Summary 1H,13C, and CP/MAS13C NMR spectra of sixp-substituted 1-aryl-3-arylamino-2-propen-1-ones in solution and in the solid state are reported and discussed. In the proteon-accepting solvent dimethylsulfoxide, electronegative substituents shift the isomeric equilibrium to the (E)-isomer. Bulky substituents promote crystallization of the (Z)-form.

In protonenakzeptierendem Dimethylsulfoxid verschieben elektronegative Substituenten das Gleichgewicht zum (E)-Isomer. Große Substituenten begünstigen die Kristallisation in der (Z)-Form.

The effects of several chemical reagents on the activity of the restriction endonucleasesPvuII andHindIII were investigated. Carbodiimide, which reacts preferentially with carboxyl groups, was found to inactivate these enzymes. This specific effect could be prevented by Mg2+ cation. pBR322 DNA, which containsPvuII andPvuII* sites andHindIII andHindIII* sites, did not protect the enzymes from the carbodiimide. On the other hand, glutaraldehyde, which reacts primarily with lysine residues, inactivatesPvuII andHindIII enzymes. This specific effect could not be prevented by pBR322 DNA. Preincubation with high concentrations ofN-ethylmaleimide, which reacts with sulfhydryl groups, caused slight inhibition ofPvuII activity, but had no effect on the activity ofHindIII enzyme. The effects of glutaraldehyde, carbodiimide, andN-ethylmaleimide on other restriction endonucleases were also investigated. Restriction endonucleasesPvuII andHindIII were immobilized by covalent coupling to various insoluble carriers. Both immobilized enzymes retained partial enzyme activities, when immobilized through phenolic groups and were stable for at least two months.  相似文献

A nonlabeling electrochemical detection method for analyzing the polymerase-chain-reaction-amplified sequence-specific p16 INK4A gene, in which the basis for the covalent immobilization of deoxyribonucleic acid (DNA) probe is described, has been developed. The self-assembly process was based on the covalent coupling of glutaraldehyde (GA) as an arm molecule onto an amino-functional surface. The p16 INK4A gene was used as the model target for the methylation detection of early cancer diagnosis. An amino-modified DNA probe was successfully assembled on the GA-coupling surface through the formation of Schiff base under potential control. The hybridization of amino-modified DNA probes with the target was investigated by means of electrochemical measurements, including cyclic voltammetry and square wave voltammetry. Furthermore, the functions of GA coupling for sequence-specific detection were compared with those obtained based on mercaptopropionic acid. Hybridization experiments indicated that the covalent coupling of GA was suitable for the immobilization of DNA probe and was sensitive to the electrochemical detection of single-base mismatches of label-free DNA targets in hybridization. Moreover, reported probe-modified surfaces exhibited excellent stability, and the hybridization reactions were found to be completely reversible and highly specific for recognition in subsequent hybridization processes. The strategy provided the potential for taking full advantage of existing modified electrode technologies and was verified in microarray technology, which could be applied as a useful and powerful tool in electrochemical biosensor and microarray technology.  相似文献

The high-resolution207Pb magnetic resonance spectra ofR 4–n PbX n (R=Methyl, Ethyl;X=h1-Cyclopentadienyl, Chloride;n=1, 2) have been studied at 16.72Mc. The207Pb- ·· -1H spin-spin coupling constants for the molecules described have been obtained by analysis of the first order NMR-patterns as AxByM and AxByCzM respectively. The experimental spectra have been verified by a computer simulation. The chemical shifts and coupling constants of the lead organyls investigated show similar dependences on moleculare structure as well as number and species of the substituents like analogous organotin compounds.

