This paper describes the development of an optimized method based on solid-phase extraction (SPE) followed by liquid chromatography–electrospray ionization tandem mass spectrometry (LC–MS/MS) for the simultaneous analysis of ten antibiotic compounds including tetracyclines, sulfonamides, macrolides and quinolones. LC–MS/MS sensitivity has been optimized by alterations to both LC and MS operations. Of the two high resolution columns tested, Waters Symmetry C18 endcapped and Agilent Zorbax Bonus-RP, the latter was found to show better performance in producing sharp peaks and clear separation for most of the target compounds. Optimization of the MS fragmentation collision and cone energy enhanced the peak areas of the target analytes. The recovery of the target compounds from water samples was most efficient on Waters Oasis HLB SPE cartridge, while methanol was shown to be the most suitable solvent for desorbing the compounds from SPE. In addition, acidification of samples prior to SPE was shown to enhance the recovery of the compounds. To ensure a satisfactory recovery, the flow rate through SPE should be maintained at ≤10 mL min−1. The method was successfully applied to the analysis of antibiotics from environmental water samples, with concentrations being <LOD in tap water, between <LOD to 28 ng L−1 in river water and between <LOD to 230 ng L−1 in sewage effluent.  相似文献

Pre-concentration and determination of 8 phenolic compounds in water samples has been achieved by in situ derivatization and using a new liquid–liquid microextraction coupled GC–MS system. Microextraction efficiency factors have been investigated and optimized: 9 μL 1-undecanol microdrop exposed for 15 min floated on surface of a 10 mL water sample at 55 °C, stirred at 1200 rpm, low pH level and saturated salt conditions. Chromatographic problems associated with free phenols have been overcome by simultaneous in situ derivatization utilizing 40 μL of acetic anhydride and 0.5% (w/v) K2CO3. Under the selected conditions, pre-concentration factor of 235–1174, limit of detection of 0.005–0.68 μg/L (S/N = 3) and linearity range of 0.02–300 μg/L have been obtained. A reasonable repeatability (RSD ≤ 10.4%, n = 5) with satisfactory linearity (0.9995 ≥ r2 ≥ 0.9975) of results illustrated a good performance of the present method. The relative recovery of different natural water samples was higher than 84%.  相似文献

Gas chromatography–mass spectrometry (GC–MS) with Cold EI is based on interfacing GC and MS with supersonic molecular beams (SMBs) along with electron ionization of vibrationally cold sample compounds in SMB in a fly-through ion source (hence the name Cold EI). Cold EI improves all the central performance aspects of GC–MS, and in this paper, we focus on its improvement of signal-to-noise ratio (S/N) and limits of detection (LODs). We found that the harder the compound for analysis with standard EI, the greater the Cold EI gain in S/N and LOD. The lower LOD and higher S/N of Cold EI emerge from a few reasons: (a) similar ionization yield as standard EI, (b) enhanced abundance of molecular ions, (c) elimination of vacuum background noise, (d) elimination of ion source-related peak tailing and degradation, (e) ability to lower the elution temperatures via the use of high column flow rates, and (f) greater range of thermally labile and low-volatility compounds that can be analyzed. We demonstrate the superior S/N and lower LOD of Cold EI versus standard EI in a range of compounds, from the simple-to-analyze octafluoronaphthalene all the way to reserpine and an organo-metallic compound that cannot be analyzed by standard EI. These compounds include methyl stearate, cholesterol, n-C32H66, large polycyclic aromatic hydrocarbons, dioctyl phthalates, diundecyl phthalate, pentachlorophenol, benzidine, lambda-cyhalothrin, and methidathion. The significantly lower Cold EI LODs that can be over 1000 times better than in standard EI further result in far superior response linearity and greater measurement dynamic range.  相似文献

n-Butyl benzyl phthalate (BBP) is an endocrine-disrupting chemical. A bacterium species capable of using BBP as the sole source of carbon and energy was isolated from mangrove sediment. Effects of BBP concentration, pH, temperature, and salinity on BBP biodegradation were studied. The optimum pH, temperature, and salinity for the BBP biodegradation were 7.0, 37°C, and 15 g L−1, respectively. BBP was completely degraded within 6 days under optimum conditions, and the biodegradation of BBP could be fitted to a first-order kinetic model. The major metabolites of BBP biodegradation were identified as mono-butyl phthalate, mono-benzyl phthalate, phthalic acid, and benzoic acid by using high-performance liquid chromatography and gas chromatography–mass spectrometry. A preliminary metabolic pathway was proposed for the biodegradation of BBP.   相似文献

