A FTIR methodology has been developed for the simultaneous determination of Cypermethrin and Chlorpyrifos in pesticide commercially available formulations. The method involves the extraction of both active principles with CHCl3 and direct measurement of the peak area values between 1747 and 1737 cm−1 corrected with a baseline defined at 2000 cm−1 for Cypermethrin and peak height values established at 1549 cm−1 corrected using a baseline situated at 1650 cm−1 for Chlorpyrifos.The limits of detection achieved were of the order of 0.7 and 0.4% (w/w), and the relative standard deviation 0.4 and 0.2% for Cypermethrin and Chlorpyrifos, respectively. The developed procedure provided statistically comparable results with those obtained by HPLC, for a series of commercial samples, which validated the FTIR method. The procedure developed reduces organic solvent consumption, per sample preparation, from 51 ml CH3CN required for HPLC to 2.5 ml CHCl3, and reduces waste generation also increasing the sample measurement frequency, from 3 to 30 samples/h, as compared with the HPLC-UV reference method.  相似文献

A series of approaches have been assayed for FTIR determination of Mancozeb in several solid commercial fungicides using different calibration strategies. The simplest procedure was based on the use of the ratio between the absorbance of a characteristic band of Mancozeb and that of a KSCN internal standard measured in the FTIR spectra obtained from KBr pellets. It was employed the quotient between peak height absorbance values at 1525 cm−1 for Mancozeb and 2070 cm−1 for KSCN. In these conditions a precision as relative standard deviation (RSD) of 0.6% and a relative accuracy error of 0.8% (w/w) were found. For complex formulations, containing other compounds with characteristic absorption bands at different wavenumbers than Mancozeb, one of them was used as internal reference being employed the standard addition approach. In this case, the Mancozeb bands at 1525 cm−1 or at 1289 cm−1 were employed, being used the ferrocyanide band at 2075 cm−1 as internal reference. RSD values between 0.7-1.4% and a relative accuracy error of 3% (w/w) were found. A third strategy was based on the use of partial least squares (PLS) calibration. A reference set was prepared mixing Mancozeb, Kaolin, Cymoxanil and KBr, being predicted the Mancozeb concentration in pesticide formulations by using the quotient between absorbance bands of Mancozeb and those of Cymoxanil. In these conditions a relative accuracy error of 0.6% (w/w) and a relative standard deviation of 1.3% were found.  相似文献

A simple and sensitive HPLC post-derivatization method with colorimetric detection has been developed for the determination of N-nitroso glyphosate in samples of technical glyphosate. Separation of the analyte was accomplished using an anionic exchange resin (2.50 mm × 4.00 mm i.d., 15 μm particle size, functional group: quaternary ammonium salt) with Na2SO4 0.0075 M (pH 11.5) (flow rate: 1.0 mL min−1) as mobile phase. After separation, the eluate was derivatized with a colorimetric reagent containing sulfanilamide 0.3% (w/v), [N-(1-naphtil)ethilendiamine] 0.03% (w/v) and HCl 4.5 M in a thermostatized bath at 95 °C. Detection was performed at 546 nm. All stages of the analytical procedure were optimized taking into account the concept of analytical minimalism: less operation times and costs; lower sample, reagents and energy consumption and minimal waste. The limit of detection (k = 3) calculated for 10 blank replicates was 0.04 mg L−1 (0.8 mg kg−1) in the solid sample which is lower than the maximum tolerable accepted by the Food and Agriculture Organization of the United Nations.  相似文献

