Aldehydes are considered potential markers for enhanced oxidative stress and have been proposed as a diagnostic measure of cancer status. Do to their volatility and activity, it is very difficult to accurately measure aldehydes in human blood. In the present work, gas chromatography/mass spectrometry (GC/MS) and solid-phase microextraction (SPME) with on-fiber derivatization was developed for determination of aldehydes in human blood. O-(2,3,4,5,6-Pentafluorobenzyl)hydroxylamine hydrochloride (PFBHA) in aqueous solution was first adsorbed by a SPME fiber, and then the aldehydes in blood samples were headspace extracted by the SPME fiber and rapidly derivatized with PFBHA on the SPME fiber. Finally, the oximes formed were desorbed and detected by GC/MS in electron ionization (EI) mode. Validation of the present method was carried out, and the method was applied to quantitative analysis of the aldehydes in lung cancer blood. The results demonstrated that GC/MS and SPME with on-fiber derivatization is a simple, rapid, sensitive and solvent-free method for the determination of aldehydes in human blood.  相似文献

The simultaneous determination of several odorous trichlorobromophenols in water has been carried out by an in-sample derivatization headspace solid-phase microextraction method (HS-SPME).The analytical procedure involved their derivatization to methyl ethers with dimethyl sulfate/NaOH and further HS-SPME and gas chromatography-mass spectrometry (GC/MS) determination. Parameters affecting both the derivatization efficiency and headspace SPME procedures, such as the selection of the SPME fiber coating, derivatization–extraction time and temperature, were studied. The commercially available polydimethylsiloxane (PDMS) 100 μm and Carboxen-polydimethylsiloxane-divinylbenzene (CAR-PDMS-DVB) fibers appeared to be the most suitable for the simultaneous determination of these compounds. The precision of the HS-SPME/GC/MS method gave good relative standard deviations (RSDs) run-to-run between 9% and 19% for most of them, except for 2,5-diCl-6-Br-phenol, 2,6-diCl-3-Br-phenol and-2,3,6-triBr-phenol (22%, 25% and 23%, respectively). The method was linear over two orders of magnitude, and detection limits were compound dependent but ranged from 0.22 ng/l to 0.95 ng/l. The results obtained for water samples using the proposed SPME procedure were compared with those found with the EPA 625 method, and good agreement was achieved. Therefore, the in-sample derivatization HS-SPME/GC/MS procedure here proposed is a suitable method for the simultaneous determination of odorous trichlorobromophenols in water.  相似文献

The solid-phase microextraction (SPME) technique with on-fiber derivatization was evaluated for the analysis of aldehydes in water. The poly(dimethylsiloxane)/divinylbenzene (PDMS/DVB) fiber was used and O-2,3,4,5,6-(pentafluorobenzyl)hydroxylamine hydrochloride (PFBHA) were first loaded onto the fiber. The aldehydes in water sample were agitated into headspace and extracted by SPME with on-fiber derivatization. Gas chromatography/mass spectrometry (GC/MS) was used for the analysis of oximes formed and the adsorption-time profiles were examined. The precision, recovery and method detection limits (MDLs) were evaluated with spiked bidistilled water, chlorinated tap water as well as well water. The relative standard deviations from different spiked water sample were all less than 10% and the recoveries were 100 +/- 15%. With 2 ml of water sample, MDLs were in the range of 0.12-0.34 microg/l. Compared with other techniques, the study shown here provided a simple, fast and reliable method for the analysis of aldehydes in water.  相似文献

Abstract

The application of single drop extraction (SDE) for isolation and enrichment of carbonyl compounds after derivatization with O‐(2,3,4,5,6‐pentafluorobenzyl)hydroxylamine in spirits and vodkas is discussed. The optimal parameters (extraction volume, drop volume, content of ethyl alcohol, sample volume, temperature and time of extraction) for isolation and preconcentration of C1–C6 aldehydes from alcoholic matrices were established. The developed SDE‐gas chromatography (GC)‐electron capture detection (ECD), an extraction method, allows the determination of low molecular aldehydes at level lower than 1 µg dm?3. The overall analysis time without derivatization is 35 minutes. The procedure was applied for the determination of aldehydes in real alcoholic beverages (vodkas). The simplicity and cost‐effectiveness of the proposed procedure makes it a good alternative to solid phase microextraction (SPME) and other more labor‐intensive methods.  相似文献

