This paper describes asymmetric synthesis of β-aminophenylpropionic acid through application of a homochiral sulfoxide auxiliary. High kinetically controlled (3R,2S,RS)-diastereoselectivity (−60°C) is achieved during addition of the lithium enolate of tert-butyl (+)-(R)-p-toluenesulfinylacetate to substituted N-(benzylidene)toluene-4-sulfonamides 2a2d. The reductive cleavage of adduct 3a with sodium amalgam yielded tert-butyl 3-(toluene-4-sulfonamido)-3-phenylpropionate 5a, which was subjected to ester hydrolysis and subsequent detosylation with sodium in liquid ammonia to yield (S)-β-aminophenylpropionic acid in good yield and high 91% e.e.  相似文献

We describe here the aminomethylation of chiral silyl enol ether derivatives using iminium ion. The choice of judicious precursors with adequate protecting groups for the silylation/aminomethylation step was required to achieve the synthesis of β2-homotryptophane in few steps. Catalytic asymmetric dehydration of β-aryl or alkyl substituted β-hydroxy esters via kinetic resolution has been investigated. A brief survey of 10 different chiral ligands is conducted to examine the effects of chiral ligand structure on selectivity of the dehydration. The kinetic resolution of a variety of rac-β-hydroxy tert-butyl esters in the presence of prolinol chiral ligand 2 and BrZnCH2CO2-t-Bu can provide highly enantioenriched β-hydroxy esters 14-21 with selectivity factors ranging from 11 to 66. Also, application of this asymmetric synthetic methodology to the preparation of enantioenriched flavane derivatives 25-29 is demonstrated.  相似文献

Diethyl (1,2-dibromoethyl)phosphonate was found to undergo cyclopropanation with nitromethane in good yield. The resulting trans β-nitrocyclopropylphosphonate was converted to the trans N-protected aminocyclopropylphosphonate through a reduction–protection sequence. Subsequent hydrolysis gave the free β-aminocyclopropylphosphonic acid without any formation of ring-opening byproduct. Cyclopropanation of 1,2-dibromoethylphosphonates with nitroalkanes and their reduction are also discussed.  相似文献

The 1,2-addition of alkyl Grignard reagents to readily available N-tert-butanesulfinyl ketimines, bearing an α-silyloxy substituent, proceeds in high yields and excellent diastereocontrol. The organocatalysed asymmetric hydrosilylation of a number of N-aryl and alkyl β-substituted enamino esters proceeds in generally good yield and enantioselectivity. Crucial to obtaining high yield and selectivity was the addition of benzoic acid as an additive and under these conditions, both N-alkyl and N-aryl substituents were well tolerated. β-Aryl and alkyl substituents were evaluated and a model proposed to account for the experimental observations based upon enamine tautomerisation and conformational preferences of the reactive ketimine intermediate.  相似文献

A new, efficient palladium-catalyzed bisfunctionalization of ethylidene malononitriles by addition of acetonitrile and allyl groups is developed for the construction of all-carbon quarternary and tertiary centers simultaneously. The four stereoisomers of Δ2-isoxazoline 2, a β-adrenergic receptor antagonist structurally related to Falintolol 1, were prepared by an enzyme-catalyzed kinetic resolution of the unsaturated secondary alcohol (±)-7 followed by its cycloaddition to pyruvonitrile oxide. Through this strategy, diastereomeric aminoalcohols (+)-2a/(−)-2b and (−)-2a/(+)-2b were obtained in 99 and 92% enantiomeric excess, respectively. The absolute configuration to the target compounds was assigned via chemical correlation to the enantiomers of epoxides 4a and 4b, whose stereochemistry had been previously established.  相似文献

The stereoselective synthesis of both cis- and trans-β-fluorostyrene derivatives from a common intermediate, (Z)-1-aryl-2-fluoro-1-(trimethylsilyl)ethenes, is described. The trans isomers are obtained by a stereospecific replacement of the silyl group in the presence of water and a fluoride source, whereas the preparation of the cis isomers is achieved by a bromination/desilicobromination sequence followed by reduction of the newly created C-Br bond. A stereoselective transformation of both stereoisomers of β-fluorostyrene is also presented.  相似文献

A simple route was developed for the synthesis of optically enriched β-amino-α-keto amides. The highly diastereoselective addition of alkyl dithiolanecarboxylates to optically enriched sulfinimines affords the corresponding β-amino-α-keto esters in which the ketone carbonyl group is protected as a dithiolane. Amidation of the ester functionality with a primary amine proceeds readily at room temperature. Treatment with HCl cleaves the N-S bond of the sulfinyl group to provide a free amine functionality, which can then be incorporated into a peptide. Removal of the dithiolane protecting group under oxidative conditions proceeds without epimerization as exemplified by a model dipeptide.  相似文献

Novel aryl-β-C-glucosidation method using glucal boronate was developed. This protocol can offer several advantages including use of non-toxic, easily handling glucal boronate as a crystalline solid and storable at room temperature for several months.
