Primary mechanism of a PdII‐catalyzed 8‐aminoquinoline‐directed C?H alkoxylation was investigated. It was understood that the PdII‐catalyzed C(sp3)?O bond formation proceeded through a concerted reductive elimination from the PdIV intermediate in the cyclic system. Deuteration experiments and related computational studies elucidate that intrinsic conformation determined the diastereoselectivity of the PdII‐catalyzed C?H alkoxylation of cyclic carboxylic acids.  相似文献

Reduction of the Pd?PEPPSI precatalyst to a Pd0 species is generally thought to be essential to drive Buchwald–Hartwig amination reactions through the well‐ documented Pd0/PdII catalytic cycle and little attention has been paid to other possible mechanisms. Considered here is the Pd?PEPPSI‐catalyzed aryl amination of chlorobenzene with aniline. A neat reaction system was used in new experiments, from which the potentially reductive roles of the solvent and labile ligand of the PEPPSI complex in leading to Pd0 species are ruled out. Computational results demonstrate that anilido‐containing PdII intermediates involving σ‐bond metathesis in pathways leading to the diphenylamine product have relatively low barriers. Such pathways are more favorable energetically than the corresponding reductive elimination reactions resulting in Pd0 species and other putative routes, such as the PdII/PdIV mechanism, single electron transfer mechanism, and halide atom transfer mechanism. In some special cases, if reactants/additives are inadequate to reduce a PdII precatalyst, a PdII‐involved σ‐bond metathesis mechanism might be feasible to drive the Buchwald–Hartwig amination reactions.  相似文献

The use of chiral transient directing groups (TDGs) is a promising approach for developing PdII‐catalyzed enantioselective C(sp3)?H activation reactions. However, this strategy is challenging because the stereogenic center on the TDG is often far from the C?H bond, and both TDG covalently attached to the substrate and free TDG are capable of coordinating to PdII centers, which can result in a mixture of reactive complexes. We report a PdII‐catalyzed enantioselective β‐C(sp3)?H arylation reaction of aliphatic ketones using a chiral TDG. A chiral trisubstituted cyclobutane was efficiently synthesized from a mono‐substituted cyclobutane through sequential C?H arylation reactions, thus demonstrating the utility of this method for accessing structurally complex products from simple starting materials. The use of an electron‐deficient pyridone ligand is crucial for the observed enantioselectivity. Interestingly, employing different silver salts can reverse the enantioselectivity.  相似文献

A mild protocol has been developed for the PdII‐catalyzed alkoxycarbonylation of terminal olefins to produce α,β‐unsaturated esters with a wide range of substrates. Key features are the use of MeCN as solvent (and/or ligand) to control the reactivity of the intermediate Pd complexes and the combination of CO with O2, which facilitates the CuII‐mediated reoxidation of the Pd0 complex to PdII and prevents double carbonylation.  相似文献

PdII‐catalyzed C(sp3)?H arylation of saturated heterocycles with a wide range of aryl iodides is enabled by an N‐heterocyclic carbene (NHC) ligand. A C(sp3)?H insertion step by the PdII/NHC complex in the absence of ArI is demonstrated experimentally for the first time. Experimental data suggests that the previously established NHC‐mediated Pd0/PdII catalytic manifold does not operate in this reaction. This transformation provides a new approach for diversifying pharmaceutically relevant piperidine and tetrahydropyran ring systems.  相似文献

A series of cyclometalated PdII complexes that contain π‐extended R? C^N^N? R′ (R? C^N^N? R′=3‐(6′‐aryl‐2′‐pyridinyl)isoquinoline) and chloride/pentafluorophenylacetylide ligands have been synthesized and their photophysical and photochemical properties examined. The complexes with the chloride ligand are emissive only in the solid state and in glassy solutions at 77 K, whereas the ones with the pentafluorophenylacetylide ligand show phosphorescence in the solid state (λmax=584–632 nm) and in solution (λmax=533–602 nm) at room temperature. Some of the complexes with the pentafluorophenylacetylide ligand show emission with λmax at 585–602 nm upon an increase in the complex concentration in solutions. These PdII complexes can act as photosensitizers for the light‐induced aerobic oxidation of amines. In the presence of 0.1 mol % PdII complex, secondary amines can be oxidized to the corresponding imines with substrate conversions and product yields up to 100 and 99 %, respectively. In the presence of 0.15 mol % PdII complex, the oxidative cyanation of tertiary amines could be performed with product yields up to 91 %. The PdII complexes have also been used to sensitize photochemical hydrogen production with a three‐component system that comprises the PdII complex, [Co(dmgH)2(py)Cl] (dmgH=dimethylglyoxime; py=pyridine), and triethanolamine, and a maximum turnover of hydrogen production of 175 in 4 h was achieved. The excited‐state electron‐transfer properties of the PdII complexes have been examined.  相似文献

