Derivatives of 1-(2-pyridyl)-4,5,6,7-tetrahydroindazole

In reaction of oxidation of 1-(2-pyridyl)3,6,6-trimethyl-4-oxo-4,5,6,7-tetrahydroindazole, the corresponding 4,5-dioxoindazole is obtained, and then 1-(2-pyridyl)-3-methyl-4-carboxy-5-(2-methyl-2-carboxypropyl)pyrazole; in bromination reaction, a series of 5-bromo and 7-bromo derivatives is obtained. From 4,5-dioxoindazole with hydrazides of acids, a series of 4-oxo-5-acylhydrazono derivatives is obtained, and also 1-(2-pyridyl)-3,6,6-trimethyl-4-oxo-5-tosylhydroindazole, which, under the action of caustic, gives the 4-oxo-5-diazo derivative.Riga Technical University, Riga LV-1658. 1- and 2-(4-Hydroxycarbonylphenyl)-4-oxo-4,5,6,7-tetrahydroindazoles

Reactions of 4-hydrazinobenzoic acid with 2-formyl- and 2-acetyldimedones were utilized to synthesize 6,6-dimethyl- and 3,6,6-trimethyl-1-(4-hydroxycarbonylphenyl)-4-oxo-4,5,6,7-tetrahydroindazoles corre-spondingly, and the reaction with 2-cyano-3-ethoxy-5,5-dimethylcyclohex-2-en-1-one gave 2-(4-hydroxy-carbonylphenyl)-3-amino-4-oxo-6,6-dimethyl-4,5,6,7-tetrahydroindazole. Reactions of 2-Amino-4-methyl-6-(2-pyridyl)- and 2-Amino-4-methyl-6-phenyl-7,8-dihydroindazolo[4,5-d]thiazoles with Aldehydes

The reaction of 2-amino-4-methyl-6-(2-pyridyl)-7,8-dihydroindazolo[4,5-d]thiazole, obtained by treating 3-methyl-4-oxo-1-(2-pyridyl)-4,5,6,7-tetrahydroindazole with pyridinium bromide perbromide and then with thiourea, and 2-amino-4-methyl-6-phenyl-7,8-dihydroindazolo[4,5-d]thiazole with 4-bromo-, 4-fluoro-, 4-dimethylamino-, 4-methoxy-, 3,4-dimethoxy-, and 3,4-methylenedioxybenzaldehydes, furfural, pyridinecarbaldehyde, and thiophenecarbaldehyde gave the corresponding Schiff bases. The products of the condensation of these aminothiazoles with cinnamaldehyde, 1-(2-pyridyl)- and 4-chloro-1-(2,4-difluorophenyl)-5-formyl-3-methyl-6,7-dihydroindazoles, 2-formyl-dimedone, and 2-formyl-1,3-indanedione were also obtained.     

4,5-Dihydropyrazolo[3,4-f]quinazolines

By reaction of 6-dimethylamino-5-oxo-2-phenyl-5,6,7,8-tetrahydroquinazoline with hydrazine, phenylhydrazine, 4-bromo-, 4-chlorophenylhydrazines, and also reaction of 6-dimethylaminomethylene-5-oxo-2-(4-pyridyl)-5,6,7,8-tetrahydroquinazoline with hydrazine, 4-methoxy- and 2-carboxyphenylhydrazines, we have obtained the corresponding 7-phenyl(4-pyridyl)-substituted 1H(2H)- or 1-aryl-4,5-dihydropyrazolo[3,4-f]quinazolines. Methylation of 7-phenyl-4,5-dihydro-1(2H)-pyrazolo[3,4-f]quinazoline by methyl iodide led to its 2-methyl derivative.     

1-(2-Quinoxalyl)-, 1-[3,5-Di(trifluoromethyl)phenyl]-, 1-(2-Carboxyphenyl)-, and 1-Ethoxycarbonyl-4-oxo-4,5,6,7-tetrahydroindazoles

The corresponding 1-(2-quinoxalyl)-, 1-[3,5-di(trifluoromethyl)phenyl]-, and 1-ethoxycarbonyl-3-methyl-4-oxo-4,5,6,7-tetrahydroindazoles have been obtained from reactions of 2-acetyl-1,3-cyclohexanedione, its 5,5-dimethyl and 5-(2-furyl) derivatives, with 2-hydrazinoquinoxaline, 3,5-di(trifluoromethyl)phenylhydrazine, and ethoxycarbonylhydrazine. On interaction with ethoxycarbonylhydrazine the intermediate 2-[1-(-ethoxycarbonyl)hydrazino]ethylidene-1,3-cyclohexanediones were also isolated. From the potassium salt of 2-formyldimedone and 2-carboxyphenylhydrazine hydrochloride, 2-(2-carboxyphenyl)hydrazinomethylene-5,5-dimethyl-1,3-cyclohexanedione was obtained, the cyclization of which in ethanol in the presence of HCl led to 1-(2-carboxyphenyl)- and 1-(2-ethoxycarbonylphenyl)-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole.     

