A new ellagic acid derivative from Polygonum runcinatum   总被引:1,自引:0,他引:1  
A new ellagic acid derivative, 3,3′-dimethylellagic acid-4′-O-(6″-galloyl)-β-d-glucoside, named runcinatside (5), together with four known compounds 3,3′-dimethylellagic acid (1), 3,3′,4′-trimethylellagic acid (2), 3,3′-dimethylellagic acid-4′-O-β-d-glucoside (3) and 3-methylellagic acid-4′-O-α-l-rhamno-pyranoside (4), was isolated from the roots of Polygonum runcinatum Buch.-Ham. ex D.Don Var. sinense Hemsl and the structures of these compounds were established by spectroscopic methods and comparison with previously reported data. All compounds showed antioxidant activities in vitro and compound 5 possessed the highest activity.  相似文献

The aim of this study was to investigate whether ellagic acid in pomegranate leaf tannins could be transported into HepG2 cells and its transport behavior. High‐performance liquid chromatography coupled with a 996 photodiode array detector at 254 nm was applied. The mobile phase was an acetonitrile–water solution (containing 0.1% triethylamine, pH 3.0; 16:64, v/v, for determining ellagic acid in cells). The flow rate was 0.8 mL/min. Cells were incubated with pomegranate leaf tannins with 100 and 50 µg/mL (containing 1.71 and 0.85 µg/mL of ellagic acid, respectively) for a specific time, then lysed and sonicated in methanol to extract intracellular ellagic acid. A 10 µL aliquot of sample was injected into the HPLC system to determine ellagic acid concentration. The results showed that ellagic acid in pomegranate leaf tannins could be transported into the cells, which was in correlation with total cholesterol alteration in the cells. This is the first time that the transport behavior of ellagic acid through HepG2 cells in vitro has been comprehensively demonstrated. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献

The absorption and fluorescence properties of the plant phenol ellagic acid have been studied in alcohol and aqueous solutions. Fluorescence is weak in all kinds of solvents, but is greatly enhanced by addition of borax. The resulting complex emits much more intensely in methanol than in water solution. UV and fluorescence data reveal that the complex formed in methanol is different from the complex formed in aqueous solution. The enormous fluorescence enhancement of ellagic acid by borax offers a simple method for the visualization and quantification of the potent anti-mutagen ellagic acid as well as a fluorimetric method for the determination of boron.The IUPAC name for ellagic acid is 2,3,7,8-tetra-hydroxy[1]benzopyrano[5,4,3—c,d,e][1]-benzopyran-5,10-dione.  相似文献

Ellagic acid, a natural polyphenol, was isolated from pomegranate peel extract by hydrophobic interaction using graphene oxide grafted cotton fiber as a stationary adsorbent. The grafted graphene oxide moieties served as hydrophobic interaction‐binding sites for ellagic acid adsorption. The graphene oxide grafted cotton fiber was made into a membrane‐like sheet in order to complete ellagic acid purification by using a binding–elution mode. The effects of operational parameters, such as the composition of the binding buffer/elution buffer, buffer pH, and buffer concentration, on the isolation process were investigated. It was found that 5 mmol/L sodium carbonate aqueous solution is a proper‐binding buffer, and sodium hydroxide aqueous solution ranging from 0.04 to 0.06 mol/L is a suitable elution solution for ellagic acid purification. Under the optimized condition, the purity of ellagic acid increased significantly from 7.5% in the crude extract to 75.0–80.0%. The pH value was found to be a key parameter that determines the adsorption and desorption of ellagic acid. No organic solvent is involved in the entire purification process. Thus, a simple and environmentally friendly method is established for ellagic acid purification using a graphene oxide‐modified biodegradable and bio‐sourced fibrous adsorbent.  相似文献

Antioxidants are gaining tremendous interest as chemopreventive as well as chemotherapeutic agents. Ellagic acid (EA) is a plant derived compound with very poor solubility in water and very low octanol/water partition coefficient and coenzyme Q10 (CoQ10) is a highly lipophilic compound, which is synthesized in the body and can be derived from food supplements as well. The new insights in the combination therapy are promising a better future in many challenging diseases. Synergism is among the key advantages of combination therapy apart from decreased intensity of unwanted effects of a compound, increased patient compliance and reduction in cost of therapy. EA and CoQ10 supplementation in combination will be beneficial in strengthening the weakened antioxidant defense system in many diseases related to oxidative stress. Here we report first derivative UV spectroscopic and HPLC methods for the simultaneous analysis of these two agents in pharmaceutical preparations. Results obtained indicate that the derivative spectroscopy is as efficient as HPLC method in quantitative analysis. Retention of ellagic acid can be increased using PEG bonded column which is poorly retained on C18 column. PEG column can be used for rapid simultaneous analysis of EA and CoQ10, which are having diverse physicochemical properties.  相似文献

