Abstract

Separations on short columns, (3 and 5 cm, packed with 3 μ ODS spherical materials) and somewhat larger ones (10 cm and 20 cm columns having 2.1 mm and 4.6 mm diameters packed with 5 μ ODS spherical materials) were compared using Aroclor 1254. With simple mixtures, the results showed that short columns can give separations comparable with those on longer columns when the percentage of the organic modifier in the mobile phase is adjusted. This was not so with more complex mixture. The results also showed that columns which have a comparable volume do not produce comparable separation. The longer column, 200 mm × 2.1 mm gave better resolution than the shorter 50 mm × 4 mm column. Also a shorter column, (100 mm × 4.6 mm), which had double the volume of a longer column. (200 mm × 2.1 mm), gave better resolution of the Aroclor 1254 test solution.  相似文献

Abstract

Utilizing phosphorus version of the Fritsch-Buttenberg-Wiechell (FBW) reaction, arylphosphaalkynes are prepared from 2,2-dihalo-1-aryl-1-phosphaalkenes, and theoretical calculations were carried out to obtain some basic insight into intramolecular rearrangement in the phosphorus system of interest.

For the first time, the synergistic effect of graphene oxide nanocolloids (nano-GO) and silicon dioxide (silica) nanoparticles (SiO2-nanoparicles) has been used to modify a glassy carbon electrode (GCE) for the determination of gallic acid (GA). The modified electrode surface was characterised by using scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDXA) and Fourier transform infrared spectroscopy (FTIR). The electrochemical behaviour of the modified electrode was then studied, using cyclic voltammetry (CV) and differential pulse voltammetry (DPV), showing that the electrode was sensitive to GA in a concentration range of 6.25 × 10−6 to 1.0 × 10−3 mol L−1, with a correlation coefficient R2 of 0.9956 and a limit of detection of 2.09 × 10−6 mol L−1 (S/N = 3). The proposed method was successfully used for the determination of GA in red wine, white wine and orange juice, with recoveries of 102.3, 95.4 and 97.6%, respectively.

A sensitive and selective reversed-phase HPLC method for analysis of ketorolac in aqueous and vitreous humor from the human eye has been developed and validated. Chromatographic separation was achieved on a 250 mm × 4.6 mm i.d., 5-μm particle, C18 analytical column. Photo diode-array detection was performed at 314 nm. Response was a linear function of ketorolac concentration from 10 to 800 ng mL−1. The limits of detection (LOD) and quantification (LOQ) were 3.0 and 10 ng mL−1, respectively. Intra-day and inter-day bias were less than 2.05 and 2.28%, respectively, and intra-day and inter-day RSD were no higher than 3.60 and 5.80%, respectively. Fluid obtained from patients eyes’ after topical application of Acular eye drops before retina decolman surgery was analyzed by use of the method. The method enabled successful quantification of levels of ketorolac in aqueous and vitreous humor.

Abstract

An improved method for developing thin layer chromatography plates in the anticircular mode is described. The apparatus consists of two glass dishes arranged concentrically. The outer dish has a diameter of 98 mm × 15 mm deep, and the inner one a diameter of 92 mm × 13 mm deep. The inner dish rests on four small pieces of glass (5 × 5 × 2 mm). A paper wick, (16 mm wide × 1.5 mm thick) sits between the two dishes and transfers the solvent from the outer dish to the thin layer plates. The plate sits on the paper wick with the adsorbent facing down. To ensure an even solvent flow, the dishes are situated on a platform which has adjustable legs, and an horizontal level. Up to 50 samples can be analyzed on one 10 × 10 cm plate in 5 minutes. Less than 10 ml of solvent are needed.  相似文献

The development of a simple reversed phase ion pair liquid chromatographic method for the assay of kanamycin A has been described. Because of the lack of a UV chromophore in the structure of kanamycin A, borate complexation was used to allow direct UV detection at 205 nm. Three columns were evaluated in this study: Zorbax Extend C18 (4.6 mm × 250 mm; 5 μm), XBridge C18 (4.6 mm × 250 mm; 5 μm) and apHera C18 (4.6 mm × 250 mm; 5 μm). The mobile phase was a mixture of 0.1 M disodium tetraborate (pH 9.0) and water (20:80, v/v) supplemented with 0.5 g L−1 sodium octanesulphonate. Final chromatographic conditions were achieved on the XBridge column at 50 °C. The method was validated according to ICH guidelines and applied to a commercially available sample. It is much faster and more specific than the current microbiological assay prescribed in the European Pharmacopoeia. No expensive equipment is necessary to perform this assay making it a viable replacement.
