1H NMR and LC-MS, commonly used metabolomics analytical platforms, were used to annotate the metabolites found in potato (Solanum tuberosum L.) irrigated with four different treatments based on FA to AMD ratios, namely: control (0% AMD; tap water), 1:1 (50% AMD), 3:1 (75% AMD is 75% FA: AMD), and 100% AMD (untreated). The effects of stress on plants were illustrated by the primary metabolite shifts in the region from δH 0.0 to δH 4.0 and secondary metabolites peaks were prominent in the region ranging from δH 4.5 to δH 8.0. The 1:3 irrigation treatment enabled, in two potato cultivars, the production of significantly high concentrations of secondary metabolites due to the 75% FA: AMD content in the irrigation mixture, which induced stress. The findings suggested that 1:1 irrigation treatment induced production of lower amounts of secondary metabolites in all crops compared to crops irrigated with untreated acid mine drainage treatment and with other FA-treated AMD solutions.  相似文献

The dynamic technique of diffusive gradients in thin films (DGT), that measures metal speciation in situ, has found wide environmental application. Simple interpretation of the metal accumulation in terms of a solution concentration has assumed that trace metals do not penetrate beyond the surface of the binding layer, but penetration, although theoretically discussed has not yet been directly measured. Multiple binding layers were used to enable analysis of different depths of a DGT binding phase (Chelex-100 or iminodiacetate resins). In simple metal solution (no ligand) at pH 7, metal penetration to the back layer was low and similar for all metals. However, at lower pH up to 42% of an individual metal accumulated in the back resin layer. This was most noticeable for Mn at pH 4 and 5, but Cd and Co were also affected at pH 4. These results were consistent with rate limited binding, particularly for Mn. A kinetic model successfully fitted the data and allowed derivation of a binding rate constant and the mean distance that metals penetrate into a resin gel (λM). Only for Mn, Co and Cd were experimentally derived λM values greater than the diameter of a Chelex-100 resin bead. For most situations, then, the penetration into the binding layer is negligible and binding of trace metal ions can be regarded as instantaneous, validating the simple use and interpretation of DGT. For weakly binding metals at low pH the slower binding allows penetration, which may affect the DGT measurement.  相似文献

This paper reports the results of an investigation on the performance of the Diffusive Gradient in Thin Films (DGT) Technique in speciation of metals in aqueous samples of municipal wastes and mine effluents. The DGT was assessed regarding its suitability for in situ determination of metal speciation in municipal wastes and aqueous mine effluents. As the thickness of the diffusive gel layer of the DGT was increased to 0.40, 0.80, and 1.60 mm, a decrease in the amount of accumulated metal mass was observed for most of the metals studied in all the effluent samples. However, the results were different from one field-study site to another. Effect of kinetics also was observed in the amount of accumulated metal mass by the DGT. The computer speciation code, Windermere Humic Aqueous Model (WHAM VI), was used to predict the metal speciation of Cd, Cu, Ni, Pb, Co, and Zn, and WHAM predictions were compared with those of the experimentally determined metal speciation by the DGT technique (free and labile metal ions). This comparison showed good similarities between the theoretically predicted WHAM VI values and the experimentally measured values by DGT. The DGT technique was found to be simple and useful for investigating chemical speciation of trace metals in aqueous samples of municipal wastes and aqueous mine effluents.  相似文献

The diffusive gradients in thin films technique (DGT) was used for uranium measurements in water. DGT devices with Dowex resin binding phase (Dow DGT) were tested in synthetic river water, which gave 84% response to total uranium concentration. The devices were also deployed in natural river water and compared to devices with other types of binding phases, Chelex 100 resin beads imbedded in polyacrylamide hydrogel (Chelex DGT) and DE 81 anion exchange membrane (DE DGT), deployed in the same location at the same time. The measurement by Dow DGT was the lowest among the different types of the DGT devices, 45% of total uranium, while measurement by DE DGT was the highest, 98% of total uranium. The results achieved by the three types of DGT devices were explained by three DGT working mechanisms, equilibrium between complexes of resin/uranyl carbonates and complexes of resin/competitive ligands in water, effective reduction of uranyl carbonate concentration by the binding phase and dissociation of UO2(CO3)22− and UO2(CO3)34− within the diffusive layer in a DGT device. This work investigated the application of diffusive gradients in thin films technique (DGT) to uranium speciation measurements in natural water. Two binding phases were examined, a commercially available affinity membrane, Whatman DE 81 (DE 81), with amino binding functional groups and the conventionally used Chelex 100 beads imbedded polyacrylamide hydrogel (Chelex) with iminodiacetate functional groups. The DGT devices assembled with the binding phases of DE 81 (DE 81 DGT) and Chelex gel (Chelex DGT) were tested both in synthetic river water solutions and in local river water. DE 81 DGT and Chelex DGT measured 80% and 75% of the total uranium in synthetic river water solution, respectively, and measured 73% and 60% of the total uranium in St. Lawrence River, Canada, respectively. The binding properties of the DE 81 membrane and Chelex gel for uranium, and the diffusion of uranyl complexes in the polyacrylamide gel (PAM) were also studied.  相似文献

