An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.  相似文献

A study has been conducted for the separation and the determination of arsenic species in plants using high-performance liquid chromatography–hydride generation atomic fluorescence spectrometry with emphasis on sample extraction procedures. Various extraction solvents have been applied to extract arsenic species from plants in order to investigate the uptake, transfer and accumulation processes of arsenic. The method was optimized with respect to the selection of extraction solvent, extraction time and the number of extraction steps. The analytical procedure has been validated by analyzing standard reference material GBW 82301 (peach leaves) and successfully used for the arsenic speciation in plants grown on contaminated soil near an arsenic mine. Inorganic arsenic, especially arsenate (As(V)) appears to be the major component in plants and organic arsenic species of monomethylarsenic acid and dimethylarsenic acid were detected at low concentrations.  相似文献

A digestion mixture of H2SO4/HNO3/H2O2/HF/V2O5 was investigated for decomposition of plant samples and sensitive detection of selenium was achieved by hydride generation atomic fluorescence spectrometry (HG-AFS). The method was found to be accurate and reproducible, with a low detection limit (DL) (0.14 ng g−1 solution). The repeatability of the determination was mostly around 10%, the reproducibility over a period of 8 months for determination of selenium in the standard reference material Trace Elements in Spinach Leaves, NIST 1570a, was 9% and the relative measurement uncertainty was 7% using a coverage factor of 2.3 at 95% probability. The average recovery of the whole procedure was 90%. The characteristics of this method are simple and inexpensive equipment, low consumption of chemicals and the ability to analyse many samples in a short time. The whole procedure was carried out in the same PTFE tube, and in addition only a simple cleaning procedure is needed. As a consequence of all these advantages, the described method is suitable for environmental and nutritional studies. The selenium content was determined in 44 vegetable samples from different regions of Slovenia and the contents found were in the range 0.3-77 ng g−1 wet weight.  相似文献

A rapid, high sensitivity method has been developed for the determination of As(III), As(V), Sb(III) and Sb(V) in milk samples by using hydride generation atomic fluorescence spectrometry. The method is based on the leaching of As and Sb from milk through the sonication of samples with aqua regia followed by direct determination of the corresponding hydrides both before and after reduction with KI. It was confirmed by recovery experiments on spiked commercially available samples that neither the reduced nor the oxidized forms of the elements under study or mixtures of the two oxidation states were modified by the room temperature sample treatment with aqua regia. The methodologies developed provided 3σ limit of detection values of 8.1, 10.3, 5.4 and 7.7 ng l−1 for As(III), As(V), Sb(III) and Sb(V) in the diluted samples. Average relative standard deviation values of 5.7, 5.5, 8.2 and 4.7% were found for determination of As(III), As(V), Sb(III) and Sb(V) in commercially available samples of different composition and origin containing from 3.5 to 13.6 ng g−1 total As and from 4.9 to 11.8 ng g−1 total Sb, it being confirmed that As(V) and Sb(V) are the main species present in the samples analyzed (62±5 and 73±5%, respectively). The time required to determine As and Sb species in milk involves 10 min sonication and 30 min prereduction but these steps can be carried out for several sample simultaneously. Additionally the fluorescence measurement step involves less than 20 min for three replicates of all the four measurements required. So, in less than 2 h it is possible to determine the content of As(III), As(V), Sb(III) and Sb(V) in four samples.  相似文献

建立了高效液相色谱-氢化物发生-原子荧光光谱砷形态分析在线联用系统,考察了不同实验条件对4种砷形态化合物(As^Ⅲ,DMA^Ⅴ,MMA^Ⅴ和As^Ⅴ)分离分析的影响,优化了实验条件.在优化的实验条件下,采用pH 5.8的磷酸盐缓冲溶液为流动相,梯度洗脱,10 min之内4种砷形态达到基线分离.进样20μL,测定4种形态的检出限分别为:As^Ⅲ2.76 ng/mL,DMA^Ⅴ7.37 ng/mL,MMA^Ⅴ2.86 ng/mL和As^Ⅴ5.22 ng/mL,相对标准偏差RSD在2.9%~4.2%之间.该联用系统灵敏度高,准确性好,分离分析了部分市售中成药中的不同砷形态化合物.  相似文献