Summary Pre-column derivatization followed by reversed-phase HPLC has become widely used as a sensitive and speedy method of amino acid analysis.The technique has been improved by the development of simple and reproducible strategies for derivatization of the amino acids with DABS and for the HPLC analysis. The extraction and separation of dyes present on textile fibers offers the possibility of enhanced discrimination between forensic trace fiber evidence. An automated liquid sample handling workstation was programmed to deliver varying solvent combinations to acid-dyed nylon samples, and the resulting extracts were analyzed by an ultraviolet/visible microplate reader to evaluate extraction efficiencies at different experimental conditions. Combinatorial experiments using three-component mixture designs varied three solvents (water, pyridine, and aqueous ammonia) and were employed at different extraction temperatures for various extraction durations. The extraction efficiency as a function of the three solvents (pyridine/ammonia/water) was modeled and used to define optimum conditions for the extraction of three subclasses of acid dyes (anthraquinone, azo, and metal complex) from nylon fibers. The capillary electrophoresis analysis of acid dye extracts is demonstrated using an electrolyte solution of 15 mM ammonium acetate in acetonitrile/water (40:60, v/v) at pH 9.3. Excellent separations and discriminating diode array spectra are obtained even for dyes of similar color. Activity of Chinese Danggui (DG), the processed root of Angelica sinensis (Oliv.) Diels, is linked to the ferulic acid content but the stability of ferulic acid during extraction for medicinal use is not known. The stabilities of ferulic acid and coniferyl ferulate were evaluated in the extracts of DG using a variety of extraction solvents. These included various combinations and proportions of methanol, water, formic acid, 1 M aqueous hydrochloric acid and 2% sodium hydrogen carbonate (NaHCO3) in water. Coniferyl ferulate was found liable to hydrolyze into ferulic acid in neutral, strongly acidic and basic solvents, where heat and water could facilitate this hydrolysis. However, the hydrolysis was relatively resisted in weakly organic acid. Based on the stability evaluation, two new terms, namely: free ferulic acid and total ferulic acid, were suggested and defined. Free ferulic acid refers to the natural content of ferulic acid in herbs. Total ferulic acid means the sum of free ferulic acid plus the amount of related hydrolyzed components. Meanwhile, the high-performance liquid chromatographic (HPLC) method was developed to assay free ferulic acid and total ferulic acid in DG using methanol-formic acid (95:5) and methanol-2% NaHCO3 in water (95:5) as extraction solvents, respectively. Ten DG samples were investigated on their contents of free and total ferulic acid. The results indicated that the amount variety of free ferulic acid was larger than that of their counterparts, and the ratio of total ferulic acid to free ferulic acid was 4.07 +/- 2.73 (mean +/- SD, n = 10). The chemical assay of DG using total ferulic acid content would be a better choice to assess the herbal quality and was recommended.  相似文献

Jua (juá in Portuguese) is an underexplored fruit from Brazil’s northeast. This fruit is rich in antioxidant substances. However, there is a dearth of information about jua’s bioactive potential. The present study evaluated two extraction methods (continuous agitation and ultrasound-assisted extraction—UAE) and employed three different solvents (water, ethanol, and acetone) to efficiently recover soluble phenolic compounds. Aqueous extracts obtained by UAE showed the highest total phenolic content (TPC) and antiradical activity. Besides being an eco-friendly procedure, extraction and/or solubility in an aqueous medium is also important for food application. Ellagic acids were the predominant phenolics (80%) found in aqueous jua pulp extract obtained by UAE, as determined by HPLC, while its TPC was 405.8 gallic acid equivalent per gram of fruit. This extract also exhibited a higher scavenging activity towards peroxyl radicals when compared to that of several other fruits from the literature, including grape, strawberry, cranberry, and walnuts, which are known references in terms of antioxidants. This is the first report that demonstrates jua pulp’s potential as an alternative source of ellagic acid and other phenolic acids and flavonoids. Therefore, the outcome of this study provides new information that can be useful for functional food and nutraceutical industries.  相似文献

Gallic acid and ellagic acid are two widely occurring phenolic compounds of plant origin, to which many biological activities including anticancer and antiviral activity have been attributed. A simple HPTLC method has been developed for the simultaneous quantification of gallic acid and ellagic acid. The method was validated for precision, repeatability, and accuracy. Instrumental precision was found to be 0.083 and 0.78, and the repeatability of the method was found to be 1.07 and 1.50 (% CV) for gallic acid and ellagic acid, respectively. The accuracy of the method was checked by a recovery study conducted at two different levels and the average percentage recovery was found to be 101.02% for gallic acid and 102.42% for ellagic acid. The above method was used for the quantification of gallic acid and ellagic acid content in seeds of Abrus precatorius Linn., whole plant of Phyllanthus maderaspatensis Linn., and flowers of Nymphaea alba Linn. The proposed HPTLC method for the simultaneous quantification of gallic acid and ellagic acid was found to be simple, precise, specific, sensitive, and accurate and can be used for routine quality control of herbal raw materials and for the quantification of these compounds in plant materials.  相似文献

