A new end-column electrochemiluminescence (ECL) detection technique coupling to capillary electrophoresis (CE) is characterized. A 300 microm diameter Pt working electrode was used to directly couple with a 75 microm inner diameter separation capillary without an electric field decoupler. The hydrodynamic cyclic voltammogram (CV) of Ru(bpy) 3 2+ showed that electrophoretic current did not affect the ECL reaction. The presence of high-voltage (HV) field only resulted in the shift of the ECL detection potential. The distance of capillary to electrode was an important parameter for optimizing detection performance as it determined the characteristics of mass transport toward the electrode and the actual concentration of Ru(bpy) 3 2+ in the detection region. The optimum distance of capillary to electrode was decided by the inner diameter of the capillary, too. For a 75 microm capillary, the working electrode should be placed away from the capillary outlet at a distance within the range of 220-260 microm. The effects of pH value of ECL solution and molecular structure of analytes on peak height and theoretical plate numbers were discussed. Using the 75 microm capillary, under the optimum conditions, the method provided a linear range for tripropylamine (TPA) between 1 x 10(-10) and 1 x 10(-5) mol/L with correlation coefficient of 0.998. The detection limit (signal-to-noise ratio S/N = 3) was 5.0 x 10(-11) mol/L. The relative standard deviation in peak height for eight consecutive injections was 5.6%. By this new technique lidocaine spiked in a urine sample was determined. The method exhibited the linear range for lidocaine from 5.0 x 10(-8) to 1.0 x 10(-5) mol/L with correlation efficient of 0.998. The limit of detection (S/N = 3) was 2.0 x 10(-8) mol/L.  相似文献

The novel phenomenon of cathodic electrochemiluminescence on a gold electrode in tris(bipyridine) ruthenium(II) (Ru(bpy)(3)(2+)) solution is described for the first time. A cathodic electrochemiluminescence (ECL) was found to mainly occur at 0.4-0.8 V with continuous potential scanning from 0.2-1.4 V and the ECL peak was observed around 0.68 V, which was quite different from generally reported Ru(bpy)(3)(2+) ECL. Our group speculated that Ru(bpy)(3)(2+) possibly reacts with the gold electrode in the acidic phosphate buffer solution (PBS) to generate luminescence. The possible ECL mechanism was discussed according to the presented results. Moreover, it is revealed that the Au as co-reactant in the Ru-system contributed dominantly to the whole ECL. Therefore, the reaction between Ru(bpy)(3)(2+) and the newly formed Au implied that the inert metal Au could become a promising material for ECL investigations.  相似文献

Tris(2,2'-bipyridyl) ruthenium(II) (Ru(bpy)(3)(2+))-roxithromycin based electrochemiluminescence (ECL) was enhanced greatly by gold nanoparticles 10 nm in diameter. Capillary electrophoresis (CE) was coupled with the resultant ECL system as a detector for roxithromycin. This ECL emission is explained by the coreactant mechanism where roxithromycin behaves as a coreactant to generate strong reducing species and gold nanoparticles act as "floating nanoelectrodes". The reaction of Ru(bpy)(3)(3+) with the generated strong reducing species on the Pt working electrode as well as on "floating nanoelectrodes" releases Ru(bpy)(3)(2+*), resulting in enhancement of ECL emission. The selectivity of this detection system towards roxithromycin was examined by CE. Under the optimized conditions, the intensity of ECL emission varies linearly with the concentration of roxithromycin from 24 nM to 0.24 mM. The detection limit is 8.4 nM, while without adding gold nanoparticles it is only 84 nM. The detection of roxithromycin in pharmaceutical and urine samples was also performed by the proposed CE-ECL method.  相似文献

It was found that stannous chloride (SnCl(2)), as a popular inorganic reducing reagent, could obviously enhance the electrochemiluminescence (ECL) of tris(2,2'-bipyridyl) ruthenium(II) (Ru(bpy)(3)(2+)) in aqueous solution. Some factors affecting the ECL reactions between Ru(bpy)(3)(2+) and Sn(2+), including pH, concentrations of coreactant, and electrode materials, were investigated by comparison with a classic ECL coreactant tripropylamine (TPA). The Ru(bpy)(3)(2+)-Sn(2+) ECL coreactant system produces stronger and more stable ECL signals, can keep its excellent ECL activity over a wider pH range and has more choices in using electrode materials than the Ru(bpy)(3)(2+)-TPA ECL coreactant system. The ECL mechanism of the Ru(bpy)(3)(2+)-Sn(2+) coreactant system was also studied in detail.  相似文献