In this paper, as novelties to the field, it is confirmed at first, that the fruits of Cirsium species, regarded as injurious weeds, do contain lignans, two, different butyrolactone-type glycoside/aglycone pairs: the well known arctiin/arctigenin and the particularly rare tracheloside/trachelogenin species. These experiences were supported by gas chromatography–mass spectrometry (GC–MS), by liquid chromatography tandem mass spectrometry (LC–MS/(MS)) and by nuclear magnetic resonance (NMR) spectroscopy. The study reflects the powerful impact of the complementary chromatographic mass fragmentation evidences resulting in the identification and quantification, the extremely rare, with on line technique not yet identified and described, tracheloside/trachelogenin pair lignans, without authentic standard compounds. Fragmentation pattern analysis of the trimethylsilyl (TMS) derivative of trachelogenin, based on GC–MS, via two different fragmentation pathways confirmed the detailed structure of the trachelogenin molecule. The complementary chromatographic evidences have been unambiguously confirmed, by 1H and 13C NMR analysis of trachelogenin, isolated by preparative chromatography. Identification and quantification of the fruit extracts of four Cirsium (C.) species (C. arvense, C. canum, C. oleraceum, and C. palustre), revealed that (i) all four species do accumulate the tracheloside/trachelogenin or the arctiin/arctigenin butyrolactone-type glycoside/aglycone pairs, (ii) the overwhelming part of lignans are present as glycosides (tracheloside 9.1–14.5 mg/g, arctiin 28.6–39.3 mg/g, expressed on dry fruit basis), (iii) their acidic and enzymatic hydrolyses to the corresponding aglycones, to trachelogenin and arctigenin are fast and quantitative and (iv) the many-sided beneficial trachelogenin and arctigenin can be prepared separately, without impurities, excellent for medicinal purposes.  相似文献

Dispersive liquid–liquid microextraction with little solvent consumption (DLLME-LSC), a novel dispersive liquid–liquid microextraction (DLLME) technique with few solvent requirements (13 μL of a binary mixture of disperser solvent and extraction solvent in the ratio of 6:4) and short extraction time (90 s), has been developed for extraction of organochlorine pesticides (OCPs) from water samples prior to gas chromatography/mass spectrometry analysis. In DLLME-LSC, much less volume of organic solvent is used as compared to DLLME. The new technique is less harmful to environment and yields a higher enrichment factor (1885–2648-fold in this study). Fine organic droplets were formed in the sample solution by manually shaking the test tube containing the mixture of sample solution and extraction solvent. The large surface area of the organic solvent droplets increases the rate of mass transfer from the water sample to the extractant and produces efficient extraction in a short period of time. DLLME-LSC shows good repeatability (RSD: 4.1–9.7% for reservoir water; 5.6–8.9% for river water) and high sensitivity (limits of detection: 0.8–2.5 ng/L for reservoir water; 0.4–1.3 ng/L for river water). The method can be used on various water samples (river water, tap water, sea water and reservoir water). It can be used for routine work for the investigation of OCPs.  相似文献

An in-syringe demulsified dispersive liquid–liquid microextraction (ISD–DLLME) technique was developed using low-density extraction solvents for the highly sensitive determination of the three trace fungicides (azoxystrobin, diethofencarb and pyrimethanil) in water samples by high performance liquid chromatography–mass spectrometry chromatography–diode array detector/electrospray ionisation mass spectrometry. In the proposed technique, a 5-mL syringe was used as an extraction, separation and preconcentration container. The emulsion was obtained after the mixture of toluene (extraction solvent) and methanol (dispersive solvent) was injected into the aqueous bulk of the syringe. The obtained emulsion cleared into two phases without centrifugation, when an aliquot of methanol was introduced as a demulsifier. The separated floating organic extraction solvent was impelled and collected into a pipette tip fitted to the tip of the syringe. Under the optimal conditions, the enrichment factors for azoxystrobin, diethofencarb and pyrimethanil were 239, 200, 195, respectively. The limits of detection, calculated as three times the signal-to-noise ratio (S N−1), were 0.026 μg L−1 for azoxystrobin, 0.071 μg L−1 for diethofencarb and 0.040 μg L−1 for pyrimethanil. The repeatability study was carried out by extracting the spiked water samples at concentration levels of 0.02 μg mL−1 for all the three fungicides. The relative standard deviations varied between 4.9 and 8.2% (n = 5). The recoveries of all the three fungicides from tap, lake and rain water samples at spiking levels of 0.2, 1, 5 μg L−1 were in the range of 90.0–105.0%, 86.0–114.0% and 88.6–110.0%, respectively. The proposed ISD–DLLME technique was demonstrated to be simple, practical and efficient for the determination of different kinds of fungicide residues in real water samples.  相似文献

