15‐Cyano‐12‐oxopentadecano‐15‐lactone was synthesized in 59% total yield starting from 2‐nitrocyclododecanone by Michael addition to acrylaldehyde, followed by reaction with trimethylsilylcyanide, hydrolysis, ring‐expansion, and Nef reaction. A two‐step, one‐pot synthesis of intermediate 2‐hydroxy‐4‐(1‐nitro‐2‐oxycyclododecyl)butanenitrile from 3‐(1‐nitro‐2‐oxocyclododecyl)propanal was developed and the conditions for the Nef reaction were studied. 15‐Cyano‐12‐oxopentadecano‐15‐lactam was synthesized in 40% total yield starting from 2‐nitrocyclododecanone by Michael addition to acrylaldehyde, followed by Strecker reaction, ring‐expansion, and Nef reaction. The conditions for the Strecker and Nef reactions were studied. The structures of the target compounds, intermediates, and by‐product were characterized by IR, 1H‐ and 13C‐NMR, and elemental analysis or MS. 相似文献

A facile synthesis of trans isomers of 4‐aryl‐3‐methyl‐6‐oxo‐4,5,6,7‐tetrahydro ‐ 2H ‐ pyrazolo[3,4‐b]pyridine‐5‐carbonitriles via three‐component condensation reaction of an aldehyde, 3‐amino‐5‐methylpyrazole and ethyl cyanoacetate in acetonitrile has been developed under microwave irradiation. This one‐pot reaction proceeds without any catalyst in short times and gives the product in high selectivities and high yields. 相似文献

An efficient method for the preparation of 2‐substituted 4‐aryl‐4,5‐dihydro‐3,1‐benzoxazepine derivatives under mild conditions has been developed. The reaction of 2‐(2‐aminophenyl)ethanols 1 with acid chlorides in the presence of excess Et3N in THF at room temperature gave the corresponding N‐acylated intermediates 2 , which were dehydrated by treatment with POCl3 to give 2‐substituted 4‐aryl‐4,5‐dihydro‐3,1‐benzoxazepines 3 in a one‐pot reaction. 相似文献

In this research, we have developed an efficient three‐component reaction for the synthesis of pyrano[3,2‐c]pyridine derivatives from the reaction of aromatic aldehydes, tert‐butyl 2,4‐dioxopiperidine‐1‐carboxylate, and N‐methyl‐1‐(methylthio)‐2‐nitroethylen‐1‐amine in [BMIM]BF4 medium. The saccharide‐based chiral bifunctional thiourea‐phosphines were developed as chiral organocatalysts for the intramolecular Morita‐Baylis‐Hillman reaction of ω‐formyl‐enones. With only 2 mol% of thiourea‐phosphine catalyst 3c , chiral functionalized cyclohexenes were achieved under mild reaction conditions with excellent yields and enantioselectivities. 相似文献

A formal [3 + 2] cyclization reaction of 3‐(2‐oxo‐2‐arylethylidene)indolin‐2‐ones and amidines has been developed, regioselectively affording a series of new 3‐(imidazol‐4‐yl)indolin‐2‐ones in good to excellent yields. The reaction was easily conducted in ethylene glycol using Cs2CO3 as a base promoter under microwave irradiation. Flexible structural modification, and broad functional group compatibility as well as mild reaction conditions make this strategy a useful and attractive process of library generation for drug discovery. 相似文献

A facile method for solid‐phase organic synthesis of 1‐substituted‐4‐vinyl‐1,2,3‐triazoles from polystyrene‐supported but‐3‐ynyl selenide has been developed. This sequential [3+2] cycloaddition and oxidation–elimination reactions could be carried out under mild reaction conditions with straightforward operation and good yield and purity of the products, and broad scope of substrates, and could be applied in this reaction system in generation of a small library of title compounds. 相似文献

