α-Chiral aminyl radicals have been generated from sulfenamides of α-amino acid esters and α-phenylethylamine using Bu3SnH. The aminyl radicals of α-amino acid esters undergo 5-exo-trig cyclisation reactions onto side chain alkenes to yield proline analogues with reasonable diastereoselectivity. Preliminary studies show urethanyl radicals generated from sulfenamides of alkenyl urethane derivatives of α-amino acid esters and α-phenylethylamine undergo 5-exo-rig cyclisations to providing a protocol for the radical amination of unactivated alkenes. The α-ester of the amino acid or the urethane groups impart electrophilic behaviour to the aminyl radicals and facilitates cyclisation onto alkenes.  相似文献

Reaction of 2-iodo-3-nitrobenzoic acid with arylalkynyl copper(I) reagent gave 3-aryl-5-nitroisocoumarins. Castro-Stephens coupling was followed by in situ Cu-catalysed ring-closure. 1H NMR and X-ray crystallography showed the cyclisations to be 6-endo, contrasting with reports of 5-exo cyclisation of analogous 2-iodobenzoate esters with alkynes. Sonogashira couplings of methyl 2-iodo-3-nitrobenzoate with phenylacetylene and with trimethylsilylacetylene gave the corresponding 2-alkynyl-3-nitrobenzoate esters. With HgSO4, the phenylalkyne underwent 6-endo cyclisation to give 5-nitro-3-phenylisocoumarin. The disubstituted alkyne esters gave 4-phenylselenylisocoumarins with PhSeCl. 5-Nitro-3-phenyl-4-phenylselenylisocoumarin shows significant sterically-driven distortion of the isocoumarin ring. Reaction of methyl 3-nitro-2-phenylethynylbenzoate with ICl gave the 4-iodoisocoumarin. Thus the nitro group tends to direct these electrophile-driven cyclisations towards the 6-endo mode.  相似文献

Brønsted acids induced the intramolecular cyclisation of 3-aryloxypropanoic esters affording 3-methyl-4-chromanones, which have been transformed into the corresponding racemic benzyl β-oxoesters. These latter esters, in the presence of hydrogen and catalytic amounts of both palladium and (endo, endo) aminoborneol, led to optically active chromanones with up to 75% ee through a deprotection-decarboxylation-protonation cascade reaction.  相似文献

Polyphosphoric acid-catalysed cyclisation of derivatives of crotonophenone and chalcone is shown to be a general reaction which occurs without migration of aryl substituents evident with other Friedel-Crafts catalysts. Two other reactions, however, may intervene: hydrogenation and α-carbonyl cleavage. Thus 4-bromochalcone gives 3 - (p - bromophenyl) -1 - phenylpropan -1 - one, 3 - (p - bromophenyl)indan -1 - one and some benzole acid. A mechanism for the cyclisation is discussed in comparison with those reported for cyclisations of α,β-unsaturated esters and divinyl ketones. The formation of intermediate Wheland complexes is predicted to be conrotatory.  相似文献

β-Keto esters and acetyl acetone on condensation with glycosylated amino esters in the presence of IR-120 resin resulted in high yields of glycosyl enamino esters or ketones. The latter on cyclisation with NaH in toluene at reflux gave 6-glycosyl-5,6-dihydro-1H-pyridin-4-ones in fair to good yields.  相似文献

The one-step high-yield synthesis of 1,2-bis(trimethylsilyl)benzene from 1,2-dibromobenzene using tert-butyllithium and trimethylsilyltriflate is reported. A mechanistic investigation shows that 1,2-dilithiobenzene is not an intermediate in this reaction; the coexistence of trimethylsilyltriflate and tert-butyllithium at very low temperatures allows a sequence of bromine-lithium exchange and subsequent derivatization reactions to operate.  相似文献

- Addition of trivalent phosphorus compounds with α, β ethylenic ketones and esters leads to examples of prototropy or cyclisation or rearrangement with ring expansion from 5 to 7 atoms.