This paper presents the quantification of Penicillin V and phenoxyacetic acid, a precursor, inline during Pencillium chrysogenum fermentations by FTIR spectroscopy and partial least squares (PLS) regression and multivariate curve resolution – alternating least squares (MCR-ALS). First, the applicability of an attenuated total reflection FTIR fiber optic probe was assessed offline by measuring standards of the analytes of interest and investigating matrix effects of the fermentation broth. Then measurements were performed inline during four fed-batch fermentations with online HPLC for the determination of Penicillin V and phenoxyacetic acid as reference analysis. PLS and MCR-ALS models were built using these data and validated by comparison of single analyte spectra with the selectivity ratio of the PLS models and the extracted spectral traces of the MCR-ALS models, respectively. The achieved root mean square errors of cross-validation for the PLS regressions were 0.22 g L−1 for Penicillin V and 0.32 g L−1 for phenoxyacetic acid and the root mean square errors of prediction for MCR-ALS were 0.23 g L−1 for Penicillin V and 0.15 g L−1 for phenoxyacetic acid. A general work-flow for building and assessing chemometric regression models for the quantification of multiple analytes in bioprocesses by FTIR spectroscopy is given.  相似文献

The thermal stability of starch cross-linked with tetraethylene glycol diacrylate was studied under nitrogen atmosphere by thermogravimetry (TG) and infrared spectroscopy (FTIR). The cross-linking reaction was confirmed by the increase in intensity of the absorption band at ca. 3330 cm−1 indicating the reinforcement of hydrogen bonds and the appearance of a new band at 1726 cm−1 associated with the carbonyl group of the cross-linking agent. After cross-linking the solubility of starch in water decreased to the range 9%-16%. The thermogravimetric curves of pure and cross-linked starches showed an initial stage of degradation (up to ca. 150 °C) associated with the loss of water. The main stage of degradation occurred in the range 250-400 °C corresponding to ca. 60%-70% mass loss. The activation energy (E) for the degradation process increased from 145 kJ mol−1 (pure starch) to 195 kJ mol−1 and 198 kJ mol−1 for starch treated for 60 min by UV (30 °C) and at 90 °C, suggesting high stability after cross-linking. A higher value (240 kJ mol−1) was obtained for starch treated by UV for 120 min. The main volatile products determined by FTIR which correspond to hydrocarbons and carbonyl groups are apparently associated with the scission of weak bonds in the chain (probably branched groups) and the scission of stronger bonds (glycosidic linkages), respectively.  相似文献

Spectrophotometric and high performance liquid chromatographic (HPLC) methods have been developed for the determination of cis-platin and carboplatin based on the pre-column derivatization of platinum(II) with 2-acetylpyridine-4-phenyl-3-thiosemicarbazone. The complex was extracted in chloroform with molar absorptivity of 2.2 × 104 L mol−1 cm−1 at 380 nm. The complex eluted from a Phenomenex C-18 (150 mm × 4.6 mm i.d.) column with methanol:water:acetonitrile:tetrabutyl ammonium bromide (1 mM) (44:30:25:1, v/v/v/v) with a flow rate of 1 ml/min and UV detection at 260 nm. Ruthenium(IV) and selenium(IV) also separated completely. The linear calibration curve was with 0.5-12.5 μg/ml and detection limit of 10 ng/ml platinum(II).The analysis of cis-platin and carboplatin injections by spectrophotometric and HPLC methods indicated relative standard deviation (R.S.D.) of 0.66-2.1%. The method was used for the determinations of cis-platin in serum and urine of cancer patients after chemotherapy and platinum contents were found 148-444 and 50-90 ng/ml with R.S.D. of 0.3-3.0 and 0.6-2.4% for the serum and urine, respectively. The recovery of platinum(II) from serum was 97% with R.S.D. 2.2%.  相似文献

