP (6.99 μg L−1), 5oxo-MEHP (6.15 μg L−1), 5OH-MEHP (5.30 μg L−1) and MEHP (4.40 μg L−1) showed intermediate levels. Finally, the lowest levels were found for MBzP (2.55 μg L−1). These data are within the same order of magnitude as those found in other similar populations.  相似文献

A new method was developed for simultaneous determination of trace arsenic and antimony in Chinese herbal medicines by hydride generation-double channel atomic fluorescence spectrometry with a Soxhlet extraction system and an n-octanol-water extraction system, respectively. The effects of analytical conditions on the fluorescence intensity were investigated and optimized. A water-dissolving and methanol-water-dissolving capability were compared. The contents of different species in five Chinese herbal medicines and their decoctions were analyzed. The concentration ratios of n-octanol-soluble As or Sb to water-soluble As or Sb were related to the kinds of medicine and the acidity of the decoction. Soxhlet extraction was found to be an effective method for plants pretreatment for determination of arsenic and antimony species in Chinese herbs; the interferences of coexisting ions were evaluated. The proposed method has the advantages of simple operation, high sensitivity and high speed, with 3σ detection limits of 0.094 μg g−1 for As(III), 0.056 μg g−1 for total As, 0.063 μg g−1 for Sb(III) and 0.019 μg g−1 for total Sb in a 1.0 g of the sample.  相似文献

Four simple, rapid, accurate, precise, reliable and economical spectrophotometric methods have been proposed for simultaneous determination of salbutamol sulphate (SS), bromhexine hydrochloride (BH) and etofylline (ET) in pure and commercial formulations without any prior separation or purification. They were first derivative zero crossing spectrophotometry (method 1), simultaneous equation method (method 2), derivative ratio spectra zero crossing method (method 3) and double divisor ratio spectra derivative method (method 4). The ranges for SS, BH and ET were found to be 1-35 μg mL−1, 4-40 μg mL−1 and 5-80 μg mL−1. For methods 1 and 2, the values of limit of detection (LOD) were 0.2314 μg mL−1, 0.4865 μg mL−1 and 0.2766 μg mL−1 and the values of limit of quantitation (LOQ) were 0.7712 μg mL−1, 1.6217 μg mL−1 and 0.9221 μg mL−1 for SS, BH and ET, respectively. For method 3, LOD values were 0.3297 μg mL−1, 0.2784 μg mL−1 and 0.7906 μg mL−1 and LOQ values were 0.9325 μg mL−1, 0.9282 μg mL−1 and 2.6352 μg mL−1 for SS, BH and ET, respectively. For method 4, LOD values were 0.3161 μg mL−1, 0.2495 μg mL−1 and 0.2064 μg mL−1 and LOQ values were 0.9869 μg mL−1, 0.8317 μg mL−1 and 0.6879 μg mL−1 for SS, BH and ET. The precision values were less then 2% R.S.D. for all four methods. The common excipients and additives did not interfere in their determinations. The results obtained by the proposed methods have been statistically compared by means of Student t-test and by the variance ratio F-test.  相似文献
