The bioassay-guided fractionation of the cytotoxic constituents of the Japanese sea hare Dollabella auricularia led to the isolation of aurilide (1), a 26-membered cyclodepsipeptide. The gross structure of 1 was established by spectroscopic analysis including 2D NMR techniques. The absolute stereostructure was determined by chiral HPLC analysis of acid hydrolysates of 1 and by the enantioselective synthesis of a degradation product arising from a dihydroxylated fatty acid portion. The enantioselective synthesis of 1 was achieved in 12% overall yield (16 steps) and confirmed the absolute stereostructure of 1. The cytotoxicity of 1 was evaluated using a synthetic sample, which was found to exhibit potent cytotoxicity against HeLa S3 cells with an IC50 of 0.011 μg/mL. Further biological and pharmacological studies of 1 have been carried out by using synthetic 1.  相似文献

The mechanism by which marine algae show resistance to pathogenic microorganisms remains poorly understood. To examine the possible role that algal secondary metabolites play in the prevention of infection, we examined the abundant green alga Penicillus capitatus, one of the major shallow water algae found in the Tropical Atlantic Ocean. Both aqueous and EtOAc extracts of this alga were found to be potent inhibitors of the well-known marine algal pathogen Lindra thallasiae. Using L. thallasiae in bioassay-guided fractionation, we isolated two new triterpene sulfate esters, capisterones A (1) and B (2). The capisterones are potent inhibitors of L. thallasiae at natural and below natural concentrations. The structures of the capisterones, with relative stereochemistry only, were assigned by comprehensive spectral analyses that relied heavily on 2D NMR methods.  相似文献

Rhizopodin was isolated as cytostatic and weakly antifungal macrolide (1) and later characterized as potent actin-depolymerizing agent. It is produced by the myxobacterium Myxococcus stipitatus, which enables a fermentative supply of the drug for biological studies. We here report a revised structure that characterizes rhizopodin (2) as the first known dimeric bis-lactone exhibiting side chains that terminate in N-methyl-vinylformamide groups, which are otherwise found in smaller marine toxins also targeting the actin cytoskeleton. Compound 2 might function as bivalent inhibitor forming ternary complexes with actin which would explain its high efficacy.  相似文献

Chromatographic fractionation of methanol extract from roots of the Piper chaba Hunter resulted in the isolation of four new dimeric alkaloids, chabamide H (1), I (2), J (3), K (4) together with 11 known compounds (5-15). Their chemical structures and relative stereochemistry were determined on the basis of the comprehensive spectroscopic techniques (IR, Mass, and NMR) and further confirmed by comparison of the data with those reported in literature. In addition, cytotoxic activities of all the dimeric amides (1-7) along with their monomers (8-10) were evaluated against cervical (HELA), breast (MCF-7), liver (HEPG2), colon (HT-29), and colon (COLO-205) cancer cell lines. Among the tested isolates, 5 and 7 exhibited potent cytotoxic activity against COLO-205 cell line with IC50 value of 3.10 μg/mL and 0.018 μg/mL, respectively. To prove biogenesis of the newly isolated compounds, biomimetic synthesis has also been carried out via Diels-Alder reaction by using copper(II) salts in aqueous medium.  相似文献

Five new dolastanes (1-5) were isolated from the brown alga Dilophus spiralis. The structural elucidation of the isolated compounds and the assignment of relative stereochemistry were based on analyses of their spectroscopic data. The structure proposed for 1 was confirmed by single crystal X-ray diffraction analysis, whereas its absolute stereochemistry was determined using the modified Mosher's method. The cytotoxicity of 1-4 was evaluated against seven cell lines.  相似文献

Chemical investigations on the acetone extract of the Formosan soft coral Nephthea erecta have afforded a new calamenene-type sesquiterpene with a mercaptan group at C-15, erectathiol (1), and a previously reported sesquiterpenoid, (+)-trans-calamenene (2). A novel sec-germacrane sesquiterpene (3), along with a novel norergosterol, chabrosterol (4), possessing a 19-norergostane skeleton, was isolated from the other soft coral Nephthea chabroli. The structures of these metabolites were elucidated through extensive spectroscopic analyses. In vitro anti-inflammatory activity of 1 and 4 (10 μM) significantly reduced the levels of the iNOS protein (58.0 ± 6.5% and 12.4 ± 2.9%) and COX-2 protein (108.7 ± 4.5% and 45.2 ± 5.4%). In addition, metabolite 1 (166 μg/disk) exhibited antimicrobial activities against a small panel of bacterial strains.  相似文献

Six novel tetranortriterpenoid derivatives were isolated from the methanolic extract of the whole bodies of Munronia henryi, namely, munronins A-F (1-6). In compounds (3-6), the side chains are rare in tetranortriterpenoids. Their structures were established by extensive NMR experiments. Based on the diversity of the side chains, possible biodegradations for the side chains of compounds 1-6 from euphane or tirucallane skeleton are proposed. Munronins A-E exhibited moderate antifeeding activity against Pieris brassicae L, while munronin F showed negative activity.  相似文献

