蛋白质结构复杂,种类多样,与各种生命活动密切相关。大部分蛋白质在生物体中含量极低,对其分析检测带来极大困难。因此实现复杂生物样品中蛋白质的选择性识别与分离,对实现蛋白质的分离分析意义重大。通过分子印迹技术制备的分子印迹聚合物含有与模板分子大小、形状一致,官能团相互匹配的三维印迹空穴,在蛋白质的选择性识别与分离领域显示出了巨大的发展潜力。但是,由于蛋白质具有尺寸较大、构型易变、结构复杂等特点,分子印迹技术在蛋白质印迹中面临着巨大挑战。该文在介绍几种新型分子印迹技术包括表面印迹、抗原决定基印迹和金属螯合物印迹的基础上,综述了近3年分子印迹技术在蛋白质分离分析方面的应用,并对其发展进行了总结与展望。  相似文献

Molecular imprinting polymers (MIPs) against N-Cbz-L-Tyr were prepared utilizing different polymer systems and evaluated in HPLC mode. It was found that MEP utilizing cocktail functional monomers, acrylamide 2-vinylpyridine showed better molecular recognition and better separation ability for the template molecule than those utilizing other functional monomers. MIP utilizing trimethylolpropane trimethacrylate as cross-linker showed higher load capacity and separation factor than those utilizing ethylene glycol dimethacrylate as cross-linker. Increasing the concentration of competing solvent, acetic acid weakened the ionic interaction and hydrogen bonding between the analyte and the functional monomers, 2-vinylpyridine and acrylamide, when the template enantiomer was separated by HPLC. Therefore increasing of the concentration of acetic acid leads to decreasing of capacity factor, separation factor and resolution.  相似文献

Methacrylic acid was first graft‐polymerized on the surfaces of micron‐sized silica gel particles in the manner of “grafting from” using 3‐methacryloxypropyl trimethoxysilane as an intermedia, obtaining the grafted particle polymethacrylic acid PMAA/SiO2. By adopting the novel surface‐molecular imprinting technique put forward by us, cytisine molecule‐imprinted material MIP‐PMAA/SiO2 was prepared with ethylene glycol diglycidyl ether as crosslinking agent. The binding characteristics of MIP‐PMAA/SiO2 towards cytisine was investigated in depth with both batch and column methods and using matrine and oxymatrine as two contrast alkaloids, which with cytisine coexist in sophora alopecuroides and their chemical structure is similar to cytisine to a certain extent. The experimental results show that the surface‐imprinted material MIP‐PMAA/SiO2 has excellent binding affinity for cytisine (20.1 g/100 g of binding capacity), and it is more important that MIP‐PMAA/SiO2 has very high recognition selectivity for cytisine in relation to the two contrast alkaloids. The selectivity coefficients of the grafted particles PMAA/SiO2 (non‐imprinted material) for cytosine in relation to matrine and oxymatrine are only 1.03 and 1.06, respectively, displaying no recognition selectivity for cytisine. However, after imprinting, the selectivity coefficient of MIP‐PMAA/SiO2 for cytisine in respect to matrine and oxymatrine are remarkably enhanced to 12.08 and 15.05, respectively.  相似文献

以猪血清白蛋白(PSA)为模板,采用分级印迹方法,制备了新型单分散多孔蛋白质表面印迹微球.将PSA吸附在5gm粒径、1000A孔径的球形硅胶表面及孔内后,将甲基丙烯酰胺、甲基丙烯酸为功能单体、甲又双丙烯酰胺为交联剂的聚合物溶液,通过真空负压引入到硅胶孔内,并在室温下聚合24h.反应完成后,用3mol/L NH4HF2刻蚀硅胶,获得形状和结构与硅胶颗粒互补的PSA印迹聚合物微球.竞争吸附实验结果表明,在非模板蛋白质存在的情况下,实现印迹颗粒对模板蛋白的高选择性吸附,选择因子达到3.6,说明该印迹聚合物材料有望成为一种可以同生物抗体相媲美的新型亲和材料.  相似文献