4,5-Dioxo-and 4-oxo-2,3-diaryl-5-diazo-6,6-dimethyl-4,5,6,7-tetrahydroindazoles

The oxidation of 2,3-diphenyl-, 3-(4-chlorophenyl)-2-phenyl-, 3-(4-dimethylaminophenyl)-2-phenyl-, 3-(4-methoxyphenyl)-2-phenyl-, 2-phenyl-3-(3-pyridyl)-, 3-phenyl-2-(2-pyridyl)-, and 3-(4-dimethylaminophenyl)-6,6-dimethyl-4-oxo-2-(2-pyridyl)-4,5,6,7-tetrahydroindazoles using H₂SeO₃ in acetic acid in the presence of sulfuric acid gave the corresponding 4,5-dioxo derivatives. Reaction of these with tosylhydrazine gave 4-oxo-5-tosylhydrazino derivatives which were decomposed by alkali to give the corresponding 2,3-diaryl-5-diazo-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazoles. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 12, pp. 1829–1833, December, 2005     

4(3H)-Quinazolinones containing heterocyclic group in position 3

The corresponding 2,3-substituted 4(3H)-quinazolinones were obtained in the reactions of 2-methyl- and 2-phenyl-4-oxo-3,1-benzoxazines with 1-amino-1,2,4-triazole, 4-amino-2,3-dimethyl-1-phenyl-5-pyrazolone, 2-amino-5-ethyl-1,3,4-thiadiazole, 3-amino-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole, 1-amino-3-cyano-4,6-dimethyl-2-pyridone, and 1-amino-3-cyano-6-phenyl-4-trifluoromethyl-2-pyridone. The formation of N-benzolyanthranilamides in the reactions of 2-phenyl-4-oxo-3,1-benzoxazine with 2-amino-5-ethyl-1,3,4-thiadiazole and 1-amino-3-cyano-6-phenyl-4-trifluoromethyl-2-pyridones was exceptional. Synthesis and reactions of 2-(4-pyridyl)-7,7-dimethyl-5-oxo-5,6,7,8-tetrahydroquinazoline

7,7-Dimethyl- and 7-phenyl-2-(4-pyridyl)-5-oxo-5,6,7,8-tetrahydroquinazolines were synthesized in the reactions of 2-formyl-5,5-dimethyl- and 5-phenyl-1,3-cyclohexanediones, respectively, with 4-amidinopyridine. The oxime, thiosemicarbazone, 4-quinazolyl-, nicotinoyl-, isonicotinoyl-, and 2-hydroxybenzoylhydrazones of 2-(4-pyridyl)-5-oxo-7,7-dimethyl-5,6,7,8-tetrahydroquinazoline were obtained. The reduction of this ketone by sodium borohydride leads to the 5-hydroxy derivative.     

Pyrazolo[5,4-h]quinazolines

The reaction of 1-phenyl- and 4-chloro-5-formyl-3-methyl-1-(2-pyridyl)-6,7-dihydroindazoles with carbamidinopyrrolidine, 4-carbamidinomorpholine, creatine, 2-carbamidinopyrazine, and 2-carbamidino-5-trifluoromethylpyridine gives the corresponding 3,8-substituted 1-methyl-4,5-dihydropyrazolo-[5,4-h]quinazolines and with 2-aminobenzimidazole gives 3-phenyl- and 3-(2-pyridyl)-1-methyl-4,5-dihydrobenzo[b]indazolo[4,5-e]imidazolo[1,2-a]pyrimidines.Riga Technical University, Riga LV-1658, Latvia; e-mail; marina@osi.lv . Synthesis and Photoisomerization of 3-Cyano-6,6-dimethyl-4-(4-nitrophenylvinyl)-5,6-dihydropyran-2-one

The condensation of 4-nitrobenzaldehyde with 3-cyano-4,6,6-trimethyl-5,6-dihydropyran-2-one leads to the formation of a crotonization product and a compound of the Michael adduct type. The main product of the photochemical conversion of (E)-3-cyano-6,6-dimethyl-4-(4-nitrophenylvinyl)-5,6-dihydropyran-2-one is the Z-isomer. Investigation of the photoisomerization of 3-cyano-6,6-dimethyl-4-(4-nitrophenylvinyl)-5,6-dihydropyran-2-one by the semiempirical AM1 method showed that in the ground state the E-isomer was thermodynamically more stable than the E-isomer. E-Z-photoisomerization is effected most probably through the lowest excited singlet state S₁.     

