A square-wave voltammetric (SWV) method using a hanging mercury drop electrode (HMDE) has been developed for determination of the herbicide molinate in a biodegradation process. The method is based on controlled adsorptive accumulation of molinate for 10 s at a potential of –0.8 V versus AgCl/Ag. An anodic peak, due to oxidation of the adsorbed pesticide, was observed in the cyclic voltammogram at ca. –0.320 V versus AgCl/Ag; a very small cathodic peak was also detected. The SWV calibration plot was established to be linear in the range 5.0×10–6 to 9.0×10–6 mol L–1; this corresponded to a detection limit of 3.5×10–8 mol L–1. This electroanalytical method was used to monitor the decrease of molinate concentration in river waters along a biodegradation process using a bacterial mixed culture. The results achieved with this voltammetric method were compared with those obtained by use of a chromatographic method (HPLC–UV) and no significant statistical differences were observed. 相似文献

运用循环伏安法(CV)考察了甘草苷在悬汞电极(HMDE)上的电化学还原行为,在-0.7~-1.7 V(vs.SCE)电位窗口及0.10 mol/L(NH4)2SO4溶液中甘草苷在HMDE上的循环伏安行为是一在低扫描速度(<100 mV/s)下受吸附控制,在高扫描速度下受扩散控制的不可逆还原过程,还原峰电位(Epc)为-1.491 V。运用计时库仑法(CC)、计时电流法(CA)测定并计算了甘草苷的电荷传递系数α、扩散系数D以及表观速率常数Kf等电极过程动力学参数。初步探讨了甘草苷在HMDE上的反应机理,同时运用方波伏安法(SWV)研究了甘草苷在HMDE上的方波伏安行为,还原峰电流与其浓度在1.2×10-6~1.2×10-5mol/L及1.2×10-5~1.2×10-3mol/L范围内呈良好的线性关系,相关系数R=0.9936及0.9966,检出限8.0×10-7mol/L,据此可建立直接电化学测定甘草苷含量的方法。 相似文献

The interaction of ethidium bromide (2,7-diamino-10-ethyl-9-phenylphenanthridinium bromide; EB) with double stranded (ds) calf thymus DNA and thermally denatured single stranded (ss) DNA was studied in solution and at the electrode surface by means of transfer voltammetry using a carbon paste electrode (CPE) as working electrode in 0.2 M acetate buffer, pH 5.0. As a result of intercalation of this dye between the base pairs of dsDNA, the characteristic peak of dsDNA, due to the oxidation of guanine residues, decreased and after a particular concentration of EB a new peak at +0.81 V appeared, probably due to the formation of a complex between dsDNA and EB. The non-intercalated EB gives another peak, but at an increased concentration of the dye. A similar behaviour was observed during the interaction of the dye with ssDNA.Furthermore, the interaction of EB with ds, ss and supercoiled (sc) DNA was studied at the hanging mercury drop electrode (HMDE) surface by means of alternating current voltammetry in 0.3 M NaCl and 50 mM sodium phosphate buffer (pH 8.5) as supporting electrolyte. dsDNA yields a smaller peak at −1.42 V (peak III) compared to the one yielded by ssDNA, since the latter is a relaxed and more accessible form. By addition of EB into the buffer solution an increase of peak III was observed in the dsDNA form as well as in ssDNA resulting from their interaction with EB. Furthermore, the appearance of peak III in covalently closed circular scDNA after exposure to increasing concentrations of EB is a result of the introduction of ‘free ends’ in DNA affecting its structural integrity. 相似文献

