Pyrazolidine‐3,5‐diones and their derivatives exhibit a wide range of biological activities. Seeking to explore the effect of combining a hydrocarbyl ring substituent, as present in sulfinpyrazone (used to treat gout), with a chlorinated aryl ring, as present in muzolimine (a diuretic), we explored the reaction between 1‐phenylpyrazolidine‐3,5‐dione and 4‐chlorobenzaldehyde under mildly basic conditions in the expectation of producing the simple condensation product 4‐(4‐chlorobenzylidene)‐1‐phenylpyrazolidine‐3,5‐dione. However, the reaction product proved to be meso‐(E,E)‐1,1′‐[1,2‐bis(4‐chlorophenyl)ethane‐1,2‐diyl]bis(phenyldiazene), C26H20Cl2N4, and a tentative mechanism is proposed. Crystallization from ethanol produces two concomitant polymorphs, i.e. a triclinic form, (I), in the space group P, and a monoclinic form, (II), in the space group C2/c. In both polymorphs, the molecules lie across centres of inversion, but in (II), the molecules are subject to whole‐molecule disorder equivalent to configurational disorder with occupancies of 0.6021 (19) and 0.3979 (19). There are no hydrogen bonds in the crystal structure of polymorph (I), but the molecules of polymorph (II) are linked by C—H...π(arene) hydrogen bonds into complex chains, which are further linked into sheets by C—H...N interactions.  相似文献

The [3,3′(4H,4′H)‐bi‐2H‐1,3‐oxazine]‐4,4′‐diones 3a – 3i were obtained by [2+4] cycloaddition reactions of furan‐2,3‐diones 1a – 1c with aromatic aldazines 2a – 2d (Scheme 1). So, new derivatives of bi‐2H‐1,3‐oxazines and their hydrolysis products, 3,5‐diaryl‐1H‐pyrazoles 4a – 4c (Scheme 3), which are potential biologically active compounds, were synthesized for the first time.  相似文献

The reaction of substituted phenyl isocyanates with 2‐amino‐2‐phenylpropanenitrile and 2‐amino‐2‐(4‐nitrophenyl)propanenitrile has been used to prepare substituted 1‐(1‐cyanoethyl‐1‐phenyl)‐3‐phenylureas. In anhydrous phosphoric acid the first products to be formed from 1‐(1‐cyanoethyl‐1‐phenyl)‐3‐phenylureas are phosphates of 4‐methyl‐4‐phenyl‐2‐phenylimino‐5‐imino‐4,5‐dihydro‐1,3‐oxazoles, which on subsequent hydrolysis give the respective ureidocarboxylic acids. On prolongation of the reaction time, the phosphates of 4‐methyl‐4‐phenyl‐2‐phenylimino‐5‐imino‐4,5‐dihydro‐1,3‐oxazoles rearrange to give phosphates of 5‐methyl‐4‐imino‐3,5‐diphenylimidazolidin‐2‐ones, and these are subsequently hydrolysed to the respective substituted 5‐methyl‐3,5‐diphenylimidazolidin‐2,4‐diones. The ureidocarboxylic acids were also prepared by alkaline hydrolysis of 5‐methyl‐3,5‐diphenylimidazolidin‐2,4‐diones. The 5‐methyl‐3,5‐diphenylimidazolidin‐2,4‐diones and ureidocarboxylic acids were characterised by their 1H and 13C NMR spectra. Structure of the 5‐methyl‐5‐(4‐nitrophenyl)‐3‐phenylimidazolidine‐2,4‐dione was verified by X‐ray diffraction. The alkaline hydrolysis of individual imidazolidine‐2,4‐diones was studies spectrophoto‐metrically in sodium hydroxide solutions at 25 °C. The rate‐limiting step of the base catalysed hydrolysis consists in decomposition of the tetrahedral intermediate. The reaction is faster if electron‐acceptor sub‐stituents are present in the 3‐phenyl group of imidazolidine‐2,4‐dione cycle. The pKa values of individual 5‐methyl‐3,5‐diphenylimidazolidine‐2,4‐diones have been determined kinetically.  相似文献