This paper reports the results of an investigation on the performance of the diffusive gradient in thin film technique (DGT) as a speciation tool for trace elements (TEs) in artificial human gastrointestinal fluids. The validity of Cd, Pb, and Zn sampling by DGT in digestive fluids was checked. The TE bioaccessibility in highly contaminated soils was determined using the in vitro Unified Barge Method (UBM) test. DGT devices were deployed in the gastrointestinal solutions obtained after carrying out the UBM test. The computer speciation code JESS (Joint Expert Speciation System) was used to predict the metal speciation of Cd, Pb, and Zn. Combining the in vitro test with the DGT technique and JESS provided an approach to the TE species available for transport across the intestinal epithelium. The gastrointestinal absorption of ingested TE ranged from 8 to 30% for Cd, 0.6 to 11% for Pb, and 0.8 to 7% for Zn and was influenced by TE speciation. In this original approach, the DGT technique was found to be simple and reliable in the investigation of TE chemical speciation in digestive fluids. Extrapolation to the in vivo situation should be undertaken very cautiously and requires further investigation.  相似文献

A preservation study has been performed for arsenic speciation in surface freshwaters affected by acid mine drainage (AMD), a pollution source characterized by low pH and high metallic content. Two sample preservation procedures described in the literature were attempted using opaque glass containers and refrigeration: i) addition of 0.25 mol L−1 EDTA to the samples, which maintained the stability of the arsenic species for 3 h; and ii) in situ sample clean-up with a cationic exchange resin, in order to reduce the metallic load, which resulted in a partial co-adsorption of arsenic onto Fe precipitates. A new proposed method was also tried: sample acidification with 6 mol L−1 HCl followed by in situ clean-up with a cationic exchange resin, which allowed a longer preservation time of at least 48 h. The proposed method was successfully applied to water samples with high arsenic content, taken from the Aguas Agrias Stream (Odiel River Basin, SW Spain), which is severely affected by AMD that originates at the nearby polymetallic sulfide mine of Tharsis. The speciation results obtained by liquid chromatography–hydride generation–atomic fluorescence spectrometry (HPLC-HG-AFS) indicated that during the summer the main arsenic species was As(V) at the hundred μg L−1 level, followed by DMA (dimethyl arsenic) and As(III) below the ten μg L−1 level. In winter, As(V) and As(III) increased at least fivefold, whereas the DMA was not detected.  相似文献

The performance of the Diffusive Gradients in Thin films (DGT) technique with Chelex®-100, Metsorb™ and Diphonix® as binding phases was evaluated in the vicinity of the former uranium mining sites of Chardon and L'Ecarpière (Loire-Atlantique department in western France). This is the first time that the DGT technique with three different binding agents was employed for the aqueous U determination in the context of uranium mining environments. The fractionation and speciation of uranium were investigated using a multi-methodological approach using filtration (0.45 μm, 0.2 μm), ultrafiltration (500 kDa, 100 kDa and 10 kDa) coupled to geochemical speciation modelling (PhreeQC) and the DGT technique. The ultrafiltration data showed that at each sampling point uranium was present mostly in the 10 kDa truly dissolved fraction and the geochemical modelling speciation calculations indicated that U speciation was markedly predominated by CaUO2(CO3)32−. In natural waters, no significant difference was observed in terms of U uptake between Chelex®-100 and Metsorb™, while similar or inferior U uptake was observed on Diphonix® resin. In turn, at mining influenced sampling spots, the U accumulation on DGT-Diphonix® was higher than on DGT-Chelex®-100 and DGT-Metsorb™, probably because their performance was disturbed by the extreme composition of the mining waters. The use of Diphonix® resin leads to a significant advance in the application and development of the DGT technique for determination of U in mining influenced environments. This investigation demonstrated that such multi-technique approach provides a better picture of U speciation and enables to assess more accurately the potentially bioavailable U pool.  相似文献