Atomic fluorescence spectrometry with a nondispersive measuring system is combined with a hydride generation technique for the determination of tellurium. Atomic fluorescence measurement is based on the reduction of tellurium by either metallic zinc or sodium borohydride, introduction of the generated tellurium hydride into a premixed argon (entrained air)-hydrogen flame, and excitation with a tellurium electrodeless discharge lamp. The comparison of the zinc and the sodium borohydride reduction methods is discussed in terms of detection limit, precision and interference. The best attainable detection limits for tellurium are 2ng (0.1 ngml) and 30 ng (1.5 ngml) with the zinc and the sodium borohydride methods respectively. Analytical working curves obtained from peak-height and peak-area measurements are linear over a range of approximately 4 orders of magnitude. Of the mineral acids examined in the range up to 2.0 m. nitric acid gives a depressing interference in the range greater than 0.5 m in the zinc method, whereas all of the acids greater than 1.0 m give a slight enhancement of the signal in the sodium borohydride method. The presence of several elements including other hydride-forming elements in 1000-fold ratio to tellurium causes a depressing interference, while enhancing interferences from tungsten and vanadium are observed in the zinc and the sodium borohydride methods, respectively. The present system coupled with the zinc method has been applied to the determination of tellurium in several samples of high-purity copper metal after separation of the analyte from copper by passing an ammoniacal solution of the sample through Chelex-100 resin. The results are in good agreement with those obtained by graphite furnace atomic absorption spectrometry.  相似文献

An electrochemical hydride generation system was developed for the detection of Te by coupling an electrochemical hydride generator with atomic fluorescence spectrometry. Since TeH2 is unstable and easily decomposes in solution, a reticular W filament cathode was used in the present system. The TeH2 generated on the cathode surface was effectively driven out by sweeping gas from the cathode chamber. In addition, a low temperature electrochemical cell (10 °C) was applied to reduce the decomposition of TeH2 in solution. The limit of detection (LOD) was 2.2 ng ml− 1 and the relative standard deviation (RSD) was 3.9% for nine consecutive measurements of standard solution. This method was successfully employed for determination of Te in soldering tin material.  相似文献

A method was proposed for the determination of zinc by atomic fluorescence spectrometry with hydride generation from surfactant-based organized media. The ability of different nature micelles to improve hydride generation of zinc was investigated using an intermittent flow hydride generator and the possible mechanism was discussed. The advantages of hydride generation from the organized media of cetyltrimethylammonium bromide (CTAB) were contrasted with that from aqueous media in sensitivity and precision of the zinc determination. The micellar media of CTAB improved the hydride generation processes of zinc both thermodynamically and kinetically. In addition some transition metals, presumably metal borides, were found to catalyze the hydrogenation of zinc. The proposed method has been validated by the determination of zinc in the certified reference materials using the standard addition method with satisfactory result.  相似文献

The optimisation of an on-line decomposition based on UV photooxidation for the analysis of organoarsenic species by coupling cation-exchange chromatography and atomic fluorescence spectrometry with hydride generation, is described. In this study, special consideration is given to the compatibility of mobile phases with post-column treatments. Results show that the most commonly used mobile phase, aqueous pyridine solutions, decreases species conversion efficiency, leading to a significant loss of sensitivity. New fully-compatible chromatographic conditions are proposed to separate arsenobetaine, arsenocholine, trimethylarsine oxide and tetramethylarsonium ion within 20 min. The very low absolute limits of detection, 4-12 pg(As), allow speciation at trace levels. Analysis of a certified reference fish tissue (DORM-2) and other seafood samples (French and Chilean oysters and mussel) highlights the robustness and the accuracy of the optimised system.  相似文献

High-performance liquid chromatography (h.p.l.c.) is used for separation of arsenite, arsenate, monomethylarsinate (MMA) and dimethylarsonate (DMA) followed by continuous sodium tetrahydroborate reduction and atomic absorption spectrometric detection. Sample preconcentration, offering improved detection limits for the individual species and the removal of matrix interferences, is achieved with a pellicular anion-exchange column. The arsenic species are then separated on a strong anion-exchange column placed in series with the preconcentration column. Detection limits of 2 ng (as arsenic) for arsenite, arsenate and MMA, and 1 ng for DMA. Results for arsenic species in soil waters and commercial bottle waters are given.  相似文献

The determinations of As, Bi, Ge, Sb and Se were performed by atomic fluorescence spectrometry following their electrochemical hydride generation. An electrochemical hydride generator based on a screw-thread seal arrangement, working in a continuous flow mode was used. The effects of cathode material, shape and area of material, catholyte, sample flow rate, applied current, catholyte solution concentration and interference of transition metals on signal intensity were studied. Five kinds of materials including lead, graphite, copper, tungsten and platinum with different shapes were tested as cathode materials. The signal obtained from a 3-dimensional electrode was higher than that from a 2-dimensional electrode under the same conditions. The signal intensity of Ge in HNO3 medium within a narrow concentration range of 0.05–0.10 mol L 1 was stronger than that in other acidic medium, such as HCl and H2SO4. However, the signal intensity of Ge was rapidly decreased with HNO3, HCl and H2SO4 concentration increasing, and then reached approximately zero. In general, limits of detection and a precision were improved using a graphite cathode in H3PO4 medium. The analysis of the reference materials showed good agreement with the certified values for As, Bi, Ge, Sb and Se. The method was successfully applied in the determination of As, Bi, Ge, Sb and Se in traditional Chinese medicine samples.  相似文献