The use of two modes for mass spectrometry (MS) detection with an ion trap instrument, selected ion storage (SIS) and tandem mass spectrometry (MS/MS), are compared for the solid-phase microextraction (SPME)–gas chromatography (GC) coupled to mass spectrometry (GC-MS) determination of 16 priority organochlorine pesticides (OCPs) in drinking water samples at the ultratrace levels (ng?L?1) required by official guidelines in the European legislation. Experimental parameters investigated for the SPME sample preparation were: the type of coating (100?µm polydimethylsiloxane, PDMS, and 65?µm poly(dimethylsiloxane)–divinylbenzene, PDMS/DVB), SPME modality, extraction and desorption times and desorption temperature and the methanol percentage in the SPME working solution. Under the calculated optimal conditions two methodologies were developed, one for SIS and the other for MS/MS modes. The detection limits, precision and accuracy were evaluated for both alternatives and were appropriate to the official guidelines requirements. The SPME–GC-MS(SIS) methodology offered LODs from 0.2–6.6?ng?L?1, precision below 13% and recoveries between 83 and 110%. The SPME–GC–MS/MS methodology provided limits of detection (LODs) ranging from 0.3 to 7.6 ng?L?1, % RSD were ≤14% and recoveries of 79–108% were achieved. After the results observed within an Interlaboratory Exercise, the latest MS methodology was selected for the pursued analysis in real drinking water samples. Also, the good results in this round-robin exercise validate the proposed SPME–GC–MS/MS methodology.  相似文献

This study focused on a comparison of three different dynamic hollow fiber-based liquid-phase microextraction (DHF-LPME) methods for extraction and preconcentration of parabens from wastewater, toothpaste, cream, and shampoo samples. The first method is two-phase DHF-LPME, in which n-octanol was used as the extraction solvent. The second is three-phase DHF-LPME, in which n-octanol and basic aqueous solution were used as the extraction solvent and acceptor phase, respectively. High-performance liquid chromatography with UV detection (HPLC–UV) was applied for determination of the parabens in both methods. The third method is a recently introduced method; three-phase DHF-LPME based on two immiscible organic solvents (n-dodecane as organic solvent and acetonitrile as an acceptor phase). The quantitative analyses were performed by the use of gas chromatography-mass spectrometry (GC–MS) after injection port derivatization. The effect of different extraction conditions (i.e., extraction solvent, pH, ionic strength, stirring rate, and dynamic and extraction times) on the extraction efficiency of the parabens was investigated and optimized. All the three procedures provide similar working parameters characterized by high repeatability (3.9–6.3 %) and good linearity (correlation coefficient ranging from 0.989 to 0.998). Results of real sample analyses obtained by these three methods were highly correlated. Although all methods provide compatible alternatives for paraben analysis, the three-phase DHF-LPME based on two immiscible organic solvents may be a more appropriate technique due to its higher extraction efficiency and thus lower limits of detection (LODs). LODs for all the parabens ranged from 0.2 to 5.0 μg L?1 using the two first methods combined with HPLC–UV. An improvement in sensitivity of several orders of magnitude was achieved using three-phase DHF-LPME based on two immiscible organic solvents followed by single-ion monitoring GC–MS analyses (0.01–0.2 μg L?1) due to compatibility of this technique with GC instrument.  相似文献

A headspace solid‐phase microextraction (HS‐SPME) method coupled to GC‐MS was developed in order to determine trace levels of tetramethyltin (TeMT) and inorganic tin (iSn) after ethylation to tetraethyltin (TeET) in various matrices. The derivatization of iSn and the extraction of both TeMT and iSn as TeET were performed in one step. Sodium tetraethylborate (NaBEt4) was used as derivatization agent and the volatile derivatives were absorbed on a PDMS‐coated fused silica fiber. The conditions for the HS‐SPME procedure were optimized in order to gain in repeatability and sensitivity. Several critical parameters of GC‐MS were also studied. The detection of TeMT and iSn as TeET peaks was performed by the SIM mode. The precision of the proposed method is satisfactory providing RSD values below 10% for both tin species and good linearity up to 10 μg/L. The developed method was successfully applied to the determination of tin species in several samples like canned fish, fish tissues, aquatic plants, canned mineral water and sea water. The proposed HS‐SPME‐GC‐MS method was proved suitable to monitor the concentration levels of toxic tin compounds in environmental and biological samples.  相似文献