The speciation measurements of trace metals by the diffusive gradients in thin-films technique (DGT) using a poly(4-styrenesulfonate) (PSS) aqueous solution as a binding phase and a cellulose dialysis membrane (CDM) as a diffusive layer, CDM-PSS DGT, were investigated and showed good agreement with computer modelling calculations. The diffusion coefficients of ethylenediaminetetraacetic acid (EDTA) complexes with Cd2+ and Cu2+ were measured and compared with those of the inorganic metal ions. CDM-PSS DGT device was tested for speciation measurement in sample solutions containing EDTA, tannic acid (TA), glucose (GL), dodecylbenzenesulfonic acid (DBS) and humic acid (HA) as complexing ligands forming organic complexes with varying stability constants. Lower percentages of DGT labile copper concentrations over total filterable copper concentrations obtained from the deployments in freshwater sites indicated that copper complexes with organic matter were basically not measured by the devices.  相似文献

A new preservation method has been proposed for the speciation of As(III) and As(V) in acid mine drainage (AMD) samples, characterised by low pH and high metallic content. Samples were taken from a polymetallic sulphides mining area in the province of Huelva (SW Spain), under exploitation until the 1960s for its Cu, Pb and Zn sulphides. The abandoned mine works and the numerous waste rocks heaps produce AMD with high As content, an aqueous pollution source for the nearby streams. Short-term (from few hours to 1 week) preservation of the two inorganic arsenic species was studied, trying different containers (polyethylene, glass), presence or absence of light, temperatures (ambient, refrigerated, frozen), preserving agents and procedures (EDTA, HCl or AcH acids, cation-exchange resin). The speciation results obtained by liquid chromatography-hydride generation-atomic fluorescence spectrometry (HPLC-HG-AFS) indicated a rapid conversion of the samples with most of the preservation procedures reported in the literature after 3 h after sample collection. A promising method for arsenic preservation has been developed in this work, which maintains the arsenic species distribution in the original samples for a longer time. It consists in the use of opaque glass containers, acidification of the samples with HCl and in situ cleanup with cationic exchange resin, which allowed to preserve the samples for As speciation for at least 48 h.  相似文献

The study was conducted to understand, (a) the effect of pH on the speciation of uranium in acid mine drainage and (b) determine the effect of uranium speciation on the removal of gross alpha activity from acid mine drainage. Speciation modeling predicted a dominance of neutral and positively charged species under acidic and neutral conditions. Negative species become dominant from pH 8. The effectiveness of the anionic exchange resin to remove gross alpha activity was lower under acidic conditions (pH < 6) compared to alkaline conditions (pH ≥ 8).

In situ measurements of copper and zinc using diffusive gradients in thin films (DGT) in two distinct natural water systems were compared to metal speciation assessed by competitive ligand exchange (CLE) and voltammetric measurements. In a dynamic river system, where dissolved metal concentrations vary significantly over short-time periods, DGT technique provided averaged values of the metal concentrations over time. In microcosms, at different total dissolved concentrations of copper and zinc, DGT technique measured a similar fraction as measurements of labile metal performed by voltammetry. The proportion of DGT and voltammetric-labile zinc to dissolved zinc was 61±4% and, respectively, 76±9%. DGT technique was measuring 81±8% of exchangeable copper (by exchange with catechol). These two fractions were similarly influenced by the addition of NTA. In the absence of NTA, copper measured by DGT represented 34±4% of dissolved copper whereas in the presence of NTA, this proportion raised to 57±2%. These measurements were compared to calculations performed with speciation programs using several models for the complexation by humic and fulvic substances, namely Model VI (WHAM), NICA-Donnan and SHM. The predicted speciation by these three models was similar. The prediction of free zinc ion and labile zinc concentrations were in agreement with experimental data. Calculated concentrations of free copper ion were overestimated because these models are not considering strong specific copper-binding ligands probably present in natural water.  相似文献