A new route for the synthesis of 3,5‐diarylcyclohex‐2‐enones is reported. The 4‐acetyl‐1,3‐diarylhexane‐1,5‐diones were obtained by the addition of pentane‐2,4‐dione to chalcones. The reaction of 4‐acetyl‐1,3‐diarylhexane‐1,5‐diones with NH4Cl/HCl in EtOH under reflux conditions gave the 3,5‐diarylcyclohex‐2‐enones in good yields. All synthesized compounds were characterized by spectroscopic methods (1H‐, 13C‐NMR, and IR), and elemental analyses.  相似文献

A direct ortho‐Csp2‐H acylmethylation of 2‐aryl‐2,3‐dihydrophthalazine‐1,4‐diones with α‐carbonyl sulfoxonium ylides is achieved through a RuII‐catalyzed C?H bond activation process. The protocol featured high functional group tolerance on the two substrates, including aryl‐, heteroaryl‐, and alkyl‐substituted α‐carbonyl sulfoxonium ylides. Thereafter, 2‐(ortho‐acylmethylaryl)‐2,3‐dihydrophthalazine‐1,4‐diones were used as potential starting materials for the expeditious synthesis of 6‐arylphthalazino[2,3‐a]cinnoline‐8,13‐diones and 5‐acyl‐5,6‐dihydrophthalazino[2,3‐a]cinnoline‐8,13‐diones under Lawesson's reagent and BF3?OEt2 mediated conditions, respectively. Of these, the BF3?OEt2‐mediated cyclization proceeded in DMSO as a solvent and a methylene source via dual C?C and C?N bond formations.  相似文献

Previously unknown 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazoline]‐2,2′‐(7′H)‐diones and their N‐substituted analogues were obtained via reaction of 6‐R1‐3‐(2‐aminophenyl)‐1,2,4‐triazin‐5‐ones with isatin and its substituted derivatives. It was shown that alkylation of 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′‐(7′H)‐diones by N‐R3‐chloroacetamides or chloroacetonitrile in the presence of а base proceeds by N‐1 atom of isatin fragment. The spectral properties (1H and 13C NMR spectra) of synthesized compounds were studied, and features of spectral patterns were discussed. The high‐effective anticonvulsant and radical scavenging agents among 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′(7′H)‐diones and their N‐substituted derivatives were detected. It was shown that compounds 2.2 , 2.8 , and 3.1 exceed or compete the activity of the most widely used in modern neurology drug—lamotrigine on the pentylenetetrazole‐induced seizures model. The aforementioned fact may be considered as a reason for further profound study of synthesized compounds using other pathology models.  相似文献

Quite unlike the reported facile ene reactions on the periphery of many related heterocyclic systems, similarly disposed moieties on the periphery of the chromen‐4‐one (=4H‐1‐benzopyran‐4‐one) system fail to undergo an ene reaction and display a rather unusual preference for an overall [1,5] shift of the allylic C‐atom. Thus, heating xylene solutions of 2‐(N‐allylanilino)‐, 2‐(N‐crotylanilino)‐, and 2‐(N‐cinnamylamino)‐substituted (E)‐(oxochromenyl)propenoates 9a – c and 2‐[allyl(benzyl)amino]‐, 2‐[benzyl(crotyl)amino]‐, and 2‐[benzyl(cinnamyl)amino]‐substituted (E)‐(oxochromenyl)propenoates 16a – c in a sealed tube at 220–230° leads to a [1,5] shift of the allylic moieties (allyl, crotyl, cinnamyl), which is followed by intramolecular cyclization involving the N‐atom and the ester function, to give the 3‐allyl‐3‐crotyl‐, and 3‐cinnamyl‐substituted‐1‐phenyl‐ or 1‐benzyl‐2H‐[1]benzopyrano[2,3‐b]pyridine‐2,5(1H)‐diones 10a – c and 17a – c . The anticipated carbonyl–ene reaction in the 2‐(N‐allylanilino)‐, 2‐(N‐crotylanilino)‐, 2‐(N‐cinnamylanilino)‐, 2‐[allyl(benzyl)amino]‐, 2‐[benzyl(crotyl)amino]‐, and 2‐[benzyl(cinnamyl)amino]‐substituted 4‐oxochromene‐3‐carboxaldehydes 8a – c and 15a – c is also not observed, and these molecules remain untransformed under identical conditions. No [1,5] shifts of benzyl, phenyl, or methyl groups are observed, even in the absence of allylic moieties, though facile [1,5]‐H shift occurs in 2‐(benzylamino)‐ and 2‐(phenylamino)‐substituted (E)‐(oxochromenyl)propenoates 23a , b , which is followed by a similar intramolecular cyclization leading to the 2H‐[1]benzopyrano[2,3‐b]pyridine‐2,5(1H)‐diones 24a , b .  相似文献