A promising and highly sensitive electrochemiluminescence (ECL) sensor for the detection of Cu(2+) based on Cu(+)-catalyzed click reaction is described in this paper. Firstly, 1-azidoundecan-11-thiol was assembled on the Au electrode surface via a simple thiol-Au reaction, then the propargyl-functionalized Ru(bpy)(3)(2+)-doped SiO(2) nanoparticles (Ru-SNPs) ECL probe was covalently coupled on the electrode surfaces via click chemistry. Cu(+), the catalyst for click chemistry, is derived from the electrolytic reduction of Cu(2+)via the Bulk Electrolysis with coulometry (BE) technique and without any reductants. It is found that the ECL intensity detected from the electrode surface has a linear relationship with the logarithm of Cu(2+) concentration in the range of 1.0 × 10(-15) to 1.0 × 10(-11) M with a detection limit of 1.0 × 10(-16) M. Also, the method is highly specific even in the presence of high concentrations of other metal cations. It has been applied to detect trace Cu(2+) in complex samples (hepatoma cell) without sample treatment.  相似文献

Tris(2,3-dibromopropyl) isocyanurate (TBC) is a heterocyclic hexabrominated flame retardant, which cannot be degraded even over a long time and may be a potential environmental pollutant. In this paper, TBC is for the first time as far as we know determined by silver nitrate-enhanced electrochemiluminescence (ECL) using a gold nanoparticles (AuNPs)-modified gold electrode. In our experiments, TBC was found to have the characteristics of increasing the ECL intensity of Ru(bpy)(3)(2+), and the ECL signal was proportional to the concentration of TBC. Based on this, we have successfully developed a novel, fast and sensitive method for the analysis of TBC. The main influencing factors including the volume ratio of acetonitrile and water, and the concentration of Ru(bpy)(3)(2+) were investigated in detail. Compared with using a bare gold electrode in MeCN without AgNO(3), the limit of detection is remarkably lowered by 20 times and the linear range is expanded by 5 times by using the AuNPs-modified gold electrode and AgNO(3). Under the optimal conditions, a limit of detection of 5.0 × 10(-8) M (S/N = 3) is achieved with a linear range of 1.0 × 10(-7) to 5.0 × 10(-5) M. The mechanism of the ECL enhancement of the system is also investigated.  相似文献

A unique multilabeling at a single-site protocol of the Ru(bpy)(3)(2+) electrochemiluminescence (ECL) system is proposed. Nanoparticles (NPs) were used as assembly substrates to enrich ECL co-reactants of Ru(bpy)(3)(2+) to construct nanoscale-enhanced ECL labels. Two different kinds of NP substrates [including semiconductor NPs (CdTe) and noble metal NPs (gold)] capped with 2-(dimethylamino)ethanethiol (DMAET) [a tertiary amine derivative which is believed to be one of the most efficient of co-reactants of the Ru(bpy)(3)(2+) system] were synthesized through a simple one-pot synthesis method in aqueous media. Although both CdTe and gold NPs realized the enrichment of ECL co-reactants, they presented entirely different ECL performances as nanoscale ECL co-reactants of Ru(bpy)(3)(2+). The different effects of these two NPs on the ECL of Ru(bpy)(3)(2+) were studied. DMAET-capped CdTe NPs showed enormous signal amplification of Ru(bpy)(3)(2+) ECL, whereas DMAET-capped gold NPs showed a slight quenching effect of the ECL signal. DMAET-capped CdTe NPs can be considered to be excellent nanoscale ECL labels of the Ru(bpy)(3)(2+) system, as even a NP solution sample of 10(-18) M was still detectable after an electrostatic self-assembly concentration process. DMAET-capped CdTe NPs were further applied in the construction of aptamer-based biosensing system for proteins and encouraging results were obtained.  相似文献

This paper reports a novel detection method for DNA hybridization based on the electrochemiluminescence (ECL) of Ru(bpy)(3)(2+) with a DNA-binding intercalator as a reductant of Ru(bpy)(3)(3+). Some ECL-inducible intercalators have been screened in this study using electrochemical methods combined with a chemiluminescent technique. The double-stranded DNA intercalated by doxorubicin, daunorubicin, or 4',6-diamidino-2-phenylindole (DAPI) shows a good ECL with Ru(bpy)(3)(2+) at +1.19 V (versus Ag/AgCl), while the non-intercalated single-stranded DNA does not. In order to stabilize the self-assembled DNA molecules during ECL reaction, we constructed the ECL DNA biosensor separating the ECL working electrode with an immobilized DNA probe. A gold electrode array on a plastic plate was assembled with a thru-hole array where oligonucleotide probes were immobilized in the side wall of thru-hole array. The fabricated ECL DNA biosensor was used to detect several pathogens using ECL technique. A good specificity of single point mutations for hepatitis disease was obtained by using the DAPI-intercalated Ru(bpy)(3)(2+) ECL.  相似文献

