When 1-[2,3,5-tri-O-TBS-4α-formyl-β-D-ribo-pentofuranosyl]uracil (5) was treated with (methylene)triphenylphosphorane in THF, an unusual ring-expansion reaction occurred to give a nucleoside (7) containing dihydrooxepine ring at the sugar moiety. A deuterium-label experiment showed that one carbon unit derived from the ylide was incorporated into the 5'-position of 7. A ring cleavage between the C-3' and C-4' of 5 during the reaction was suggested.  相似文献

Six new β-carboline-type indole alkaloid glycosides, ophiorrhisides A–F (16), were isolated from Ophiorrhiza trichocarpon (Rubiaceae) collected in Thailand. Ophiorrhisides A (1) and B (2) possess a lactam function in the C ring and a unique biose residue in the molecule. Ophiorrhiside F (6) has a highly oxidized C ring with a 1,2-dicarbonyl function at C-5 and C-6 positions as well as a double bond between C-3 and C-14.  相似文献

Summary.  Reaction of a series of 21 glycosides unprotected at O-2 and featuring various configurations with DAST (diethylaminosulfur trifluoride) was monitored by 19F NMR spectroscopy. By means of the diacritical set of data (shift values and coupling constants) thus obtained for each product, identification of the operative mechanisms was possible. By correlation of these findings with stereochemical details from the structure of the educts, new parameters governing the choice of the reaction paths could be deduced. This evaluation led to the result that ring contraction after attack at C-2 of the ring oxygen and entry of the fluoride at C-1 is strongly favoured over all other possibilities. Exceptions are all derivatives of the manno series as well as all members of the trans-decalin type structure as present after 4,6-O-benzylidene acetal formation with educts having their ring substituents at C-4 and C-5 in diequatorial (trans) orientation. From these, α-D-mannopyranosides generally and of the trans-decalin types those with an additional 1,2-trans-configuration are prone to 1,2-aglycon migration and, again, entry of the fluoride at C-1. Additional pathways like alkoxy group migration, substitution under retention of configuration, or orthoester formation, are possible by participation of a suitably located neighbouring group at C-3 inasmuch as an alkoxy group interferes from an antiperiplanar orientation to the leaving group at C-2 and an acyloxy functionality attacks in a diequatorial relationship to the latter. Six new withanolide-type steroids from the leaves of Solanum cilistum   总被引:2,自引:0,他引:2  
Six new withanolide-type steroids, designated cilistols v, t, i, j, y and w (1-6, respectively), were obtained from the leaves of Solanum cilistum. Their respective structures were characterized by spectroscopic means as follows: Cilistol v (1) was (22R,24Z)-1alpha,3beta,22,26-tetrahydroxyergost-5,24-diene 26-O-beta-D-glucopyranoside, which is regarded as the precursor of other withanolide-type steroids included in this title plant. Cilistol t (2) was (22R,24S,25R,26S)-24,25;22,26-diepoxy-1alpha,3beta,26-trihydroxyergost-5-ene 26-O-beta-D-glucopyranoside, and cilistols j (3) and i (4) corresponded to the substances probably formed by the cleavage of the epoxy ring at C-24 and 25 of 2. Cilistol y (5) was 3-O-sulphonyl (22R,24S,25R,26R)-1-oxo-24,25; 22,26-diepoxy-3beta,17alpha,26-trihydroxyergost-5-ene 26-O-beta-D-glucopyranoside, and cilistol w (6) corresponded to the substance obtained by the fission of the epoxy ring at C-24 and 25 of 5. The occurrence of these withanolide type steroids from Solanum genera is rare and worthy of note.  相似文献

New taxane diterpenoids from the roots of Taiwanese Taxus mairei   总被引:2,自引:0,他引:2  
Five new taxane diterpenoids, taxumairols G (1), H (2), I (3), J (4), and L (5) were isolated from extracts of the roots of Taiwanese Taxus mairei (LEMEE & LEVL.) S. Y. Hu. Compounds 1-4 belong to the new 11(15-->1)-abeo-taxene system, having a tetrahydrofuran ring along carbons C-2, C-3, C-4 and C-20. Compounds 3 and 4 contain an isopropenyl group at C-1 while compounds 1 and 2 are attached with a benzoxyl group at C-15. The structures of compounds 1-5 were determined on the basis of two-dimensional (2D)-NMR techniques including correlation spectroscopy (COSY), heteronuclear single quantum coherence (HSQC), heteronuclear multiple bond correlation (HMBC), and nuclear Overhauser effect spectroscopy (NOESY) experiments.  相似文献