In this work, the determination of total As in seawater by hydride generation atomic fluorescence spectrometry was studied. The influence of the chemical, flow and instrumental parameters were investigated and optimized. The pre-reduction of As(V) to As(III) was performed using KI plus ascorbic acid in 3.5 mol L− 1 HCl medium. No multiplicative interference was present and external aqueous calibration could be used. The limit of detection was 36 ng L− 1, while the repeatability was 2% (n = 10), at a 500 ng L− 1 concentration level. The sample throughput was 15 h− 1 if triplicate measurements were made. The accuracy was assessed by the analysis of a seawater certified reference material and excellent agreement between the obtained and certified values was verified. The procedure was used for the analysis of seawater offshore samples collected at the Brazilian coast and results ranging from 860 to 1200 ng L− 1 were found.  相似文献

Twelve commercially available edible marine algae from France, Japan and Spain and the certified reference material (CRM) NIES No. 9 Sargassum fulvellum were analyzed for total arsenic and arsenic species. Total arsenic concentrations were determined by inductively coupled plasma atomic emission spectrometry (ICP-AES) after microwave digestion and ranged from 23 to 126 μg g−1. Arsenic species in alga samples were extracted with deionized water by microwave-assisted extraction and showed extraction efficiencies from 49 to 98%, in terms of total arsenic. The presence of eleven arsenic species was studied by high performance liquid chromatography–ultraviolet photo-oxidation–hydride generation atomic–fluorescence spectrometry (HPLC–(UV)–HG–AFS) developed methods, using both anion and cation exchange chromatography. Glycerol and phosphate sugars were found in all alga samples analyzed, at concentrations between 0.11 and 22 μg g−1, whereas sulfonate and sulfate sugars were only detected in three of them (0.6-7.2 μg g−1). Regarding arsenic toxic species, low concentration levels of dimethylarsinic acid (DMA) (<0.9 μg g−1) and generally high arsenate (As(V)) concentrations (up to 77 μg g−1) were found in most of the algae studied. The results obtained are of interest to highlight the need to perform speciation analysis and to introduce appropriate legislation to limit toxic arsenic species content in these food products.  相似文献

Hydride generation atomic fluorescence spectrometry was for the first time utilized to determine trace toxic element arsenic in the skeleton fossils of four dinosaurs unearthed in Sichuan Province of China. The instrumental limit of detection (LOD) for arsenic was 0.03 μg/L under optimal experimental conditions, which compared favorably to that by ICP-AES and ETAAS. The samples were digested with aqua regia in boiling water bath. The recoveries of standard addition were found to be from 97 to 109%, and the analytical results were found in good agreement with those by ICP-AES. It is a simple, reliable, sensitive yet relatively inexpensive analytical method, compared to ICP-AES, ICP-MS or ETAAS. Interesting analytical results were found that the arsenic concentrations were all abnormally high in the skeleton fossils. The established analytical method and the analytical results may be helpful in revealing the mystery of the mass extinction of the dinosaur fauna. The analytical results, together with other data available to date, supported the argument that the arsenic toxicosis could be a contributing factor for the mass extinction of the dinosaur fauna in Sichuan Province of China.  相似文献

A highly sensitive procedure has been developed for total arsenic and antimony determination in milk samples by hydride generation atomic fluorescence spectrometry after microwave-assisted sample digestion. The discrete introduction of 2 ml of digested sample in the automated continuous flow hydride generation system allows us to reduce drastically the sample and HCl consume and to determine several elements from a same sample digestion. The method provides detection limits of 0.006 and 0.003 ng ml−1, a sensitivity of 2390 and 2840 fluorescence units per ng ml−1 for As and Sb respectively, and average relative standard deviation of 2.3% for As and 4.8% for Sb. The analysis of cow milk samples, obtained from the Spanish market evidenced the presence of As at concentration levels from 3.4 to 11.6 ng g−1 and Sb levels from 3.5 to 11.9 ng g−1, thus in a proportion near to 1:1, which is in contrast with the 10:1 natural ratio between As and Sb and could evidence the effect of the introduction of new alloys and polymer materials in the industrial process of milk. The method was validated by the comparison of data found for commercial samples by using the proposed procedure and reference methods based on dry-ashing and AFS, and microwave-assisted digestion and inductively coupled plasma mass spectrometry determination.  相似文献
