This review article provides a critical overview of several different synthetic approaches developed for asymmetric preparation of fluorine-containing amines, amino alcohols, α- and β-amino acids. The common feature of these methods is the application of sulfinyl group as a chiral auxiliary to control the stereochemical outcome of the reactions under study. In particular, the following general methods are critically discussed: diastereoselective methylene transfer from diazomethane to the carbonyl of β-keto-γ-fluoroalkyl sulfoxides as a general approach for preparation of various α-fluoroalkyl α-sulfinylalkyl oxiranes. The resulting compounds were used as true chiral synthons for their further elaboration via oxidative or reductive desulfurization, to numerous fluorine-containing and biologically relevant amino- and hydroxy-containing derivatives. Another general approaches discussed here are asymmetric additions to CN double bond. One of them is addition of chiral sulfoxide stabilized carbon nucleophiles to fluorine-containing imines, leading to convenient preparation of alpha-fluoroalkyl derivatives of alpha amino acids and amines. Another approach is asymmetric Reformatsky reaction between N-sulfinyl imines and ethyl bromodifluoroacetate allowing operationally convenient preparation of α,α-difluoro-β-amino acids in enantiomerically pure form. Finally, structurally similar but mechanistically different addition reactions of diethyl difluoromethylphosphonate to N-sulfinyl imines, as a general approach to asymmetric synthesis of α,α-difluoro-β-aminophosphonates and phosphonic acids, are discussed. Effect of fluorine on the mechanism and stereochemical outcome of these reactions is discussed in detail and compared, where it is possible, with that of the analogous reactions of fluorine-free substrates.  相似文献

The addition of DCPA to several α-olefins and α,β-unsaturated esters has been studied. The reaction was found to proceed in boiling benzene and was evidently accelerated by UV irradiation. N-Chloro adducts, formed in reasonable yields, afforded the corresponding diethyl N-(β-chloroalkyl) phosphoroamidates upon reduction with sodium bisulphite solution. β-Chloroamine hydrochlorides or α-chloro-β-aminoacid ester hydrochlorides could be produced from the latter compounds by PN bond cleavage with hydrogen chloride in benzene. The full regioselectivity for anti-Markovnikov addition, as proven by NMR evidence, is in accord with the photolytically initiated radical-chain mechanism proposed for the reaction. This reaction offers an easy, two-step approach to aminochlorination of a double bond in α-olefins and α,β-unsaturated esters.  相似文献

Mannich-type reactions of α-amido p-tolylsulfones with diethyl fluoromalonates and diethyl malonates, respectively in presence of catalytic amount of DBU have been developed. A variety of α-amido p-tolylsulfones prepared from aromatic and aliphatic aldehydes reacts with diethyl fluoromalonates and diethyl malonates, respectively under mild reaction conditions to afford α-fluoro β-amino esters and β-amino esters in moderate to good yield.  相似文献

Addition of diethyl lithiodifluoromethylphosphonate to enantiomerically pure aromatic, heteroaromatic, and aliphatic aldehyde-derived sulfinimines afforded N-sulfinyl α,α-difluoro-β-aminophosphonates with generally good enantioselectivity and in high yield. The reaction with acetophenone-derived sulfinimine resulted in the formation of the addition product with high diastereoselectivity and in only moderate yield. A two-step deprotection involving treatment of diastereomerically pure N-sulfinyl α,α-difluoro-β-aminophosphonates with trifluoroacetic acid in EtOH followed by refluxing with 10 N HCl provided enantiopure α,α-difluoro-β-aminophosphonates and α,α-difluoro-β-aminophosphonic acids. The N-Cbz derivative of (R)-2-amino-1,1-difluoro-2-phenylethylphosphonate was a convenient starting point for the preparation of corresponding difluorophosphonate monoester, difluorophosphonic acid, and difluorophosphonamidic acid. At 21 °C difluorophosphonamidic acid was stable in aqueous solution at pH above 5.  相似文献

