The relationships between the ionization profile, sensitivity, and structures of 64 exogenous anabolic steroids (groups I–IV) was investigated under electrospray ionization (ESI) conditions. The target analytes were ionized as [M + H]+ or [M + H–nH2O]+ in the positive mode, and these ions were used as precursor ions for selected reaction monitoring analysis. The collision energy and Q3 ions were optimized based on the sensitivity and selectivity. The limits of detection (LODs) were 0.05–20 ng/mL for the 64 steroids. The LODs for 38 compounds, 14 compounds and 12 compounds were in the range of 0.05–1, 2–5 and 10–20 ng/mL, respectively. Steroids including the conjugated keto‐functional group at C3 showed good proton affinity and stability, and generated the [M + H]+ ion as the most abundant precursor ion. In addition, the LODs of steroids using the [M + H]+ ion as the precursor ion were mostly distributed at low concentrations. In contrast, steroids containing conjugated/unconjugated hydroxyl functional groups at C3 generated [M + H ? H2O]+ or [M + H ? 2H2O]+ ions, and these steroids showed relatively high LODs owing to poor stability and multiple ion formation. An LC‐MS/MS method based on the present ionization profile was developed and validated for the determination of 78 steroids (groups I–V) in human urine. ;A simple and reproducible method was developed for the quantification of ketamine and S(+)‐ketamine in dog plasma using a high‐performance liquid chromatography system coupled to a positive ion electrospray mass spectrometric analysis. Solid‐phase extraction was used for extracting analytes from dog plasma samples. The analytes were separated on a Zorbax SB C18 column (100 × 2.1 mm, 3.5 μm) with acetonitrile–formate buffer (10 mM ammonium formate and 0.3% formic acid) (17 : 83, v/v) as mobile phase at a flow‐rate of 0.2 mL/min. Detection was operated under selected ion monitoring mode. [M + H]+ at m/z 238 for ketamine and S(+)‐ketamine and [M + H]+ at m/z 180 for phenacetin (internal standard) were selected as detecting ions, respectively. The method was linear in the concentration range 51.6–2580 ng/mL. The intra‐ and inter‐day precisions (RSD %) were within 11.3% and the assay accuracies ranged from 80.0 to 101.4%. Their average recoveries were greater than 91.1% at all test concentrations. The analytes were proved to be stable during all sample storage, preparation and analysis procedures. The method was successfully applied to the toxicokinetics study and comparison of ketamine and S (+)‐ketamine following intravenous administration to dogs. Anabolic androgenic steroids are widely abused substances in sports doping. Their detection present limitations regarding the use of soft ion sources such as electrospray or atmospheric pressure chemical ionization by liquid chromatography–tandem mass spectrometry. In the current study, a novel derivatization method was developed for the ionization enhancement of selected anabolic androgenic steroids. The proposed method aims at the introduction of an easily ionizable moiety into the steroid molecule by converting the hydroxyl groups into imidazole carbamates using 1,1′‐carbonyldiimidazole as derivatization reagent. The proposed method was applied to water and urine samples spiked with exogenous anabolic androgenic steroids in various concentration levels. Steroid imidazole carbamate derivatives have shown intensive [M+H]+ signals under electrospray ionization and common fragmentation patterns in tandem mass spectrometry mode with [M‐CO2+H]+ and [M‐ΙmCO2+H]+ as major ions with low collision energy. The obtained results showed that the majority of steroids were detectable at concentrations equal or lower to their minimum required performance level according to the World Anti‐Doping Agency technical document. The proposed method is sensitive with a preparation procedure that could be easily applied to the analysis of doping control samples.  相似文献