Stereochemistry of 7-aryl-1,7,8,8a-tetrahydro-3(2H)-indolizinones was studied by1H and13C NMR. Complete assignment of1H NMR signals and analysis of1H-1H coupling constants were performed using the iterative PANIC program. Values of3 J 6,7,3 J 7,8endo, and4 J 5,7 allow one to unambiguously identify the correspondingexo- andendo-stereoisomers. Solid-state 13C nuclear magnetic resonance (13C NMR) with magic-angle spinning (MAS) and with cross-polarization and magic-angle spinning (CP/MAS) spectra, and differential scanning calorimetry (DSC) techniques were used to obtain structural data from a sample of acetyl aleuritolic acid (AAA) extracted from the stem bark of Croton cajucara Benth. (Euphorbiaceae) and recrystallized from acetone. Since solid-state 13C NMR results suggested the presence of more than one molecule in the unitary cell for the AAA, DSC analysis and molecular modeling calculations were used to access this possibility. The absence of phase transition peaks in the DSC spectra and the dimeric models of AAA simulated using the semi-empirical PM3 method are in agreement with that proposal.  相似文献

Summary Eleven compounds have been prepared by azeotropic destillation of water from toluene solutions of bis(tri-n-butyltin)oxide and N-acetyl amino acids. All derivatives are white solids.119Sn-NMR-spectra of the tri-n-butyltin compounds have been studied in coordinating and non coordinating solvents. The chemical shifts and the coupling constants1 J(119Sn,13C) depend significantly on the coordination number of the tin atom and on the properties of the substituents. The data for the compounds are discussed in comparison with those for other tri-n-butyltin compounds.

Molar excess volumes, VE, molar excess enthalpies, HE, and speeds of sound data, u, of chloroform (i) + aniline or o-toluidine (j) binary mixtures have been measured as a function of composition at 308.15 K. Isentropic compressibility changes of mixing, have been determined by employing speed of sound data. Topological investigations of VE data reveals that aniline, chloroform and o-toluidine are associated entities and these (i + j) mixtures contain a 1:1 molecular complex. The IR studies lend further support to the nature and extent of interaction for the proposed molecular entity in the mixtures. HE and values have also been calculated by employing Moelwyn-Huggins concept [Polymer 12 (1971) 387] taking topology of the constituents of the mixtures. It has been observed that calculated HE and values compare well with their corresponding experimental values. The observed VE, HE and data have also been analyzed in terms of Flory theory.  相似文献

Nitropyrazoles     
The structures of substitutedN-aminonitropyrazoles and 1- and 2-amino-4-nitro-1,2,3-triazoles as well as the site of protonation of 1-aminopyrazole were determined based on the1H,13C, and15N (14N) NMR spectra. The13C NMR spectra were recorded under conditions of13C-{1H,14N} triple resonance. Effects of substituents in the pyrazole ring on the13C and14N chemical shifts were studied. Molar excess volumes, V ijk E, and speeds of sound, U ijk , of o-toluidine (i) + benzene (j) + cyclohexane or n-hexane or n-heptane (k) ternary mixtures have been measured as a function of composition at 308.15 K. The observed speed of sound data have been utilized to determine the excess isentropic compressibilities, (K S E) ijk , of the ternary (i+j+k) mixtures. The Moelywn-Huggins concept (Huggins in Polymer 12: 389–399, 1971) of connectivity between the surfaces of the binary mixture constituents has been extended to ternary mixtures (using the concept of a connectivity parameter of third degree of molecules, 3 ξ, which in turn depends on its topology) to obtain an expression that describes well the measured V ijk E and (K S E) ijk data. The observed speeds of sound data have been utilized to predict excess isentropic compressibilities, of the investigated binary mixtures. The observed excess thermodynamic properties VE, HE and have been analyzed in terms of Graph theory. The analysis of VE data by the Graph theory suggests that pyrrolidin-2-one exists mainly as a mixture of cyclic and open dimer; ethanol as a mixture of dimer and trimer; butan-1-ol and propan-2-ol as mixture of monomer and dimer and propan-1-ol as a dimer in the pure state, and their mixtures contain 1:1 molecular complex. The IR studies lend additional credence to the nature and extent of interactions for the proposed molecular entities in the mixtures. Also, it has been observed that VE, HE and values predicted by the Graph theory compare well to with their corresponding experimental values.  相似文献