A semi-automatic flow-based method for the simultaneous determination of 9 pharmaceuticals and 3 hormones in water samples in a single analytical run is proposed. The analytes were retained on a solid-phase extraction sorbent column and 1 μL of the eluate analysed by gas chromatography in combination with electron impact ionization mass spectrometry in the SIM mode. The sorbent used, Oasis-HLB, provided near-quantitative recovery of all analytes. The proposed method was validated with quite good analytical results including low limits of detection (0.01–0.06 ng L−1 for 100 mL of water) and good linearity (r2 > 0.993) throughout the studied concentration ranges. The method provided good accuracy (recoveries of 85–103%) and precision (between- and within-day RSD values less than 7%) in the determination of the pharmaceuticals and hormones in tap, river, pond, well, swimming pool and wastewater.  相似文献

The alterations produced by microbiological attack on terpenoid resin-based varnishes from panel and canvas paintings have been evaluated using pyrolysis–gas chromatography–mass spectrometry (Py–GC–MS) and gas chromatography–mass spectrometry (GC–MS). The proposed methods include the on-line derivatisation of drying oils and diterpenoid resins using hexamethyldisilazane during pyrolysis and the application of methyl chloroformate as a derivatisation reagent for triterpenoid resins in GC–MS. Two types of specimens, consisting of model oil medium prepared from linseed oil and model spirit varnishes prepared from colophony and mastic resins dissolved in turpentine, have been used as reference materials. For a series of specimens upon which different genera of bacteria and fungi were inoculated and encouraged to grow, analyses indicated that no mechanisms that commonly occur during the attack of enzymes on drying oils and terpenoid biodegraders were observed to occur in the oil medium and varnishes studied. Thus, the degradation pathways observed in the performed trials usually occur as consequence of natural ageing. Specific trials consisting of the application of biocides to uninoculated colophony varnish resulted in the identification of processes that produce undesirable degradation of the varnish due to interactions between the biocide and the varnish components. Finally, the studied biocides—Biotin, New-Des and Nipagine—generally exhibited good inhibiting effects on the microorganisms studied, although some interesting differences were found between them regarding the application method and type of biocide.  相似文献

This paper describes a novel, simple and environmentally friendly method for rapid determination of the amide herbicides metoalchlor, acetochlor, and butachlor. It is based on dispersive liquid-liquid microextraction and gas chromatography–mass spectrometry. Factors that may influence the enrichment efficiency, such as type and volume of extraction solvent, type and volume of dispersive solvent, extraction time, and content of NaCl, were investigated and optimized in detail. Under the optimum conditions, the limits of detection of metoalchlor, acetochlor, and butachlor were 0.02, 0.04, and 0.003 μg L−1, respectively. The experimental results indicated that there was linearity over the range 0.1–50 μg L−1 and good reproducibility with relative standard deviations over the range 1.6–3.0% (n = 5). The proposed method has been applied for the analysis of real-world water samples, and satisfactory results were achieved. Average recoveries of spiked herbicides were in the range 80.3–108.8%. All of these indicated that the developed method would be an efficient method for simultaneous determination of the three herbicides in environmental water samples.  相似文献

Single-drop microextraction (SDME) followed by gas chromatography–mass spectrometry detection was used for the determination of some carbamate pesticides in water samples. The studied pesticides were thiofanox, carbofuran, pirimicarb, methiocarb, carbaryl, propoxur, desmedipham and phenmedipham. Two alternative sample introduction methods have been examined and compared; SDME followed by cool on-column injection (without derivatization) and SDME followed by in-microvial derivatization and splitless injection. Acetic anhydride was used as derivatization reagent. Parameters that affect the derivatization reaction yield and the extraction efficiency of the SDME method were studied and optimized. The analytical performances and possible applications of both approaches were investigated. Relative standard deviations for the studied compounds ranged from 3.2 to 8.3%. The detection limits obtained by the derivatization method were found to be in the range 3–35 ng/L. Using cool on-column injection (without derivatization), the detection limits were between 30 and 80 ng/L.  相似文献

This paper describes a method for the determination of a group of personal care products including four UV filters, four preservatives and two antimicrobials in sewage sludge. The method combines pressurized liquid extraction and ultra high performance liquid chromatography–tandem mass spectrometry. Most of the parameters that affect the extraction step such as temperature, pressure, static extraction time, number of cycles, purge time and flush volume were optimized using a fractional experimental design. In the chromatographic step, the compounds were detected by using tandem mass spectrometry with a triple quadrupole analyzer with electrospray ionization in positive and negative modes. The use of small diameter particles (1.8 μm) in the chromatographic column allowed the compounds to be eluted in 9 min. The entire process took a total of 39 min. All recoveries were higher than 72% except for 2,4-dihydroxybenzophenone (a UV filter), whose recovery was 30%. The repeatability and reproducibility between days expressed as RSD (%) (n = 3) were less than 8% and 13%, respectively. The LODs and LOQs were lower than 8 μg/kg and 12.5 μg/kg of dry weight (d.w.), respectively. When the method was applied to determine the compounds in sewage sludge from a domestic sewage treatment plant, triclosan (an antimicrobial) and octocrylene (a UV filter) showed the highest levels, 1490 μg/kg (d.w.) and 1842 μg/kg (d.w.), respectively. This paper describes for the first time the determination of parabens and two UV filters (octyldimethyl-p-aminobenzoic acid and benzophenone-3) in sewage sludge.  相似文献