The accurate and precise measurement of endogenous corticosteroids in urine is a powerful tool to understand the biochemical state in several diseases. In this study, a rapid, accurate, and sensitive method based on liquid chromatography-tandem mass spectrometry (LC–MS/MS) for the quantification of 67 endogenous gluco- and mineralo-corticosteroids and progestins has been developed and validated. Sample preparation, chromatographic separation, and mass spectrometric detection were optimized. Urine samples (0.5 mL) were hydrolyzed with β-glucuronidase and the released analytes were extracted by liquid–liquid extraction. The chromatographic separation was performed in 20 min after redisolution of the extract. MS behavior of endogenous corticosteroids was evaluated in order to select the most specific precursor ion ([M+H]+, [M+NH4]+, or [M+H-nH2O]+) for the detection. MS/MS determination was performed under selected reaction monitoring mode using electrospray ionization in positive mode. The method was shown to be linear (r > 0.99) in the range of endogenous concentrations for all studied metabolites. Limits of detection (LOD) below 1 ng mL−1 were typically obtained for analytes with a 3-oxo-4-ene structure whereas LODs below 15 ng mL−1 were common for the rest of analytes. Recoveries were higher than 80% and intra-assay precisions below 20%, evaluated at three concentration levels, were found for most steroids. No significant or moderate matrix effect, ranging from 54 to 155%, was observed for most of the analytes. The applicability of the method was confirmed by analyzing 24 h urine samples collected from twenty healthy volunteers and comparing the results with previously established normal ranges. The wide coverage of corticosteroid metabolism, together with short analysis time, low sample volume, simple sample preparation, and satisfactory quantitative results make this method useful for clinical purposes.  相似文献

A gas chromatography-microchip atmospheric pressure photoionization-mass spectrometric (GC-μAPPI-MS) method was developed and used for the analysis of three 2-quinolinone-derived selective androgen receptor modulators (SARMs). SARMs were analyzed from spiked urine samples, which were hydrolyzed and derivatized with N-methyl-N-(trimethylsilyl)trifluoroacetamide before analysis. Trimethylsilyl derivatives of SARMs formed both radical cations (M+•) and protonated molecules ([M + H]+) in photoionization. Better signal-to-noise ratios (S/N) were obtained in MS/MS analysis using the M+• ions as precursor ions than using the [M + H]+ ions, and therefore the M+• ions were selected for the precursor ions in selected reaction monitoring (SRM) analysis. Limits of detection (LODs) with the method ranged from 0.01 to 1 ng/mL, which correspond to instrumental LODs of 0.2–20 pg. Limits of quantitation ranged from 0.03 to 3 ng/mL. The mass spectrometric response to the analytes was linear (R ≥ 0.995) from the LOQ concentration level up to 100 ng/mL concentration, and intra-day repeatabilities were 5%–9%. In addition to the GC-μAPPI-MS study, the proof-of-principle of gas chromatography-microchip atmospheric pressure chemical ionization-Orbitrap MS (GC-μAPCI-Orbitrap MS) was demonstrated.  相似文献

The present study provides a comparison of two liquid chromatography–tandem mass spectrometry methods for ginsenosides analysis. The two methods have the same liquid chromatography separation procedure, and both use tandem mass spectrometry detection. However, one method uses multiple reaction monitoring transitions commonly recommended in the literature starting with [M + Na]+ as the molecular ions and with detection of specific fragment ions from the molecules M, while the other is an original method using [M + Cs]+ as molecular ions and Cs+ as fragment ion. The method using [M + Cs]+ as molecular ion has a very high sensitivity allowing the measurement of concentrations in the injecting solutions as low as 4 ng/ml with peaks at this concentration showing signal to noise ratio of 20 or higher. The procedures were utilized for the measurement of eight ginsenosides (Rb1, Rb2, Rc, Rd, Re, Rf (S), Rg1, and Rg2), although the method using [M + Cs]+ has the potential for measuring other ginsenosides. As an application, the ginsenosides were measured in several types of ginseng root, several dietary supplements containing ginseng extracts, four energy drinks, and a sample of ashwagandha.  相似文献