N-((S)-1-Phenylethyl)halofluoroethanamides have been investigated as precursors to N-protected α-fluoro-α-amino amides by nucleophilic displacement of halide with nitrogen nucleophiles such as potassium phthalimide, sodium succinimide, sodium glutarimide, trimethylamine and sodium azide. With single diastereoisomers of the iodofluoroethanamide, clean inversion of configuration occurs at room temperature, but subsequent epimerisation may occur as a result of the liberated iodide. The α-fluoro-α-amino amides made underwent a wide variety of reactions depending on conditions, but in many cases the carbon-fluorine bond was compromised. However, reacting trimethylamine and N-((S)-1-phenylethyl)iodofluoroethanamide gave the corresponding α-fluorobetaine amide, and subsequent acidic hydrolysis led to α-fluorobetaine as the first example of an ‘unprotected’ α-fluoroamino acid.  相似文献

The formation of P -- C bond is one of the important problems in synthetic organophosphorous chemistry. This paper describes the study of the formation of P -- C linkage via phosphorylation of carbon atom containing active hydrogen. p-Substituted phenylacetonitrile possessing an active methylene group undergoes stepwise C -- phosphorylation smoothly with diethyl phosphoryl chloride in the presence of naphthalene-sodium. Phosphonyl chloride reacts analogously. Being a deprotonizing agent naphthalene-sodium is distinguished by its availability, effectiveness and mild reaction conditions. The presence of an electron-withdrawing group in the benzene ring reduces the yield of phosphorylation seriously. The chemical shift of ^3^1P NMR of the resulted α-cyano-benzylphosphonates (1) correlates linearly with the Hammett \s\ constants of the substituents. As indicated by ^1H NMR studies the protons in the ester ethyl groups are magnetically nonequivalent. Compound 1 can be alkylated with alkyl halide or condensed with aromatic aldehyde by Hornor-Wittig type reaction due to the presence of active hydrogen atom. The structure-reactivity studies of diethyl phosphoryl chloride on reacting with various β- and β-diketones and β-keto esters (5) as regiospecific reaction were reported. The influences of metal ions in the enolates on the reactivity of the latter were examined and discussed. Only vinyl phosphates with predominantly Z configuration have been isolated exclusively. The C atom with an active hydrogen in the β-diketones investigated is inert to phosphorylation. These experimental results are well supported by reaction selectivity in terms of Eqo/Ecoc based on EHMO calculation.  相似文献

Application of water as a sole solvent promotes the aza-Michael reaction of diethyl vinylphosphonate and diphenylvinylphosphine oxide with α-substituted amino acid sodium salts generated in situ to afford the corresponding β-aminophosphonates and β-aminophosphine oxides in excellent yields and of high purity. The approach is equally suitable for the synthesis of both racemic and optically active compounds. In the case of glycine, the mono and bis(phosphonoethyl)-substituted products are formed in 6:4 ratio and when using a stoichiometric amount of the reactants, N,N-bis[2-(diethoxyphosphoryl)ethyl]glycine was the only product. In contrast, to perform the double phosphonoethylation of d,l-alanine, prolonged heating of the reaction mixture was required.  相似文献

The synthesis of a Δ1-carbapenem and two β-lactams possessing a Br-atom at the N-substituting center not involved in the lactam ring and bearing the carboxyl group is described. The β-lactams having this kind of Br-substitution are more susceptible to nucleophilic attack than those having a conjugated double bond with the N-atom of the β-lactam ring. DBU is found to be an excellent reagent for the elimination of the silyloxy function. Moreover, a simple method for the addition of diethyl phosphite to an α, β-unsaturated double bond using a catalytic amount of NaH is described.  相似文献