A sequential one‐pot two‐step reaction for efficient synthesis of pyrrolo[2,1‐a]isoquinoline and pyrrolo[1,2‐a]quinoline derivatives in good yields has been successfully developed. A site‐selective hydroxylative dearomatization of 2‐substituted phenols to either 1,2‐benzoquinols or their cyclodimers, catalyzed by 4,5‐dimethyl‐2‐iodoxybenzenesulfonic acid with Oxone, has been developed. Natural products such as biscarvacrol and lacinilene C methyl ether could be synthesized efficiently under mild reaction conditions. Furthermore, both the reaction rate and site selectivity could be further improved by the introduction of a trialkylsilylmethyl substituent at the 2‐position of phenols. The corresponding 1,2‐quinols could be transformed into various useful structural motifs by [4+2] cycloaddition cascade reactions. 相似文献

An efficient method for the synthesis of N‐alkylated 2‐(4‐substituted‐1H‐1,2,3‐triazol‐1‐yl)‐1H‐indole‐3‐carbaldehyde has been developed starting from oxindole and indole using Huisgen's 1,3‐dipolar cycloaddition reaction of organic azides to alkynes. The effect of catalysts and solvent on these reactions has been investigated. Among all these conditions, while using CuSO4·5H2O, DMF was found to be the best system for this reaction. It could also be prepared in a one‐pot three‐component manner by treating equimolar quantities of halides, azides, and alkynes. The Huisgen's 1,3‐dipolar cycloaddition reaction was performed using CuSO4·5H2O in DMF with easy work‐up procedure. 相似文献

An eco‐friendly method has been developed for the synthesis of 2‐amino‐4‐(9H‐carbazole‐3‐yl)thiophene‐3‐carbonitriles from preliminary carbazole ( 1 ) through an intermediate of 2‐(1‐(9H‐carbazole‐3‐yl)ethylidene)malononitriles using the Knoevenagel condensation followed by the Gewald reaction. On the other hand, the target compounds could also be prepared in a one‐pot three‐component manner by treating equimolar quantities of 1‐(9H‐carbazole‐3‐yl)ethanone ( 3 ), malononitrile, and elemental sulfur. The merits of this preparation are mild reaction conditions. The Gewald reaction is executed with inorganic base NaHCO3 (H2O) in tetrahydrofuran, easy work‐up procedure with good yields. 相似文献

A new and convenient method for the preparation of 3,4‐dihydroquinazolines 5 with aryl and Ac groups at C(2) and N(3), respectively, has been developed. The key sequence is the formation of aza‐phosphorane intermediates by the reaction of N‐[2‐(azidomethyl)phenyl]benzamides 1 with Ph3P, followed by intramolecular aza‐Wittig reaction and 3‐acetylation, which can be conducted in one‐pot. 相似文献

We report an environmentally friendly, efficient and practical method for the synthesis of 2‐aminobenzothiazoles by a copper(II)‐catalyzed tandem reaction of 2‐haloanilines with isothiocyanates under ligand‐ and solvent‐free conditions in air. The developed methodology conforms to the principles of 'green chemistry' and addresses the shortage of such methods for the synthesis of 2‐aminobenzothiazoles. The reaction is quite general and generates a variety of 2‐aminobenzothiazoles in good to excellent yields. Copyright © 2012 John Wiley & Sons, Ltd. 相似文献

A simple and easy synthesis of 2‐(3‐nitro‐phenyl)‐quinazoline‐4‐carboxylic acid ( 3 ) has been successfully developed through a one‐pot three‐component condensation reaction of (2‐amino‐phenyl)‐oxo‐acetic acid sodium salt ( 1 ) obtained from the hydrolysis of isatin with ammonium acetate and 3‐nitrobenzaldehyde. Some novel quinazoline‐ester derivatives 4‐7 were then obtained by the reaction between the new compound 3 and various alcohols. Then, quinazoline‐amide derivatives 10‐14 were synthesized from the reaction of various amines and 2‐(3‐nitro‐phenyl)‐quinazoline‐4‐carbonyl chloride ( 8 ), obtained by the reaction of compound 3 with SOCl2. Finally, some novel quinazoline‐azo derivatives 17‐19 were synthesized by the coupling reaction between β‐dicarbonyl compounds and the novel amino‐quinazoline derivative compound 15 , obtained by reduction of nitro‐quinazoline derivative compound 11 . Thus, a new series of quinazoline‐4‐carboxylic acid, ester, amide, and azo derivatives was synthesized and fully characterized by 1H NMR, 13C NMR, IR, and mass spectrometry analysis. 相似文献