A novel plant tissue-based bioelectrode obtained by incorporating sunflower (Helianthus annuus L.) leaves tissue as a source of glycolate oxidase and peroxidase into a ferrocene-mediated carbon paste electrode for the determination of glycolic acid was developed. It was coupled with the flow-injection (FI) system and used as the basis to develop a novel FI amperometric procedure for glycolic acid determination. The flow-injection amperometric measurements were performed by injecting aliquot of glycolic acid solution into the flowing stream of 0.05 mol L−1 of phosphate buffer solution having pH 8.0 with a flow rate of 0.3 mL min−1. The bioelectrode consisted of 20% (w/w) of sunflower leaves tissue and 5% (w/w) of ferrocene at 0.00 V (vs Ag/AgCl). The bioelectrode exhibited a linear response from 1.0 × 10−6 up to 2.0 × 10−3 mol L−1 glycolic acid with a detection limit (S/N = 3) and a quantitation limit (S/N = 10) of 1 × 10−6 and 3.3 × 10−6 mol L−1, respectively. The sampling rate of 12 h−1 and a relative standard deviation of 1.67% (n = 15) were achieved. The bioelectrode response decreased to 70% of the original value within 90 continuous injections. The proposed bioelectrode was satisfactorily applied to glycolic acid determination in human urine samples after appropriate sample pretreatment. Results obtained by the FI method were compared favorably with those obtained by HPLC. It offers advantages, which included rapidity, high activity, limited stability, ease of preparation and low cost.  相似文献

Spectrophotometric determination of titanium(IV) was accomplished with o-carboxyphenylfluorone (OCPF) in the presence of hexadecyltrimethyl ammonium chloride (HTAC) under strongly acidic media. In the determination of titanium(IV), Beer's law was obeyed in the range of 24-340 ng mL−1 with an effective molar absorption coefficient (at 530 nm) and relative standard deviation of 2.24 × 105 dm3 mol−1 cm−1 and 0.64% (n = 8), respectively. The severe interference of iron ions was easily eliminated by the addition of ethylenediaminetetraacetic acid (EDTA); the effects of other foreign substances were low. Equilibrium and kinetic studies under analytical conditions were investigated to quantitatively evaluate the reaction mechanism. The obtained orange complex is considered to be Ti(OCPF)4. Its stability log Kf and rate constant Kobs are 16.88 and 1.65 × 10−2 s−1, respectively. It is suggested that the color of the complex is related to the species of OCPF in the solution.  相似文献

The present paper describes a procedure to isolate volatiles from rock-rose (Cistus ladanifer L.) using simultaneous distillation-extraction (SDE). High-value volatile compounds (HVVC) were selected and the influence of the extraction conditions investigated. The effect of the solvent nature and extraction time on SDE efficiency was studied. The best performance was achieved with pentane in 1 h operation. The extraction efficiencies ranged from 65% to 85% and the repeatability varied between 4% and 6% (as a CV%).The C. ladanifer SDE extracts were analysed by headspace solid phase microextraction (HS-SPME) followed by gas chromatography with flame ionization detection (GC-FID). The HS-SPME sampling conditions such as fiber coating, temperature, ionic strength and exposure time were optimized. The best results were achieved with an 85 μm polyacrylate fiber for a 60 min headspace extraction at 40 °C with 20% (w/v) of NaCl. For optimized conditions the recovery was in average higher than 90% for all compounds and the intermediate precision ranged from 4 to 9% (as CV %). The volatiles α-pinene (22.2 mg g−1 of extract), 2,2,6-trimethylcyclohexanone (6.1 mg g−1 of extract), borneol (3.0 mg g−1 of extract) and bornyl acetate (3.9 mg g−1 of extract) were identified in the SDE extracts obtained from the fresh plant material.  相似文献

In this paper, the feasibility and advantage of employing FTIR spectroscopy and the corresponding second-derivative spectroscopy combined with 2D-IR spectroscopy for the analysis of water extracts of Ganoderma lucidum with different storage durations were investigated and demonstrated for the first time. In order to compare the second-derivative microscopic fingerprint spectra from the four periods of storage, the range from 1200 to 500 cm−1 has to be ignored due to the detection of starch in the samples and the standard as well. This is crucial because the main characteristic band of polysaccharide was assigned within this range. The addition of starch as outer constituent was considered spoilage of polysaccharide content investigation. In fact, polysaccharide content in G. lucidum plays a main role as anti-cancer properties. The ranges from 1480 to 1200 cm−1 and from 1700 to 1480 cm−1 were interpreted and directly compared. For the range (from 1800 to 400 cm−1), the 22 months sample was closest to the control, followed by 38 months, 13 months and 6 months product. The matching of the extract spectra with the control was not consistent and depended on which range of spectra had been chosen. Many factors can be considered which may have possibly affected inferences of the composition of the extract from different storage durations.  相似文献

