(S)-ortho-(p-Tolylsulfinyl)benzyl alkyl (and aryl) ketones 1a-e were prepared in good yields by reaction of esters or nitriles with the lithium benzyl carbanion derived from 2-(p-tolylsulfinyl) methylbenzene. α-Methylbenzyl ketones 2 were prepared as ca. 1:1 diastereoisomeric mixtures by methylation of the unsubstituted ketones 1 with NaH/MeI. The use of the ethylbenzene derivative as the starting material afforded complex mixtures. The obtention of pure (S,(S)S)-2 diastereoisomers could be attained in good yields by oxidation with PCC of the alcohols (epimeric mixtures at the hydroxylic carbon) obtained from reactions of aldehydes with the lithium carbanion derived from 2-(p-tolylsulfinyl)ethylbenzene.  相似文献

2-(tert-Butyldimethylsilyloxymethyl)-4-(methoxymethoxy)pent-2-enyl(tributyl)stannane is transmetallated by tin(IV) chloride stereoselectively to give a pent-1-en-3-yltin trichloride that reacts with aldehydes with excellent (E)-1,5-syn-stereocontrol, e.g. (3E)-1,5-syn-3-(tert-butyldimethylsilyloxymethyl)-5-(methoxymethoxy)-1-phenylhex-3-en-1-ol was the dominant product with benzaldehyde. The products from these reactions were taken through to more complex 2-substituted alk-2-enyl(tributyl)stannanes but only very low yields of the expected products were obtained from tin(IV) chloride mediated reactions of these stannanes and aldehydes. Nevertheless a stereoselective synthesis of 2-substituted 4-[(E)-2-alkoxypropylidene]tetrahydrofurans was developed.  相似文献

Racemic 2-trimethylsilyl- and 2-trimethylstannyl-1-(3-phenyl-2-propynyl)ferrocene (rac-1a,b) as well as the dextrorotatory specimen of the latter, (+)-1b, were synthesized in two steps from racemic 1-formyl-2-trimethylsilyl- and 1-formyl-2-trimethylstannylferrocenes (2a,b) or from the levorotatory specimen of the latter, (–)-2b, respectively. On the contact with strongly alkaline alumina compounds1a,b and (+)-1b undergo diastereoselective prototropic acetylene-allene rearrangement to give predominantly one of the two possible stereoisomers of 2-trimethylsilyl- or 2-trimethylstannyl-1-(3-phenyl-1,2-propadienyl)ferrocenes,rac-4a,b or (+)-4b, depending on the starting material (d. e. 30–40 %). The extent of intramolecular asymmetric induction in the formation of the axially chiral fragment during the transformation of (+)-1b to (+)-4b is estimated at 38 %.Translated fromIzvestiya Akademii Nauk. The regio- and stereoselectivity of the reactions of carbanions, generated from alkanals, carboxylates of the type XYCHCOOEt (X = EtOOC, CN, Ac; Y = H, Br), or derivatives of 3-methyl-4-phosphono-2-butenoic acid using PTC techniques, with aldehydes of various types (alkanals, ,-enals, cross-conjugated enedials, benzaldehydes,etc.) are reviewed. The factors affecting the outcome of these reactions are discussed. The carbanion analogs, triphenylphosphorus ylides, are shown to attack selectively at one of the aldehyde groups of aromatic dialdehydes. The catalytic enantioselective Mannich reaction promoted by chiral palladium complexes is described. The treatment of α-fluoro-β-ketoesters with N-Boc-aldimines under mild reaction conditions afforded the corresponding β-aminated α-fluoro-β-ketoesters with excellent enantioselectivities (up to 99% ee).  相似文献

L -2-Hydroxyglutaric aciduria is a recently discovered inherited neurometabolic disease for which diagnostic identification of the right enantiomer is required. A method is described where alkyl esters of both L -and D -2-hydroxyglutaric acids are prepared without side lactonization. Esterification is achieved at room temperature by reaction with appropriate alkyl chloroformates. This method avoids lactonization, in contrast to esterification with alcohols in acidic media or with diazomethane. The identity of the derivatives is established by HRGC-FTIR and HRGC-MS. Chiral HRGC separation with capillaries coated with 1(R)-trans-N-N'-1,2-cyclohexylenebisbenzamideoligodimethyL-siloxane and with a cyclodextrin is compared. The method is adequate for quantitative determinations.  相似文献