Desorption atmospheric pressure photoionization‐mass spectrometry (DAPPI‐MS) is a versatile surface analysis technique for a wide range of analytes, especially for neutral and non‐polar analytes. Here, a set of analytes typically found in environmental or food samples was analyzed by DAPPI‐MS. The set included five polyaromatic hydrocarbons (PAHs), one N‐PAH, one brominated flame retardant, and nine pesticides, which were studied with three different spray solvents: acetone and toluene in positive ion mode, and anisole in negative ion mode. The analytes showed [M + H]+, M+?, and [M–H]? ions as well as fragmentation and substitution products. Detection limits for the studied compounds ranged from 30 pg to 1 ng (from 0.14 to 5.6 pmol). To demonstrate the feasibility of the use of DAPPI‐MS two authentic samples – a circuit board and orange peel – and a spiked soil sample were analyzed. Tetrabromobisphenol A, imazalil, and PAHs were observed from the three above‐mentioned samples, respectively. The method is best suited for rapid screening analysis of environmental or food samples. A single LC–MS/MS assay has been developed and validated for the simultaneous determination of metformin and dapagliflozin in human plasma using ion‐pair solid‐phase extraction. Chromatographic separation of the analytes and their internal standards was carried out on a reversed‐phase ACE 5CN (150 × 4.6 mm, 5 μm) column using acetonitrile–15 mm ammonium acetate, pH 4.5 (70:30, v/v) as the mobile phase. To achieve higher sensitivity and selectivity for the analytes, mass spectrometric analysis was performed using a polarity switching approach. Ion transitions studied using multiple reaction monitoring mode were m/z 130.1 [M + H]+/60.1 for metformin and m/z 467.1 [M + CH3COO]?/329.1 for dapagliflozin in the positive and negative modes, respectively. The linear calibration range of the assay was established from 1.00 to 2000 ng/mL for metformin and from 0.10 to 200 ng/mL for dapagliflozin to achieve a better assessment of the pharmacokinetics of the drugs. The limit of detection and limit of quantitation for the analytes were 0.39 and 1.0 ng/mL for metformin and 0.03 and 0.1 ng/mL for dapagliflozin, respectively. There was no interference of plasma matrix obtained from different sources, including hemolyzed and lipemic plasma. The method was successfully applied to study the effect of food on the pharmacokinetics of metformin and dapagliflozin in healthy subjects.  相似文献

Gambogic acid and gambogenic acid are two major bioactive components of Garcinia hanburyi, and play a pivotal role in biologic activity. In this study, a specific and sensitive liquid chromatography–tandem mass spectrometry was developed and validated for simultaneous determination of gambogic acid and gambogenic acid in rat plasma. Chromatographic separation was achieved on a C18 column using an isocratic elution with methanol–10 m m ammonium acetate buffer–acetic acid (90:10:0.1, v/v/v) as the mobile phase. The detection was performed on a triple–quadrupole tandem mass spectrometer equipped with electrospray positive ionization using multiple reaction monitoring modes. The transitions monitored were m/z 629.3 [M + H]+ → 573.2 for gambogic acid, m/z 631.2 [M + H]+ → 507.2 for gambogenic acid and m/z 444.2 [M + NH4]+ → 83.1 for IS. Linear calibration curves were obtained in the concentration range of 2.00–1000 ng/mL for gambogic acid and 0.500–250 ng/mL for gambogenic acid. The lower limits of quantification of gambogic acid and gambogenic acid in rat plasma were 2.00 and 0.500 ng/mL, respectively. The intra‐ and inter‐day precision (RSD) values were <11.7% and accuracy (RE) was ?10.6–12.4% at three QC levels for both analytes. The chemical warfare agent O-ethyl S-(2-diisopropylaminoethyl) methyl phosphonothiolate (VX) and many related degradation products produce poorly diagnostic electron ionization (EI) mass spectra by transmission quadrupole mass spectrometry. Thus, chemical ionization (CI) is often used for these analytes. In this work, pseudomolecular ([M+H]+) ion formation from self-chemical ionization (self-CI) was examined for four VX degradation products containing the diisopropylamine functional group. A person-portable toroidal ion trap mass spectrometer with a gas chromatographic inlet was used with EI, and both fixed-duration and feedback-controlled ionization time. With feedback-controlled ionization, ion cooling (reaction) times and ion formation target values were varied. Evidence for protonation of analytes was observed under all conditions, except for the largest analyte, bis(diisopropylaminoethyl)disulfide which yielded [M+H]+ ions only with increased fixed ionization or ion cooling times. Analysis of triethylamine-d15 provided evidence that [M+H]+ production was likely due to self-CI. Analysis of a degraded VX sample where lengthened ion storage and feedback-controlled ionization time were used resulted in detection of [M+H]+ ions for VX and several relevant degradation products. Dimer ions were also observed for two phosphonate compounds detected in this sample.  相似文献

