Even after emergence of most advanced instrumental techniques for the final separation, detection, identification and determination of analytes, sample handling continues to play a basic role in environmental analysis of complex matrices. In fact, sample preparation steps are often the bottleneck for combined time and efficiency in many overall analytical procedures. Thus, it is not surprising that, in the last two decades, a lot of effort has been devoted to the development of faster, safer, and more environment friendly techniques for sample extraction and extract clean up, prior to actual instrumental analysis. This article focuses on the state of the art in sample preparation of environmental solid biological samples dedicated to persistent organic pollutants (POPs) analysis. Extraction techniques such as Soxhlet extraction, sonication-assisted extraction, supercritical fluid extraction (SFE), microwave-assisted extraction (MAE), pressurised liquid extraction (PLE) and matrix solid-phase dispersion (MSPD) are reviewed and their most recent applications to the determination of POPs in biota samples are provided. Additionally, classical as well as promising novel extraction/clean-up techniques such as solid phase microextraction (SPME) are also summarized. Finally, emerging trends in sample preparation able to integrate analytes extraction and their adequate clean-up are presented.  相似文献

Green chemistry principles for developing methodologies have gained attention in analytical chemistry in recent decades. A growing number of analytical techniques have been proposed for determination of organic persistent pollutants in environmental and biological samples. In this light, the current review aims to present state-of-the-art sample preparation approaches based on green analytical principles proposed for the determination of polybrominated diphenyl ethers (PBDEs) and metabolites (OH-PBDEs and MeO-PBDEs) in environmental and biological samples. Approaches to lower the solvent consumption and accelerate the extraction, such as pressurized liquid extraction, microwave-assisted extraction, and ultrasound-assisted extraction, are discussed in this review. Special attention is paid to miniaturized sample preparation methodologies and strategies proposed to reduce organic solvent consumption. Additionally, extraction techniques based on alternative solvents (surfactants, supercritical fluids, or ionic liquids) are also commented in this work, even though these are scarcely used for determination of PBDEs. In addition to liquid-based extraction techniques, solid-based analytical techniques are also addressed. The development of greener, faster and simpler sample preparation approaches has increased in recent years (2003–2013). Among green extraction techniques, those based on the liquid phase predominate over those based on the solid phase (71% vs. 29%, respectively). For solid samples, solvent assisted extraction techniques are preferred for leaching of PBDEs, and liquid phase microextraction techniques are mostly used for liquid samples. Likewise, green characteristics of the instrumental analysis used after the extraction and clean-up steps are briefly discussed.  相似文献

Polybrominated diphenyl ethers (PBDEs) are a group of persistent organic pollutants. They are used as flame retardants in plastics, paints, varnishes and textile materials. PBDEs pose great risk to the environment because of their high persistence and ability to get into the environment easily due to the lack of chemical bonds with the matrix of materials, to which they are added. Global research studies confirmed the occurrence of those compounds in the majority of elements of water and land environment. Analysis of PBDEs in environmental samples is one of the specific analytical methods of criteria that comprise low detection limits and high selectivity. In this article, a literature review of methods for environmental sample preparation and analysis of the PBDE content was presented. The article discusses the potential of modern extraction techniques such as: solid-phase microextraction, single-drop microextraction, dispersive liquid-liquid microextraction, microwave-assisted extraction, cloud point extraction, hollow fibre-liquid phase microextraction and others for the separation of PBDEs from environmental samples with a complex matrix. Among the methods for qualitative and quantitative determination of PBDEs, a particular focus was put on gas chromatography/mass spectrometry with various injection techniques and different types of sample ionisation.  相似文献

