Radical cyclizations of enantiomerically enriched N-allyl-o-iodoanilides provide N-acyl-3-alkyl-2,3-dihydroindoles in good yields and with good to excellent levels of chirality transfer from the N-Ar axis to the new stereocenter. In competitive cyclizations of N-acryloyl-N-allyl-o-iodoanilides, the addition of an o-methyl group reverses the regioselectivity of the radical cyclization from the acryloyl group to the allyl group. Approximate rate constants for representative radical cyclizations have been measured to provide insight into the origin of these observations.  相似文献

Pd(II)-catalyzed intramolecular Wacker-type cyclizations of o-trisubstituted allylphenols were studied. The chelation-induced axially chiral catalytic system, Pd(CF3COO)2-8 (1:1 molar ratio), showed excellent catalytic activities and enantioselectivities in the Wacker-type cyclizations of o-trisubstituted allylphenols with up to 94% ee.  相似文献

Various biologically important saccharin skeletons and their N-alkyl derivatives have been efficiently prepared by chromium(VI) oxide catalyzed H5IO6 oxidation of N-alkyl-o-methyl-arenesulfonamides in acetonitrile. N-tert-Butyl saccharin skeletons were easily prepared by H5IO6-CrO3 oxidation of N-tert-butyl-o-methyl arenesulfonamides in the presence of acetic anhydride. The method that furnished the novel fluoro and trifluoromethyl substituted saccharin skeletons is characterized by two steps, a simple work-up procedure, a single purification and good overall yields from substituted toluene derivatives.  相似文献

Substituted 5-hexen-1-oxyl radicals have been generated from N-(5-hexen-1-oxy)-5-(p-methoxyphenyl)-4-methylthiazole-2(3H)-thiones under tin-free conditions and have been successfully applied as reactive intermediates in a mechanistic study on the formation of bromomethyl-substituted tetrahydropyrans via 6-exo-trig selective cyclizations.  相似文献

Treatment of N-alkyl- and N-aryl-imines of 2,3-diaryl- and 2-alkyl-3-aryl-3-(benzotriazol-1-yl)propenals with trifluoroacetic anhydride in THF at room temperature gave 5-alkyl-4-aryl-6-[N-alkyl (and aryl)-N-trifluoroacetyl]amino-2,3-benzo-1,3a,6a-triazapentalenes in moderate to good yields. On heating triazapentalenes having R2=aryl in MeOH at reflux, detrifluoroacetylation of triazapentalene occurred to give title compounds in good yields. However, the same treatment of triazapentalenes having R2=alkyl did not give the corresponding detrifluoroacetylation product. The title compounds and 5-alkyl-4-aryl-6-(N-alkyl-N-trifluoroacetyl)amino-2,3-benzo-1,3a,6a-triazapentalenes were found to be good precursors for the synthesis of 1-(o-aminophenyl)-3-arylamino-4-alkyl (and aryl)-5-arylpyrazoles and 1-(o-aminophenyl)-3-(N-alkyl-N-trifluoroacetyl)amino-4-alkyl (and aryl)-5-arylpyrazoles, respectively.  相似文献

Two-step cyclizations of 2-alkylthiazolines or 2-alkyloxazolines with α,α-disubstituted diacid chlorides produce excellent yields of 6,6-dialkyl-2,3-dihydrothiazolo[3,2-a]pyridine-5,7-diones and 6,6-dialkyl-2,3-dihydrooxazolo[3,2-a]pyridine-5,7-diones in refluxing acetonitrile containing Et3N. Similar cyclizations using N-(chlorocarbonyl) isocyanate in place of diacid chlorides produced 2,3-dihydrothiazolo[3,2-c]pyrimidine-5,7-diones or 2,3-dihydrooxazolo[3,2-c]pyrimidine-5,7-diones, respectively. Each cyclization proceeded through cyclic ketene-N,X-acetal (X = O or S) intermediates.  相似文献

Oxidative radical cyclizations starting from easily accessible N-allylic β-alanine esters are reported. Deprotonation generates the corresponding enolates, which are transformed efficiently into α-ester radicals by single electron transfer mediated by ferrocenium hexafluorophosphate. The stereochemistry of the radical 5-exo cyclization can be switched by the configuration of the enolate precursor. First examples of asymmetric oxidative radical cyclizations using N-(1-phenylethyl)-substituted β-amino esters are reported. Only two of the four possible diastereomers are formed from the (E)-enolate with high cis-selectivity. From (Z)-enolates, an additional diastereomer is formed, which is likely to be formed only under chelation control.  相似文献

A variety of benzimidazo[1,2-c]quinazolin-6-ones derivatives can be accessed in moderate to good yields under simple and metal-free reaction conditions using indoles and o-benzenediamines oxidized by iodine and TBHP. This procedure works in reasonable yields for different indoles as well as o-benzenediamines thus may provide a good synthesis of quinazolinones. A TBHP oxidized ring expansion reaction mechanism that explains the synthesis of benzimidazo[1,2-c]quinazolin-6-ones were reported.  相似文献