Analyses of a series of nitroaromatic compounds using fast atom bombardment (FAB) mass spectrometry are discussed. An interesting ion-molecule reaction leading to [M + O ? H]? ions is observed in the negative ion FAB spectra. Evidence from linked-scan and collision-induced dissociation spectra proved that [M + O ? H]? ions are produced by the following reaction: M + NO2? → [M + NO2]? → [M + O ? H]?. These experiments also showed that M ions are produced in part by the exchange of an electron between M and NO2? species. All samples showed M, [M ? H]? or both ions in their negative ion FAB spectra. Not all analytes studied showed either [M + H]+ and/or M+˙ in the positive ion FAB spectra. No M+˙ ions were observed for ions having ionization energies above ~9 eV.  相似文献

2,3‐Dimethyl‐2,3‐dinitrobutane (DMNB) is an explosive taggant added to plastic explosives during manufacture making them more susceptible to vapour‐phase detection systems. In this study, the formation and detection of gas‐phase [M+H]+, [M+Li]+, [M+NH4]+ and [M+Na]+ adducts of DMNB was achieved using electrospray ionisation on a triple quadrupole mass spectrometer. The [M+H]+ ion abundance was found to have a strong dependence on ion source temperature, decreasing markedly at source temperatures above 50°C. In contrast, the [M+Na]+ ion demonstrated increasing ion abundance at source temperatures up to 105°C. The relative susceptibility of DMNB adduct ions toward dissociation was investigated by collision‐induced dissociation. Probable structures of product ions and mechanisms for unimolecular dissociation have been inferred based on fragmentation patterns from tandem mass (MS/MS) spectra of source‐formed ions of normal and isotopically labelled DMNB, and quantum chemical calculations. Both thermal and collisional activation studies suggest that the [M+Na]+ adduct ions are significantly more stable toward dissociation than their protonated analogues and, as a consequence, the former provide attractive targets for detection by contemporary rapid screening methods such as desorption electrospray ionisation mass spectrometry. An HPLC method permitting the simultaneous determination of fourteen analytes (phenylalkanoids and monoterpenoids) from the roots of Rhodiola rosea was developed. A separation was achieved within 35 min using C18 column material and a water–acetonitrile mobile phase, both containing a 0.05% phosphoric acid gradient system and a temperature of 53°C. The method was validated for linearity, repeatability, limits of detection and limits of quantification. The limits of detection and limits of quantification of 14 phenylalkanoids and monoterpenoids were found to be 0.20–1.0 and 0.5–3.5 µg/mL, respectively. The wavelengths used for quantification of phenylalkanoids and monoterpenoids with a diode array detector were 205, 220 and 251 nm. The method was used to analyze the roots of two species of Rhodiola and commercial extracts of R. rosea and provides preliminary evidence of phytochemical differences between North American and Eurasian populations of R. rosea. LC–mass spectrometry coupled with electrospray ionization (ESI) interface method is described for the identification of phenylalkanoids and monoterpenoids in various Rhodiola samples. Summary Fragmentation patterns of the essential amino acids (AAs) as their silyl derivatives have been obtained with the aid of ion trap detection (ITD). Three derivatizing reagents, hexamethyldisilazane+trifluoroacetic acid (HMDS+TFAA),bis-(trimethylsilyl)trifluoroacetamide (BSTFA) andN-methyl-N-(t-butyldimethylsilyl)trifluoroacetamide (MTBSTFA) were used. Simple and multiple derivatives obtained with each reagent have been investigated, with regard to their sensitivity and selectivity. Our study performed in the concentration range of 5-2000 ng amino acids has shown that, contrary to literature data, thirteen of the twenty-two AAs investigated including the TBDMS derivatives give rise to more than one peak when eluted. As a result of ion/molecule interaction the very informative ions of high masses, ([M]+, [M+TMS/(TBDMS)]+, [M+1]+) are formed with considerable intensities. The fragments [M-CH3]+, [M-C4H9]+, [M-(CH3)2Si]+, [M-TMS/(TBDMS)COO]+, [M-TBDMSOH]+, [M-TBDMSO]+, [M-TBDMSNH]+ and numerous others could be utilized for identification purposes. Selective and Sensitive LC-MS Determination of Avenanthramides in Oats   总被引:1,自引:0,他引:1  
A new liquid chromatographic – mass spectrometric (LC-MS) method suitable for the determination of avenanthramides in oats has been developed. The avenanthramides were detected using positive ion electrospray with selective ion monitoring of protonated ions [M+H]+. The separation of avenanthramides was achieved on Genesis C18 column by using acetonitrile – aqueous formic acid as mobile phase and gradient elution. The effects of buffer nature and concentration on separation, peak shape and intensity of mass spectrometric signal were investigated. The detection limits ranged between 10 and 30 ng mL?1 for standard solutions when using 1 μL injection. The developed LC-MS method was found to be superior over commonly used liquid chromatographic methods with UV detection in terms of selectivity and sensitivity. The applicability of the method to real samples (oat groats) was demonstrated.  相似文献