The Rh(II)-catalyzed intramolecular C H insertion reactions of N,N-dialkyl-α-diazo-α-(diethylphosphono)acetamides 2a , f–j in CHCl3 or ClCH2CH2Cl were found to give monocyclic and bicyclic α-phosphono-β-lactams, 3a and 3f–j , in 43–67% yields via regiospecific α-C H insertion of the N-alkyl groups. Similar treatment of N-benzyl-N-isopropyl-α-diazo-α-(diethylphosphono)acetamide ( 2b ) and the corresponding N-isobutyl-N-methylacetamide 2d in ClCH2CH2Cl afforded mixtures of β-lactams 3b (35%) and and 3b ′ (16%), β-lactam 3d (47%), and γ-lactam 4d (10%), respectively, each of which is formed by the competitive C H insertion reaction between benzylic and isopropyl α-C H bonds and between methyl α-C H and methine β-C H bonds, respectively. For the formation of β-lactams, the selectivity in the rhodium-mediated C H insertion in ClCH2CH2Cl follows the order methyl > methine > benzylic α-C H bond on N-substituents. The N,N-dibutyl-α-diazo homologue 2c and Nα[α-diazo-α-(diethylphosphono)acetyl]-2-methylindoline ( 2k ) exclusively produced γ-lactams 4c (67%) and 4k (81%) via insertion into the methylene β-C H and methyl β-C H bonds. tert-Butyl N-[α-diazo-α-(dibenzylphosphono)acetyl]-piperidine-2-carboxylate ( 2m ) on similar treatment, followed by deprotection of the benzyl ester afforded the 7-phosphono carbacepham 6 in 32% overall yield. Similar Rh(II)-catalyzed cyclization of N-methyl-N[4-benzyloxy-α-(diethylphosphono)-phenyl(diethyl-phosphono)methyl]-α-diazo-acetamide ( 2n ) led to 1-[4′-benzylphenyl(diethylphosphono)methyl] -3-(diethyl-phosphono)azetidin-2-one ( 3n ) in 78% yicld. The phosphono group at C-7 of 3f was converted into the acetylamino group via a four-step reaction. Application of chiral rhodium(II) carboxylates 12a–c to the insertion reactions of 2b , c produced α-phosphono-β-and γ-lactams, 3b and 4c , in 6–24% ee and 25–29% ee, respectively.  相似文献

A simple, efficient, and general method has been developed for the one-pot synthesis of α-aminophosphonic esters in water using β-cyclodextrin as an efficient catalyst. α-Aminophosphonic esters were obtained in good yields (45-82%) and purity under mild conditions by the reaction of diethyl phosphite with a mixture of aldehyde and amine in water at reflux.  相似文献

Aminophosphonic acids have become increasingly important in different fields of chemistry, medicine and agriculture. This account outlines the results obtained in the author’s laboratory on the asymmetric synthesis of acyclic and cyclic aminophosphonic acids mediated by chiral sulfinyl auxiliary. A key reaction in the synthesis of enantiopure α- and β-aminoalkanephosphonic acids involving a highly diastereoselective addition of phosphite anions or α-phosphonate carbanions to enantiopure sulfinimines is discussed. The asymmetric cyclopropanation of enantiopure α-phosphorylvinyl sulfoxides with sulfur ylides is presented as a platform for developing a new approach to optically active β-aminocyclopropanephosphonic acids. It is exemplified by the total synthesis of enantiopure β-amino-γ-phenylcyclopropanephosphonic acid - a constrained analogue of the GABAB antagonist phaclofen.  相似文献

Tetra-n-butylammonium peroxydisulfate was prepared and found to be a good source of tetra-n-butylammonium sulfate radical by its oxygen - oxygen bond cleavage. The sulfate radical can be utilized for the efficient organic synthesis in organic solvents. Electron deficient olefins such as α,β-unsaturated ketones or lactones were smoothly β-masked formylated by treatment of the olefins with 1,3-dioxolane in the presence of tetra-n-butylammonium peroxydisulfate. Extremely high diastereofacial selectivity (?100% de) was obtained in β-masked formylation of α,β-unsaturated lactone, (S)-5-(t-butyldiphenyl silyloxymethyl)-2(5H)-furanose.  相似文献