Bridgehead lithiations have successfully been carried out on substrates derived from catechinic acid, which possess the core bicyclo[3.3.1]nonane-1,3,5-trione structure present in garsubellin A. Using an external quench method, various electrophiles have been incorporated at the C-5 bridgehead position in a one-step process that appears to be sensitive to the substitution pattern on the bicyclic system. Regioselective lithiation at the C-3 sp(2) centre was achieved by changing the base used from LDA to LTMP. Following the introduction of a prenyl substituent by bridgehead substitution, annulation of a THF ring, analogous to that in garsubellin A, was possible via an epoxidation-ring opening sequence. Oxidative modification of the catechol substituent of the catechinic acid core was possible to give systems with muconic acid, ortho-quinone or furan 2-carboxylic acid side chains.  相似文献

Clarithromycin (6-O-methylerythromycin A) is a 14-membered macrolide antibiotic which is active in vitro against clinically important gram-positive and gram-negative bacteria. The selectivity of the methylation of the C-6 OH group is studied on erythromycin A derivatives. To understand the effect of the solvent on the methylation process, detailed molecular dynamics (MD) simulations are performed in pure DMSO, pure THF and DMSO:THF (1:1) mixture by using the anions at the C-6, C-11 and C-12 positions of 2',4"-[O-bis(TMS)]erythromycin A 9-[O-(dimethylthexylsilyl)oxime] under the assumption that the anions are stable on the sub-nanosecond time scale. The conformations of the anions are not affected by the presence of the solvent mixture. The radial distribution functions are computed for the distribution of different solvent molecules around the 'O-' of the anions. At distances shorter than 5 A, DMSO molecules are found to cluster around the C-11 anion, whereas the anion at the C-12 position is surrounded by the THF molecules. The anion at the C-6 position is not blocked by the solvent molecules. The results are consistent with the experimental finding that the methylation yield at the latter position is increased in the presence of a DMSO:THF (1:1) solvent mixture. Thus, the effect of the solvent in enhancing the yield during the synthesis is not by changing the conformational properties of the anions, but rather by creating a suitable environment for methylation at the C-6 position.  相似文献

C-2-unsubstituted imidazoline-3-oxides were reduced with NaBH4 in THF to give the corresponding trans-3,5-diarylimidazolidin-1-ols, while under the same conditions C-2-substituted derivatives gave the corresponding ring–chain–ring tautomers. Treatment of the crude reaction mixture from the reduction of C-2-unsubstituted imidazoline-3-oxides with a MeOH–H2O mixture provided reductive C–N bond cleavage to give hydroxylamines, while under the same conditions ring–chain–ring tautomers remained unchanged.  相似文献

The wood of Anodendron paniculatum contains a large number of cardenolides; 14 of such compounds could be spottet in PC. and TLC. with Kedde reagent. Five of these (A, E 1, E 2, F, and G) could be isolated in crystalline form and two other ones (B 1 and B 2) as an amorphous mixture. A crystalline aromatic ketone (compound α) was obtained as by-product. The isolated cardenolides, named as anodendrosides A-G, were found to be glycosides containing unusual sugars. According to UV. and IR. spectra the genin portion in anodendroside E 1 (probably also in F and B 2) contains a cardenolide structure with saturated ring D and normal butenolide side-chain, while the anodendrosides A, E 2 and G (probably also B 1) contain an additional double bond at C-16. The four compounds for which IR. spectra could be taken (A, E 1, E 2, and G) possess a keto group.  相似文献