The sensitive differential pulse anodic stripping voltammetry (DPASV) proposed originally by Ishiyama et al. (2001) has been revised and improved to allow the accurate measurement of silicon on a hanging mercury drop electrode (HMDE) instead of a glassy carbon electrode. We assessed the rate of formation of the partially reduced β-silicododecamolybdate and found that metallic mercury promotes the reaction in the presence of a large concentration of Fe3+. The scope of the method has been broadened by carrying out the measurements in the presence of a constant amount of Fe3+. The limit of detection (LOD) of the method described in the present paper is 100 μg Si g−1 of steel, with a relative precision ranging from 5% to 12%. It can be further enhanced to 700 ng Si g−1 of steel provided the weight of the sample, the dilution factors, the duration of the electrolysis and the ballast of iron are adequately revised. The tolerance to several interfering species has been examined, especially regarding Al3+, Cr3+ and Cr VI species. The method was validated using four low-alloy ferritic steels certified by the National Institute of Standards and Technology (NIST). Its application to nickel base alloys as well as to less complicated matrixes is straightforward. The interaction of cyclophosphamide (CP) with calf thymus double-stranded DNA (dsDNA) and thermally denatured single-stranded DNA (ssDNA) immobilized at the carbon paste (CPE) and pencil graphite electrodes (PGE), was studied electrochemically based on oxidation signals of guanine and adenine using differential pulse voltammetry (DPV).As a result of the interaction of CP with DNA, the voltammetric signals of guanine and adenine increased in the case of dsDNA while a slight increase was observed in ssDNA. The effect of experimental parameters such as the interaction time between CP and DNA forms and the concentration of CP, were studied using DPV with CPE and PGE. Additionally, reproducibility and detection limits were determined using both electrodes. A comparison of the analytical performance between CPE and PGE was done. Our results showed that these two different DNA biosensors could be used for the sensitive, rapid and cost effective detection of CP itself as well as of CP-DNA interaction.Furthermore, the interaction of CP with dsDNA and ssDNA was studied in solution and at the electrode surface by means of alternating current voltammetry (ACV) in 0.3 M NaCl and 50 mM sodium phosphate buffer (pH 8.5) supporting electrolyte, using a hanging mercury drop electrode (HMDE) as working electrode.The conclusions of this study were mainly based on tensammetric peaks I (at −1.183 V) and II (−1.419 V) of DNA. This study involved the interaction of CP with surface-confined and solution phase DNA where experimental parameters, such as the concentration of CP and the interaction time, were studied. By increasing the concentration of CP, an increase of peak II was observed in both ds and ssDNA, while an increase of peak I was observed only in the case of dsDNA. An overall conclusion of the study using HMDE was that the interaction of CP with surface-confined DNA significantly differed from that with solution phase DNA. The increase of peaks I and II was lower in the case of interaction of CP with surface-confined DNA, probably due to steric positioning of DNA at the electrode surface. 相似文献

Dimenhydrinate exhibits a single adsorptive stripping peak at a hanging mercury drop electrode after accumulation at 0.0V vs Ag/AgCl electrode at pH 3.8 (acetate buffer). The addition of trace amounts of copper ions enhanced the dimenhydrinate peak and its height depends on the concentration of each dimenhydrinate and Cu2+. The adsorptive stripping response was evaluated with respect to accumulation time and potential, concentration dependence, electrolyte, the presence of other purines, surfactants and other metal ions, and some variables. The calibration graph for dimenhydrinate determination is linear over the range 2.0×10–8–2.0×10–7 M (pre-concentration for 60s). The correlation factor is found to be 0.985 and RSD is 3.2% at 1.0×10–7 M. Detection limit is 1.0×10–8 M after 5 min accumulation. The determination of dimenhydrinate in pharmaceutical formulations by the proposed method is also reported. 相似文献

A method for the pesticide DNOC (4,6-Dinitro-o-cresol) quantification using hanging mercury drop electrode (HMDE) and stripping square wave voltammetry (SSWV) optimisation is proposed. As a continuous cathodic current decrease was observed during the experiments, a waiting time, together with solution pH and SWV instrumental variables were optimised by factorial designs. From the two reduction current peaks values, only one was considered as dependent variable in the optimisation process. While the cathodic current peak and standard deviation were used in the waiting time and solution pH optimisation process, the instrumental parameters for SWV were optimised by using only current peak values. With the optimal parameters, a calibration curve from (0.01–0.55) × 10?6 mol L?1 with LOD of 2 × 10?8 mol L?1 was obtained. The proposed method was checked for DNOC quantification in different water samples obtained from Cordoba area and very good results, with recovery values around 102% were observed. 相似文献