Synthesis of 2-(4-Hydroxy-6-methyl-2-oxo-2H-pyran-3-carbonyl)-6,6-dimethyl-3-phenyl-3,5,6,7-tetrahydro-2H-benzofuran-4-one Derivatives via Multicomponent Reaction

A sequential one-pot, two-step reaction for an efficient preparation of 2-(4-hydroxy-6-methyl-2-oxo-2H-pyran-3-carbonyl)-6,6-dimethyl-3-phenyl-3,5,6,7-tetrahydro-2H-benzofuran-4-one derivatives has been described. One-pot reaction of in situ–formed benzopyran-substituted pyridinium ylides with aromatic aldehydes and dimedone gives corresponding 2,3-dihydrofurans in good yields. The structures of all the newly synthesized compounds were confirmed from their analytical and spectral data.     

Alkylation of 4-oxo-3-methyl-4,5,6,7-tetrahydroindazoles

The alkylation of 4-oxo-3,6,6-trimethyl-4,5,6,7-tetrahydroindazole with alkyl halides forms mixtures of 1-alkyl and 2-alkyl derivatives which have been separated by fractional crystallization from n-hexane. The 4-oxo-1-alkyl-,-1-aryl-, and -1-acyl-4,5,6,7-tetrahydroindazoles have characteristic frequencies in their IR spectra at 1540-1530 cm^–1, and the corresponding 2-substituted derivatives have frequencies at 1565-1555 cm^–1. The introduction of an alkyl substituent at a nitrogen atom decreases the basicity of 4,5,6,7-tetrahydroindazole by 0.6 orders of magnitude, the 1-alkyl isomers being somewhat stronger bases than the 2-alkyl isomers.     

Pyrimido[4,5-f]quinazolines

The corresponding 6-dimethylaminomethylen-5-oxo-5,6,7,8-tetrahydroquinazolines were obtained from reactions of 2-substituted and 2,7-disubstituted 5-oxo-5,6,7,8-tetrahydroquinazolines with the dimethylacetal of DMF. Sixteen 2,8-disubstituted 5,6-dihydropyrimido[4,5-f]quinazolines were obtained from the reaction of 2-phenyl- and 6-(dimethylamino)methylen-5-oxo-2-(4-pyridyl)-5,6,7,8-tetrahydroquinazolines with guanidine and eight amidines. The corresponding 6-hydroxymethylene derivative was obtained by hydrolysis of 6-(dimethylamino)methylen-5-oxo-1-phenyl-5,6,7,8-tetrahydroquinazoline.     

Syntheses Based on 4-Chloro-1-[3,5-di(trifluoromethyl)phenyl]-, 4-Chloro-1-(2,4-difluorophenyl)-5-formyl-3-methyl-6,7-dihydroindazoles,and 4-Chloro-5-formyl-3-methyl-1-(2-pyridyl)-6,7-dihydroindazoles

Vilsmeier formylation of 1-[3,5-di(trifluoromethyl)phenyl]- and 1-(2,4-difluorophenyl)-3-methyl-4-oxo-4,5,6,7-tetrahydroindazoles gave the corresponding 1-aryl-4-chloro-5-formyl-3-methyl-6,7-dihydroindazoles. Reaction of the latter with amidines, o-phenylenediamine, hydrazine, or hydroxylamine gave a series of 1-aryl-3-methyl-6,6-dihydroindazoles annelated at positions 4 and 5. The reaction of 4-chloro-5-formyl-3-methyl-1-(2-pyridyl)-6,7-dihydroindazole with substituted anilines gave 5-arylaminomethylene-4-oxo- or 5-arylaminomethylene-4-arylimino-3-methyl-1-(2-pyridyl)-4,5,6,7-tetrahydroindazoles depending on the molar ratio of reagents and the nucleophilicity of the amines.__________Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 5, pp. 740–750, May, 2005.     

