Key metabolites for the diagnosis of the genetic disorder trimethylaminuria are trimethylamine (TMA) and trimethylamine N-oxide (TMAO). A rapid, automatable flow injection ESI-MS/MS method for their measurement in urine has been developed. The TMA was derivatized with ethyl bromoacetate to form ethyl betaine bromide. The 2 min ESI-MS/MS analysis employed four multiple reaction monitoring (MRM) ion pairs for derivatized TMA (146.1, 118.1), derivatized (2)H(9)-TMA (155.1, 127.1), TMAO (76.1, 58.1) and (2)H(9)-TMAO (85.1, 66.1). In control urine samples (n = 27) referred for suspected metabolic problems TMA was 0.11-1.19 mmol/mol creatinine, TMAO was 13.5-181 mmol/mol creatinine and the TMA/TMAO ratio was 0.0025-0.055. In five patients with diagnosed trimethylaminuria, TMA was 5.3-230 mmol/mol creatinine, TMAO was 0.36-607 mmol/mol creatinine and the TMA/TMAO ratio was 0.20-134.  相似文献

Background: Trimethylaminuria (TMAU) is a rare genetic disease characterized by the accumulation of trimethylamine (TMA) and its subsequent excretion trough main body fluids, determining the characteristic fish odour in affected patients. We realized an experimental study to investigate the role of several coding variants in the causative gene FMO3, that were only considered as polymorphic or benign, even if the available literature on them did not functionally explain their ineffectiveness on the encoded enzyme. Methods: Mutational analysis of 26 TMAU patients was realized by Sanger sequencing. Detected variants were, subsequently, deeply statistically and in silico characterized to determine their possible effects on the enzyme activity. To achieve this goal, a docking prediction for TMA/FMO3 and an unbinding pathway study were performed. Finally, a TMAO/TMA urine quantification by 1H-NMR spectroscopy was performed to support modelling results. Results: The FMO3 screening of all patients highlighted the presence of 17 variants distributed in 26 different haplotypes. Both non-sense and missense considered variants might impair the enzymatic kinetics of FMO3, probably reducing the interaction time between the protein catalytic site and TMA, or losing the wild-type binding site. Conclusions: Even if further functional assays will confirm our predictive results, considering the possible role of FMO3 variants with still uncertain effects, might be a relevant step towards the detection of novel scenarios in TMAU etiopathogenesis.  相似文献

Use of a thermomechanical analyzer, TMA, affords a rapid, precise method for determining linear expansion coefficients and linear expansivities of cross-linked polymeric insulation material. Completed cable samples were dissassembled and insulation specimens were cut longitudinally and examined with repeated temperature cycling: ethylene—propylene copolymer rubbers and filled and unfilled crosslinked polyethylenes. The TMA results indicated that all specimes were oriented, likely incurred from extrusion processing. The facility of TMA for investigating potential anisotropic behavior from prior orientation is illustrated by examples of TMA measurements on radial specimens, similarly free of mechanical constraints.  相似文献

The recent introduction of the PhastSystem, an automatic electrophoresis and staining system with precast gradient-gels, allows rapid and reproducible analysis of proteinuria in patients suffering from renal injury. A routine method for sodium dodecyl sulfate-polyacrylamide gradient gel electrophoresis (SDS-PAGE) and silver staining of unconcentrated urine specimens in the PhastSystem is described and compared to our conventional "macro"-method with self-cast SDS-polyacrylamide gradient gels. The method described for the PhastSystem using 0.3 microL sample volumes and an 8-25% polyacrylamide gradient gel leads to highly reproducible results within 1.5 h. Before electrophoresis urine specimens were neither concentrated nor dialyzed. Samples with a protein concentration exceeding 5 mg/mL had to be diluted 1:5 (v/v). Analysis and documentation of PhastGels appeared as easy as with our conventional SDS-PAGE. Protein bands could reliably be identified by Western blotting. Urine and serum proteins, separated in PhastGels, were electrophoretically transferred to nitrocellulose and detected with specific antibodies against human albumin, transferrin, alpha-1-antitrypsin and IgG. Comparison of several standard kits for molecular weight determination revealed considerable differences concerning the quality of protein separation patterns. Availability of precast gels and automatization of SDS-PAGE and staining allows easy standardization of urine SDS-PAGE among clinical routine laboratories.  相似文献