When trivalent phosphorus compounds react with acetylenic ketones and esters, the 1, 3 dipolar species can be trapped with an electrophilic reagent (aldehydes or the original acetylenic compound) or a protic reagent (alcohol, acid, amide, phenol, etc…) Ylides, phosphoranes, spirophos-phoranes and phospholes can be obtained.  相似文献

Antimony(III) chloride (20 mol %) in refluxing acetonitrile efficiently catalyses the synthesis of dihydropyrimidinones (50-90% yields) by the Biginelli reaction of aromatic aldehydes, acetoacetate esters and urea. Vinylogous esters bearing para or meta methoxy benzyl groups undergo oxidative cyclisation with 5-20 mol% iodobenzene and m-CPBA to give spirofuran or benzopyran containing heterocycles. New tricyclic quinoxalinone skeletons with bridge-head nitrogen atoms and containing sulphur in a fully-reduced five-membered ring C were obtained. 3,3a-Dihydrothiazolo[3,4-α]quinoxalin-4-ones I-III were prepared by metal-acid reductive cyclisation of N-(nitrophenyl)- and N-(dinitrophenyl)thiazolidine-4-carboxylic acids IVa,b,c. 2,2-Dimethyl-5-(triphenylphosphoranylidene)acetyl-1,3-dioxan-4,6-dione (3) is a new activated β-ketoacyl equivalent, readily prepared in quantitative yield by reaction of Meldrum’s acid with the stable ylide Ph3PCCO. Its reaction with α-amino esters affords the corresponding N-(β-ketoacyl)amino ester ylides which, when treated with aldehydes and KOtBu, undergo a simultaneous Wittig olefination cum Dieckmann cyclisation to yield the respective 3-enoyltetramic acids.  相似文献

A synthesis of aziridine esters based on the cyclisation of amino selanyl esters induced by the selanyl group activation was developed with either the Meerwein salt or NBS. Two asymmetric approaches are proposed: the diastereoselective reductions of α-selanyl β-iminoesters derived from α-oxoesters, which lead to cis chiral aziridine esters 6 and 6′; and the diastereoselective conjugate additions of a chiral amide to α,β-unsaturated esters providing trans chiral aziridine esters 6 and 6″.  相似文献

The conjugate addition of lithium (R)-N-benzyl-N-(α-methylbenzyl)amide to δ-(N-allylamino)-α,β-unsaturated esters, followed by N-deallylation and cyclisation of the resultant β,δ-diamino esters, gives the corresponding 4-aminopiperidin-2-ones as single diastereoisomers (>99:1 dr). Subsequent deprotonation with LiHMDS and functionalisation of the resultant lithium enolate gives 3,4-anti-3-substituted-4-aminopiperidin-2-ones in >99:1 dr. Alternatively, in situ oxidation of the intermediate lithium (Z)-β-amino enolates formed upon conjugate addition gives α-hydroxy-β,δ-diamino esters, which after N-deallylation and cyclisation gives the corresponding 3,4-syn-3-hydroxy-4-aminopiperidin-2-ones in >99:1 dr. The utility of this methodology was successfully demonstrated in a concise asymmetric synthesis of the gastroprokinetic agent (+)-(3S,4R)-cisapride {(+)-(3S,4R)-N(1)-[3′-(4″-fluorophenoxy)propyl]-3-methoxy-4-(2?-methoxy-4?-amino-5?-chlorobenzamido)piperidine} in nine steps from commercially available starting materials with an overall yield of 19%.  相似文献

The use of chiral, binaphthyl-based phosphepines as catalysts improves previous results for the enantioselective [3+2] cyclisation reactions between allenic esters and N-tosylimines, both in terms of conversion rate and enantioselectivity. Pyrrolines bearing 1-naphthyl, phenyl, o-tolyl and p-MeO-phenyl substituents on the stereogenic alpha-carbon have been obtained with enantiomeric excesses up to 64-80%.  相似文献