A new water-soluble iridium(III) diimine complex with appended sugar was synthesized and characterized. The electrochemiluminescent behavior of the new complex in aqueous buffer was first studied and the ECL signal was found to be much higher than that of [Ru(bpy)(3)](2+) at a Pt working electrode. Tri-n-propylamine (TPA) and antibiotics were determined by the ECL of the iridium(III) complex in aqueous buffer at the Pt electrode and the method was found to show good sensitivity and reproducibility. The new iridium(III) complex was found to display good solubility in aqueous solution and a strong ECL signal at the Pt electrode, which might open up the possibility of its application in analysis.  相似文献

Two water soluble Re(i) tricarbonyl diimine complexes containing cationic 2,2'-bipyridyl ligands [Re(L1)(CO)(3)(AN)](2+) (1) and [Re(L2)(CO)(3)(AN)](3+) (2) (L1 and L2: a cationic 2,2'-bipyridyl ligand, AN: acetonitrile) were synthesized and characterized. Their photophysical, electrochemical and electrochemiluminescent properties were investigated. The crystal structures of the two complexes have also been determined. Electrochemiluminescence (ECL) of the two complexes 1 and 2 have been studied in aqueous buffer solution in the presence of co-reactant tri-n-propylamine (TPrA) or 2-(dibutylamino)ethanol (DBAE) at a Au working electrode. The ECL behavior of the complexes was also studied in the presence of several surfactants such as Triton X-100 and Zonyl FSN. The ECL signals of the rhenium(i) complex were enhanced about 190-fold and 70-fold at a Au electrode in the presence of Triton X-100 for the [Re(L1)(CO)(3)(AN)](2+)/TPrA and [Re(L1)(CO)(3)(AN)](2+)/DBAE systems, respectively.  相似文献

The electrochemiluminescence (ECL) of Ru(bpy)(3)(2+) at bare and single-wall carbon nanotube (SWNT) modified glassy carbon (GC) electrodes has been employed for the determination of melamine for the first time, giving a linear response (R(2) = 0.99682) for melamine concentration from 1.0 × 10(-10) to 1.0 × 10(-5) M at a bare GC electrode in pH 10 borate buffer, and the detection limit is 1.0 × 10(-10) M. However, the detection limit can be reduced further to 1.0 × 10(-13) M after modification of the GC electrode by SWNTs. This is much lower compared to other detection methods. The proposed method was applied to the determination of melamine added to a commercial milk sample; the recovery is quite satisfactory with good reproducibility and stability. All of these results provide the possibility of developing a novel ECL detection method for melamine.  相似文献

In this paper, a novel micro-electrochemiluminescent (μ-ECL) cell with volume less than 100 μl was designed and fabricated. A special configuration of the three-electrode system, in which the counter and reference electrodes were integrated on a Perspex optical window while the working electrode was placed opposite to the optical window, has greatly decreased the amount of reagents required for ECL measurements. In the μ-ECL cell, large size working electrode was still applicable, which maintained ECL detect sensitivity while consuming a small amount of sample. In addition, the immobilization of the working electrode by a special adjusting device facilitated not only the detachment of the working electrode for treatment but also accurate orientation of the electrode, consequently improved ECL reproducibility. Finally, ECL of luminol in ionic liquid 1-butyl-3-methylimidazolium hexafluorophosphate was used to evaluate this μ-ECL cell.  相似文献

采用滴涂法将氧化锌纳米颗粒滴涂在自制铅笔芯电极上制成氧化锌修饰铅笔芯电极,当以过硫酸根为共反应剂时,该修饰电极在氢氧化钠溶液中具有良好的电致化学发光(ECL)行为,对其发光机理进行了考察。基于苯酚对该修饰电极的ECL具有抑制作用,建立了一种测量苯酚的新方法,当苯酚浓度为2×10~(-8)~2×10~(-6)mol/L时,发光强度与苯酚浓度的对数呈线性关系,检出限为1×10~(-8)mol/L。该方法具有灵敏度高、方法简单、快速、稳定性好等优点,将其应用于工业废水中苯酚浓度的检测,回收率为96.5%~104.5%。  相似文献