A convergent synthesis of an analogue of (1α)‐1,25‐dihydroxyvitamin D3 ( 1b ) with a C7 side chain at C(12), i.e., of 5 (Fig.), is described. A key step of the synthesis is the assembly of the triene system by a PdII‐catalyzed ring closure of an enol triflate (‘bottom’ fragment) followed by coupling of the resulting PdII intermediate with an alkenylboronate (‘upper’ fragment) (Scheme 2). The synthetic strategy allows isotopic labelling at the end of the synthesis.  相似文献

The Pd0 complex 1 that bears the Trost ligand 2 undergoes a facile redox reaction with 1,4‐biscarbonates 5 b – d and rac‐ 22 under formation of the diamidato–PdII complex 7 and the corresponding 1,3‐cycloalkadienes 8 b – d . The redox deactivation of complex 1 was the dominating pathway in the reaction of 5 b – d with HCO3? at room temperature. However, at 0 °C the six‐membered biscarbonate 5 b , catalytic amounts of complex 1 , and HCO3? mainly reacted in an allylic alkylation, which led to a highly selective desymmetrization of the substrate and gave alcohol 6 b with ≥99 % ee in 66 % yield. An increase of the catalyst loading in the reaction of 5 b with 1 and HCO3? afforded the bicyclic carbonate 12 b (96 % ee, 92 %). Formation of carbonate 12 b involves two consecutive inter‐ and intramolecular substitution reactions of the π‐allyl–PdII complexes 16 b and 18 b , respectively, with O‐nucleophiles and presumably proceeds through the hydrogen carbonate 17 b as key intermediate. The intermediate formation of 17 b is also indicated by the conversion of alcohol rac‐ 6 b to carbonate 12 b upon treatment with HCO3? and 1 . The Pd0‐catalyzed desymmetrization of 5 b with formation of 12 b and its hydrolysis allow an efficient enantioselective synthesis of diol 13 b . The reaction of the seven‐membered biscarbonate 5 c with ent‐ 1 and HCO3? afforded carbonate ent‐ 12 c (99 % ee, 39 %). The Pd0 complex 1 is stable in solution and suffers no intramolecular redox reaction with formation of complex 7 and dihydrogen as recently claimed for the similar Pd0 complex 9 . Instead, complex 1 is rapidly oxidized by dioxygen to give the stable PdII complex 7 . Thus, formation of the PdII complex 10 from 9 was most likely due to an oxidation by dioxygen. Oxidative workup (air) of the reaction mixture stemming from the desymmetrization of 5 c catalyzed by 1 gave the PdII complex 7 in high yield besides carbonate 12 c .  相似文献

A palladium‐catalyzed carbene insertion into C(sp3)?H bonds leading to pyrrolidines was developed. The coupling reaction can be catalyzed by both Pd0 and PdII, is regioselective, and shows a broad functional group tolerance. This reaction is the first example of palladium‐catalyzed C(sp3)?C(sp3) bond assembly starting from diazocarbonyl compounds. DFT calculations revealed that this direct C(sp3)?H bond functionalization reaction involves an unprecedented concerted metalation–deprotonation step.  相似文献

The first example of PdII‐catalyzed enantioselective C?H olefination with non‐chiral or racemic sulfoxides as directing groups was developed. A variety of chiral diaryl sulfoxides were synthesized with high enantioselectivity (up to 99 %) through both desymmetrization and parallel kinetic resolution (PKR). This is the first report of PdII‐catalyzed enantioselective C(sp2)?H functionalization through PKR, and it represents a novel strategy to construct sulfur chiral centers.  相似文献

A Pd‐catalyzed efficient reductive cross‐coupling reaction without metallic reductant to construct a Csp2?Csp3 bond has been reported. A PdIV complex was proposed to be a key intermediate, which subsequently went through double oxidative addition and double reductive elimination to produce the cross‐coupling products by involving Pd0/II/IV in one transformation. The oxidative addition from PdII to PdIV was partially demonstrated to be a radical process by self‐oxidation of substrate without additional oxidants. Furthermore, the solvent was proved to be the reductant for this transformation through XPS analysis.  相似文献

Pincer PdII–isocyanide complexes are described that display intermolecular interactions and emissive 3MMLCT excited states in aggregation state(s) at room temperature. The intermolecular PdII?PdII and ligand–ligand interactions drive these complexes to undergo supramolecular polymerization in a living manner. Comprehensive spectroscopic studies reveal a pathway with a kinetic trap that can be modulated by changing the counteranion and metal atom. The PdII supramolecular assemblies comprise two different aggregation forms with only one to be emissive. DFT/TDDFT calculations lend support to the MMLCT absorption and emission of these pincer PdII–isocyanide aggregates.  相似文献