An improved approach to the anodic stripping voltammetric (ASV) determination of heavy metals, using the hanging mercury drop electrode (HMDE), is reported. It was discovered that using very cathodic accumulation potentials, at which the solvent reduction occurs (overpotential deposition), the voltammetric signals of zinc(II), cadmium(II), lead(II) and copper(II) increase. When compared with the classical methodology a 5 to 10-fold signal increase is obtained. This effect is likely due to both mercury drop oscillation at such cathodic potentials and added local convection at the mercury drop surface caused by the evolution of hydrogen bubbles. 相似文献

A carbon paste electrode modified with 2-mercapto-4(3H)-quinazolinone was used for the voltammetric determination of mercury(II). Mercury was preconcentrated onto the surface of the modified electrode only by the complexing effect of the modifier without application of potential (i.e. in open-circuit conditions). After exchange of the medium, the accumulated amount of mercury(II) was determined by differential pulse anodic stripping voltammetry. The response depended on the concentration of mercury in the bulk solution, preconcentration time, and other parameters. The detection limit was 0.1 g 1–1 Hg(II) for a preconcentration time of 15 min. Preconcentration for suitable times yielded a linear calibration graph from 0.5 to 6000 g 1–1 Hg(II). For multiple determinations (5 runs), the relative standard deviation was 5% for a concentration of 100 g 1–1 Hg(II). An indirect voltammetric method is described for determination of cyanide ions and hydrogen cyanide, using the effect of cyanide on cathodic adsorptive stripping peak height of Cu-adenine. The method is based on competitive Cu complex formation reaction between adenine at the electrode surface and CN− ions in solution. Under the optimum experimental conditions (pH=6.42 Britton-Robinson buffer, 1×10−4 M copper and 8×10−7 M adenine), the linear decrease of the peak current of Cu-adenine was observed, when the cyanide concentration was increased from 5×10−8 to 8×10−7 M. The detection limit was obtained as 1×10−8 M for 60 s accumulation time. The relative standard deviations for six measurements were 4 and 2% for the cyanide concentrations of 5×10−8 and 2×10−7 M, respectively. The method was applied to the determination of cyanide in various industrial waste waters such as electroplating waste water and also for determination of hydrogen cyanide in air samples. 相似文献

The refreshable mercury film silver based electrode Hg(Ag)FE applied for determination of Cr(VI) traces using catalytic adsorptive striping voltammetry (CAdSV) will be presented. The film electrode is characterized by its very good surface reproducibility (not less than 2%) and long-term stability (1500–2000 measurement cycles). The mechanical refreshing of mercury film is realized in the specially constructed device, in a time shorter than 1–2 s.
In the paper, it will be proved that a mechanically weak hanging mercury drop electrode (HMDE) may be substituted by mercury film Hg(Ag)FE electrode with a surface area adjustable from 1.5 to 12 mm2. For the electrode surface 4 mm2 the detection limit obtained for Cr(VI) was 0.19 nM, while the linearity range measured for a 20 s accumulation time was between 0.5 and 50 nM. The relative standard deviation (R.S.D.) in determination of Cr(VI) varied from 1 to 5%. The influence of the excess of Cr(III) on determination of Cr(VI) was analyzed using samples from the Dobczyce reservoir spiked with known amounts of Cr(VI) and Cr(III). 相似文献

Capabilities for heavy metal speciation of anodic stripping voltammetry (ASV) and constant-current stripping chronopotentiometry (SCP) in both mercury drop (HMDE) and mercury film rotating disk (MFE-RDE) electrodes are compared. For this purpose, the Cd(II)–glycine and Cd(II)–polymethacrylate (PMA) systems are used as models of simple labile and macromolecular labile complexes adsorbing onto the electrode, respectively. The results suggest that SCP could be a valuable alternative to the more widespread ASV in this kind of study. Concerning the electrode, the MFE-RDE is less user-friendly than the HMDE, but presents a better definition of both the hydrodynamic conditions during the deposition step and the stripping regime during the oxidation. An important interference in SCP is the dissolved oxygen, which can be minimised by combining relatively large oxidation currents and low stirring rates. Moreover, for Cd–PMA, double peaks have been observed in both ASV and SCP, which seems to be due to the lack of enough ligand excess to complex the metal ions released by the amalgam oxidation. Anyway, this problem can be minimised by optimising the rotation rate of the electrode and ensuring enough ligand excess. 相似文献