Efficient and stable quenching of electrochemiluminescence (ECL) of tris(2,2'-bipyridine)ruthenium(II) by oxidizing ferrocene methanol (FcMeOH) at the electrode is reported. Bimolecular energy or electron transfer between Ru(bpy)(3)(2+*) and ferrocenium (Fc(+)), the oxidized species of Fc, along with suppression of radical reactions is suggested as the mechanism for quenching ECL. Fc shows more efficient quenching of ECL compared with the known quenchers phenol and 1,1-dimethyl-4,4'-bipyridine dication (MV(2+)). The ECL quenching rate constant was 5.6 x 10(10) M(-)(1) s(-)(1). Using Fc as a quencher label on a complementary DNA sequence, an intramolecular ECL quenching in hybridized oligonucleotide strands has been realized. With essentially complete quenching efficiency, this system has the potential for application to sequence-specific DNA detection.  相似文献

Electrogenerated chemiluminescence (ECL) with tris(4,7-diphenyl-1,10-phenanthrolinedisulfonic acid)rathenium(II) (RuBPS) in solution and immobilized on an electrode surface is investigated. Flow injection analysis with a thin layer electrochemical cell modified for ECL detection is used to determine the analytical utility of solution phase RuBPS and RuBPS immobilized in a cationic polypyrrole derivative. The solution phase reaction of RuBPS with oxalate is investigated with regard to the dependence of ECL emission on RuBPS concentration, carrier stream flow rate, and pH. In the parameter range studied, ECL intensity is not linear with the concentration of RuBPS in the sample. A maximum ECL intensity is observed with a RuBPS concentration of approximately 250 M. Slower linear velocities give greater ECL intensities which is the opposite of what is observed for Ru(bpy) 3 3+ and oxalate. Greater ECL intensity is observed at lower pHs for oxalate and at higher pHs for proline. RuBPS ECL with oxalate yields a working curve with a linear range from 0.1–100 M oxalate. Solution phase ECL is observed for RuBPS and other amines such as NADH, proline, tripropylamine, and antibiotics including streptomycin and gentamicin. RuBPS is also immobilized by electrochemical polymerization of 1-methyl-3-(pyrrol-1-ylmethyl)pyridinium chloride (MPP) in the presence of RuBPS. A simple, rapid and sensitive electrochemiluminescence (ECL) sensor was proposed for direct measurements of methyl parathion (MP) based on the strong affinity of a nano zirconia particles (ZrO(2) NPs) modified film on the electrode to the phosphoric group. ZrO(2) NPs, which could provide a larger absorption area to immobilize organophosphorus, was firstly modified on the glassy carbon electrode surface to prepare the proposed ECL sensor (ZrO(2)/GC). Subsequently, the ZrO(2)/GC electrode was scanned from -0.8 to +0.6 V to obtain the background signal at 0.44 V in a luminol/KCl solution. Then, a certain concentration of MP was added to an aqueous solution for 240 s, which was absorbed onto the ZrO(2)/GC electrode surface. Moreover, the MP absorbed on the surface of the ZrO(2)/GC electrode enhanced the ECL signal of luminol in the luminol/KCl solution, which increased with the concentration of MP. As a result, a novel ECL sensor was obtained in a luminol/KCl solution. The MP was determined in the range of from 3.8 × 10(-11) to 3.8 × 10(-6) mol L(-1), with a low detection limit of 1.27 × 10(-11) mol L(-1) (S/N = 3). The proposed ECL sensor performance for MP detection will open a new field in the application of rapid and screen detection of ultra-trace amounts of organ phosphorus pesticides (OPs) of vegetables used in farm markets.  相似文献

The major goal of this work was to develop a new solid-state electrochemiluminescence (ECL) detector suitable for capillary electrophoresis (CE). The detector was fabricated by coating a sol-gel derived zirconia (ZrO(2))-Nafion composite film on a graphite electrode, then the zirconia-Nafion modified electrode was immersed in tris(2,2'-bipyridyl)ruthenium(II) (Ru(bpy)(3) (2+)) solution to immobilize this active chemiluminescence reagent. The voltammetric and ECL behaviors of the detector were investigated and optimized in tripropylamine solution. The ratio of 53% for zirconia in the zirconia-Nafion composite provided the highest luminescence intensity of immobilized Ru(bpy)(3) (2+). The ECL can maintain its stability very well in the phosphate solution in the period of 5-90 h when the solid-state ECL detector was immersed in the solution all the time. The optimum distance of capillary outlet to the solid-state ECL detector has been found to be ca. 50-80 microm for a 75 microm capillary. The effects of ionic strength and pH of ECL solution on peak height were investigated. The CE with solid-state ECL detector system was successfully used to detect tripropylamine, lidocaine, and proline. The detection limits (S/N = 3) were 5 x 10(-9) mol.L(-1) for tripropylamine, 1 x 10(-8) mol.L(-1) for lidocaine and 5 x 10(-6) mol.L(-1) for proline, and the linear ranges were from 1.0 x 10(-8) to 1.0 x 10(-5) mol.L(-1) for tripropylamine, 5.0 x 10(-7) mol.L(-1) to 1.0 x 10(-5) mol.L(-1) for lidocaine and 1.0 x 10(-5) to 1.0 x 10(-3) mol.L(-1) for proline, respectively.  相似文献