Industrial processing of raspberry juice and wine generates considerable byproducts of raspberry pomace. Ellagic acids/ellagitannins, being characterized by their antioxidant and antiproliferation properties, constitute the majority of polyphenolics in the pomace and are valuable for recovery. In the present study, we developed a novel procedure with sodium bicarbonate assisted extraction (SBAE) to recover ellagic acid from raspberry wine pomace. Key parameters in the procedure, i.e., sodium bicarbonate concentration, temperature, time and solid/liquid (S/L) ratio, were investigated by single factor analysis and optimized subsequently by Response Surface Methodology (RSM). Optimal parameters for the SBAE method here were found to be 1.2% (w/v) NaHCO3, 1:93 (w/v) S/L ratio, 22 min and 100 °C. Under these conditions, the ellagic acid yield was 6.30 ± 0.92 mg/g pomace with an antioxidant activity of 79.0 ± 0.96 μmol Trolox eq/g pomace (DPPH assay), which are 2.37 and 1.32 times the values obtained by extraction with methanol–acetone–water solvent, respectively. The considerable improvement in ellagic acid extraction efficiency could be highly attributed to the reactions of lipid saponification and ellagitannin hydrolysis resulted from sodium bicarbonates. The present study has established an organic solvent-free method for the extraction of ellagic acid from raspberry wine pomace, which is feasible and practical in nutraceutical applications.  相似文献

建立了一种基于纯水提取的新型烟熏鲣鱼中肌苷酸的提取方法。样品前处理采用95 ℃的纯水作为提取液,提取5 min。采用的色谱条件: 色谱柱为Agilent Zorbax SB-C18柱(150 mm×4.6 mm, 5 μm),柱温为30 ℃,流动相为甲醇-50 mmol/L乙酸铵水溶液(5:95, v/v),流速为1.0 mL/min,检测波长为254 nm。结果表明,该方法测定烟熏鲣鱼中肌苷酸含量的线性范围为1.0~120.0 mg/L;在0.40、2.00、4.00 g/kg (n=3)的添加水平下,回收率为84.6%~100.2%,测定结果的相对标准偏差(RSD)为1.00%~6.12%;检出限(信噪比(S/N)为3)为4 mg/kg;定量限(S/N=10)为12 mg/kg。该方法与常规采用的基于高氯酸的提取方法相比较,提取效率提高了37.0%,并且该方法操作安全、与环境友好,对烟熏鲣鱼中肌苷酸含量测定准确度良好,对其他食品中肌苷酸含量的测定具有借鉴意义,是一种值得推广的样品预处理方法。  相似文献

Tears have a significant role in antioxidant defense in ocular tissues and since their collection is quick and noninvasive, their analysis would facilitate monitoring of pathophysiological changes. However, their low volume and low content of antioxidants makes analysis difficult; methods of high sensitivity are needed. In this paper, we present a method for tear analysis of two antioxidant molecules (ascorbic and uric acid) and of a lipid peroxidation indicator (malondialdehyde) with capillary electrophoresis. Tears were collected with Schirmer strips, extracted with a low‐pH phosphate buffer, centrifuged through membrane filters and an antioxidant was added. They were stable at ?70°C for 15 days. After pilot experiments, optimum electrophoretic separation was achieved in a 25 mm borate buffer, pH 10.0, containing 100 mm sodium dodecyl sulfate at 25°C and 20 kV. The developed method has good repeatability (<5% RSD), precision (<15% relative error values) and high sensitivity (LLOQ values of 20, 2.3 and 2.5 μM for ascorbate, urate and malondialdehyde, respectively). It was applied to the analysis of tears from healthy individuals and the antioxidant levels are in agreement with those obtained with other techniques. A novel europium complex, Eu(Hdinic)(dinic)(H2O)5·4H2O (dinic = 2,5-pyridinedicarboxylic acid), has been synthesized under hydrothermal conditions and characterized structurally by X-ray diffraction methods. The complex has a mononuclear molecular structure, is monoclinic, space group C2/c, with a?=?13.877(5), b?=?9.632(3), c?=?32.991(11)?Å, β?=?93.414(4)°, V?=?4402(2)?Å3, D c?=?1.948?Mg?m?3, Z?=?8, F(000)?=?2576, R 1?=?0.0320. Arjunic acid and arjunolic acid are main bioactive components of Terminalia arjuna stem bark and reported for various biological activities. In this study, microwave-assisted extraction (MAE) of arjunic and arjunolic acid from stem bark of T. arjuna was investigated with developed and validated HPLC-PDA method, which resulted in the isolation of a novel anticancer molecule i.e. arjunic acid. Effects of several experimental parameters, such as type and volume of extraction solvents, microwave power, microwave extraction time, on the extraction efficiencies of arjunic, and arjunolic acid from stem bark of T. arjuna were evaluated. The optimal extraction conditions identified were 5.0 g quantity of stem bark powder, 20 mL of ethyl acetate, preleaching time 10 min, microwave power 600 W, temperature 65°C, and microwave irradiation time 5 min. The results showed that MAE is a more rapid extraction method with higher yield and lower solvent consumptions than reported methods. The HPLC-PDA analysis method was developed and validated to have good linearity, precision, sensitivity, and accuracy. MAE-HPLC-PDA is a faster, convenient, and appropriate method for isolation and determination of arjunic acid and arjunolic acid in the stem bark of T. arjuna.  相似文献