A headspace solid phase microextraction (HS-SPME) method coupled with gas chromatography and MS detection (GC/MS) was optimized for the simultaneous determination of 21 target Pharmaceuticals and Personal Care Products (PPCPs) in water samples. The analytes included fragrances, UV-filters, antiseptics, estrogens, anti-inflammatory drugs, and pesticides. An on-fiber SPME derivatization, using silyl reagents, was performed for the analysis of more polar acidic compounds. An experimental design approach was applied to systematically investigate and optimize the operative parameters affecting the extraction recovery, namely: extraction temperature and time, derivatization time, desorption temperature and time. The optimum operating conditions were: extraction time of 125?min at a temperature of 40?°C; derivatization time of 30.5?min; desorption time of 2?min at a temperature of 300?°C. Under these conditions, good reproducibility was assessed as RDS% values ≤10% for underivatized PPCPs and ≤20% for derivatized compounds. The method detection limits (LOD) were between 0.7 and 9.0?ng?L?1, with the highest values in the range 2.5–9.0?ng?L?1 for the derivatized analytes. Method accuracy was evaluated on spiked tap water samples: recoveries varied from 85 to 103% and from 75 to 110% for non-derivatized and derivatized compounds, respectively.  相似文献

A GC/MS-MS method for the determination of Hg(II) and alkylated Hg, Pb, and Sn species in human urine is described. Separation and identification of the metal species are performed by capillary gas chromatography coupled with an ion-trap mass spectrometer with electron impact ionization in the tandem-MS mode. For sample preparation a very promising technique was applied that is based on a derivatization with sodium tetraethylborate followed by headspace solid phase microextraction (SPME). Operation of the used ion trap in the tandem-MS mode yields in improved detection limits because of a signal-to-noise ratio that is at least one order of magnitude better than in the MS mode. The detection limits in real matrices like urine are between 7 and 22 ng/L for all species investigated. Urinary levels of inorganic Hg in non-occupationally exposed persons with and without dental amalgam were found to be between 0.1 and 1.4 μg/L. A reference material (“ClinRep, Level I”) was used for quality assurance. Compared to the coupling of GC with ICP-MS (“inorganic” MS), the advantage of the proposed method using an “organic” MS is that (i) the species can be directly identified via their precursor and daughter ions and (ii) analysis can be performed with a commercially available hyphenated technique at moderate costs and needs no lab-made interfacing. Moreover, it offers a real multi-element/multi-species capability with low detection limits and a minimum of sample preparation.  相似文献

A rapid method for the analysis of biomass feedstocks was established to identify the quality of the pyrolysis products likely to impact on bio-oil production. A total of 15 Lolium and Festuca grasses known to exhibit a range of Klason lignin contents were analysed by pyroprobe–GC/MS (Py–GC/MS) to determine the composition of the thermal degradation products of lignin. The identification of key marker compounds which are the derivatives of the three major lignin subunits (G, H, and S) allowed pyroprobe-GC/MS to be statistically correlated to the Klason lignin content of the biomass using the partial least-square method to produce a calibration model. Data from this multivariate modelling procedure was then applied to identify likely “key marker” ions representative of the lignin subunits from the mass spectral data. The combined total abundance of the identified key markers for the lignin subunits exhibited a linear relationship with the Klason lignin content. In addition the effect of alkali metal concentration on optimum pyrolysis characteristics was also examined. Washing of the grass samples removed approximately 70% of the metals and changed the characteristics of the thermal degradation process and products. Overall the data indicate that both the organic and inorganic specification of the biofuel impacts on the pyrolysis process and that pyroprobe–GC/MS is a suitable analytical technique to asses lignin composition.  相似文献

In this study, a simple, rapid, and sensitive method was developed and validated for the quantification of valproic acid (VPA), an antiepileptic drug, in human plasma, which was based on water-phase derivatization followed by headspace solid-phase microextraction (HS-SPME) and gas chromatography/mass spectrometry (GC/MS). In the proposed method, VPA in plasma was rapidly derivatized with a mixture of isobutyl chloroformate, ethanol and pyridine under mild conditions (room temperature, aqueous medium), and the VPA ethyl ester formed was headspace-extracted and simultaneously concentrated using the SPME technique. Finally, the analyte extracted on SPME fiber was analyzed by GC/MS. The experimental parameters and method validations were studied. The optimal conditions were obtained: PDMS fiber, stirring rate of 1100 rpm, sample temperature of 80 degrees C, extraction time of 20 min, NaCl concentration of 30%. The proposed method had a limit of quantification (0.3 microg/mL), good recovery (89-97%) and precision (RSD value less than 10%). Because the proposed method combined a rapid water-phase derivatization with a fast, simple and solvent-free sample extraction and concentration technique of SPME, the sample preparation time was less than 25 min. This much shortens the whole analysis time of VPA in plasma. The validated method has been successfully used to analyze VPA in human plasma samples for application in pharmacokinetic studies. All these results show that water-phase derivatization followed by HS-SPME and GC/MS is an alternative and powerful method for fast determination of VPA in biological fluids.  相似文献