In this review sample preparation strategies used for crude oil digestion in last ten years are discussed focusing on further metals and non-metals determination. One of the main challenges of proposed methods has been to overcome the difficulty to bring crude oil samples into solution, which should be compatible with analytical techniques used for element determination. On this aspect, this review summarizes the sample preparation methods for metals and non metals determination in crude oil including those based on wet digestion, combustion, emulsification, extraction, sample dilution with organic solvents, among others. Conventional methods related to wet digestion with concentrated acids or combustion are also covered, with special emphasis to closed systems. Trends in sample digestion, such as microwave-assisted digestion using diluted acids combined with high-efficiency decomposition systems are discussed. On the other hand, strategies based on sample dilution in organic solvents and procedures recommended for speciation analysis are reported as well as the use of direct analysis in view of the recent importance for crude oil field. A compilation concerning sample preparation for crude oil provided by official methods as well as certified reference materials available for accuracy evaluation is also presented and discussed.  相似文献

Methodology for detection of native (underivatized) amino acids (AA) in atmospheric aerosols has been developed. This article describes the use of LC-MS (Q-TOF) and microwave-assisted gas phase hydrolysis for detection of free and combined amino acids in aerosols collected in a Southeastern U.S. forest environment. Accurate mass detection and the addition of isotopically labeled surrogates prior to sample preparation allows for sensitive quantitation of target AA in a complex aerosol matrix. A total of 16 native AA were detected above the reporting threshold as water-soluble free AA, with an average concentration of 22 ± 9 ng m−3 (N = 13). Following microwave-assisted gas phase hydrolysis, the total AA concentration in the forest environment increased significantly (70 ± 35 ng m−3) and additional compounds (methionine, isoleucine) were detected above the reporting threshold. The ability to quantify AA in aerosol samples without derivatization reduces time-consuming preparation procedures while providing the advancement of selective mass determination for important organic nitrogen (ON) species. Details on sample preparation that eliminates the freeze-drying approach typically practiced for water removal with biological samples, and vapor phase microwave hydrolysis parameters are provided. In this study, a novel solid-liquid-solid extraction approach, which was termed ‘microwave-assisted micro-solid-phase extraction’ (MAE-μ-SPE), was developed. Target analytes were extracted from the sample into extraction solvent enhanced by microwave field, following adsorption by the adsorbent in the micro-solid-phase extraction device. Without any clean-up steps, the extracts were analysed by high performance liquid chromatography coupled with ultra-violet detector. The MAE-μ-SPE approach was developed for the extraction of four tetracycline antibiotics residues in environmental soil, sludge and atmospheric particulate matters. Variables affecting extraction procedures were systematically investigated. Low detection limits of 0.1–6.3 ng/g and low quantification limits of 0.33–20.7 ng/g were achieved under optimised conditions. The recoveries of antibiotics ranged from 70.6% to 110.5% with relative standard deviation of less than 15.1%. The predominance was showed when compared to conventional MAE and μ-SPE method. According to the results, MAE-μ-SPE was proved to be a simple and effective sample preparation method for the analysis of trace organic contaminants in environmental samples.  相似文献

大气颗粒物中有机物成分分析对深入研究大气颗粒物对人类健康、环境、气候、生态的影响,解析气溶胶来源,制定颗粒物控制相关法规,以及风险管理方法具有重要意义。由于颗粒物中的有机组分种类繁多,分析复杂,目前仅10%~20%的有机物得到了定性和定量分析。因此,大气细颗粒中有机物的分析已成为环境分析领域的优先发展方向。色谱是大气颗粒物中有机物分析的主要方法,而样品制备则是影响分析速度和精度的关键步骤。本文对颗粒物中有机组分色谱分析前的样品制备方法进行了综述,介绍了索氏提取、超声辅助提取、微波辅助提取、加压溶剂提取等溶剂提取方法以及热解吸提取方法,并重点介绍了这些方法在大气颗粒物样品处理中的应用,总结了各种方法的优缺点。  相似文献