In this paper we report a transparent bipolar electrode based microfluidic chip-electrochemiluminescence (ECL) system for sensitive detection of folate receptors (FR) on cell membranes. This integrated system consists of a poly(dimethylsiloxane) (PDMS) layer containing a microchannel and a glass bottom sheet with indium tin oxide (ITO) strips as bipolar detectors. The ITO strips are fabricated using a PDMS micromold with carbon ink as a protective layer in place of traditional photoresist. The configuration of the bipolar electrode has great influence on the ECL intensity of Ru(bpy)(3)(2+)/tripropylamine(TPA) system. Further studies show that folic acid (FA) can strongly inhibit the ECL of the Ru(bpy)(3)(2+)/TPA system. Based on specific recognition between FA and FR on cell membrane, this microfluidic chip-ECL system is successfully applied for detecting the level of FR on human cervical tumor (HL-60) cells and MEF cells. It is found that the ECL intensity increases with the number of HL-60 cells in the range of 21 to 3.28 × 10(4) cells/mL. The average level of FR on HL-60 cells is calculated to be 8.05 ± 0.75 × 10(-18) mol/cell. While for MEF cells, it shows a much slower ECL increment than HL-60 cells due to the much lower FR level on MEF cells (5.30 ± 0.61 × 10(-19) mol/cell). Moreover, exocytosis of FA after FR mediated endocytosis was observed according to the change of the ECL signal with the incubation time of HL-60 cells in the FA- Ru(bpy)(3)(2+)/TPA system.  相似文献

A simple, novel electrochemiluminescence (ECL) method for the detection of dichlorvos pesticide (DDVP) with high sensitivity was discovered. Detection was carried out in a static ECL system, in which a glassy carbon electrode was selected as the working electrode. ECL parameters, including the concentrations of cetyrltrimethylammonium bromide and luminol, the solution pH, and the scan rate of the applied potential, were optimized. Under these optimal conditions, the linear response of ECL-emission versus DDVP concentration was valid in the range 5-8000 ng/L (r(2)=0.9982) with a relative standard deviation of 4.3% at 2000 ng/L (n=10), yielding a detection limit (S/N=3) of 0.42 ng/L. The ECL emission was caused by a radical reaction process, in which the dissolved oxygen in the luminol solution reacted with the DDVP and generated free radicals. The free radicals reacted with the luminol anion and yielded the luminol radical. The approach presented was successfully applied to the determination of DDVP residues in vegetable samples.  相似文献

The electrogenerated chemiluminescence (ECL) behavior of N-(4-aminobutyl)-N-ethylisoluminol (ABEI) was studied and it was found that ABEI could produce emission light when oxidized at a +1.0 V (vs. Ag/AgCl) potential in alkaline solution. The addition of H2O2 markedly improved the ECL sensitivity. The pH value of the solution as well as the H2O2 concentration and working potential all have influences on the ECL response. Under optimal conditions, ABEI can be detected in the range 1.3 x 10(-6)-6.5 x 10(-12) mol L(-1). A detection limit of 2.2 x 10(-12) mol L(-1) for ABEI was obtained at a signal-to-noise ratio of 3. ABEI was then used as a marker to label a known sequence oligonucleotide, which was used as a DNA probe for identifying a target ssDNA immobilized on a PPy modified electrode based on a specific hybridization reaction. The hybridization events were evaluated by the ECL measurements. The results showed that only a complementary sequence could form a double-stranded DNA with the DNA probe and give a strong ECL response. A three-base mismatch sequence and non-complementary sequence have no response. The intensity of the ECL was linearly related to the concentration of the complementary sequence in the range 9.6 x 10(-11)-9.6 x 10(-8) mol L(-1), the detection limit was 3.0 x 10(-11) mol L(-1).  相似文献