A cost-effective sequential injection system incorporating with an in-line UV digestion for breakdown of organic matter prior to voltammetric determination of Zn(II), Cd(II), Pb(II) and Cu(II) by anodic stripping voltammetry (ASV) on a hanging mercury drop electrode (HMDE) of a small scale voltammetric cell was developed. A low-cost small scale voltammetric cell was fabricated from disposable pipet tip and microcentrifuge tube with volume of about 3 mL for conveniently incorporated with the SI system. A home-made UV digestion unit was fabricated employing a small size and low wattage UV lamps and flow reactor made from PTFE tubing coiled around the UV lamp. An in-line single standard calibration or a standard addition procedure was developed employing a monosegmented flow technique. Performance of the proposed system was tested for in-line digestion of model water samples containing metal ions and some organic ligands such as strong organic ligand (EDTA) or intermediate organic ligand (humic acid). The wet acid digestion method (USEPA 3010a) was used as a standard digestion method for comparison. Under the optimum conditions, with deposition time of 180 s, linear calibration graphs in range of 10-300 μg L−1 Zn(II), 5-200 μg L−1 Cd(II), 10-200 μg L−1 Pb(II), 20-400 μg L−1 Cu(II) were obtained with detection limit of 3.6, 0.1, 0.7 and 4.3 μg L−1, respectively. Relative standard deviation were 4.2, 2.6, 3.1 and 4.7% for seven replicate analyses of 27 μg L−1 Zn(II), 13 μg L−1 Cd(II), 13 μg L−1 Pb(II) and 27 μg L−1 Cu(II), respectively. The system was validated by certified reference material of trace metals in natural water (SRM 1640 NIST). The developed system was successfully applied for speciation of Cd(II) Pb(II) and Cu(II) in ground water samples collected from nearby zinc mining area. 相似文献

This paper describes the development of a sequential injection analysis method to automate the determination of atrazine by square wave voltammetry exploiting the concept of monosegmented flow analysis to perform in-line sample conditioning and standard addition. To perform these tasks, an 800 μL monosegment is formed, composed by 400 μL of sample and 400 μL of buffer/standard solution. To obtain an efficient homogenization, the sample solution is divided in five zones intercalated by four zones of the Britton-Robinson buffer (pH 2.0) in presence of appropriate concentration of NaNO3 and varying atrazine standard concentrations. This mixture zone is isolated from the carrier solution by two 100 μL air bubbles. After homogenization in an auxiliary reaction coil the mixture zone is injected toward the flow cell, which is adapted to the capillary of a hanging drop mercury electrode, at a flow rate of 50 μL s−1. After a suitable delay time, the potential is scanned from −0.5 to −1.2 V versus Ag/AgCl using a frequency of 300 Hz and pulse height of 25 mV. The linear dynamic range is observed for atrazine concentrations between 1.16 × 10−7 and 2.32 × 10−6 mol L−1, obeying the linear equation ip = (−6.91 ± 0.07) × 108[atrazine] + (4 ± 8), with r2 = 0.9996, for which the slope is given in nA L mol−1. The detection and quantification limits of the method are 2.1 × 10−8 and 7.0 × 10−8 mol L−1, respectively. The sampling frequency is 37 h−1, when the standard addition protocol is followed. This frequency can be increased to 42 h−1 if the protocol to obtain in-line calibration curve is used for quantification. The method was applied for determination of atrazine in spiked river water samples and its accuracy was evaluated by comparison with the batch standard addition approach, which revealed that there is no evidence of statistically significant differences between the two methods. 相似文献

