The synthesis and spectral properties (ir, ms, nmr) of a substituted 2‐methyl‐2H‐pyrazolo[4,3‐d]‐pyrimidin‐7‐one ( 3 ), an isomer of Viagra®, are described. The key synthon, 4‐amino‐1‐methyl‐5‐propyl‐3‐pyrazolecarboxamide ( 7 ), is prepared via the reaction of ethyl 2,4‐dioxoheptanoate with methylhydrazine, followed by cyclization, nitration, amidation, and nitro group reduction. Interaction of 7 with 2‐ethoxyben‐zoyl chloride yielded the respective bis‐amide ( 8 ) which was cyclized in polyphosphoric acid to the corresponding pyrazolo[4,3‐d]pyrimidin‐7‐one derivative 9 . Chlorosulfonylation of 9 , and subsequent treatment with 1‐methylpiperazine furnished iso Viagra ( 3 ).  相似文献

The condensation of 4‐hydroxy‐6‐methyl‐2H‐pyran‐2‐one and substituted 2‐hydroxybenzaldehydes with ammonium acetate gave the title heterocycles. Synthesis of 1,5‐dihydro‐2‐methyl‐4H‐[1]naphtho‐[1′,2′:5,6]pyrano[4,3‐b]‐pyridine‐4,5‐dione is also described. A reaction mechanism is discussed.  相似文献

The isomeric 2‐substituted‐7(5)‐methyl‐2,3‐dihydro‐5(7)H‐oxazolo[3,2‐a]pyrimidin‐5‐ones 3a‐b and 7‐ones 2a‐b,7a were synthesized by cyclocondensation from the 5‐substituted‐2‐amino‐2‐oxazolines 1a‐b with biselectrophiles. In boiling ethanol, the reaction of 1a‐b with acetylenic esters led to a mixture of 2a‐b,7a with a small amount of (E)‐2‐N‐(2‐ethoxycarbonylethylene)‐5‐substituted‐2‐iminooxazolines 5a‐b . The ring annulation between 1a‐b and diketene gave the 2‐substituted‐7‐hydroxy‐7‐methyl‐2,3,6,7‐tetrahydro‐5H‐oxazolo[3,2‐ a ]pyrimidin‐5‐ones 4a‐b which can be easily dehydrated to provide the 2‐substituted‐7‐methyl‐2,3‐dihydro‐5H‐oxazolo[3,2‐a]pyrimidin‐5‐ones 3a‐b .  相似文献

Synthesis of {3‐[1‐(ethoxycarbonyl)‐[1,2,4]triazolo[4,3‐a]quinoxalin‐4‐yl]‐1‐phenyl‐1H‐pyrazol‐5‐yl}methyl ethyl oxalate ( 2 ), ethyl 4‐[5‐(acetoxymethyl)‐1‐phenyl‐1H‐pyrazol‐3‐yl]‐[1,2,4]triazolo[4,3‐a]quioxaline‐1‐carboxylate ( 4 ), [4‐halo‐1‐phenyl‐3‐(1‐phenyl‐[1,2,4]triazolo[4,3‐a]quioxalin‐4‐yl)‐1H‐pyrazol‐5‐yl]methyl acetate ( 11 ), {4‐halo‐3‐[1‐methyl‐[1,2,4]triazolo[4,3‐a]quinoxalin‐4‐yl]‐1‐phenyl‐1H‐pyraz‐ol‐5‐yl}methyl acetate ( 13 ), and [3‐([1,2,4]triazolo‐[4,3‐a]quinoxalin‐4‐yl)‐4‐halo‐1‐phenyl‐1H‐pyrazol‐5‐yl] methyl formate ( 15 ) was accomplished. The reaction of 4‐aryl‐6‐bromomethyl‐5‐nitro‐3,4‐dihydropyrimidin‐2(1H)‐ones, containing three possible combinations of substituted and unsubstituted nitrogen atoms with anilines depending on the conditions leads to the products of ring contraction of the pyrimidinone ring into an imidazolone, as well as to the formation of 7‐aryl‐6,7‐dihydroisoxazolo[4,3‐d]pyrimidin‐5(4H)‐one derivatives, and in some cases to the 5‐aminopyrimidinones. The mechanisms of these unusual ring transformations are discussed.  相似文献