Studies on steroid metabolism are of utmost importance to improve the detection capabilities of anabolic androgenic steroids (AASs) misuse in sports drug testing. In humans, glucuronoconjugates are the most abundant phase II metabolites of AAS. Bisglucuronidation is a reaction where two separated functional groups on the same molecule are conjugated with glucuronic acid. These metabolites have not been studied in depth for steroids and could be interesting markers for doping control. The aim of the present work was to study the ionization and collision‐induced dissociation of steroid bisglucuronides to be able to develop mass spectrometric analytical strategies for their detection in urine samples after AAS administration. Because steroid bisglucuronides are not commercially available, 19 of them were qualitatively synthesized to study their mass spectrometric behavior. Bisglucuronides ionized as [M+NH4]+ in positive mode, and as [M–H] and [M–2H]2− in negative mode. The most specific product ions of steroid bisglucuronides in positive mode resulted from the neutral losses of 387 and 405 Da (corresponding to [M+NH4–NH3–2gluc–H2O]+ and [M+NH4–NH3–2gluc–2H2O]+, respectively, being “gluc” a dehydrated glucuronide moiety), and in negative mode, the fragmentation of [M–2H]2− showed ion losses of m /z 175 and 75 (gluc and HOCH2CO2, respectively). On the basis of the common behavior, a selected reaction monitoring method was developed to detect bisglucuronide metabolites in urine samples. As a proof of concept, urines obtained after administration of norandrostenediol were studied, and a bisglucuronide metabolite was detected in those urines. The results demonstrate the usefulness of the analytical strategy to detect bisglucuronide metabolites in urine samples, and the formation of these metabolites after administration of AAS.  相似文献

Two carbonyl compounds, nabumetone and testosterone, were derivatized with pentafluorophenyl hydrazine (PFPH) and analyzed by atmospheric-pressure chemical-ionization mass spectrometry. The PFPH derivatives underwent dissociative electron capture in negative-ion APCI (ECAPCI) and gave intense [M–20] ions in the mass spectra. In positive-ion APCI, the PFPH derivatives underwent efficient protonation and gave intense [M+H]+ ions in the mass spectra. In CID, the major product ions of the [M–20] ions in ECAPCI corresponded to the partial moiety of PFPH. In contrast, the major product ions of [M+H]+ corresponded to the partial moiety of the analyte. By using selected reaction monitoring (SRM) detection, low pg of nabumetone (1 pg) and testosterone (7 pg) could be detected in both ECAPCI and positive-ion APCI. In comparison with the detection limits (SRM) of the underivatized analytes, use of the PFPH derivatives resulted in 2500-fold and 35-fold sensitivity enhancements for nabumetone and testosterone, respectively. The PFPH derivatives were applied to the analysis of nabumetone and testosterone in human plasma by both ECAPCI and positive-ion APCI and were found to enable detection of 0.1 ng mL–1 nabumetone in spiked plasma. For testosterone, endogenous testosterone in female plasma was detected in both ECAPCI and positive-ion APCI.  相似文献