A sensitive and precise high performance liquid chromatography (HPLC)-electrochemical detection (ECD) method has been developed for the simultaneous determination of four isoquinoline alkaloids including berberine, jatrorrhizine, coptisine and palmatine in Chinese medicine Coptis chinensis. The typical HPLC analysis was performed on WondaSil® C18-WR column (250 × 4.6 mm, 5 μm) with the mobile phase comprising 40 mM phosphate buffer (pH 7.0)–acetonitrile (40:60, v/v) at the flow rate of 0.8 mL min−1. The electrochemical detection employed a three electrode system with a bare glassy carbon electrode at +1.3 V versus the Ag/AgCl reference electrode. The limits of detection (LODs) of four alkaloids ranged from 0.01 to 0.03 μmol L−1 and the LOD of berberine was 80 times lower than LOD obtained by UV detection. The rat plasma samples were assayed after oral administration of the traditional Chinese medicine Coptis chinensis by the proposed HPLC-ECD method. The recoveries of this method were ranging from 88.0 to 116%, with the relative standard deviation lower than 3.1% for intra-day precision and 5.7% for inter-day precision. These results show that HPLC-ECD is a useful tool for the quality control of herbal medicine Coptis chinensis and also for pharmacokinetic studies.  相似文献

According to the data from calorimetric adsorption of methylamine, under the best experimental conditions, 10 mg ETS-10 was saturated with 20 μl portions of an aqueous solution of methylamine at 0.05 mol dm−3 concentration corresponding to 0.490 mmol of methylamine per gram of solid. From calorimetric titration data, the thermodynamic values of ΔH, ΔG and ΔS were calculated, showing favourable n-alkylamine-ETS-10 interactions, from which the most favourable occurs for n-butylamine. The adsorption of these bases did not affect the structure of the titaniunsilicate as the XRD profile of ETS-10 is not altered. The FTIR spectra showed that with the increase in the amine mass the bands corresponding to the symmetric and asymmetric deformations at 1390 and 1467 cm−1 increase in intensity along with the NH2 deformation at 1530 cm−1 as expected. The thermogravimetry shows two mass losses for ETS-10 that correspond to physisorbed water and water in smaller channels and cavities. After methylamine adsorption, the same behaviour was observed, however for butyl- and pentylamine three other mass losses were observed corresponding to decomposition and/or release of n-alkylamines.  相似文献

A highly sensitive and relatively interference-free spectrophotometric method for determination of calcium is described. The method is based on the reaction between calcium ions and carboxyazo-p-CH3 in aqueous citrate medium of pH 7, to form a blue complex with maximum absorption at 716 nm. The calibration is linear up to 0.12 μg ml−1 calcium with a repeatability (R.S.D.) of 1.0% at a concentration of 0.04 μg ml−1 (n=5). The molar absorptivity of the complex and Sandell’s sensitivity are 3.5×105 l mol−1 cm−1 and 0.11 ng cm−2, its 10σ limit of quantification and the 3σ limit of detection were found to be 0.3 ng ml−1 and 0.09 ng ml−1 respectively. The influence of reaction variables and the effect of interfering ions are studied; no interference was observed in clinical samples. The proposed method has been applied directly to the determination of calcium in clinical samples without the need for pre-concentration, masking metal ions and digesting samples.  相似文献