A series of new 2‐substituted 3‐(4‐chlorophenyl)‐5,8,9‐trimethylthieno[3′,2′: 5,6]pyrido[4,3‐d]pyrimidin‐4(3H)‐ones 8 were synthesized via an aza‐Wittig reaction. Phosphoranylideneamino derivatives 6a or 6b reacted with 4‐chlorophenyl isocyanate to give carbodiimide derivatives 7a or 7b , respectively, which were further treated with amines or phenols to give compounds 8 in the presence of a catalytic amount of EtONa or K2CO3. The structure of 2‐(4‐chlorophenoxy)‐3‐(4‐chlorophenyl)‐5,8,9‐trimethylthieno[3′,2′: 5,6]pyrido[4,3‐d]pyrimidin‐4(3H)‐one ( 8j ) was comfirmed by X‐ray analysis.  相似文献

Chemical transformations of chromone‐3‐carbonitrile ( 1 ) with some substituted hydrazines, namely, thiosemicarbazide, S‐methyl/benzyldithiocarbazate, 7‐chloro‐4‐hydrazinoquinoline, and 3‐hydrazino‐5,6‐diphenyl‐1,2,4‐triazine, led to substituted pyrazoles 2 , 5 – 8 . Ring opening of carbonitrile 1 followed by recyclization with 3‐amino‐1,2,4‐triazole and 2‐aminobenzimidazole gave triazolo[1,5‐a]pyrimidine 9 and pyrimido[1,2‐a]benzimidazole 10 , respectively. Treatment of carbonitrile 1 with some heterocyclic amines produced 2‐amino‐3‐substituted‐chromones 11 and 12 . The novel 3‐hydroxychromeno[4,3‐b]pyrazolo[4,3‐e]pyridin‐5(1H)‐one ( 13 ) was efficiently synthesized from the ring conversion of carbonitrile 1 with cyanoacetohydrazide. A mixture of chromeno[2,3‐b]naphthyridine 14 and chromeno[4,3‐b]pyridine 15 was obtained from base catalyzed transformation of carbonitrile 1 with malononitrile dimer. A diversity of novel annulated chromeno[2,3‐b]pyridines 16 – 22 was also synthesized. Chromeno[2,3‐b]pyrrole‐2‐carboxylate 23 was obtained from the reaction of carbonitrile 1 with ethyl chloroacetate. Structures of the new synthesized products were deduced on the basis of their analytical and spectral data.  相似文献

5-Oxo-5H,7H-[2]benzopyrano[4,3-b][1]benzopyran ( 2 ) has been synthesized from 3-(o-hydroxybenzylidene)isochroman-1,4-dione ( 3 ) by reductive cyclization, and from 5H,7H[2]benzo-pyrano[4,3-b][1]benzopyran-5,7-dione ( 4 ) by selective hydro-genation. This second method affords the dihydro compound 6 or 6a,12a-dihydro-5H,7H-[2]benzopyrano[4,3-b] [1 ]benzopyran-5-one as reaction time increases.  相似文献