A new approach for the detection of trimethylamine (TMA) using recombinant Xenopus laevis melanophores was developed. The cells were genetically modified to express the mouse trace amine-associated receptor 5 (mTAAR5), a G protein-coupled receptor from the olfactory epithelium, which conferred high sensitivity to TMA. A focused chemical screen allowed the discovery of additional, previously unknown stimuli of mTAAR5. The cell-based sensor demonstrated no sensitivity to trimethylamine N-oxide (TMAO), making it suitable for a convenient evaluation of TMA levels in fish tissue extracts. The developed gas measurement platform was able to detect TMA from 1 to 100 ppm within thirty-five minutes.  相似文献

A simple method is presented for the routine analysis of iodide in urine. After a one-step sample clean-up, iodide was separated by ion-pair reversed-phase liquid chromatography and detected electrochemically with a silver electrode. The coefficient of variation of a single analysis of iodide in a pooled urine sample (530 nmol/l) was 7.6%. The detection limit, derived from a signal-to-noise ratio of 3, was 3 pmol, corresponding to 0.06 mumol/l. The recovery of iodide added to urine was 96 +/- 7%. The accuracy of the method was assessed by analysing ten different samples with neutron activation analysis. The data obtained with the two methods showed a high correlation (r = 0.991) and did not differ significantly. Excretion of iodide in samples of 24-h urine from a free-living population was shown to have a log-normal distribution and to be higher in men than in women. The iodide/creatinine ratio was independent of sex and increased with age.  相似文献

A method for the direct determination of alkaloid profiling in plant tissues by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was developed. The alkaloid profiles of the herbs were obtained without the need for complicated sample preparation. Experimental results demonstrated that the direct MALDI-TOFMS analysis allowed rapid and reliable characterization of the components in plant tissues. Four commonly used Chinese medicinal herbs were studied, including Aconitum Carmichaeli Debx. (Fuzi in Chinese) and Processed Fuzi, for herb differentiation and explanation of the significant difference in their toxicities. The direct analysis method proved valuable for the preliminary study of plant component profiles. The rapid collection of information from the direct analysis on plant tissues could be valuable for supporting the discovery of new compounds and for the quality control of medicinal herbs.  相似文献

A rapid, sensitive and reliable gas chromatographic-mass spectrometric (GC-MS) screening procedure for diuretics in human urine has been developed. The procedure uses derivatisation by extractive methylation directly from the urine. The suitability of a number of phase transfer reagents and solvents were studied for the detection of sixteen diuretics. The results obtained indicate that the screening procedure employing tetrahexylammonium hydrogensulphate at pH 12 with methyl iodide in toluene at room temperature was the most effective. This method gives selectivity and sensitivities down to 0.03-0.1 microgram/ml and provides a substrate suitable for GC-MS confirmation without further manipulation. The application of the method is demonstrated by the screening of urine for bumetanide, ethacrynic acid, acetazolamide, chlorothiazide and hydrochlorothiazide.  相似文献

A matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) method for quantification of D-glucosamine-6P (GlcN-6P) that allows the kinetic study of glucosamine-6P synthase (Glms) is presented. The present report describes the optimization of the different steps of a new enzymatic assay for Glms based on in situ N-acetylation of GlcN-6P and MALDI-TOFMS analysis using N-(13C2)acetylglucosamine-6P as internal standard. Since no isotopically substituted GlcN-6P was available, the N-(13C2)acetyl derivative, easily obtained from (13C4)-acetic anhydride, was used as internal standard. Validation of the assay was achieved by measuring the fructose-6P Michaelis constant, in full agreement with reported values, and by studying the inhibition properties of arabinose-5P oxime.  相似文献

17-hydroxyprogesterone (17OHP) is the most important plasma parameter for diagnosing and monitoring congenital adrenal hyperplasia (CAH) caused by 21-hydroxylase deficiency. A rapid, simple, and specific method based on microbore high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (micro-HPLC/ESI-MS/MS) was developed to determine the presence of 17OHP on dried filter-paper blood samples from patients with CAH caused by 21-hydroxylase deficiency. 17OHP from dried blood spots formed by the action of Girard reagent P (GirP) turned out to be a water-soluble hydrazone complex. Derivatization with GirP led to higher ESI sensitivity for 17OHP. The LC/MS/MS detection of GirP-derivatized 17OHP (GirP-17OHP) was rapid (<3 min). The method is repeatable and reproducible, with CVs <7% and 12%, respectively. This new method was used for direct determination of 17OHP in dried blood specimens obtained from abnormal children and infants of various ages with a detection limit of 10 ng/mL ( approximately 12 microL blood). The method described allows for rapid and reliable measurements of 17OHP in dried blood specimens from patients affected by CAH.  相似文献