Eight platinum(II) complexes with the new chiral ligands, (1R,2R)-N 1-(pyridine-2-ylmethyl) cyclohexane-1,2-diamine (R) or (1S,2S)-N 1-(pyridine-2-ylmethyl) cyclohexane-1,2-diamine (S) as the carrier groups were designed, synthesized, and spectrally characterized. All platinum(II) complexes showed much better aqueous solubility than cisplatin and oxaliplatin. In vitro cytotoxicity of the compounds against human HepG-2, MCF-7, A549, and HCT-116 cell lines was evaluated. Results indicate that all compounds with R as the carrier group showed cytotoxicity against HCT-116, A549, and MCF-7 cell lines; however, all compounds with S as carrier group exhibited disappointing cytotoxicity against tested cell lines. Compound R2, bearing ClCH2COO- as leaving group, exhibited better cytotoxicity than that of carboplatin against A549 and MCF-7 cell lines and also showed close activity to oxaliplatin against HCT-116 cell line.  相似文献

A number of novel chiral diamines 3 , (1R,2R)‐N‐monoalkylcyclohexane‐1,2‐diamines, were designed and synthesized from trans‐cyclohexane‐1,2‐diamine and applied to the catalytic asymmetric Henry reaction of benzaldehyde and nitromethane to provide β‐nitroalcohol in high yield (up to 99%) and good enantiomeric excess (up to 89%). By using ligand (1R,2R)‐N1‐(4‐methylpentan‐2‐yl)cyclohexane‐1,2‐diamine ( 3g ), the reaction was optimized in terms of the metal ion, temperature, solvent and base. Further experiments indicated that the complex, 3g –Cu(OAc)2, was an efficient catalyst in the asymmetric Henry reaction between different aldehydes and nitromethane, and the desired products have been obtained with high chemical yields (up to 99%) and high enantiomeric excess (up to 93%). The optimized catalyst promoted the diastereoselective Henry reaction of various aldehyde substrates and nitroalkane, which gave the corresponding anti‐selective adduct with up to 99% yield and 83:17 anti/syn selectivity. Upon scaling up to gram quantities, the β‐nitroalcohol was obtained in good yield (96%) with excellent selectivities (93% ee). The use of chiral, binaphthyl-based phosphepines as catalysts improves previous results for the enantioselective [3+2] cyclisation reactions between allenic esters and N-tosylimines, both in terms of conversion rate and enantioselectivity. Pyrrolines bearing 1-naphthyl, phenyl, o-tolyl and p-MeO-phenyl substituents on the stereogenic alpha-carbon have been obtained with enantiomeric excesses up to 64-80%.  相似文献

The addition reaction of carbon nucleophiles to cis-substituted cyclopropanecarbaldehydes was systematically investigated. Ab initio calculations of model cyclopropanecarbaldehydes suggested that the bisected s-cis and s-trans conformers are the only two minimum energy conformers, which are stabilized due to the π-donating stereoelectronic effect of the cyclopropane ring. The experimental results of a series of substrates, that is, cyclopropanecarbaldehydes 1-5 bearing a cis-(tert-butyldiphenylsilyloxy)methyl group, a cis-benzyloxymethyl group, a cis-(p-methoxybenzyloxy)methyl group, cis-N,N-diethylcarbamoyl and trans-phenyl groups, and cis-(tert-butyldiphenylsilyloxy)methyl and trans-phenyl groups, respectively, showed that highly anti-selective Grignard additions could be realized. It turned out that it occurred via an unusual 7-membered 1,4-chelation-controlled pathway. Highly stereoselective Grignard addition via the chelation-controlled pathway occurred even in the reaction of the usually non-chelating silyl ether-type substrate 5. The results have great importance because the 1,4-chelation-controlled stereoselective addition reactions can indeed be realized. Under non-chelation conditions, the syn-products were produced with moderate stereoselectivity, which are likely to be formed via the bisected s-cis conformation-like transition state stabilized by the characteristic orbital interaction. These reactions, especially the chelation-controlled reaction, should be useful because of their t stereoselectivity and stereochemical predictability.  相似文献