2‐Hydrazino‐5,7‐di‐p‐tolylpyrido[2,3‐d ]pyrimidin‐4(3H )‐one ( 4 ) was prepared and condensed with different aldehydes 5a , 5b , 5c , 5d , 5e , 5f , 5g to give the corresponding hydrazone derivatives 6a , 6b , 6c , 6d , 6e , 6f , 6g . Oxidative cyclization of the latter compounds 6a , 6b , 6c , 6d , 6e , 6f , 6g gave the corresponding pyrido[2,3‐d ][1,2,4]triazolo[4,3‐a ]pyrimidin‐5(1H )‐ones 7a , 7b , 7c , 7d , 7e , 7f , 7g . Furthermore, compound 4 reacted with benzoyl chloride, triethyl orthoformate, acetyl chloride, ethyl chloroformate, and carbon disulphide in alcoholic KOH solution to afford the corresponding pyrido[2,3‐d ][1,2,4]triazolo[4,3‐a ]pyrimidinones ( 7a , 8 , 9 , 10 , 11 ). The reaction of thione 3 or its 2‐methylthio derivative 16 with hydrazonoyl halides 12a , 12b , 12c , 12d , 12e , 12f , 12g , 12h , 12i , 12j , 12k , 12l , 12m yielded the corresponding pyrido[2,3‐d ][1,2,4]triazolo[4,3‐a ]pyrimidinones 15a , 15b , 15c , 15d , 15e , 15f , 15g , 15h , 15i , 15j , 15k , 15l , 15m . The structures of all the products were confirmed by elemental and spectral analyses (1H NMR, 13C NMR, IR, and MS). In addition, the anticancer activity of 20 pyridotriazolopyrimidinones against two cancer cell lines namely MCF‐7 and HepG2 was evaluated, and the results revealed that compounds 7d and 9 have promising activity , compared with doxorubicin, which used as standard reference drug.  相似文献

A series of novel 4‐(4‐ethylphenyl)‐1‐substituted‐4H‐[1,2,4]triazolo[4,3‐a]quinazolin‐5‐ones were synthesized by the cyclization of 2‐hydrazino intermediate with various electrophile. The starting material 2‐hydrazino compound was synthesized from 2‐ethyl aniline by a new innovative route with improved yield. When tested for their in vivo H1‐antihistaminic activity on conscious guinea pigs, all the test compounds significantly protected the animals from histamine induced bronchospasm. The compound II emerged as the most active compound of the series and it is more potent (73.93% protection) when compared to the reference standard, chlorpheniramine maleate (71% protection), it showed negligible sedation (10%) when compared to chlorpheniramine maleate (30%). Therefore compound II will serve as prototype molecule for further development as a new class of H1‐antihistamines  相似文献

Pyrazolo[4,3‐d]pyrimidines are of interest as potential kinase inhibitors. This article describes the formation of a novel highly conjugated, condensed, centrosymmetric heteroaromatic compound, 4,9‐dichloro‐2,7‐diisopropyl‐1,3,5,5b,6,8,10,10b‐octaazacyclopenta[h,i]aceanthrylene ( 3 ), during the chlorination of 5,7‐dihydroxypyrazolo[4,3‐d]pyrimidine ( 1 ) with phenylphosphonic dichloride. The nucleophilic attack of benzylamine on 3 afforded N‐benzyl‐5‐chloro‐3‐isopropyl‐1H‐pyrazolo[4,3‐d]pyrimidin‐7‐amine ( 6 ), which was further substituted to yield a pyrazolo[4,3‐d]pyrimidine analogue of roscovitine, a well‐known cyclin‐dependent kinase inhibitor.  相似文献

A novel class of 5‐amino‐N′‐(1‐(pyridin‐4‐yl)ethylidene)‐1H‐pyrazole‐4‐carbohydrazides and 8‐(pyridin‐4‐yl)pyrido[2,3‐d][1,2,4]triazolo[4,3‐a]pyrimidin‐5(1H)‐ones was synthesized from reaction of 2‐cyano‐N′‐(1‐(pyridin‐4‐yl)ethylidene)‐acetohydrazide and 7‐(pyridin‐4‐yl)‐2‐thioxo‐2,3‐dihydropyrido[2,3‐d]pyrimidin‐4(1H)‐one with the appropriate hydrazonoyl halides. Moreover, 2‐cyano‐N′‐(1‐(pyridin‐4‐yl)‐ethylidene)‐acetohydrazide was used for the synthesis of 2‐cyano‐N′‐(1‐(pyridin‐4‐yl)ethylidene)‐acrylohydrazides and 2‐oxo‐2‐(2‐(1‐(pyridin‐4‐yl)ethylidene)‐hydrazinyl)‐acetohydrazonoyl cyanides. The structures of the newly prepared compounds were confirmed by both elemental and spectral analyses as well as by alternate synthesis. The anticancer activities of the prepared compounds were screened against the hepatocellular carcinoma (HepG2) cell line, and the results showed that most of the compounds exhibit considerable activities.  相似文献