Wall paintings typically contain low concentrations of organic materials within a largely inorganic matrix and are characterised by their high porosity and long-term exposure to severe environmental conditions. The identification of organic materials within specific paint or plaster layers is challenging and the inherent characteristics of wall painting samples present further complications. Embedding materials (such as epoxy, polyester and acrylic-based resins) used to produce cross-sections often infiltrate porous and leanly bound samples, and compromise the interpretation of Fourier transform infrared attenuated total reflectance (FTIR-ATR) spectra and the qualitative identification of natural organic materials. An alternative method for the preparation of cross-sections of wall painting samples was developed using cyclododecane (C(12)H(24)) as a temporary consolidant and barrier coating to encapsulate the sample, and to provide necessary support to produce a cross-section through microtoming. Impacts of traditional and novel sample preparation techniques on the identification of organic materials with micro-FTIR-ATR were examined for both replica and real wall painting samples.  相似文献

The application of microwave-assisted extraction (MAE) to the work-up of environmental and biological samples in the study of mercury speciation analysis has increased in recent years and is now increasingly accepted as a standard approach. The review provides a brief theoretical background of microwave heating and the basic principles of microwave energy used for extraction. The advantages and disadvantages of (a) MAE techniques, (b) the influence of the main parameters affecting the extraction, (c) statistical optimization approaches, and (d) strategies for method validation also are highlighted. Recent applications of MAE to mercury species analyses in biological samples, soils, sediments, and crude oil samples are surveyed and critically reviewed. In addition, comparisons of its use with other well-established extraction procedures are discussed.

The miniaturization and improvement of sample preparation is a challenge that has been fulfilled up to a point in many fields of analytical chemistry. Particularly, the hyphenation of microextraction with advanced analytical techniques has allowed the monitoring of target analytes in a vast variety of environmental samples. Several benefits can be obtained when miniaturized techniques such as solid-phase microextraction (SPME) or liquid-phase microextraction (LPME) are applied, specifically, their easiness, rapidity and capability to separate and pre-concentrate target analytes with a negligible consumption of organic solvents. In spite of the great acceptance that these green sample preparation techniques have in environmental research, their full implementation has not been achieved or even attempted in some relevant environmental matrices.  相似文献

This paper presents a new approach for complete and systematic analysis of organic additives in polyolefins. The proposed procedure is a convenient combination of sample preparation, performed by microwave-assisted extraction (MAE), and direct chromatographic evaluation of extract by high-performance liquid chromatography coupled with ultraviolet and evaporative light scattering detection. In particular two microwave-assisted processes are reported and discussed: the one-step MAE, useful for additives with low-medium dipolarity (like stabilizers, flame retardant, antistatics, slip and processing agents), and the two-step MAE, useful for additives with either high dipolarity (like organic salts, antigasfading, antiacid, nucleating agent) or high molecular mass (like polymeric hindered amine light stabilizers). Both the proposed processes have been tested on representative additives in five commercially common polymeric matrices, demonstrating their satisfactory analytical results, in terms of repeatability and percentage recoveries, and their good performances, in terms of safety and time/solvent consumption, in comparison with those of traditional extraction methods.  相似文献

Three non-specific methods for the extraction of total petroleum hydrocarbons (TPH) from soil into organic solvent were compared. The techniques used for sample preparation were Soxhlet extraction, closed-vessel microwave-assisted extraction, and CEN shake extraction. The total concentrations of extracted compounds in the boiling point range of C10–C40 were determined by gas chromatography with flame ionization detection. The best recovery (99%) and repeatability (±3%) from standard oil mixtures were obtained with microwave-assisted extraction. However, the different extraction methods exhibited different behaviour when spiked soil samples were extracted. The best repeatability was obtained with CEN shake extraction (±6%) but the repeatability values for Soxhlet and microwave-assisted methods were quite high (>20%). However, the larger uncertainties of the latter extraction methods does not necessarily limit the applicability of these methods to the determination of petroleum hydrocarbons in soil, as in the assessment of soil contamination the expanded uncertainty of the result is usually not limited by analytical uncertainty, but rather by the uncertainty of the primary sampling stage. However, distinctive variation found in the chromatographic profiles illustrates that discretion should be obeyed when chromatograms obtained after application of different extraction methods on petroleum contaminated samples are to be used in the fingerprinting or age dating studies. Otherwise, misleading conclusions concerning the age of spillage could be drawn.  相似文献

