Electrochemical reduction of 4-methyl-4-(trichloromethyl)cyclohexa-2,5-dien-1-one

The properties of 4-methyl-4-(trichloromethyl)cyclohexa-2,5-dien-1-one which contains the trichloromethyl substituent at a quaternary carbon atom have been studied by cyclic voltammetry, rotating disk electrode voltammetry, and preparative electrolysis. Reductive dehalogenation involves initial two-electron reductive elimination of one of the geminal chlorine atoms followed by consecutive addition of proton, simultaneous elimination of the two residual halogen atoms to form carbene, and rearrangement of cyclohexadienone into the corresponding 4-methylcyclohepta-2,4,6-trien-1-one.     

Photorearrangements in spiro-conjoined cyclohexa-2,5-dien-1-one

The unexpected formation of cyclohexa-2,5-dien-1-one (6) spiro-conjoined with a dihydrobenzofuran framework, and the photochemical behavior of this compound in solution as well as in the solid state are described. The photoreaction of 6 in solution affords two rearranged products, one (7) accompanied by the enlargement of the oxygen heterocyclic ring and the other (8) accompanied by cyclopentadienone fragmentation. In the solid state, the former is the sole photoproduct of both solvated and desolvated crystals. The desolvated crystals were obtained as polycrystalline solids by thermal release of solvent molecules, and its structure was elucidated by ab initio determination from X-ray powder diffraction data followed by the Rietveld refinement.     

Bromination of 6-dibromomethyl-6-methylcyclohexyl-2,4-dien-1-one

Bromination of 6-dibromomethyl-6-methylcyclohexyl-2,4-dien-1-one (I) with molecular bromine was studied at different ratios of reactants and varied reaction time. A number of tri-, tetra-, penta-, and hexabromo derivatives was obtained, which were characterized by ¹H, ¹³C, IR, UV spectroscopy and elemental analysis. The molecular structure of (2R,3R,4S,5S,6R)-2-dibromomethyl-2-methyl-3,4,5,6-tetrabromocyclohexan-1-one (V) was revealed using XRD. It was shown that 2-bromo-6-dibromomethyl-6-methylcyclohexyl-2,4-dien-1-one (III) upon standing for six months at room temperature dimerized quantitatively via Diels-Alder reaction to form IX. Dimer IX was investigated by the methods of cyclic voltammetry (CVA) and rotating disk electrode (RDE) in a solution of DMF on glassy carbon electrode.

Folic acid analogs. III. N-(2-[2-(2,4-diarnino-6-quinazolinyl)ethyl]benzoyl)-l-glutamic acid

A trideaza analog of aminopterin, N-(4[2-(2,4-diamino-6-quinazolinyl)ethyl]benzoyl)-L-glutamic acid, was prepared by a Wittig condensation of 2,4-diaminoquinazoline-6-carboxaldehyde and [P-(N-[1,3-bis(ethoxycarbonyl)propan-1-yl]aminocarbonyl)phenylmethyl]triphenylphosphonium bromide followed by catalytic reduction and mild hydrolysis. This compound was found to have confirmed inhibitory activity against leukemia L1210 in mice.