Complementary methods using liquid chromatography–tandem quadrupole mass spectrometry (LC–MS/MS) and comprehensive two-dimensional gas chromatography–time-of-flight mass spectrometry (GC × GC–TOF-MS) were developed and applied to determine targeted metabolites involved in central carbon metabolism [including tricarboxylic acid cycle, serine cycle, ethylmalonyl-coenzyme A (ethylmalonyl-CoA) pathway and poly-β-hydroxybutyrate cycle] of the bacterium Methylobacterium extorquens AM1 grown on two carbon sources, ethylamine (C2) and succinate (C4). Nucleotides, acyl-CoAs and a few volatile metabolites in cell extracts of M. extorquens AM1 were readily separated using either hydrophilic interaction liquid chromatography or reversed-phase liquid chromatography, and detected with good sensitivity by MS/MS. However, volatile intermediates within a low mass range (<300 m/z), especially at low abundance (such as glyoxylic acid and others <500 nM), were more effectively analyzed by GC × GC–TOF-MS which often provided better sensitivity, resolution and reproducibility. The complementary nature of the LC-based and GC-based methods allowed the comparison of 39 metabolite concentrations (the lowest level was at 139.3 nM). The overlap between the LC-based and GC-based methods of seven metabolites provided a basis to check for consistency between the two methods, and thus provided some validation of the quantification accuracy. The abundance change of 20 intermediates further suggested differences in pathways linked to C2 and C4 metabolism.  相似文献

The performance of the dispersive liquid–liquid microextraction (DLLME) technique for the determination of eight UV filters and a structurally related personal care species, benzyl salicylate (BzS), in environmental water samples is evaluated. After extraction, analytes were determined by gas chromatography combined with mass spectrometry detection (GC-MS). Parameters potentially affecting the performance of the sample preparation method (sample pH, ionic strength, type and volume of dispersant and extractant solvents) were systematically investigated using both multi- and univariant optimization strategies. Under final working conditions, analytes were extracted from 10 mL water samples by addition of 1 mL of acetone (dispersant) containing 60 μL of chlorobenzene (extractant), without modifying either the pH or the ionic strength of the sample. Limits of quantification (LOQs) between 2 and 14 ng L−1, inter-day variability (evaluated with relative standard deviations, RSDs) from 9% to 14% and good linearity up to concentrations of 10,000 ng L−1 were obtained. Moreover, the efficiency of the extraction was scarcely affected by the type of water sample. With the only exception of 2-ethylhexyl-p-dimethylaminobenzoate (EHPABA), compounds were found in environmental water samples at concentrations between 6 ± 1 ng L−1 and 26 ± 2 ng mL−1.  相似文献

In this work,a reliable and sensitive method for detecting polybrominated diphenyl ethers(PBDEs) has been developed by the combination of liquid–liquid extraction and gas chromatography–mass spectrometry.PBDEs were extracted from a large volume of water by liquid–liquid extraction and purified by silica gel chromatography.In order to reduce the deviation,dibromobiphenyl was exploited as the internal standard to minimize differences among the injections.The quantification was performed using an external standard.Good linear correlation coefficients(0.991) and a wide linearity range(1.0–500.0 ng/L) indicated the steadiness of the proposed method.Moreover,the satisfactory recovery(75%)suggested that successful determination of PBDEs in river water had been achieved.Furthermore,the deduction behavior of PBDEs in river water could be inferred according to the results.  相似文献

An ultra high performance liquid chromatography–tandem mass spectrometry method (UPLC–MS/MS) is proposed for the simultaneous quantification of inosine, adenosine, guanosine, uridine, hypoxanthine, xanthine and uric acid in pork meat, dry-cured and cooked ham. Samples were added with 15N2-xanthine (internal standard) and extracted with boiling water for 30 min. Supernatants were washed with hexane, added with formic acid 10% in water, methanol:acetone (1:1, v/v), evaporated to dryness under N2, and finally re-dissolved in water prior to injection. Chromatographic separation was carried out with a HSS T3 column with a total time of analysis of 15 min. Two specific transitions for each compound were used for identification and quantification (with matrix matched calibration curves). Linearity, limit of detection, repeatability and accuracy were evaluated. The method was used to quantify the seven purines and pyrimidines in 15 commercial samples.  相似文献