Theoretical studies on the α- and β-forms nitroguanidine were carried out using ab initio theoretical methods, at the MP2/6-31G(d,p) level. The predicted geometrical parameters were in good agreement with the available theoretical values, which calculated by other author. The three C-N bond lengths in α-form nitroguanidine were different, the longest bond length was 1.430 A, the shortest was 1.283 A. But they were almost similar in β-form, the longest was 1.375 A, the shortest was 1.322 A. Therefore there were conjugative effects in β-form but not in α-form. The calculated results also show that the β-form is stable with respect to the α-form from energetically, lower 28.16 kJ/mol corrected with zero point vibrational energy. The transition-state for the unimolecular isomerization was conformed by the IRC calculation. The calculated energy barrier for the direct intramolecular hydrogen atom transfer isomerization process was 132.95 kJ/mol. The isomerization reaction, exothermal reaction, is a typical intramolecular hydrogen atom synfacial transfer reaction. Rate constants of the isomerization reaction were evaluated within the temperature range of 200-1773 K by the classical transition state theory. The rate constant was 1.99×10-11 s-1 and the equilibrium constant was 1.00×105 at 298 K. With the temperature increasing, the equilibrium value decayed and the reaction process was more difficult.  相似文献

The asymmetric Michael addition of diethyl malonate and α-substituted diethyl malonates to aromatic nitroalkenes was carried out under mild reaction conditions in a solid–liquid phase transfer reaction in the presence of α-d-glucopyranoside- and α-d-mannopyranoside-based crown ethers as the catalysts. The use of d-glucose-based lariat ether 1 gave the best results. The substituents of the β-nitrostyrene and the diethyl malonate had a significant impact on the chemical yields and enantioselectivity. The addition of diethyl-2-acetamidomalonate to aromatic nitroalkenes afforded the corresponding Michael adducts in moderate to high enantiomeric excess (ee up to 99%). The reaction of diethyl-2-methylmalonate with 2-nitro-β-nitrostyrene gave the adduct with 93% enantiomeric excess in the presence of crown catalyst 1.  相似文献

The general synthesis of a new class of non-natural diamino acids, 2-amino-3-[(2′-aminoalkyl)seleno]propanoic acids, or Se-(aminoalkyl)selenocysteines, is reported. Under the conditions devised, enantiopure N-Boc-protected β-l-iodoamines, which are readily generated from proteinogenic α-amino acids, were treated with the selenolate anion obtained from NaBH4 splitting of the Se-Se bond in commercial l-selenocystine. The Se-alkylation products were enantiomerically pure and the reaction is high yielding (92-98%), without any detectable traces of accompanying by-products.  相似文献

Copper(II) or tin(II) trifluoromethanesulfonate in combination with TMSCl effectively activates a C-H bond for the imino-ene reaction of N-tosylarylaldimines with α-methylstyrene. A wide variety of N-tosylarylaldimines were used to give homoallylamines in good to excellent yields under mild conditions. The imino-ene adduct was converted into a β-amino ketone. The synthesis of a 2,4-substituted pyrrolidine and a piperidine was also achieved from the imino-ene product via a Mitsunobu reaction and a Grubbs cyclization, respectively.  相似文献

Treatment of lithium ester enolates with magnesium alkylidene carbenoids, generated from 1-chlorovinyl p-tolyl sulfoxides with isopropylmagnesium chloride via the sulfoxide-magnesium exchange reaction, gave β,γ-unsaturated esters in moderate to good yields. When this reaction was conducted with the lithium ester enolates of α-chlorocarboxylic acid esters, allenic esters were obtained. This procedure provides an unprecedented way for the synthesis of β,γ-unsaturated esters and allenic esters from ketones with the construction of a carbon-carbon bond between α- and β-positions.  相似文献