Chiral ligand (A)‐N,N′‐Bis(2‐hydroxy‐3,5‐di‐tert‐butyl‐arylmethyl)‐1,1′‐binaphthalene‐2,2′‐diamine derived from the reduction of Schiff base (R)‐2,2′‐bis (3,5‐di‐tert‐butyl‐2‐hydroxybenzylideneamino)‐1, 1′‐binaphthyl with LiAlH4, is fairly effective in the asymmetric addition reaction of diethylzinc to aldehydes by which good yields (46%‐94%) of the corresponding sec‐alcohols can be obtained in moderate ee (51%‐79%) with R configuration for a variety of aldehydes.  相似文献

Quinazoline‐2,4(1H,3H)‐diones are core structural subunits frequently found in many biologically important compounds. The reaction of 2‐​aminobenzonitrile and CO2, which was frequently studied, only provided N3‐unsubstituted quinazoline‐2,4(1H,3H)‐dione compounds. Herein we report palladium‐catalyzed cyclization reactions of o‐haloanilines, CO2 and isocyanides to prepare N3‐substituted quinazoline‐2,4(1H,3H)‐diones. Electron‐rich o‐bromoanilines participated in the cyclization reaction using Cs2CO3 at high temperature, and electron‐deficient o‐bromoaniline or o‐iodoaniline substrates conducted the reaction using CsF as base to deliver corresponding quinazoline‐2,4(1H,3H)‐dione products in good yields.  相似文献

The Zn atom in dichloro­[(3,5‐dimethyl‐1H‐pyrazol‐1‐yl)­methane]zinc(II), [ZnCl2(C11H16N4)], (I), is tetra­hedrally coordinated by two N atoms from one bis­(3,5‐dimethyl­pyrazol­yl)methane ligand and two terminal Cl atoms. The mol­ecule has no crystallographic symmetry. One H atom of the CH2 group of the bis­(3,5‐dimethyl­pyrazol­yl)methane ligand inter­acts with a Cl atom of an adjacent mol­ecule to yield inter­molecular C—H⋯Cl contacts, thereby forming a one‐dimensional zigzag chain extending along the b axis. On the other hand, in di‐μ‐chloro‐bis­{chloro­[(3,5‐dimethyl‐1H‐pyrazol‐1‐yl)methane]cadmium(II)}, [Cd2Cl4(C11H16N4)2], (II), each of the two crystallographically equivalent Cd atoms is penta­coordinated by two N atoms from one bis­(3,5‐dimethyl­pyrazol­yl)methane ligand, and by one terminal and two bridging Cl anions. The mol­ecule has a crystallographic centre of symmetry located at the mid‐point of the Cd⋯Cd line. One H atom of the CH2 group of the bis­(3,5‐dimethyl­pyrazolyl)­methane ligand inter­acts with a Cl atom of an adjacent mol­ecule to produce pairwise inter­molecular C—H⋯Cl contacts, thereby affording chains of mol­ecules running along the c axis.  相似文献