An organocatalyst prepared from (2R,3R)-diethyl 2-amino-3-hydroxysuccinate and L-proline exhibited high regio- and enantioselectivities for the direct aldol reactions of hydroxyacetone and fluoroacetone with aldehydes in aqueous media. It was found that water could be used to control the regioselectivity. The presence of 20-30 mol% of the catalyst afforded the direct aldol reactions of a wide range of aldehydes with hydroxyacetone to give the otherwise disfavored products with excellent enantioselectivities, ranging from 91 to 99% ee, and high regioselectivities. Aldolizations of fluoroacetone with aldehydes mediated by 30 mol% of the organocatalyst in aqueous media preferentially occurred at the methyl group, yielding products with high enantioselectivities (up to 91% ee); however, these additions took place dominantly at the fluoromethyl group in THF. Optically active 3,5-disubstituted tetrahydrofurans and (2S,4R)-dihydroxy-4-biphenylbutyric acid were prepared by starting from the aldol reaction of hydroxyacetone. Theoretical studies on the role of water in controlling the regioselectivity revealed that the hydrogen bonds formed between the amide oxygen of proline amide, the hydroxy of hydroxyacetone, and water are responsible for the regioselectivity by microsolvation with explicit one water molecule as a hydrogen-bond donor and/or an acceptor.  相似文献

To value a stoichometric asymmetric synthesis it is important for a synthetic chemist to gain information about those structural elements of a chiral auxiliary which are responsible for high diastertomeric excesses in a particular reaction. Furtheron it is desirable to have detailed knowledge about the mechanism of the chirality transfer. In this paper an empirical relation is formulated for the correlation of structural variations of a chiral auxiliary and its influence of the diastereomeric excess in the photochemical oxetane formation in correspondance to the Ugi/Ruch concept. For this purpose we use a Linear Free Energy Relationship (LFER) which we have adjusted to reaction (2) in order to obtain quantitative information about the selectivity of this reaction on the basis of special parameters of the auxiliary applied.  相似文献

Lewis acid catalyzed asymmetric halohydrin reactions—(halohydroxylation as well as halomethoxylation) of chiral α,β-unsaturated carboxylic acid derivatives were performed using N-halosuccinimide (NXS; X = Br, I) as the halogen source. Regio- and anti-selectivity of 100% and moderate to good diastereoselectivity with good yields were observed when Oppolzer’s sultam was used as the chiral auxiliary. Among the Lewis acids, Yb(OTf)3 was found to be the best catalyst. Alkenoyl and cinnamoyl substrates smoothly underwent bromohydrin reactions and the more electron-rich cinnamoyl substrates preferred to undergo iodohydrin reactions. However, electron-deficient cinnamoyl substrates did not respond to this Lewis acid catalyzed halohydrin reaction with NXS (X = Cl, Br, I).  相似文献

Cyclopolymerization of benzaldehyde divinyl acetal ( 1 ) and 1,2-divinyloxybenzene ( 4 ) was carried out using (+)- or (−)-camphorsulfonic acid [(+)- or (−)- 2 ]/ZnCl2 initiating system. Polymer 3 obtained from monomer 1 consisted essentially of cyclic repeating units, whereas polymer 5 from 4 possessed a small amount of residual vinyloxy groups. Both polymers 3 and 5 using (+)- 2 /ZnCl2 showed optical activity with a negative sign of optical rotation. The molar optical rotation values of polymer 3 using ZnCl2 · OEt2 were larger than those using ZnCl2. The CD spectra of the polymer with [Φ] = −30.1° ((−)- 3 ) and (4S,6S)-4,6-dimethyl-2-phenyl-1,3-dioxane showed a negative Cotton effect at 210 nm, whereas the polymer with [Φ] = +24.1° ((+)- 3 ) and (4R,6R)-4,6-dimethyl-2-phenyl-1,3-dioxane had mirror image CD curves with a positive Cotton effect at 210 nm.