This paper presents a contact distance dependence analysis scheme and an abinitio calculation application for the electron transfer (ET) reactivity of Co2+OH2/Co3+OH2 reacting pair. The applicability of these schemes and the corresponding models has been discussed. The contact distance (R CoCo) dependence of the relevant quantities has been analyzed. The results indicate that the activation energy from the accurate PES method agrees well with that from the anharmonic potential method, and they are obviously better than that from the harmonic potential method. The pair distribution function varies from 10−2 to 10−5 along withR CoCo changing from 1.20 to 0.35 nm. The coupling matrix element exponentially decays along with the increase ofR CoCo, and the effective electronic coupling requiresR CoCo smaller than 0.75 nm. In the range from 0.50 to 0.75 nm forR CoCo, the corresponding electronic transmission coefficient falls within 1.0—10−6. The local ET rate also exponentially decays along with the increase ofR CoCo due to the electronic factor. Since the contribution from the pair distribution function to the total ET rate is an inverse measure of that from the electronic factor, the variation of the spherically averaged local ET rate along withR CoCo exhibits a parabola with a maximum at 0.50 nm ofR CoCo. This maximum is close to the overall observed ET rate value. For this mono-hydrated transition metal ionic system, the ET rate generally is about 106 L·mol−1·s−1 in gaseous process. Further, since it is impossible to experimentally determine the structures and their PESs of these hydrated systems, especially for the unstable intermediate species,ab initio calculations can play an effective auxiliary role in discussing the ET reactivities of these kinds of reacting systems.  相似文献

The interaction energyE of the systems Cu+-H2O and Cl-H2O has been computed over a wide range of distances and orientations with the MINI-1 basis set in the SCF approximation. The interaction energy has been decomposed according to the Kitaura-Morokuma scheme, with and without counterpoise (CP) corrections to the basis set superposition error. The importance of this correction is analysed by its effect upon Monte Carlo calculations of the Cu+-water and Cl-water systems, using two-body potentials without and with CP corrections. The effect of CP corrections on theE analysis is similar to that found in other systems of analogous composition (of the general type ion plus neutral ligands), but with significant differences in the details. The effect of the CP corrections to the interaction potential, and then on the results of the Monte Carlo simulations, is small for the Cu+ ion, but remarkable for the Cl ion.  相似文献

Summary The use of hydrophobic-interaction chromatography (HIC) is proposed for the simultaneous determination of more than one thiol-protein after formation of the corresponding mercury mercaptides withp-hydroxymercuribenzoate (PHMB). The new chromatographic procedure, based on the HIC separation of the modified proteins from each other and from excess organomercury reagent has been successfully applied to the quantitative determination of phosphoglucose isomerase (PGI) and phosphoglucose mutase (PGM) in crude PGI powder, and of L-lactate dehydrogenase, PGM and aldolase in crude pyruvate kinase from rabbit muscle. The suitability of203Hg-labelled PHMB has been tested in the analysis of mixtures, which give barely distinguishable UV-peaks owing to the presence of other non-thiol components in the sample. For this purpose glyceraldehyde 3-phosphate dehydrogenase (GAPDHy) and PGIy from bakers yeast have been considered. Results obtained in experiments performed by both procedures are reported.  相似文献