A general and practical PdII‐catalyzed intermolecular silylation of primary and secondary C?H bonds of α‐amino acids and simple aliphatic acids is reported. This method provides divergent and stereoselective access to a variety of optical pure β‐silyl‐α‐amino acids, which are useful for genetic technologies and proteomics. It can also be readily performed on a gram scale and the auxiliary can be easily removed with retention of configuration. The synthetic importance of this method is further demonstrated by the late‐stage functionalization of biological small molecules, such as (?)‐santonin and β‐cholic acid. Moreover, several key palladacycles were successfully isolated and characterized to elucidate the mechanism of this β?C(sp3)‐H silylation process.  相似文献

A palladium‐catalyzed cascade reaction based on the trapping of transient alkyl–PdII intermediates with arynes encompassing a C?H activation step has been developed. This synthetic pathway gives rise to hetero‐spirocyclic scaffolds containing a biaryl motif, and opens up new synthetic strategies in the design of cascade reactions since it gathers several aspects of Pd chemistry, i.e., intra‐ and intermolecular carbopalladation of unsaturated species, C?H activation and C?C coupling processes.  相似文献

Palladium‐catalyzed intermolecular coupling of o‐carborane with aromatics by direct cage B?H bond activation has been achieved, leading to the synthesis of a series of cage B(4,5)‐diarylated‐o‐carboranes in high yields with excellent regioselectivity. Traceless directing group ‐COOH plays a crucial role for site‐ and di‐selectivity of such intermolecular coupling reaction. A PdII–PdIV–PdII catalytic cycle is proposed to be responsible for the stepwise arylation.  相似文献

Mono‐N‐protected amino acids (MPAAs) are increasingly common ligands in Pd‐catalyzed C?H functionalization reactions. Previous studies have shown how these ligands accelerate catalytic turnover by facilitating the C?H activation step. Here, it is shown that MPAA ligands exhibit a second property commonly associated with ligand‐accelerated catalysis: the ability to support catalytic turnover at substoichiometric ligand‐to‐metal ratios. This catalytic role of the MPAA ligand is characterized in stoichiometric C?H activation and catalytic C?H functionalization reactions. Palladacycle formation with substrates bearing carboxylate and pyridine directing groups exhibit a 50–100‐fold increase in rate when only 0.05 equivalents of MPAA are present relative to PdII. These and other mechanistic data indicate that facile exchange between MPAAs and anionic ligands coordinated to PdII enables a single MPAA to support C?H activation at multiple PdII centers.  相似文献

The first example of PdII‐catalyzed γ‐C(sp3)?H functionalization of aliphatic and benzoheteroaryl aldehydes has been developed using a transient ligand and an external ligand, concurrently. A wide array of γ‐arylated aldehydes were readily accessed without preinstalling internal directing groups. The catalytic mechanism was studied by performing deuterium‐labelling experiments, which indicated that the γ‐C(sp3)?H bond cleavage is the rate‐limiting step during the reaction process. This reaction could be performed on a gram scale, and also demonstrated its potential application in the synthesis of new mechanofluorochromic materials with blue‐shifted mechanochromic properties.  相似文献

The first asymmetric PdII‐catalyzed aminofluorination of unactivated alkenes using chiral quinoline‐oxazolines (Quox) as ligands has been developed. This reaction provides easy access to a wide array of enantiomerically enriched β‐fluoropiperidines in good yields and with excellent enantioselectivity. Notably, Et4NF?3 HF as a readily accessible nucleophilic fluoride source was found to play an essential role in the enantioselective control, and CsOCF3 also acts a key additive to improve the excellent ee value of products.  相似文献

The palladium‐catalyzed desymmetrization of silacyclobutanes using electron‐deficient alkynes proceeds with high enantioselectivity in the presence of a chiral P ligand; this provides a facile approach for the synthesis of novel silicon‐stereogenic silanes. In this work, we used hybrid density functional theory (DFT) to elucidate the mechanism of the palladium‐catalyzed desymmetrization of silacyclobutanes with dimethyl acetylenedicarboxylate. Full catalytic cycle including two different initiation modes that were proposed to be a possible initial step to the formation of the 1‐pallada‐2‐silacyclopentane/alkyne intermediate—the oxidative addition of the palladium complex to the silacyclobutane Si?C bond (cycle MA) or coordination of the Pd0 complex with the alkyne C≡C bond (cycle MB)—have been studied. It was found that the ring‐expansion reaction began with cycle MB is energetically more favorable. The formation of a seven‐membered metallocyclic PdII intermediate was found to be the rate‐determining step, whereas the enantioselectivity‐determining step, oxidative addition of silacyclobutane to the three‐membered metallocyclic PdII intermediate, was found to be quite sensitive to the steric repulsion between the chiral ligand and silacyclobutane.  相似文献