An enatioselective surface-imprinted polymer for an amino acid derivative (N-benzyloxycarbonyl-glutamic acid, Z-Glu) was prepared using the bifunctional molecule, benzyldimethyl-n-tetradecylammonium chloride (Zeph). The long-chain quaternary ammonium chloride was found to serve the dual function of an emulsifier and host molecule, while conventional functional molecules possess only the latter function. Chiral recognition ability and ligand specificity of the imprinted polymer were demonstrated by several batchwise tests using different four amino acid derivatives. The surface-imprinted polymer could recognize the chirality of N-protected glutamic acid; therefore, it preferentially adsorbed the corresponding enantiomer that was imprinted in the preparation. The pH and buffer concentration in the aqueous solution are the key factors enhancing enantioselectivity. The molecularly imprinted polymer could distinguish the specific structure from other molecular analogues, even though the structural difference was the only methylene group. The high interfacial activity of the functional molecule and the low swelling property of the imprinted polymer were important in ensuring high imprinting effect. The mechanism of chiral recognition on the polymer was also discussed.  相似文献

天然产物体系复杂,大分子和小分子、生命和非生命物质共存,多存在结构相近的异构体,且有效成分含量低,采用一般的分离方法富集难度较大。分子印迹技术是制备高选择性分离介质的有效技术手段,分子印迹聚合物(MIP)的选择性强,分离操作简单,在各种分离纯化中展现了良好的应用前景。本文对近年来分子印迹技术在天然产物活性组分(如生物碱、甾体、多元酚、黄酮等)的分离纯化中的应用进行了综述,并介绍了本课题组利用MIP从天然产物中分离纯化莽草酸等活性成分的研究进展。  相似文献

利用非共价分子印迹法,采用4-乙烯基吡啶为功能单体,以二甲基丙烯酸乙二 醇酯为交联剂,在模板分子(S)-酮洛芬的存在下,制备出(S)-酮洛芬的分子印 迹聚合物(MIPs),并用作HPLC固定相,对其进行了高效液相色谱评价。结果表明 外消旋酮洛芬在制备的印迹柱上得到了有效的分离,(S)-酮洛芬的容量因子 k_s'为9.52,选择性因子α为1.52,分离度R为0.88。此外,HPLC分析表明制备的 MIPs能够分离结构相似的酮洛芬、布洛芬和萘普生。研究了流动相中乙酸浓度、流 速及温度对拆分效果的影响,测定了分离过程中的焓变、熵变和自由能变化,对分 子印迹分离过程作了进一步解释。  相似文献

以分子印迹技术合成分子印迹聚合物,作为手性色谱柱的固定相来拆分苯丙氨酸衍生物的对映异构体。采用了新型的交联剂及光引发剂。模板分子和可聚合的功能单体形成配合物是分子印迹聚合物必不可少的条件。模板分子与功能单体的比例为1:4时,获得了良好的分离效果,分离因子α为1.69。光聚合方式合成的分子印迹聚合物比热聚合方式合成的拆分能力要高。且聚合的温度越低,聚合物分离效果越好。  相似文献

A double-labelled synthetic oligonucleotide is used as a fluorescent molecular aptamer beacon for the reagentless determination of total hardness in tap and bottled waters. Modified thrombin binding aptamer (5′-NH-C3-GGTTGGTGTGGTTGG-C3-SH-3′) carrying 6-carboxyfluorescein (FAM) and 7-amino-4-methyl-coumarin labels at 5′ and 3′, respectively, was used for the simultaneous combined measurement of Mg2+ and Ca2+ cations. Interference from the K+ cation is eliminated via selective tuning of the assay conditions, increasing the temperature beyond the melting point of the potassium-stabilised quadruplex facilitating its liberation from the quadruplex, whilst maintaining the integrity of the magnesium/calcium-stabilised structure. No interference from other cations found in tap or bottled water was observed. The detection limit of the aptamer beacon is 0.04 mmol L−1, with a dynamic linear range of 0-0.5 μM and is very reproducible, with an R.S.D. = 8%, n = 3. The fluorescent molecular beacon is applied to the determination of total hardness in tap and bottled waters and its’ performance compared to that of the standard method of complexiometric titration and atomic absorption spectroscopy, with an excellent correlation observed. Further work is focused on the immobilization of the aptamer for the development of a re-usable fluorescent/electrochemical aptasensor, for the determination of water hardness.  相似文献

In this work,the quercetin imprinted core-shell microspheres were prepared using silica surface imprinting technique.A simple sol-gel procedure was used for the synthesis of the imprinted materials with 3-aminopropyltriethoxysilane as functional monomer and tetraethyl orthosilicate as crosslinker.The SEM images indicated that the MIPs shell was successfully grafted onto the silica surface.The characteristics of the molecularly imprinted polymers such as capacity,selectivity and absorption dynamic were investigated by rebinding experiments.The results showed that the prepared MIPs had good imprinting effect and adsorption amount of quercetin.  相似文献