1H‐Quinazoline‐2,4‐diones, which are key intermediates in the synthesis of medicines, were successfully synthesized from 2‐aminobenzonitriles by the fixation of CO2 in the presence of a polystyrene derivative bearing amidine moiety [poly(amidine)]. A model reaction, that is, the reaction of 2‐aminobenzonitrile ( 1a ) with CO2 in the presence of N‐methyltetrahydropyrimidine ( MTHP ) revealed that a catalytic amount of MTHP afforded 1H‐quinazoline‐2,4‐dione ( 2a ) quantitatively at atmospheric pressure. Several 1H‐quinazoline‐2,4‐diones ( 2a ‐ 2c ) were successfully synthesized from the corresponding 2‐aminobenzonitriles ( 1a ‐ 1c ) in the presence of poly(amidine). The poly(amidine) could easily be separated from the reaction mixture by filtration and reused in subsequent reactions owing to the heterogeneous system. These demonstrated that poly(amidine) is a useful heterogeneous polymer‐supported reagent for the synthesis of 1H‐quinazoline‐2,4‐diones from CO2. © 2008 Wiley Periodicals, Inc. Bright yellow N,N'‐carbonyl‐bridged dipyrrinones (substituted 3H,5H‐dipyrrolo[1,2‐c:2′,1′‐f]pyrimidine‐3,5‐diones) were synthesized by reaction of the parent dipyrrinone with carbonyldiimidazole. Three photoluminescent complexes containing either ZnII or CdII have been synthesized and their structures determined. Bis[4‐amino‐3,5‐bis(pyridin‐2‐yl)‐1,2,4‐triazole‐κ2N 1,N 5]bis(dicyanamido‐κN 1)zinc(II), [Zn(C12H10N6)2(C2N3)2], (I), bis[4‐amino‐3,5‐bis(pyridin‐2‐yl)‐1,2,4‐triazole‐κ2N 1,N 5]bis(dicyanamido‐κN 1)cadmium(II), [Cd(C12H10N6)2(C2N3)2], (II), and bis[4‐amino‐3,5‐bis(pyridin‐2‐yl)‐1,2,4‐triazole‐κ2N 1,N 5]bis(tricyanomethanido‐κN 1)cadmium(II), [Cd(C12H10N6)2(C4N3)2], (III), all crystallize in the space group P , with the metal centres lying on centres of inversion, but neither analogues (I) and (II) nor CdII complexes (II) and (III) are isomorphous. A combination of N—H…N and C—H…N hydrogen bonds and π–π stacking interactions generates three‐dimensional framework structures in (I) and (II), and a sheet structure in (III). The photoluminescence spectra of (I)–(III) indicate that the energies of the π–π* transitions in the coordinated triazole ligand are modified by minor changes of the ligand geometry associated with coordination to the metal centres.  相似文献

Homophthalic anhydride 1 reacts with different aromatic amines to produce N‐substituted homophthalimides 2 under microwave irradiation. A rapid microwave‐assisted chemical synthesis of condensed 4‐substituted furo[2,3‐c]isoquinoline‐1,5(2H,4H)‐diones 3 and 5‐substituted‐2,3‐dihydro‐1H‐pyrano[2,3‐c]isoquinoline‐1,6(5H)‐diones 4 involving the condensation of a variety of alkanoyl chlorides with 2‐arylisoquinoline‐1,3‐diones 2 in the presence of base and aprotic solvent is described for the first time. By contrast, the facile ring opening reaction of furo[2,3‐c]isoquinoline‐1,5(2H,4H)‐dione 3 with Vilsmeier–Haack reagent under microwave irradiation yielded the α‐β unsaturated carboxyaldehyde 5 . This novel and clean one‐pot methodology, which is characterized by very short reaction time and easy workup procedure, can be exploited to generate some novel condensed isoquinoline derivatives.  相似文献

Pyrrolo[3,4‐c]pyrazole‐4,6‐diones, pyrazoles, pyrazolo[3,4‐d]pyridazines, and pyrazolo[3,4‐d]pyrimidines were prepared via 1‐bromo‐2‐(5‐chlorobenzofuran‐2‐yl)ethanedione‐1‐phenylhydrazone with N‐arylmalemides and active methylene. The heteroscorpionate ligand 2,2‐bis(3,5‐dimethylpyrazol‐1‐yl)‐1,1‐diphenylethanol, C24H26N4O, features in the solid state an intramolecular O—H…N hydrogen bond. A heteroscorpionate tungsten complex, cis‐[2,2‐bis(3,5‐dimethylpyrazolyl)‐1,1‐diphenylethanolato]chloridodioxidotungsten(VI) tetrahydrofuran monosolvate, [W(C24H25N4O)ClO2]·C4H8O, was prepared by the simple mixing of solutions of the ligand and WOCl4 in tetrahydrofuran. The tungsten complex was isolated after standing for several weeks. The complex exhibits a κ3N,N′,O‐coordination of the ligand. This simple synthetic procedure allows access to the cis isomer in high yield without additional purification steps. The Hirshfeld surface analysis shows a change of the intermolecular contacts due to the coordination of the WO2Cl unit with the ligand molecule.  相似文献