CE with indirect UV and mass-spectrometric detection was used for the simultaneous determination of arsenic acid (As(V)), arsenous acid (As(III)), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), trimethylarsine oxide (TMAO), tetramethylarsonium ion (TMA(+)), arsenobetaine (AB), and arsenocholine (AC). In the CE-indirect UV analysis, a baseline separation of arsenic species was successfully achieved by using a basic background solution (BGS) for anions and an acidic BGS for cations, respectively. The LOD values in CE-indirect UV for the individual analytes were 7.8, 12.5, 7.8, 12.5, 62.5, 125, 250, and 62.5 ppm, respectively. To achieve sensitive and selective analysis, CE coupled with ESI-MS was applied to the determination of arsenic compounds. The organic arsenic species were successfully separated with a higher sensitivity by CE-MS using the acidic BGS. The LODs in CE-MS for MMA, DMA, TMAO, TMA(+), AB, and AC were 1.0, 0.1, 0.01, 0.1, 0.01, and 0.01 ppm, respectively. In contrast, the analysis of inorganic arsenic species (As(V) and As(III)) resulted in a lower detectability in CE-MS compared to that obtained with the CE-indirect UV analysis. However, the speciation of eight arsenics by CE-MS was successfully achieved in a single run by switching the ESI polarity during MS detection.  相似文献

Studies of amine N-oxides in urine are important for the evaluation of occupational exposure to amines. These thermolabile compounds are difficult to handle by either gas or liquid chromatography, so a device for controlled thermal degradation has therefore been developed. It consists of a short precolumn with shut-off valves at both ends and an aluminum block for heating, and it was connected to the injection port of a gas chromatograph. After injection of amine N-oxides onto the precolumn and thermal degradation, the degradation products were allowed to enter the analytical column. Trimethylamine N-oxide (TMAO) and triethylamine N-oxide (TEAO) were investigated. Their thermal degradation patterns could be used for identification and quantification in aqueous solutions and in urine. Linear calibration graphs based on degradation product peaks (trimethylamine and O,N,N-trimethylhydroxylamine from TMAO and diethylamine and triethylamine from TEAO) were obtained for concentrations up to 500 ppm. Detection limits in aqueous solutions were 0.2 ppm (ca. 1 ng) for TMAO and 1 ppm for TEAO and the precisions were 6% and 9%, respectively. In urine, similar values were obtained for TEAO. The detection limit for TEAO corresponds to the expected concentration in urine after an 8-h exposure to air containing 0.8 mg/m3 of triethylamine.  相似文献

Bacteria frequently found in equine urine samples may cause degradation of 17beta-OH steroids. A simple liquid chromatography/tandem mass spectrometry (LC/MS/MS) method has been developed to evaluate the microbiological contamination of equine urine as a marker of poor storage conditions. Norethandrolone was used as the internal standard, and the linearity, sensitivity, precision and accuracy of the method were evaluated. 17beta-OH oxidation was demonstrated for testosterone, nandrolone, trenbolone and boldenone, but did not occur in alpha-epimers such as alpha-boldenone and epitestosterone, demonstrating the stereoselectivity of the reaction. A rapid test was performed by spiking one of the four 17beta-OH steroids in samples of diluted equine urine. The steroids were transformed into their respective ketones in the presence of bacterial activity. The test allows direct injection of diluted samples into the LC/MS system, without the need for prior extraction. Results show that the best method of storage is freezing at -18 degrees C. Urine specimens should be analyzed as soon as possible after thawing. This allows bacterial degradation of equine urine to be arrested temporarily, so that the urine can be used for qualitative or quantitative analysis of 17beta-OH steroids.  相似文献

Streptocidins, a family of tyrocidine-like cyclic decapeptides, are an ideal demonstration object for the detection and in situ structure analysis of natural compounds directly in microbial cells using whole cell matrix-assisted laser desorption/ionization time-of-flight-mass spectrometry (MALDI-TOFMS), an emerging technique that can be used for rapid sensitive metabolic profiling of microorganisms. Five main members of the streptocidin family (A-E) were detected in Brevibacillus cells picked from agar plates and identified by in situ structure analysis with post-source decay MALDI-TOFMS. This efficient modern method allows the precise detection of metabolites within minutes without the need to isolate and purify the target compounds. The generated mass spectra are of similar quality to those obtained for the purified peptides. In addition, surface extracts were prepared by treating Brevibacillus cells with 70% acetonitrile in the presence of 0.1% trifluoroacetic acid and fractionated by high-resolution reversed-phase high-performance liquid chromatography (HPLC). In this way ten minor streptocidins were detected demonstrating the full biosynthetic variety of streptocidin production on the cellular level. The streptocidins differ from the well-known tyrocidines essentially in position 3 of the decapeptide chain by replacement of the aromatic amino acid (F/W) found in tyrocidines by L-leucine or L-valine.  相似文献