Electrophilic heterocyclization of 5‐alkenyl‐1‐methyl‐6‐thioxopyrazolo[3,4‐d]pyrimidin‐4‐ones and 3‐alkenyl‐2‐thioxoquinazoline‐4‐ones under the action of p‐alkoxyphenyltellurium trichloride leads to annulation of thiazoline cycle with formation of 7‐[(p‐alkoxyphenyl)telluromethyl]‐1‐methyl‐6,7‐dihydropyrazolo[3,4‐d][1,3]thiazolo[3,2‐a]pyrimidin‐4(1H)‐ones hydrochlorides and 2‐(p‐alkoxyphenyl)dichlorotelluromethyl‐2,3‐dihydro ‐ 5H‐[1,3]thiazolo[2,3‐b]quinazolin‐5‐ones hydrochlorides. Reduction of salts by the action of excess of sodium sulfite leads to formation of arylhetaryl telluride.  相似文献

When equimolar quantities of salicylaldehyde 2 and ethyl 3-amino-2-butenoate 3 or its constituents (ethyl 3-oxobutanoate and ammonia) were refluxed on a steam-bath for 6 hours with a trace of acetic acid, two products, a pentacyclic pyridine dilactone 4 and 3-acetyl coumarin 5 , resulted in 15% and 45% yields, respectively. The structure of 4 was elucidated as 7-methyl[1]benzopyrano[4,3-d][1]benzoxacino[4,3-b]-pyridine-6,16-dione on the basis of its spectral data. The mechanism of its formation has been discussed. The reaction has been extended to three more substituted salicylaldehydes.  相似文献

Some inimitable and therapeutic coumarin‐substituted fused[1,2,4]triazolo‐[3,4‐b][1,3,4]thiadizole derivatives were synthesized by the cyclocondensation reaction of 2‐oxo‐2H‐chromene‐3‐carboxylic acid ( 1 ) and 4‐amino‐5‐hydrazinyl‐4H‐[1,2,4]‐triazole‐3‐thiol ( 2 ) by using phosphorous oxychloride as a cyclizing agent. This cyclized intermediate 3‐(3‐hydrazino‐[1,2,4]triazolo[3,4‐b][1,3,4]thiadiazol‐6‐yl)‐chromen‐2‐one ( 3 ) later condensation with various ethyl 2‐(2‐arylhydrazono)‐3‐oxobutanoates ( 4 ) in NaOAc/MeOH under reflux conditions afforded the corresponding new series of aryl‐substituted hydrazono‐pyrazolyl‐[1,2,4]triazolo[3,4‐b][1,3,4][thiadiazol]‐coumarin derivatives ( 5 ) in good to excellent yields. The structures of newly synthesized compounds were established on the basis of elemental analysis, IR, 1H NMR and mass spectroscopic studies.  相似文献

Sodium m-nitrobenzene sulfonate, widely used in dehydrogenation of 4,5-dihydro-3(2H)-pyridazinones to their corresponding aromatic derivatives, behaves in an unexpected way when 4,4a-dihydro-5H[1]benzopyrano[4,3-c]pyridazin-3(2H)-ones are employed as substrate. The synthesis of a new class of 5-hydroxy[1]benzopyrano[4,3-c]pyridazin-3(2H)-ones is described.  相似文献