This contribution presents a research aimed at the study of the commercial products Laropal K80, Keton N and MS2A, ascribed to ketone resins prepared as thin films, in an attempt to reproduce the pictorial layers and protective finishes that are commonplace in contemporary paintings. For this purpose, a new method based on “online” silylation–pyrolysis using hexamethyldisilazane as a derivatisation reagent in pyrolysis–gas chromatography–mass spectrometry (Py–GC–MS) has been proposed. This procedure leads to the unambiguous identification of such varnishes and improves direct Py–GC–MS. Chemical changes due to the degradation effect of environmental agents have been especially considered. To that end, three different accelerated ageing processes were applied to a series of specimens prepared from the studied commercial products: thermal, UV light and ageing in an SO2-polluted chamber. Chemical changes due to UV light ageing of Keton N resin are in good agreement with those previously reported in the literature.  相似文献

This critical overview covers current analytical methods and future developments in quantitative determination of fatty acids (FAs), emphasizing sample extraction, derivatization and instrumental analysis with gas chromatography/mass spectrometry (GC/MS) and high-performance liquid chromatography/tandem mass spectrometry (HPLC/MS2). We compare the benefits and the drawbacks of these two analytical techniques.We consider the well-established GC/MS method with pre-derivatization to be a traditional technique in terms of highly standardized sample-preparation procedures, affordability and readily available library searching for compound identification. However, the complicated derivatization steps required prior to instrumental analysis with GC/MS take a long time, with loss and transformation of FAs, low recovery and poor reproducibility.HPLC/MS2 without derivatization shows the benefits of simple, mild sample-processing conditions, satisfactory recovery, short running time and high selectivity and sensitivity, which may allow it to become a viable alternative to GC/MS for the analysis of FAs in the years ahead.  相似文献

In order to identify the most suitable method for the estimation of nanosulfur for studying its residue dynamics, the present work was taken up. HPLC and GC methods were explored for its analysis. A comparative study of the existing analytical methods for the quality control of nanosulfur was undertaken. UV spectrophotometry and HPLC methods were superior with lower LOD when compared to GC–MS, which was not satisfactory due to breakage of catenated S20 into S6 and S8. The method has been validated by analyzing various nanosulfur formulations of known concentrations. The recovery of the UV and HPLC methods ranged from 80.71 to 109.51% and 82.31 to 109.84%, respectively. The LOD of UV, GC–MS, and HPLC is 4, 20, and 1 ppm, respectively. The retention time of sulfur was 13.77 (HPLC), 2.89 (ultra high performance liquid chromatography), and 12.715 + 21.524 min (GC–MS). The method was successfully utilized for estimating sulfur in natural samples such as water from a sulfur hot spring and wastewater. The method has been validated by following the method recommended by the American Society for Testing and Materials. The HPLC method emerged as the best analytical method for the estimation of elemental sulfur.  相似文献

A method for the extraction and determination of methylmercury (MeHg) in solid matrices is presented. Combining the advantages of two extraction techniques—subcritical water extraction (subWE) and solid‐phase microextraction (SPME)—selective separation of MeHg from soils is possible. The procedure is based on extraction with subcritical water without using organic solvents, followed by in situ aqueous‐phase derivatization with sodium tetraethylborate and headspace SPME with a silica fiber coated with poly(dimethylsiloxane). The optimization of the extraction parameters is described. The identification and quantification of the extracted alkylmercury compounds from spiked soil samples is performed by GC–MS after thermal desorption. Solid-phase microextraction (SPME) coupled with gas chromatography–mass spectrometry (GC–MS) and selected ion monitoring (SIM) was used for the analytical determination of priority pesticide residues. Fibers coated with a 65-µm film thickness of polydimethylsiloxane divinylbenzene (PDMS-DVB) were used to extract 31 pesticides of different chemical groups. The quality parameters of the method demonstrated a good precision with detection limits of 1–56?ng/L. Linearity was controlled in the range of 0.1–50?µg/L. The proposed method was applied for the trace-level determination of the target pesticides in surface water samples including three rivers and one lake at the Epirus region (north-west Greece) for a period of one year. The results demonstrate the suitability of the SPME–GC–MS approach for the analysis of multi-residue pesticides in environmental water samples.  相似文献