Twenty-five new highly oxygenated prieurianin-type limonoids, zaphaprinins A–Y (125), were isolated from the fruits of Aphanamixis grandifolia. Their structures were elucidated by extensive spectroscopic analyses. Absolute configurations for the key structures like 1 were assigned by a comprehensive method, such as comparing the calculated electronic circular dichroism (ECD) with the experimental one, single-crystal X-ray and Mosher ester method were also involved for other compounds' study. The solid data from X-ray diffraction allowed revision of the stereochemistry of the C1–C14 tetrahydropyran ring-type limonoid. A new pigment, laetiporic acid, has been isolated from fruit-bodies of the wood-rotting fungus Laetiporus sulphureus (sulfur shelf). Structure elucidation by application of extensive 2D NMR techniques permitted its identification as a polyene of non-isoprenoid origin. Laetiporic acid, which represents the main pigment in L. sulphureus basidiocarps, bears an unprecedented decaene skeleton as part of its chromophore and, interestingly, contains double bonds with a stable cis configuration.  相似文献

Three novel dimeric sesquiterpenoids, named parviflorenes G-I (1-3), have been isolated from Curcuma parviflora (Zingiberaceae), and their structures were elucidated by means of spectroscopic studies. Absolute stereochemistry of parviflorene G (1) as well as previously isolated related compounds, parviflorenes A (4), B (5), D (6), and F (7), was revealed by CD spectral data and chemical means. Parviflorenes G (1) and I (3) were cytotoxic against HeLa cells, while parviflorenes A (4) and F (7) were cytotoxic against all tested tumor cell lines in the human cancer cell line panel assay.  相似文献

A rare alkaloid, shahidine (1), having an unstable oxazoline core has been isolated as a major constituent from the fresh leaves of Aegle marmelos. It is moisture-sensitive, and found to be the parent compound of aegeline and other amides, however, it is stable in dimethyl sulfoxide. Its structure was established by spectroscopic analysis. Biogenetically, oxazolines may be considered as the precursor of hydroxy amides and oxazoles found in plants. Shahidine (1) showed activity against a few Gram-positive bacteria.  相似文献

Parallel chemical investigations of the hexane and chloroform extracts of the sea plumes Pseudopterogorgia bipinnata and Pseudopterogorgia kallos has led to the discovery of seven new diterpenoids of the pseudopterane class, namely, kallolide D (2), kallolide C acetate (3), kallolide E (4), kallolide F (5), kallolide G (6), kallolide H (7), and kallolide I (8), in addition to nine previously described compounds of the pseudopterane and gersolane families of diterpenes. The chemical structures of the new metabolites were established by 1D and 2D NMR, IR, UV, HRMS, and, in some instances, by single-crystal X-ray crystallographic analyses. Biological screening of these metabolites revealed significant anti-parasitic activity albeit marginal anti-tubercular activity.  相似文献

Cochinchinones I-L (1-3 and 13) along with 11 known xanthones (4-12, 14, and 15) were isolated from the resin and green fruits of Cratoxylum cochinchinense. In addition, four new acetylated compounds (16-19) were derivatized from 7-geranyloxy-1,3-dihydroxyxanthone (14) and 3-geranyloxy-1,7-dihydroxyxanthone (15). All compounds were characterized on the basis of spectroscopic analyses. The structures of cochinchinone I (1), a monoacetate (18) and a dibrosylate (20), were also confirmed by X-ray diffraction analysis. A new metabolite, infectopyrone (1), has been isolated from the filamentous fungus Alternaria infectoria. The structure of 1 was elucidated by analysis of 2D NMR spectroscopic data. Compound 1 is an α-pyrone resembling known toxins, and is a useful phenotaxonomic marker for the A. infectoria species-group. Infectopyrone (1) was also produced by species within Stemphyllium and Ulocladium, and found in mouldy food.  相似文献

Pressurized fluid extraction (PFE) was examined as an alternative technology for the extraction of carotenoids in the green algae Haematococcus pluvialis and Dunaliella salina and kavalactones in Piper methysticum. The extraction process was optimized by varying the key extraction factors of solvent, sample-solvent ratio, temperature, and time. The selectivity and efficiency of extraction parameters were determined with high performance liquid chromatography (LC) and LC-mass spectrometry (LC-MS). Results showed that PFE utilization of conventional solvents under controlled temperature and pressure in an oxygen and light-free environment could result in the use of less solvent in a shorter period of time. PFE showed higher or equal extraction efficiencies as compared with traditional solvent extractions while maintaining the integrity of chemical components. PFE showed high potential for extraction of natural products and nutraceuticals, particularly labile and light sensitive chemicals.  相似文献