The recombination kinetics of a series of flavonoids with stable DPPH radicals are studied. Flavonols are the most reactive. Polarization of the carbonyl in flavones reduces the reactivity. Substitution on C-7 and the B ring has practically no effect on the reactivity. Glycosylation of C-3 reduces the reactivity if the sugar can form two intramolecular H-bonds. The reactivity of the flavonoid phenol hydroxyls, with the exception of the hydroxyl on C-5, is proportional to their number. The most promising antioxidants are lespedin, kaempferitrin, kaempferol, kaempferol-7-rhamnoside, kaempferol-3-robinobioside, and robinin.I. G. In addition to 19-dydroxybaccatin III, 1beta-hydroxy-5 alpha-deacetylbaccatin I, taxayuntin G and 13-O-deacetyltaxumairol Z (4), three new taxane diterpenoids, taxumairols X (1), Y (2), Z (3) have been isolated from extracts of the Formosan Taxus mairei (LEMEE & LEVL.) S. Y. HU. Compounds 1-2 belong to the 11(15-->1)-abeo-taxane system, having a tetrahydrofuran ring at C-2, C-3, C-4 and C-20. The new compound 3 and 4, which was misidentified previously are derivatives of 11(15-->1)-abeo-taxane with an intact oxirane system. The structures of compounds 1-4 were elucidated on the basis of extensive two dimensional (2D)-NMR analysis.  相似文献

Using 500 MHz NMR, we have carried out a stable ion protonation and model nitration study of the methoxy-substituted hydrocarbon 6, its 15-ol 7, and the dimer 10, in order to evaluate OMe substituent effects on directing electrophilic attack and on charge delocalization mode/conformational aspects in the resulting carbocations. It is found that the C-11 methoxy group directs the electrophilic attack to C-12 and C-14. Thus protonation of 6 with FSO(3)H/SO(2)ClF gives a 4:1 mixture of monoarenium ions 6H(+)()/6aH(+)(). Prolonged reaction times and increased temperature induced fluorosulfonylation at C-14 (6(+)-SO(2)()F), whereas ambient nitration with NO(2)(+)BF(4)(-) occurred at C-12. The 15-ol derivative 7 is cleanly ionized to 11(+)(), providing the first example of an alpha-phenanthrene-substituted carbocation from phenanthrene C-1 position. Contrasting behavior of the D-ring methyl-substituted 9 and the C-11 methoxy-substituted 10 dimers is remarkable in that unlike 9 which is readily cleaved to produce the monomeric arenium ion 3H(+)(), 10 is diprotonated at the two C-12 sites and at C-12/C-14 in each unit. The latter dication-dimer exists as a mixture of diastereomers. Reactivity of 7 underscores the importance of 11(+)(). Attack at the C-14 ring junction is in concert with the proposal that electrophilic oxygen would attack at C-14/C-15 (epoxidation) followed by ring opening to give the biologically active 15-ol as a major metabolite.  相似文献

Thirty three C19-diterpenoid alkaloids, twenty-two prepared from known C19-diterpenoid alkaloids and eleven isolated from Aconitum and Delphinium spp. were evaluated for their cardiac activity in the isolated bullfrog heart assay. Among them, eleven compounds exhibited cardiac activity, with average rate of amplitude increase in the range of 16-118%. Compound 7, mesaconine (17), hypaconine (25), and beiwutinine (26) exhibited strong cardiac activities relative to the reference drug. The structure-activity relationship data acquired indicated that an alpha-hydroxyl group at C-15, a hydroxyl group at C-8, an alpha-methoxyl or hydroxyl group at C-1, and a secondary amine or N-methyl group in ring A are important structure features necessary for the cardiac activities of the aconitine-type C19-diterpenoid alkaloids without any ester groups. In addition, an alpha-hydroxyl group at C-3 is also helpful for the cardiac activity of these alkaloids.  相似文献

Armatol F, isolated from the red alga Chondria armata as a polyether triterpene, has a fused tricyclic ether moiety (BCD-ring) with an unusual cis ring junction at C18-C19 between the C- and D-rings. En route to the total synthesis of armatol F, the stereoselective construction of the C18 and C19 stereocenters by Ireland-Claisen rearrangement and the formation of the C-ring by relay ring-closing olefin metathesis were established through the synthesis of monocyclic (C-ring) and bicyclic (BC-ring) model compounds.  相似文献