In this work, an on-line system with vapor-phase generation (VPG) and Fourier transform infrared (FTIR) spectrometric detection has been developed as a direct and highly selective analytical technique for the assay of penicillamine (PA). Potassium iodate solution was injected into a reactor, heated at 75 °C, containing PA. The CO generated under these conditions was transported by means of N2 gas carrier stream to an infrared gas cell and corresponding FTIR spectra were acquired in a continuous mode. The maximum absorbance of CO band at 2170 cm−1, corrected by a baseline established between 2240 and 2000 cm−1 at a nominal resolution of 2 cm−1, was selected as a measurement criterion. Initially, the effect of different chemical, physical and spectroscopic parameters, such as concentration and volume of oxidant, pH, equilibrium time, reactor temperature, reactor volume, N2 carrier flow rate and number of scans on the analytical signals were evaluated by using a short path length (10 cm) IR gas cell. At optimum experimental conditions, the method provided a relatively broad linear dynamic range of 4-380 mg L−1, a limit of detection of 0.5 mg L−1, a sampling frequency of 15 h−1 and a relative standard deviation (R.S.D.) of 1.6%. Further, the method was successfully applied to the determination of PA in pharmaceutical formulations and results compared well with those obtained by a reference colorimetric method.  相似文献

Artemisinin isolated from the aerial parts of Artemisia annua L., is a promising and potent antimalarial drug, which meets the dual challenge posed by drug-resistant parasites and rapid progression of malarial illness. The aim of the current study was to develop a reliable and fast analytical procedure for the determination of artemisinin in A. annua using high performance liquid chromatography (HPLC) with evaporative light scattering detection (ELSD) in couple with microwave-assisted extraction (MAE) as an efficient sample preparation technique. The HPLC conditions were Agilent C18 column using water:acetonitrile (40:60 v/v) mixture as mobile phase at a flow rate of 1 mL min−1. ELSD conditions were optimized at nebulizer-gas flow rate of 2.0 L min−1 and drift tube temperature of 70 °C under the impactor off-mode, and the gain was set at 2. Afterwards, method validation system for HPLC-ELSD analysis was developed. Calibration range was 0.2-1.0 mg mL−1 and correlation coefficient r was above 0.9990. Precision experiments showed relative standard deviation (R.S.D.) of retention time was less than 0.5% and R.S.D. of peak area was less than 1.30%. Inter-day and intra-day variabilities showed that R.S.D. was ranged from 1.01% to 4.66%. Limit of detection was less than 40 μg mL−1 and limit of quantification was less than 100 μg mL−1. Accuracy validation showed that average recovery was between 98.23% and 104.97%. The developed analytical procedure was successfully applied to determine the contents of artemisinin in the different parts of A. annua plants.  相似文献

This work describes the construction of a polyallylamine modified tubular glassy carbon electrode and its application in the electroreduction of food azo colorants (tartrazine, sunset yellow and allura red) by square wave voltammetry. The electrode modification prevented the surface fouling and, simultaneously, enhanced the analytical signal intensity. The developed unit was coupled to a multicommutated flow system which, given the complexity of samples, was designed to allow the implementation of the standard additions method in an automatic way, using only one standard solution.The described method presented a linear range up to about 2.0 × 10−4 mol l−1 for the referred colorants, with a detection limit of 1.8 × 10−6 mol l−1 for tartrazine, 3.5 × 10−6 mol l−1 for sunset yellow and 1.4 × 10−6 mol l−1 for allura red. The method was applied in the analysis of these colorants in several food samples, and no statistically significant difference between the results obtained by the proposed and the comparative method (HPLC) was found, at a 95% confidence level. Repeatability in the analysis of samples (expressed in R.S.D.) was about 3% (n = 10).  相似文献