Arsenous acid, dimethylarsinic acid (DMA), methylarsonic acid (MA), arsenic acid, arsenobetaine bromide (AB), trimethylarsine oxide (TMAO), arsenocholine iodide (AC), and tetramethylarsonium iodide (TETRA) were heated in a microwave autoclave with nitric acid to 100-300 degrees C. The arsenic compounds in the digests were separated with anion- and cation-exchange chromatography and determined with an inductively coupled plasma mass spectrometer as arsenic-specific detector. Arsenous acid was completely oxidized to arsenic acid at 100 degrees C. For a complete oxidation of MA and DMA to arsenic acid temperatures > 220 degrees C and > 280 degrees C were necessary. AB decomposed to arsenic acid via TMAO. Complete conversion was only obtained after heating the sample for 90 min to 300 degrees C. For a complete conversion of TMAO similar harsh conditions were necessary. AC was already substantially degraded to TMAO, TETRA and two unknown compounds at 100 degrees C. The unknown arsenic compounds were found only in the digests up to 160 degrees C. Quantitative conversion of AC to arsenic acid went also via TMAO. At temperatures above 220 degrees C TETRA started to convert to TMAO, which then was further converted to arsenic acid. To investigate whether the results obtained for the arsenic standards are transferable to real samples, the certified reference material DORM-2 was also heated in nitric acid with variable digestion temperatures and times. For an almost complete conversion of the AB present in DORM-2 90 min at 300 degrees C were necessary. Total organic carbon (TOC) was less < 0.2% when DORM-2 was heated at temperatures > or = 260 degrees C for 60 min. UV photo-oxidation of DORM-2 was investigated as an alternative sample decomposition. Only 6% of AB was converted to arsenic acid when DORM-2 was irradiated for 2 h at 1000 W. In contrast to microwave heating substantial amounts of MA were observed as degradation product.  相似文献

A simple and rapid method using reversed-phase liquid chromatography/tandem mass spectrometry (LC/MS/MS) for the simultaneous determination of the urinary metabolites of benzene, toluene, xylene and styrene in human urine specimens and standard solutions is described. A hybrid quadrupole/time-of-flight (QqTOF) mass spectrometer was compared for the determination of metabolite of aromatic solvents in urine samples. The metabolites selected were: trans,trans-muconic acid, hippuric acid, o-, m- and p-methylhippuric acid and phenylglyoxylic acid. The compounds were well separated from each other on narrow-bore 1-mm i.d. reversed-phase LC C-18 columns. Average recoveries for loading 100 microL of urine samples varied from 88-110% and the quantification limits were less than 30 ng/mL for each analyte (3 ng/mL for trans,trans-muconic acid). The qualitative information obtained (mass accuracy, resolution and full-scan spectra) with the QqTOF mass spectrometer allows a secure identification of analytes in biological matrices.  相似文献

A simple head-space (HS) flow injection (FI) system with chemiluminescence (CL) detection for the determination of iodide as iodine in urine is presented. The iodide is converted to iodine by potassium dichromate under stirring in the closed HS vial, and the iodine is released from urine by thermostatting and is carried in a nitrogen flow through an iodide trapping solution. The concomitant introduction of aliquots of iodine, luminol and cobalt(II) solutions by means of a time-based injector into an FI system allowed its mixing in a flow-through cell in front of the detector. The emission intensity at 425 nm was recorded as a function of time. The salting-out of the standard solutions affected the gas-liquid distribution coefficient of iodine in the HS vial. The typical analytical working graphs obtained under the optimized experimental conditions were rectilinear from 0 to 5 mg l(-1) iodine, achieving a precision of 2.3 and a relative standard deviation of 1.8 for ten replicate analyses of 50 and 200 microg l(-1) iodine. However, a second-order process becomes significant at higher iodine concentrations (from 10 to 40 mg l(-1)). The detection limit of the method is 10 microg l(-1) (80 ng) iodine when 8 ml samples are taken. Data for the iodide content of 10 urine samples were in good agreement with those obtained by a conventional catalytic method, and recoveries varied between 101 and 103% for urine samples spiked with different amounts of iodide. The analysis of one sample takes less than 20 min. In the present study the iodide levels found for 100 subjects were 86.8 +/- 19.0 (61-125) microg l(-1), which is lower than the WHO's optimal level (150-300 microg per day).  相似文献