In-vivo and in-vitro metabolism of atropine has been investigated by use of a highly specific and sensitive LC–MS n method. Feces, urine, and plasma samples were collected separately after ingestion of 25 mg kg−1 atropine by healthy rats. Rat feces and urine samples were cleaned by liquid–liquid extraction and by solid-phase extraction (on C18 cartridges), respectively. Methanol was added to rat plasma samples to precipitate plasma proteins. Atropine was incubated, in vitro, with homogenized liver and with intestinal flora from rats. The metabolites in the incubation solution were extracted with ethyl acetate. These pretreated samples were then analyzed by reversed-phase high-performance liquid chromatography on a C18 column with methanol–ammonium acetate (2 mm, adjusted to pH 3.5 with formic acid), 70:30 (v/v), as mobile phase. Detection was by on-line MS n . Identification and elucidation of the structure of the metabolites were achieved by comparing molecular mass (ΔM), retention-times, and full-scan MS n spectra with those of the parent drug. Ten new metabolites (aponoratropine, apoatropine, hydroxymethoxyatropine, trihydroxyatropine, dimethoxyatropine, dihydroxymethoxyatropine, hydroxydimethoxyatropine, trihydroxymethoxyatropine, dihydroxydimethoxyatropine, and tropic acid) were identified in rat urine after ingestion of atropine. Nine metabolites (nortropine, tropine, aponoratropine, apoatropine, noratropine, hydroxyatropine, hydroxyatropine N-oxide, hydroxymethoxyatropine, and tropic acid) and the parent drug were detected in rat feces. Five metabolites (nortropine, tropine, tropic acid, apoatropine, and hydroxyatropine) and the parent drug were detected in rat plasma. Only two metabolites (apoatropine and noratropine) were detected in the homogenized liver incubation mixture. The hydrolyzed metabolites (tropine and tropic acid) and dehydrated metabolite apoatropine were found in the rat intestinal flora incubation mixture.  相似文献

Binary blends and pseudo complexes of cellulose acetate (CA) with vinyl polymers containing N-vinyl pyrrolidone (VP) units, poly(N-vinyl pyrrolidone) (PVP) and poly(N-vinyl pyrrolidone-co-vinyl acetate) [P(VP-co-VAc)], were prepared, respectively, by casting from mixed polymer solutions in N,N-dimethylformamide as good solvent and by spontaneous co-precipitation from solutions in tetrahydrofuran as comparatively poor solvent. The scale of miscibility and intermolecular interaction were examined for the blends and complexes by solid-state 13C-NMR spectroscopy. It was revealed that the formation of complexes was due to a higher frequency of hydrogen-bonding interactions between the residual hydroxyl groups of CA and the carbonyl groups of VP residues in the vinyl polymer component. From measurements of CP/MAS spectra and proton spin-lattice relaxation times (TH) in the NMR study, the existence of the hydrogen-bonding interaction was also confirmed for the miscible blends and the homogeneity of the mixing was estimated to be substantially on a scale within a few nanometers.  相似文献

The 13C, 15N CP MAS NMR and FT-IR spectra of dioxomolybdenum (VI) complexes of trans-N,N′-bis-(R-salicylidene)-1,2-cyclohexanediamine (R=H, R=3,5-diCl, R=3,5-diBr, R=4,6-diOCH3), trans-N,N′-bis-(2-OH-naphthylidene)-1,2-cyclohexanediamine and trans-N-(salicylidene)-N′-(2-OH-naphthylidene)-1,2-cyclohexanediamine have been measured. Comparative analysis of the NMR and IR spectra of the complexes with those of the corresponding ligands has shown that the complexation of the di-Schiff bases leads to changes in the conformation of the ligands and the charge redistribution. The asymmetric structure and non-planar structure of the complexes have been suggested.  相似文献

Citric acid (CA) production has been conducted through a careful strain selection, physical–chemical optimization and mutation. The aim of this work was to optimize the physical–chemical conditions of CA production by solid-state fermentation (SSF) using the Aspergillus niger LPB BC strain, which was isolated in our laboratory. The parental and mutant strain showed a good production of CA using citric pulp (CP) as a substrate. The physical–chemical parameters were optimized and the best production was reached at 65% moisture, 30 °C and pH 5.5. The influence of the addition of commercial and alternative sugars, nitrogen sources, salts, and alcohols was also studied. The best results (445.4 g of CA/kg of CP) were obtained with sugarcane molasses and 4% methanol (v/w). The mutagenesis induction of LPB BC was performed with UV irradiation. Eleven mutant strains were tested in SSF where two mutants showed a higher CA production when compared to the parental strain. A. niger LPB B3 produced 537.6 g of CA/kg of CP on the sixth day of fermentation, while A. niger LPB B6 produced 616.5 g of CA/kg of CP on the fourth day of fermentation, representing a 19.5% and 37% gain, respectively.  相似文献