Spin‐labeled nitroxide derivatives of podophyllotoxin had better antitumor activity and less toxicity than that of the parent compounds. However, the 2‐H configurations of these spin‐labeled derivatives cannot be determined by nuclear magnetic resonance (NMR) methods. In the present paper, a high‐performance liquid chromatography‐diode array detection (HPLC‐DAD) and a high‐performance liquid chromatography‐electrospray ionization tandem mass spectrometry (HPLC‐ESI/MS/MS) method were developed and validated for the separation, identification of four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 position. In the HPLC‐ESI/MS spectra, each pair of diastereoisomers of the spin‐labeled derivatives in the mixture was directly confirmed and identified by [M+H]+ ions and ion ratios of relative abundance of [M‐ROH+H]+ (ion 397) to [M+H]+. When the [M‐ROH+H]+ ions (at m/z 397) were selected as the precursor ions to perform the MS/MS product ion scan. The product ions at m/z 313, 282, and 229 were the common diagnostic ions. The ion ratios of relative abundance of the [M‐ROH+H]+ (ion 397) to [M+H]+, [A+H]+ (ion 313) to [M‐ROH+H]+, [A+H‐OCH3]+ (ion 282) to [M‐ROH+H]+ and [M‐ROH‐ArH+H]+ (ion 229) to [M‐ROH+H]+ of each pair of diastereoisomers of the derivatives specifically exhibited a stereochemical effect. Thus, by using identical chromatographic conditions, the combination of DAD and MS/MS data permitted the separation and identification of the four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 in the mixture.  相似文献

Normal-phase HPLC and hydrophilic interaction HPLC (HILIC) were investigated for the separation of selenometabolites in a water extract of Se-rich yeast prior to their detection by ICP-MS and identification by electrospray MS/MS. The targeted fraction was a low-abundant fraction co-eluting with salt and sulfur analogues in size-exclusion chromatography which has so far been inaccessible to Se speciation studies. The optimization of the separation conditions resulted in the highest separation efficiency when HILIC was used and elution was carried out isocratically with a low concentration ammonium acetate buffer (1 mM ammonium acetate/10 mM acetic acid) in 80% acetonitrile. Out of 15 peaks observed with the Se-specific ICP-MS detection 12 was identified by electrospray Q-TOF MS/MS (2,3-dihydroxypropionyl (DHP)-Se-methylselenocysteine [M+H]+: 272, Se-methyl-γ-glutamyl-selenocysteinylglycine dioxide [M+H]+: 402, γ-glutamyl-Se-methylselenocysteine [M+H]+: 313; isomers of γ-glutamylselenocystathionine [M+H]+: 400; Se-methyl-selenoglutathione [M+H]+: 370, isomers of N-acetylselenocystathionine [M+H]+: 313, 2,3-DHP-selenohomolanthionine [M+H]+: 373, isomers of 2,3-DHP-selenocystathionine [M+H]+: 359, 2,3-DHP-selenolanthionine [M+H]+: 345 and selenohomolanthionine [M+H]+: 285).  相似文献

Organophosphate esters (OPEs) are chemical compounds incorporated into materials as flame‐proof and/or plasticizing agents. In this work, 13 non‐halogenated and 5 halogenated OPEs were studied. Their mass spectra were interpreted and compared in terms of fragmentation patterns and dominant ions via various ionization techniques [electron ionization (EI) and chemical ionization (CI) under vacuum and corona discharge atmospheric pressure chemical ionization (APCI)] on gas chromatography coupled to mass spectrometry (GC‐MS). The novelty of this paper relies on the investigation of APCI technique for the analysis of OPEs via favored protonation mechanism, where the mass spectra were mostly dominated by the quasi‐molecular ion [M + H]+. The EI mass spectra were dominated by ions such as [H4PO4]+, [M–R]+, [M–Cl]+, and [M–Br]+, and for some non‐halogenated aryl OPEs, [M]+● was also observed. The CI mass spectra in positive mode were dominated by [M + H]+ and sometimes by [M–R]+, while in negative mode, [M–R] and more particularly [X] and [X2]‐● were mainly observed for the halogenated OPEs. Both EI and APCI techniques showed promising results for further development of instrumental method operating in selective reaction monitoring mode. Instrumental detection limits by using APCI mode were 2.5 to 25 times lower than using EI mode for the non‐brominated OPEs, while they were determined at 50‐100 times lower by the APCI mode than by the EI mode, for the two brominated OPEs. The method was applied to fish samples, and monitored transitions by using APCI mode showed higher specificity but lower stability compared with EI mode.