This article introduces a simple, rapid, and reliable solid‐phase microextraction (SPME) method coupled with GC‐MS for the quantitative determination of 16 polycyclic aromatic hydrocarbons in water. In this study, the Taguchi experimental design was used to optimize extraction conditions of polycyclic aromatic hydrocarbons using SPME method to obtain highly enriched analytes. Consequently, quantitative determination of polycyclic aromatic hydrocarbons in water was achieved by GC‐MS technique. The selected parameters affecting enrichment of polycyclic aromatic hydrocarbons were sample extraction time, stirring speed, temperature, ionic strength, and pH. The study revealed that optimal operating conditions were found to be 90‐min extraction time, 1400 rpm stirring speed, and 60°C sample temperature. The effect of ionic strength and pH were shown to be insignificant. Optimized conditions were also reevaluated by placing the 16 polycyclic aromatic hydrocarbons into several subgroups based on their molecular weight. The extraction efficiency of polycyclic aromatic hydrocarbons with low molecular weight was shown to be a function of only the extracting temperature. Satisfactory results were obtained for linearity (0.983–0.999), detection limits (2.67–18.02 ng/L), accuracy (71.2–99.3%), and precision (4.3–13.5%). The optimum conditions reported by other design approaches were evaluated and generalized optimum conditions were suggested.  相似文献

A GC–MS method is proposed for the analysis of camphor and menthol in “over-the-counter” (OTC) products. Sample preparation was achieved by head-space solid phase microextraction using a polydimethylsiloxane fibre. GC analysis was performed using a Phenomenex ZB-5 column and a temperature program was adopted. The method was validated by studying linearity (range 0.1–15.0% w/w), accuracy, reproducibility and sensitivity. Applications were directed to the determination of the active ingredients in four different OTC-products; the mean recoveries were in the ranges 91.30–99.74% and 94.34–102.89% for camphor and menthol, respectively and the LOD was in the order of 0.005% (w/w). Other important terpenoids (α- and β-pinene, 1,8-cineole, menthone, isomenthone, α-terpineole, t-cinnamaldehyde, eugenole) were identified by mass spectra and Kovats retention indices and quantified by peak area normalization to 100%. Further information for a comprehensive characterization of the OTC-products was achieved by the GC chiral analysis on a Cyclosil B capillary column.  相似文献

The transformation of renewable biomass into valuable products as alternatives to fossil fuels is essential for sustainable energy in sustainable society. This work systematically investigates the pyrolysis of sorghum bagasse biomass into bio-char and bio-oil products and studies the effect of temperature (623–823 K) on the conversion of sorghum bagasse and products yields. The physicochemical properties of bio-char were thoroughly studied using powder X-ray diffraction, elemental analysis (CHNSO), scanning electronic microscope, calorific value (CV), and Fourier transform infrared (FTIR) spectroscopy techniques. Also, gas chromatography–mass spectrometry (GC–MS), CV, and FTIR were used to understand the properties of bio-oil. The results obtained indicate that an increase in the pyrolysis temperature from 623 to 823 K leads to a decrease in the bio-char yield from 42.55 to 30.38%. On the other hand, the maximum bio-oil yield of 15.94% was obtained at 723 K. The bio-char obtained at 673 and 773 K was found by FTIR analysis to be composed of a highly ordered aromatic carbon structure. The calorific value of bio-oil, which contains a greater amount of acidic compounds, was found to be 6740 kcal/kg. The GC–MS analyses revealed the presence of octadecenoic acid, p-cresol, 2,6-dimethoxy phenol, 4-ethyl 2-methoxy phenol, phenol, o-guaiacol, and octadecanoic acid in the bio-oil obtained from the pyrolysis of sorghum bagasse biomass. The present study provides useful information for understanding the quality of bio-oil and bio-char obtained from high biomass sorghum bagasse.  相似文献

A new approach has been developed for the extraction and determination of aldehydes such as veratraldehyde, m-nitrobenzaldehyde, cinnamaldehyde, benzaldehyde, and p-chlorobenzaldehyde by using solid-phase microextraction (SPME) and high-performance liquid chromatography with UV detection (HPLC/UV). The method involves adsorption of the aldehydes on polydimethylsiloxane/divinylbenzene-coated fiber, followed by desorption in the desorption chamber of the SPME-HPLC interface, using acetonitrile-water (70 + 30) as the mobile phase; UV detection was at 254 nm. A good separation of 5 aldehydes was obtained on a C18 column. The detection limits of veratraldehyde, m-nitrobenzaldehyde, cinnamaldehyde, benzaldehyde, and p-chlorobenzaldehyde are 25, 41, 13, 12, and 11 pg/mL, respectively, which are about 100 times better than the detection limits for other SPME methods using gas chromatography. The proposed method was validated by determining benzaldehyde in bitter almonds and cinnamaldehyde in cinnamon bark. The recoveries of the 5 analytes were determined by analysis of spiked drinking water.  相似文献