A set of synthetic procedures was developed to yield functionalized pyrido‐, pyrimido‐, and thiazo‐annulated thiadiazine‐1,1‐dioxides on a preparative scale. In all cases the thiadiazine‐1,1‐dioxide ring closure was carried out through a reaction of hetaryl‐sulfonyl chlorides with amidines under mild noncatalytic conditions. In the case of 2‐chloropyridine‐3‐sulfonyl chloride derivatives and 2,4‐dichlorothiazole‐5‐sulfonyl chloride open‐chain sulfonylated amidine intermediates were isolated and then subjected to the cyclization step. The reaction with 2,4‐dichloropyrimidine‐5‐sulfonyl chloride gave rise to the corresponding thiadiazine‐1,1‐dioxides in one‐pot. Similarly, a reaction of 2‐chloropyridine‐3‐sulfonamide with lactime ethers proceeded in one‐pot readily giving the corresponding thiadiazine‐1,1‐dioxides. Remaining chlorine atoms on the prepared hetaryl‐annulated benzothiadiazine‐1,1‐dioxides readily undergo aromatic nucleophilic displacement reactions serving thus as additional variation points for the design of biologically potent compounds. 相似文献

New high yield preparation methods were developed for the pharmaceutically interesting compounds, 1‐benzyl‐, 1‐methyl‐, and 1H‐5‐[(2‐oxo‐2‐phenyl)ethyl]imidazoles 1a‐c , respectively. The title compounds were synthesized by four different methods using various starting materials. Two of the methods involved transformation reactions of the key intermediates, 1‐substituted‐5‐[(2‐nitro‐2‐phenyl)ethenyl]imidazoles 2a‐c and 1‐substituted‐5‐[(2‐nitro‐2‐phenyl)ethyl]imidazoles 3a‐c , while the other two utilized the oxidation of 1‐substituted‐5‐[(2‐hydroxy‐2‐phenyl)ethyl]imidazoles 4a‐c , with chromic oxide, and the umpolung reaction of benzaldehyde followed by a condensation reaction of the umpolung intermediate with imidazolecarboxaldehydes 6a‐c. 相似文献

Air stable and easily accessible, 1‐(α‐aminobenzyl)‐2‐naphthols are used as efficient phosphine‐free ligands in palladium‐catalyzed Suzuki reaction for a variety of substrates under conventional heating as well as ultrasonic conditions. Multi‐brominated aromatic substrates were successfully converted to corresponding arylated moieties with good conversion and selectivity. A novel one‐pot two‐step cascade reaction strategy involving Wittig and Suzuki reactions is developed for efficient synthesis of 4‐styryl biphenyls (C6‐C2‐C6‐C6 unit). Generally, amine‐catalyzed enantioselective transformations rely on chiral enamine or unsaturated iminium intermediates. Herein, we report a protocol involving dual activation by an aromatic iminium and hydrogen‐bonding. An enantioselective aza‐Michael–Henry domino reaction of 2‐aminobenzaldehydes with nitroolefins has been developed through this protocol using primary amine thiourea catalysts to provide a variety of 3‐nitro‐1,2‐dihydroquinolines in moderate yields and with up to 90 % ee. The mechanism for the catalytic enantioselective reaction was confirmed by ESI mass spectrometric detection of the reaction intermediates. The products formed are substructures found in skeletons of important biological and pharmaceutical molecules. 相似文献

Polymer‐supported 2‐azidoethyl sulfonate and 3‐azidopropyl sulfonate reagents have been developed and applied to the solid‐phase organic synthesis of 1‐vinyl‐ and 1‐allyl‐1,2,3‐triazoles, respectively, by CuI‐mediated azide‐alkyne cycloadditions and subsequent cleavage from the polymer support through elimination reaction promoted by DBU. The advantages of this new synthetic method include simple operation and high yield of the products, as well as good stability of the reagents. 相似文献