Binding resin beads used in DGT (diffusion gradients in thin films) tend to settle to one side of the resin during casting. This phenomenon might be relevant for metal accumulation when partially labile complexes dominate the metal speciation, especially after recognizing the important role played by complex dissociation in the resin domain. The influence of the inhomogeneity of the binding agent distribution on metal accumulation is here assessed by numerical simulation of DGT devices with binding beads in only one half of the resin disc, as a reasonable model of the standard resin discs. Results indicate that a decrease in mass accumulation of less than 13% can arise in these inhomogeneous devices (as compared with an ideal disc with homogeneous dispersion of the resin beads) when complexes with stability constant K < 102 m3 mol−1 (K < 105 L mol−1) dominate the metal speciation. The loss increases as K increases, but the percentage of mass loss always remains lower than the volume fraction of resin disc without beads. For very labile or inert complexes, the impact of the inhomogeneous distribution of binding resin beads is negligible. As kinetic dissociation constants of complexes can be estimated from the distribution of the metal accumulation in a DGT device with a stack of two resin discs, the influence of the inhomogeneity on the recovered kinetic constant is also assessed. For the cases studied, the recovered kinetic dissociation constant, kd,recovered, retains the correct order of magnitude, being related to the true kd by kd ≈ f−1 kd,recovered, quite independently of K and kd values, being f the fraction of volume of the resin disc where resin beads are dispersed.  相似文献

The structure of iron and managanese ions substituted in the framework of nanoporous AlPO-5 is determined by ex situ and in situ X-ray absorption spectroscopy. Fe K-edge XANES and EXAFS studies clearly indicate that iron ions are present as Fe(III) in octahedral coordination in the assynthesised material and tetrahedral coordination in the calcined material in both pure FeAlPO-5 and FeMnalPO-5. XANES and EXAFS results also indicate that reaction with hydrogen peroxide causes the removal of Fe(III) ions from the framework. Mn K-edge XANES and EXAFS of FeMnAlPO-5 samples indicate that Mn(II) ions are present in the framework, tetrahedrally coordinated, in the as-synthesised material but upon calcination it is found that the Mn(II) ions are removed from the framework, suggesting a different synthesis strategy is necessary to stabilise the Mn(II) ions in the framework simultaneously with Fe(III) ions.  相似文献

Since its invention in the mid-1990s, the diffusive gradients in thin films (DGT) technique has rapidly become one of the most promising in situ sampling techniques for trace metal measurement in natural waters. We investigated here the possibility of using DGT devices with different binding phases to determine different DGT labile fractions of Cd and Cu in laboratory solutions and in natural waters. Several binding phases were studied, including conventional Chelex 100 resin imbedded polyacrylamide hydrogel (Chelex) and several recently developed binding phases, poly(acrylamide-co-acrylic acid) (PAM-PAA) gel, poly(acrylamidoglycolic acid-co-acrylamide) (PAAG-PAM) gel, Whatman P81 cellulose phosphate ion-exchange membrane (P81), and poly(4-styrenesulfonate) (PSS) aqueous solution. Laboratory testing in metal solutions spiked with EDTA or humic acid suggested that all the DGT devices measured only free metal ions and inorganic metal complexes. Upon field testing at both freshwater and seawater sites it was found that the DGT labile metal concentrations measured by different binding phases can be significantly different, suggesting that the DGT labile metal fractions were dependent on binding strength of the binding phase. By designing binding phases that can compete with different natural water complexing ligands to varying extents, it is possible to use these different DGT devices to measure metal speciation in natural waters.  相似文献

Mineral mining generates acidic, saline, metal-rich mine waters, often referred to as acid mine drainage (AMD). Treatment of AMD and recovering saleable products during the treatment process are a necessity since water is, especially in South Africa, a scarce commodity. The aim of the study presented here was to investigate the effect of zero valent iron (Fe0) on the biological removal of sulphate from AMD in batch reactors. The performance of the reactors was assessed by means of sulphate reduction, chemical oxygen demand (COD), volatile fatty acid (VFA) utilisation and volatile suspended solids (VSS) concentration. To this end, three batch reactors, A, B and C (volume 2.5 L), were operated similarly with the exception of the addition of grass cuttings and iron filings. Reactors A and B received twice as much grass (100 g) as C (50 g). Reactor A received no iron filings to act as a control, while reactors B and C received 50-g iron filings for the experimental duration. The results showed that Fe0 appears to provide sustained sulphate removal when sufficient grass substrate is available. In reactors A and C, sulphate removal efficiency was higher when the COD concentration was lower due to utilisation. In reactor B, sulphate removal efficiency was accompanied by an accumulation of COD as hydrogen (H2) provided by the Fe0 was utilised for sulphate reduction. Furthermore, these results showed the potential of Fe0 to enhance the participation of microorganisms in sulphate reduction.  相似文献