Catechol-containing biomolecules and related synthetic materials are of interest for a broad range of applications. Our group has been interested in the potential of eumelanin, a catechol-containing biomaterial and the black-to-brown pigment in humans, and synthetic eumelanin analogues for water purification applications. In an attempt to apply the strategy of molecular imprinting towards lead-binding synthetic eumelanin coatings, we developed a PbO2-mediated synthesis that was subsequently tested with a number of catechols in addition to the eumelanin and polydopamine monomers l-dopa and dopamine. Although this strategy did not provide significant improvements in affinity or selectivity, we found that the PbO2-mediated oxidative polymerisation of 1,2-dihydroxybenzene generates a coating that darkens visibly upon binding Pb2+ and other metal ions. Oxidative polymerisation of catechol with sodium periodate also yields a coating with this colorimetric response. Of eight metal ions tested, the coatings are most responsive to Pb2+ and Cu2+, as analysed by quantitative colorimetry. With further optimisation, these coatings could prove useful for the colorimetric sensing of metal ions.  相似文献

以壳聚糖(CTS)为功能基体,以L-脯氨酸(L-Pro)为模板分子,采用分子印迹技术,在水溶液中合成了在空间结构和结合位点上与L-脯氨酸匹配的分子印迹聚合物(L-Pro-M IPs)。利用红外、扫描电镜和热失重分析,对相关化合物进行了表征,并对合成机理进行初步探索;通过对pH值、反应时间、交联剂用量的调节,获得最佳的反应条件为:制备壳聚糖-L-脯氨酸复合物的pH=10.7,干复合物1.0 g,交联剂8 mL,室温下反应18 h。结果表明,在水相中,L-Pro-M IPs对L-Pro具有良好的吸附选择性和高效分离特性,分离因子为4.67。  相似文献

An ultrathin molecularly imprinted polymer film was anchored on an Au surface for fabricating a surface plasmon resonance sensor sensitive to acephate by a surface-bound photo-radical initiator. The polymerization in the presence of acephate resulted in a molecular-imprinted matrix for the enhanced binding of acephate. Analysis of the SPR wavenumber changes in the presence of different concentrations of acephate gave a calibration curve that included the ultrasensitive detection of acephate by the imprinted sites in the composite, Kass for the association of acephate to the imprinted sites, 7.7 × 1012 M−1. The imprinted ultrathin film revealed impressive selectivity. The selectivity efficiencies for acephate and other structurally related analogues were 1.0 and 0.11-0.37, respectively. Based on a signal to noise ratio of 3, the detection limits were 1.14 × 10−13 M for apple sample and 4.29 × 10−14 M for cole sample. The method showed good recoveries and precision for the apple and cole samples spiked with acephate solution. This suggests that a combination of SPR sensing with MIP film is a promising alternative method for the detection of organophosphate compounds.  相似文献

Firstly,we synthesized N-methacryloyl-histidine monomer and N-methacryloyl-histidine-Cu2+ complex(MAH-Cu2+).Then the molecular imprinting polymers(MIP) has been prepared by surface grafting on uniform polystyrene(PS) core using reversible addition-fragmentation transfer polymerization(RAFT) with MAH-Cu2+ as the functional monomer,methyl paraoxon as the template to simulate phosphodiesterase(PTE).Finally,we have investigated the catalytic hydrolytic activities of MIP and non-imprinting polymers(NIP) to the template methyl paraoxon and the template analogue ethyl paraoxon respectively by UV spectrophotometry.The results showed that the catalytic hydrolytic activity of MIP to the template methyl paraoxon was highest and the value of k is 8.67×10-5 mmol L-1 min-1,3.89-fold higher than MIP to the template analogue ethyl paraoxon,2.79-fold higher than NIP to the template methyl paraoxon.The KM,rm of MSP are also determined,and KM = 3.95×10-4mol/L,rm = 2.12μmol/ min.The MIP can be reused with only lose 7%of catalytic activity for four cycles.  相似文献

 采用原位分子印迹技术 ,单步制备了一种辛可宁印迹的手性整体柱。为了提高柱效和选择性 ,选择了相对低极性的甲苯 /十二醇复合致孔体系。在等度及梯度洗脱条件下 ,非对映异构体辛可宁与辛可尼丁被完全分离。等度洗脱中相对较宽的峰可以在梯度洗脱中得到改善。同时考察了流动相中醋酸浓度、流速以及温度对分离的影响。由于柱中存在大的流通孔 ,大大降低了分离过程中的柱压降 ,从而使这种柱能够在相对高的流速下使用。提高温度可以提高分离因子 ,在 60℃获得最大分离因子 5 40。  相似文献