Tagitinin C, an antiplasmodial compound, identified as one major compound of the subtropical medicinal plant, Tithonia diversifolia, was determined by FT-IR spectroscopy method. The crude ether extracts from aerial parts of the plant were evaporated to dryness and re-dissolved in tetrachloroethylene (C2Cl4) before analysis.The magnitude of the absorbance of the very specific CO stretching vibration (νCO) at 1664.8 cm−1 was exploited in order to quantify tagitinin C. The determination coefficient (r2) of the calibration scale was 0.9994, the detection limit was lower than 3 μg ml−1 and the quantification limit was lower than 10 μg ml−1.Recovery values from 100.5 to 101.7% were found for spiked concentration levels from 19.91 to 89.95 μg ml−1. The main characteristics of the curves obtained from the calibration standards and from the standard addition technique were not statistically different (Student t-test) suggesting that matrix effects were negligible.The results obtained for the determination of tagitinin C in the crude ether extract from aerial parts of T. diversifolia by LC and FT-IR spectroscopic method agreed well: 0.76±0.02 and 0.773±0.009, of tagitinin C in dried plant respectively.  相似文献

A novel sensitive chromogenic reagent, N,N′-bis(3-methylsalicylidene)-ortho-phenylene diamine (MSOPD), has been synthesized and used in the spectrophotometric determination of nickel. At pH 8, MSOPD can react with nickel ion at room temperature to form a 1:1 complex. The apparent molar absorptivity is 9.5 × 104 l mol−1 cm−1 at 430 nm. Beer's low is obeyed over the range 0-1.0 × 10−5 M of nickel with a detection limit of 1.36 × 10−8 M. The relative standard deviation for measurement of 3.41 × 10−6 M nickel is 1.3% (n = 10). The method has successfully been applied to determination of trace amounts of nickel in some natural food samples.  相似文献

We report the synthesis of a novel bistriazene, 4,4′-bis(3-(4-phenylthiazol-2-yl)triazenyl)biphenyl (BPTTBP), and its highly sensitive color reaction with Hg2+. The new reagent was synthesized in good yield by coupling 2-amino-4-phenylthiazole with 4,4′-biphenyldiamine bisdiazonium salt. Using a blend of surfactants N-cetylpyridinium chloride (CPC) and polyethylene glycol n-octanoic phenyl ether (OP) as a micelle sensitizer, the red colored reagent assembles with Hg2+ in pH 9.8 borate buffer according to a 1:1 stoichiometry, forming a blue oligomeric/polymeric chelating complex with a high apparent stability constant (1.1 × 108 M−1). Whereas the maximum absorption of reagent occurs at 510 nm with an extinct coefficient of 1.35 × 104 M−1 cm−1, the complex absorbs at 611 nm, with an apparent extinct coefficient of 1.04 × 105 M−1 cm−1. Beer's law is obeyed in the range of 0-15 μg/25 mL Hg2+, and Sandell's sensitivity is 1.92 × 10−3 μg/cm2. In the presence of thiourea and Na4P2O7 as masking agents, the method was found free from interferences of foreign ions commonly occurring with mercury. The optimized protocol has been successfully applied to spectrophotometric determination of mercury in waste water samples. The features of the new reagent associated with its special structure were discussed, and an unprecedented “domino effect” was proposed to account for its unique chelating stoichiometry with Hg2+.  相似文献

The characteristics, performance, and application of an electrode, namely, Pt|Hg|Hg2(PABzt)2| graphite, where PABzt stands for p-aminobenzoate ion, are described. This electrode responds to PABzt with sensivity of (58.1±1.0) mV per decade over the range 1.0×10−4 to 1.0×10−1 mol l−1 at pH 6.5-8.0 and a detection limit of 3.2×10−5 mol l−1. The electrode shows easy construction, fast response time (within 10-30 s), low-cost, and excellent response stability (lifetime greater than 6 months, in continuous use). The proposed sensor displayed good selectivity for p-aminobenzoate in the presence of several substances, especially, concerning carboxylate and inorganic anions. It was used to determine p-aminobenzoate in pharmaceutical formulations by means of the standard additions method. The results obtained by using this electrode compared very favorably with those given by an HPLC procedure.  相似文献