The technique of diffusive gradients in thin films (DGT) is a newly developed analytical technique capable of measuring in situ concentrations of trace metals in the environment. The technique employs a thin film diffusive hydrogel (with well-defined diffusion properties) in contact with a binding phase capable of binding metal ions of interest. In this work, we demonstrate, for the first time, the use of a commercially available solid ion exchange membrane (Whatman P81) as the binding phase in DGT analysis. The cellulose phosphate-based Whatman P81 membrane is a strong cation exchange membrane. Its performance characteristics as a new binding phase in DGT measurement of Cu2+ and Cd2+ were systematically investigated. Several advantages over the conventional ion exchange resin-embedded hydrogel binding phases used in DGT were observed including simple preparation, ease of handling, and reusability. The binding capacities of the material to various metal ions were examined both collectively and individually. The binding phase preferentially binds to transition metal ions rather than matrix ions such as potassium, sodium, calcium and magnesium, which are competitive species in natural waters. Within the optimum pH range (pH 4.0-9.0), the maximum non-competitive binding capacities of the membrane for Cu2+ and Cd2+ were 3.22 and 3.07 μmol cm−2, respectively. The suitability of the new membrane-based binding phase for DGT applications was validated experimentally. The experimental results demonstrated excellent agreement with theoretically predicted trends. The measurement was not degraded after four consecutive reuses of the cellulose phosphate binding phase.  相似文献

The possible adverse effect of competitive binding on DGT (diffusive gradients in thin films) measurements of metals in marine situations was investigated. Of the divalent transition metals, manganese binds most weakly to Chelex resin and is most likely to be affected by competition. In media resembling seawater, the Chelex within DGT devices becomes saturated with Ca and Mg within 2 h, and at pH 5–6 the sensitivity of the DGT measurement for Mn is substantially reduced, due to the appreciable concentration of Mn in contact with the resin. For media resembling freshwater DGT gives a near theoretical response for Mn. Iron was shown to be capable of displacing Mn and to a more limited extent Cd from the resin when its capacity for Fe is approached. Vertical profiles of Mn in a mesocosm sediment, obtained by deploying DGT for different times, could be explained by this displacement effect. The problem only occurs when Fe concentrations are exceptionally high and can be avoided by using short deployment times, typically less than 12 h. Whilst most trace metals can be simply measured by deploying DGT in marine systems, for Mn consideration should be given to possible effects associated with the capacity of the Chelex binding layer being approached by accumulation of the other cations present.  相似文献

The diffusive gradient in the thin films (DGT) technique was tested to measure dissolved mercury (Hg) both in laboratory aqueous solutions and in situ in river water. For this purpose, a commercial ready-to-use and specific-for-Hg DGT device was used. Each sampler consisted of a filter membrane-agarose gel as the diffusive layer and a Spheron-Thiol resin in polyacrylamide gel as the binding agent. Basic performance assays at the laboratory with this type of DGT unit confirmed the applicability of Fick's first law for DGT measurements. The diffusion coefficient of MeHg in the agarose diffusive gel was 8.50?×?10?6?cm2 s?1 at 25°C. Several field studies were also carried out in two different rivers of the Ebro River basin (NE Spain) affected by Hg wastes released by the chlor-alkali industry. Hg concentrations determined by DGT were generally much lower than the results obtained through direct measurements of the river water. In addition, the results of a time series experiment also performed in the field show that the amount of Hg accumulated in the resin does not increase at all with the exposure time. This may be explained by the underestimation of the truly dissolved Hg fraction due to the formation of a biofilm layer on the surface of the samplers, thus clogging the filter and preventing Hg species from diffusing through it. Consequently, it was demonstrated that the DGT technique presents important limitations for measuring Hg in polluted rivers characterised by a high biomass load (eutrophic), whereas its performance was demonstrated to be correct in oligotrophic waters.  相似文献