The synthesis of several ABE tricyclic analogues of the alkaloid methyllycaconitine 1 is reported. The analogues contain two key pharmacophores: a homocholine motif formed from a tertiary N-ethyl amine in a 3-azabicyclo[3.3.1]nonane ring system and a 2-(3-methyl-2,5-dioxopyrrolidin-1-ly)benzoate ester 4. The synthesis of the ABE tricyclic analogues of MLA 1 began with selective allylation at C-3 of 3 to produce allyl beta-keto ester 4. Double Mannich reaction of 4 with ethylamine and formaldehyde produced bicyclic amine 5 The C-9 ketone of bicyclic amine 5 was selectively reduced to form bicyclic alcohols 6 and 7 which were subsequently allylated to form dienes 8 and 9. Ring closing metathesis of dienes 8 and 9 afforded tricyclic ethers 11 and 12, respectively, the C-8 ester of which was reduced to a hydroxymethyl group to form ABE tricyclic analogues 13 and 14. Addition of allylmagnesium bromide to the C-9 ketone of 20 afforded dienes 21 and 22, which underwent ring closing metathesis to form tricyclic esters 23 and 24, respectively. Reduction of the C-8 ethyl ester of 23 and 24 to a hydroxymethyl group afforded diols 25 and 26 respectively. The 2-(3-methyl-2,5-dioxopyrrolin-1-ly)benzoate ester was introduced by conversion of alcohols 13, 14, 25 and 26, to the anthranilate esters 16, 17, 27 and 28 using N-(trifluoroacetyl)anthranilic acid 15 followed by fusion with methylsuccinic anhydride to afford the substituted anthranilates 18, 19, 29 and 30 containing the key 2-(3-methyl-2,5-dioxopyrrolidin-1-ly)benzoate ester pharmacophore.  相似文献

The total synthesis of protein kinase C activator (-)-indolactam V (IL-V) has been successfully completed with two separate approaches: From known 4-nitrotryptophan derivative 3 in 8 steps (49% overall yield) and from L-glutamic acid in 12 steps (18% overall yield), where 4-nitrotryptophanol derivative 4 served as a key intermediate. Derivatives 3 and 4, both incorporating indole 4-substitution and the C-9 stereocenter in IL-V, were synthesized via the Pd-catalyzed indole synthesis from 3-nitro-2-iodoaniline 5 with aldehydes 6 and 7, respectively. Aldehyde 7 was, meanwhile, synthesized from l-glutamic acid in 5 steps (68% yield). Lactamization of the 9-membered ring was achieved using HATU in THF in good yield.  相似文献

2-丙烯酰胺基-2-甲基丙磺酸(AMPS)与甲基丙烯酸-2-羟基丙酯(HPM)在乙二醇/水(1:1,质量比)中70℃下进行共聚,AMPS/HPM(8:2,摩尔比)为该体系的恒比共聚点.在此组成加入交联剂N,N'-亚甲基双丙烯酰胺2%、3%和5%(摩尔分数)制备了凝胶试样GO2、GO3、GO5.它们在DMSO/THF混合溶剂中THF达55%~60%(体积百分数)时发生体积相变;在乙醇/THF混合溶剂中GO3的体积随THF的加入连续缩小,但不出现体积相变.此现象可用高分子链溶剂化层的变化以及低极性介质中离子对之间的偶极-偶极相互作用来说明.  相似文献

Rhodopsin, the pigment responsible for vision in animals, insect and fish is a typical G protein (guanyl-nucleotide binding protein) consisting of seven transmembrane alpha helices and their interconnecting extramembrane loops. In the case of bovine rhodopsin, the best studied of the visual pigments, the chromophore is 11-cis retinal attached to the terminal amino group of Lys296 through a protonated Schiff base linkage. Photoaffinity labeling with a 3-diazo-4-oxo-retinoid shows that C-3 of the ionone ring moiety is close to Trp265 in helix F (VI) in dark inactivated rhodopsin. Irradiation causes a cis to trans isomerization of the 11-cis double bond giving rise to the highly strained intermediate bathorhodopsin. This undergoes a series of thermal relaxation through lumi-, meta-I and meta-II intermediates after which the retinal chromophore is expelled from the opsin binding pocket. Photoaffinity labeling performed with 3-diazo-4-oxoretinal at -196 degrees C for batho-, -80 degrees C for lumi-, -40 degrees C for meta-I, and 0 degrees C for meta-II rhodopsin showed that in bathorhodopsin the ring is still close to Trp265. However, in lumi-, meta-I and meta-II intermediates crosslinking occurs unexpectedly at A169 in helix D (IV). This shows that large movements in the helical arrangements and a flip over of the ring moiety accompanies the transduction (or bleaching) process. These changes in retinal/opsin interactions are necessarily accompanied by movements of the extramembrane loops, which in turn lead to activation of the G protein residing in the cytoplasmic side. Of the numerous G protein coupled receptors, this is the first time that the outline of transduction pathway has been clarified.  相似文献