An integrated sample preparation method, termed “imFASP”, which combined in-situ filter-aided sample pretreatment and microwave-assisted trypsin digestion, was developed for preparation of microgram and even nanogram amounts of complex protein samples with high efficiency in 1 h. For imFASP method, proteins dissolved in 8 M urea were loaded onto a filter device with molecular weight cut off (MWCO) as 10 kDa, followed by in-situ protein preconcentration, denaturation, reduction, alkylation, and microwave-assisted tryptic digestion. Compared with traditional in-solution sample preparation method, imFASP method generated more protein and peptide identifications (IDs) from preparation of 45 μg Escherichia coli protein sample due to the higher efficiency, and the sample preparation throughput was significantly improved by 14 times (1 h vs. 15 h). More importantly, when the starting amounts of E. coli cell lysate decreased to nanogram level (50–500 ng), the protein and peptide identified by imFASP method were improved at least 30% and 44%, compared with traditional in-solution preparation method, suggesting dramatically higher peptide recovery of imFASP method for trace amounts of complex proteome samples. All these results demonstrate that the imFASP method developed here is of high potential for high efficient and high throughput preparation of trace amounts of complex proteome samples.  相似文献

In this review, a general discussion of sample preparation techniques based on combustion reactions in closed vessels is presented. Applications for several kinds of samples are described, taking into account the literature data reported in the last 25 years. The operational conditions as well as the main characteristics and drawbacks are discussed for bomb combustion, oxygen flask and microwave-induced combustion (MIC) techniques. Recent applications of MIC techniques are discussed with special concern for samples not well digested by conventional microwave-assisted wet digestion as, for example, coal and also for subsequent determination of halogens.  相似文献

Flame retardants, surfactants, pharmaceutical and personal care products, among other compounds, have been the object of numerous environmental studies. In this chapter, the application of microwave-assisted extraction (MAE) in the development of analytical methods for several groups of organic compounds with growing concern as emerging pollutants has been considered. Compared to other extraction techniques, optimization of MAE experimental conditions is rather easy owing to the low number of influential parameters (i.e. matrix moisture, nature of the solvent, time, power, and temperature in closed vessels). The great reduction in the extraction time and solvent consumption, as well as the possibility of performing multiple extractions, increasing the sample throughput, can also be highlighted among MAE advantages. In summary, the study of several applications of MAE to environmental problems demonstrates that this technique constitutes a good alternative for the determination of organic compounds in environmental samples. It can be used as a rapid screening tool, and also to obtain detailed information on the sources, behaviour and fate of emerging pollutants in environmental matrices.  相似文献

An efficient method for extracting pentachlorophenol (PCP) from organic solid matrices and soils using direct excitation of the fresh samples by microwave energy is presented. Steam-volatile PCP partitioned into the organic solvent n-hexane within the closed extraction vessel, resulting in recovery rates of 85-92% from aged samples. Condensed water provided a boundary layer between extract and sample thus preventing any contact of PCP with and re-partitioning into the sample. Duration of the microwave-assisted process was 35 min. Crude extracts were derivatized by heptafluorobutyric anhydride (HFBA) to improve selectivity and sensitivity resulting in detection limits of 1-2 micro g kg(-1). GC-MS/MS analyses proved that suitable extracts with only minor impurities were obtained. Common sample treatment steps like drying, thorough grinding, frequent transferring, and tedious clean-up, and concentration procedures which all can cause certain losses of analyte were minimized. The efficiency of the method was verified by comparison with an established ultrasonic extraction procedure. This microwave-assisted pressurized steam distillation with simultaneous partition into an organic phase thus provides a streamlined and efficient strategy which requires no additional investment in standard equipment for microwave-assisted extractions (MAE). Degradation of analytes at longer extraction times must be taken into account.  相似文献