A chemically modified carbon paste electrode with 3,4-tetra pyridinoporphirazinatocobalt(II) (Co(3,4 tppa) was applied to the determination of free cyanide ion. The electrode has a linear range between 1.5 × 10−5 M and 1.0 × 10−2 M with a Nernstian slope of 60 ± 1.5 mV/decade and its detection limit is 9 × 10−6 M. The response time of electrode is 5 min. The proposed electrode was applied successfully for the determination of cyanide in commercially available spring water. Some anions, such as SCN−, I−, Cl−, Br− and oxalate that are usually serious interfering species for most of cyanide selective electrodes, did not have any interfering effect for this proposed electrode. 相似文献

The effects of the proximity of the signals of two heavy metal ions in stripping voltammetry (SV) and constant‐current stripping chronopotentiometry (SCP) is studied at mercury drop (HMDE) and mercury film (MFE) electrodes. For this purpose, the Cd(II)‐Pb(II)‐phthalate system is used, taking advantage of the approaching of the signals corresponding to Cd(II)‐phthalate and Pb(II)‐phthalate labile complexes as phthalate is added to mixtures of Cd(II) and Pb(II)‐ions. The results are compared with those obtained by differential pulse polarography (DPP) and by stripping measurements on the Pb(II)‐phthalate system alone, showing discrepancies in SCP data under nondepletive conditions and negligible differences in the other cases. 相似文献

A catalytic adsorptive stripping voltammetric method for the determination of copper(II) on a carbon paste electrode (PCE)
in an alizarin red S (ARS)-K2S2O8 system is proposed. In this method, copper(II) is effectively enriched by both the formation and adsorption of a copper(II)-ARS
complex on the PCE, and is determined by catalytic stripping voltammetry. The catalytic enhancement of the cathodic stripping
current of the Cu(II) in the complex results from a redox cycle consisting of electrochemical reduction of Cu(II) ion in the
complex and subsequent chemical oxidation of the Cu(II) reduction product by persulfate, which reduces the contamination of
the working electrode from Cu deposition and also improves analytical sensitivity. In Britton-Robinson buffer (pH 4.56±0.1)
containing 3.6×10−5 mol L−1 ARS and 1.6×10−3 mol L−1 K2S2O8, with 180 s of accumulation at −0.2 V, the second-order derivative peak current of the catalytic stripping wave was proportional
to the copper(II) concentration in the range of 8.0×10−10 to ∼3.0×10−8 mol L−1. The detection limit was 1.6×10−10 mol L−1. The proposed method was evaluated by analyzing copper in water and soil. 相似文献

A new procedure was elaborated to determine mercury(II) using an anodic stripping square-wave voltammetry at the antimony film carbon paste electrode (SbF-CPE). In highly acidic medium of 1 M hydrochloric acid, voltammetric measurements can be realized in a wide potential window. Presence of cadmium(II) allows to separate peaks of Hg(II) and Sb(III) and apparently catalyses reoxidation of electrolytically accumulated mercury, thus allowing its determination at ppb levels. Calibration dependence was linear up to 100 ppb Hg with a detection limit of 1.3 ppb. Applicability of the method was tested on the real river water sample. 相似文献

This work describes the electroanalytical determination of pendimethalin herbicide levels in natural waters, river sediment and baby food samples, based on the electro-reduction of herbicide on the hanging mercury drop electrode using square wave voltammetry (SWV). A number of experimental and voltammetric conditions were evaluated and the best responses were achieved in Britton-Robinson buffer solutions at pH 8.0, using a frequency of 500 s− 1, a scan increment of 10 mV and a square wave amplitude of 50 mV. Under these conditions, the pendimethalin is reduced in an irreversible process, with two reduction peaks at − 0.60 V and − 0.71 V, using a Ag/AgCl reference system. Analytical curves were constructed and the detection limit values were calculated to be 7.79 μg L− 1 and 4.88 μg L− 1, for peak 1 and peak 2, respectively. The precision and accuracy were determinate as a function of experimental repeatability and reproducibility, which showed standard relative deviation values that were lower than 2% for both voltammetric peaks. The applicability of the proposed methodology was evaluated in natural water, river sediments and baby food samples. The calculated recovery efficiencies demonstrate that the proposed methodology is suitable for determining any contamination by pendimethalin in these samples.