Cation exchange and anion exchange liquid chromatography were coupled to an ICP-MS and optimised for the separation of 13 different arsenic species in body fluids (arsenite, arsenate, dimethylarsinic acid (DMAA), monomethylarsonic acid (MMAA), trimethylarsine oxide (TMAO), tetramethylarsonium ion (TMA), arsenobetaine (AsB), arsenocholine (AsC), dimethylarsinoyl ethanol (DMAE) and four common dimethylarsinoylribosides (arsenosugars). The arsenic species were determined in seaweed extracts and in the urine and blood serum of seaweed-eating sheep from Northern Scotland. The sheep eat 2-4 kg of seaweed daily which is washed ashore on the most northern Island of Orkney. The urine, blood and wool of 20 North Ronaldsay sheep and kidney, liver and muscle from 11 sheep were sampled and analysed for their arsenic species. In addition five Dorset Finn sheep, which lived entirely on grass, were used as a control group. The sheep have a body burden of approximately 45-90 mg arsenic daily. Since the metabolism of arsenic species varies with the arsenite and arsenate being the most toxic, and organoarsenic compounds such as arsenobetaine the least toxic compounds, the determination of the arsenic species in the diet and their body fluids are important. The major arsenic species in their diet are arsenoribosides. The major metabolite excreted into urine and blood is DMAA (95 +/- 4.1%) with minor amounts of MMAA, riboside X, TMA and an unidentified species. The occurrence of MMAA is assumed to be a precursor of the exposure to inorganic arsenic, since demethylation of dimethylated or trimethylated organoarsenic compounds is not known (max. MMAA concentration 259 microg/L). The concentrations in the urine (3179 +/- 2667 microg/L) and blood (44 +/- 19 microg/kg) are at least two orders of magnitude higher than the level of arsenic in the urine of the control sheep or literature levels of blood for the unexposed sheep. The tissue samples (liver: 292 +/- 99 microg/kg, kidney: 565 +/- 193 microg/kg, muscle: 680 +/- 224 microg/kg) and wool samples (10470 +/- 5690 microg/kg) show elevated levels which are also 100 times higher than the levels for the unexposed sheep.  相似文献

Bacillus subtilis synthesizes the lanthionine containing 32-amino-acid peptide antibiotic (lanti-biotic) subtilin from a ribosomally generated 56-amino-acid precursor pre-propeptide by extensive posttranslational modifications. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was used to monitor the production of matured subtilin within crude samples taken from B. subtilis culture media without prior fractionation. The processing reaction of subtilin was blocked with the serine protease inhibitor phenylmethylsulfonyl fluoride and different subtilin precursor peptides in the molecular mass range up to 6220 were observed. Two of these species were isolated by reversed-phase high-performance liquid chromatography (HPLC) and structurally analyzed by post-source decay MALDI-TOFMS. We provide evidence that the precursor species comprise the posttranslational modified C-terminal part of subtilin to which leader peptide moieties with different chain lengths are attached. These antimicrobial-inactive species could be processed to antibiotic-active subtilin by incubation with culture media of different subtilin-nonproducing B. subtilis strains as indicated by a combination of antimicrobial growth assays and MALDI-TOFMS analyses. These achievements are strong evidence for the sensitivity of MALDI-TOFMS methodology that allows straightforward investigations of analytes even in complex mixtures without time-consuming sample preparations.  相似文献

Iodide plays a vital role in many biological processes, including neurological activity and thyroid function. Due to its physiological relevance, a method for the rapid, sensitive, and selective detection of iodide in food, pharmaceutical products, and biological samples such as urine is of great importance. Herein, we demonstrate a novel and facile strategy for constructing a fluorescence turn-on sensor for iodide based on a T-Hg(II)-T complex (T=thymine). A fluorescent anthracene-thymine dyad (An-T) was synthesized, the binding of which to a mercury(II) ion lead to the formation of a An-T-Hg(II)-T-An complex, thereby quenching the fluorescent emission of this dyad. In this respect, the dyad An-T constituted a fluorescence turn-off sensor for mercury(II) ions in aqueous media. More importantly, it was found that upon addition of iodide, the mercury(II) ion was extracted from the complex due to the even stronger binding between mercury(II) ions and iodide, leading to the release of the free dyad and restoration of the fluorescence. By virtue of this fluorescence quenching and recovery process, the An-T-Hg(II)-T-An complex constitutes a fluorescence turn-on sensor for iodide with a detection limit of 126 nM. Moreover, this sensor is highly selective for iodide over other common anions, and can be used in the determination of iodide in drinking water and biological samples such as urine. This strategy may provide a new approach for sensing some other anions.  相似文献