Synthesis and reactions of 1-(2-pyridyl)-3-methyl-4-chloro-5-formyl-6,7-dihydroindazoles

The Vilsmeier formylation of 1-(2-pyridyl)-3-methyl-4-oxo-4,5,6,7-tetrahydroindazole and its 6-phenyl derivative gives 1-(2-pyridyl)-3-methyl-4-chloro-5-formyl-6,7-dihydroindazoles. Synthesis and reactions of 1-(4-chloro-, 3-chloro-, 2-chloro-, 2,4-dichloro-, and 2,4-difluorophenyl)-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazoles

Reaction of the potassium salt of 2-formyldimedone with hydrochlorides of 4-chloro-, 3-chloro-, 2-chloro-, 2,4-dichloro-, and 2,4-difluorophenylhydrazines gave the corresponding 2-arylhydrazinomethylene-dimedones which cyclized in acid media to 1-substituted 6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazoles. Oxidation of the latter with selenious acid gave the corresponding 4,5-dioxo-4,5,6,7-tetrahydroindazoles which were further converted into 3-aryl-5,5-dimethyl-4,5-dihydro-3H-pyrazolo[4,3-a]phenazines and 2,6-diaryl-4,4-dimethyl-4,5-dihydro-1H(3H)-indazolo[4,5-g]imidazoles.Riga Technical University, Riga LV-1658, Latvia; e-mail: marina@osi.lv. A study of the Claisen rearrangement of 7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one derivatives

Summary The Claisen rearrangement of 7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 a) gave 7-hydroxy-8-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (3 a) and 2,3-dihydro-2,6-diphenyl-3-methyl-(7H)furo[2,3-h]-1-benzopyran-7-one (7 a). 2-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 b) afforded4 b and7 b. 8-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (12) gave only the alkali soluble product 7-hydroxy-8-methyl-6-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (13).3 a,4 b, and13 were further cyclized in acidic medium to9 a,10 b, and14 followed by dehydrogenation.This paper is dedicated to Dr. F. 2-Substituted 5-oxo-5,6,7,8-tetrahydroquinazolines

Twenty novel 2-substituted 5-oxo-5,6,7,8-tetrahydroquinazolines in reactions of 2-formyl-1,3-cyclohexanedione and its 5,5-dimethyl- and 5-phenyl derivatives with 4-chloro- and 4-carbonylaminobenzamidines, 3- and 4-carbamidinopyridines, 2-carbamidinopyrazine, 2-carbamidino-5-trifluoromethylpyridine, 1-carbamidinopyrrolidine, 4-carbamidinomorpholine, and 1-carbamidino-3,5-dimethylpyrazole have been obtained.Riga Technical University, Riga LV-1658, Latvia; e-mail: marina@osi.lv. The Use of 4-(Bromomethylene)-5,5-dimethyl-1,3-dioxolan-2-one as “Masked” α-Bromo-α'-Hydroxy Ketone in the Synthesis of Heterocyclic Systems

4-(Bromomethylene)-5,5-dimethyl-1,3-dioxolan-2-one was obtained on the basis of the readily obtainable 4-methylene-5,5-dimethyl-1,3-dioxolan-2-one. It forms 2-imidazo[1,2-a]pyridin-2-yl-2-propanol with 2-aminopyridine, 11a-hydroxy-1,1-dimethyl-3-oxo-1,5,11,11a-tetrahydro[1,3]oxazolo[3,4-a]pyrido[1,2-d]-10-pyrazinium bromide with 2-(aminomethyl)pyridine, and the corresponding derivative of 4-hydroxyoxazolidin-2-one with 2-(3,4-dimethoxyphenyl)ethylamine. The last product was converted by intramolecular amidoalkylation without isolation into 10b-(bromomethyl)-8,9-dimethoxy-1,1-dimethyl-1,5,6,10b-tetrahydro[1,3]oxazolo[3,4-a]isoquinolin-3-one.     

Synthesis of 4-hetaryl-5,6-(2,5-dimethyl-3-thienyl)-2-phenyl-4h-thiazines and investigation of their photochromism

The polar [4+2] cycloaddition reaction of 1,2-bis(2,5-dimethyl-3-thienyl)acetylene with thiobenzamide and certain aldehydes of the thiophene and furan series catalyzed by boron trifluoride etherate gives the corresponding 4-hetaryl-5,6-di(2,5-dimethyl-3-thienyl)-2-phenyl-4H-1,3-thiazines.