The scope of an alkoxyl radical version of the classical bromine cyclization was explored. Oxygen-centered radicals were generated in photochemically initiated radical chain reactions from N-alkenoxy-4-(p-chlorophenyl)thiazole-2(3H)-thiones 5ac, N-alkenoxy-4-methylthiazole-2(3H)-thiones 13, 14, 21, rac-23, and N-alkenoxypyridine-2(1H)-thiones 6df, 16, rac-25. Thus, 2-(2-bromopropyl)-substituted tetrahydrofurans 4ac, which are minor compounds (< 10 %) in NBS-mediated bromine cyclizations of corresponding alkenols 1ac, were prepared in 87–90 % yield and with good to excellent diastereoselectivities. Further, photochemical conversions of O-alkyl thiohydroxamates 14, 16 and 21 in benzene and BrCCl3 afforded β-oxy-functionalized bromomethyl substituted tetrahydrofurans 29, 34, and 36. The results of this study indicate, that efficient 5-exo-trig cyclizations of β-oxy-substituted 4-penten-1-oxyl radical require an electronwithdrawing substituent at the β-heteroatom substituent. In the third part of the study a synthetically useful new access to oxabicyclo〚4.3.0〛nonanes rac-38 and rac-40 is reported which takes profit from highly diastereoselective 5-exo-trig-ring closures and, in case of the formation of rac-40, stereoselective bromine atom transfer.  相似文献

The reaction of the Baylis-Hillman adducts 1b-f derived from o-nitrobenzaldehydes in trifluoroacetic acid in the presence of triflic acid (0.2 equiv.) afforded 3-substituted-4-hydroxyquinoline N-oxides 2b-e and 2a in good to moderate yields. The reaction mechanism was evidenced by the experiment with 1f, the Baylis-Hillman adduct of 2-nitrobenzaldehyde N-tosylimine, as the one involving N-hydroxyisoxazoline as the key intermediate.  相似文献

Arylpalladium or aminopalladium species bearing a 2,2-difluorovinyl group undergo an unusual 5-endo alkene insertion followed by β-fluorine elimination. These processes provide a facile access to ring-fluorinated five-membered carbocyclic and heterocyclic compounds starting from an o-(3,3-difluoroallyl)phenyl trifluoromethanesulfonate and 3,3-difluoroallyl ketone O-pentafluorobenzoyloximes. In both systems, the two vinylic fluorine atoms are essential for Heck-type 5-endo-trig cyclizations.  相似文献

The chemical synthesis of peptide nucleic acid (PNA) monomers was accomplished using various combinations of the o-nitroveratryloxycarbonyl (NVOC) group (N-aminoethylglycine backbone) and base labile acyl-type nucleobase protecting groups (anisoyl for adenine and cytosine; isobutyryl for guanine), thus offering a photolithographic solid-phase PNA synthetic strategy compatible with photolithographic oligonucleotide synthesis conditions and allowing the in situ synthesis of PNA microarrays in an essentially neutral medium, by avoiding the use of the commonly used deprotection reagents such as trifluoroacetic acid or piperidine. Convenient methods were also explored to prepare 1-(carboxymethyl)-4-N-(4-methoxybenzoyl)cytosine and 9-(carboxymethyl)-2-N-(isobutyryl)guanine with good yields.  相似文献

The base-catalysed hydrolysis of N-ethyl-2-methylbenzazol-3-ium iodides was re-examined performing the reaction in boiling 96% ethanol, in the presence of triethylamine. The resulting unstable intermediates were isolated as the corresponding ether, thioether or selenoether derivatives, depending on the starting benzazole salt, by trapping via alkylation with ethyl and hexyl iodides, in moderate to good yields. Reduction of the o-(alkylchalcogen)acetanilides so obtained afforded the corresponding o-(alkylchalcogen)anilines. This methodology provides potential access to o-(alkylchalcogen)anilines bearing up to three different N-alkyl groups introduced in an unambiguous and regioselective way.  相似文献

NMR study of the reactivity of multifunctional ligand cis,cis-C6H9(NHCH2C6H4-o-PPh2)3 (1) with GaMe3 and Zr(NMe2)4 was carried out, yielding [cis,cis-(κN-NHCH2C6H4-o-PPh2)(κN-NCH2C6H4-o-PPh2)2C6H9]GaMe (2) and [cis,cis-(NCH2C6H4-o-PPh2)3C6H9]Ga2Me3 (3), and [cis,cis-(NCH2C6H4-o-PPh2)3C6H9]Zr(NMe2) (4), respectively. The spectral properties of 2 and 3 are very similar to that observed for the equivalent aluminum species already reported, but form at a much slower rate which allows for the observation of a GaMe31 adduct. Species 4 undergoes coordination/displacement of one of the phosphine arms, which was observed using both NMR spectroscopy and DFT analyses.  相似文献

A series of novel N-benzylcarboxamide derivatives of bicyclic compounds, 3,4-dihydropyrido[3,2-f][1,4]oxazepin-5(2H)-one and 2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-one, were synthesized by cyclization of N-benzyl-2-chloro-N-(2-hydroxyethyl)- [and -(3-hydroxypropyl)-] nicotinamides, respectively. Atropisomerism was observed in 5-[3,5-bis(trifluoromethyl)benzyl]-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-ones due to steric hindrance of the carboxamide moiety and restriction of its rotation. Cyclization of N-[3,5-bis(trifluoromethyl)benzyl]-2-chloro-N-[(2S)-3-hydroxy-2-methylpropyl]-5-methyl-4-phenylnicotinamide gave (3S)-5-[3,5-bis(trifluoromethyl)benzyl]-3,8-dimethyl-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3b][1,5]oxazocin-6-one, which exists predominantly in the thermodynamically stable aR-conformer in CDCl3. This compound showed excellent NK1-antagonistic activity with IC50 value (in vitro inhibition of [125I]-Bolton-Hunter-substance P binding in human IM-9 cells) of 0.47 nM, which is ca. 200-fold more potent than that of its enantiomer, indicating that the atropisomer chirality affects NK1-receptor recognition.  相似文献
