d ‐Glucaric acid (GA) is an aldaric acid and consists of an asymmetric acyclic sugar backbone with a carboxyl group positioned at either end of its structure (i.e., the C1 and C6 positions). The purpose of this study was to conduct a conformation analysis of flexible GA as a solution in deuterium oxide by NMR spectroscopy, based on J‐resolved conformation analysis using proton–proton (3JHH) and proton–carbon (2JCH and 3JCH) coupling constants, as well as nuclear overhauser effect spectroscopy (NOESY). The 2JCH and 3JCH coupling constants were measured using the J‐resolved heteronuclear multiple bond correlation (HMBC) NMR technique. NOESY correlation experiments indicated that H2 and H5 were in close proximity, despite the fact that these protons were separated by too large distance in the fully extended form of the chain structure to provide a NOESY correlation. The validities of the three possible conformers along the three different bonds (i.e., C2? C3, C3? C4, and C4? C5) were evaluated sequentially based on the J‐coupling values and the NOESY correlations. The results of these analyses suggested that there were three dominant conformers of GA, including conformer 1 , which was H2H3:gauche, H3H4:anti, and H4H5:gauche; conformer 2 , which was H2H3:gauche, H3H4:anti, and H4H5:anti; and conformer 3 , which was H2H3:gauche, H3H4: gauche, and H4H5:anti. These results also suggested that all three of these conformers exist in equilibrium with each other. Lastly, the results of the current study suggested that the conformational structures of GA in solution were ‘bent’ rather than being fully extended. The effect of hydroxymethyl conformation (gg, gt, and tg rotamers about the C4-C5 bond) on the conformational energies and structural parameters (bond lengths, bond angles, bond torsions) of the 10 envelope forms of the biologically relevant aldopentofuranose, 2-deoxy-beta-D-erythro-pentofuranose (2-deoxy-D-ribofuranose) 2, has been investigated by ab initio molecular orbital calculations at the HF/6-31G level of theory. C4-C5 bond rotation induces significant changes in the conformational energy profile of 2 (2gt and 2tg exhibit one global energy minimum, whereas 2gg exhibits two nearly equivalent energy minima), and structural changes, especially those in bond lengths, are consistent with predictions based on previously reported vicinal, 1,3- and 1,4-oxygen lone pair effects. HF/6-31G-optimized envelope geometries of 2gg were re-optimized using density functional theory (DFT, B3LYP/6-31G), and the resulting structures were used in DFT calculations of NMR spin-spin coupling constants involving 13C (i.e., J(CH) and J(CC) over one, two, and three bonds) in 2gg according to methods described previously. The computed J-couplings were compared to those reported previously in 2gt to assess the effect of C4-C5 bond rotation on scalar couplings within the furanose ring and hydroxymethyl side chain. The results confirm prior predictions of correlations between 2J(CH), 3J(CH), 2J(CC) and 3J(CC), and ring conformation, and verify the usefulness of a concerted application of these couplings (both their magnitudes and signs) in assigning preferred ring and C4-C5 bond conformations in aldopentofuranosyl rings. The new calculated J-couplings in 2gg have particular relevance to related J-couplings in DNA (and RNA indirectly), where the gg rotamer, rather than the gt rotamer, is observed in most native structures. The effects of two additional structural perturbations on 2 were also studied, namely, deoxygenation at C5 (yielding 2,5-dideoxy-beta-D-erythro-pentofuranose 4) and methyl glycosidation at O1 (yielding methyl 2-deoxy-beta-D-erythro-pentofuranoside 5) at the HF/6-31G level. The conformational energy profile of 4 resembles that found for 2gt, not 2gg, indicating that 4 is an inappropriate structural mimic of the furanose ring in DNA. Glycosidation failed to induce differential stabilization of ring conformations containing an axial C1-O1 bond (anomeric effect), contrary to experimental data. The latter discrepancy indicates that either the magnitude of this differential stabilization depends on ring configuration or that solvent effects, which are neglected in these calculations, play a role in promoting this stabilization.  相似文献

We present a quantum chemical ab initio study which demonstrates a new combined experimental and theoretical approach, whereby a comparison of calculated and experimental (1)H NMR chemical shifts allows the elucidation of structural arrangements in solid-state molecular ensembles, taking advantage of the marked sensitivity of the (1)H chemical shift to intermolecular interactions. Recently, Brown et al. have shown that, under fast magic-angle spinning (MAS) at 35 kHz, the resolution in a (1)H NMR spectrum of the solid phase of an alkyl-substituted hexabenzocoronene (HBC) derivative is sufficient to observe the hitherto unexpected resolution of three distinct aromatic resonances ( J. Am. Chem. Soc. 1999, 121, 6712). Exploiting the additional information about proton proximities provided by (1)H double-quantum (DQ) MAS NMR spectroscopy, it was shown that the results are qualitatively consistent with the aromatic cores packing in a manner similar to that in unsubstituted HBC. Using the HBC-C(12) molecule as an example, we show here that the new combined experimental and theoretical approach allows the observed (1)H chemical shifts to be related in a quantitative manner to the intermolecular structure. In the quantum chemical calculations, a series of model systems of stacked HBC oligomers are used. On account of the marked dependence of the (1)H chemical shift to ring currents arising from nearby aromatic rings, the calculated (1)H chemical shifts are found to be very sensitive to the stacking arrangement of the HBC molecules. Moreover, the ring current effect is found to be particularly long range, with a considerable influence of the second neighbor, at a distance of 700 pm, being observed.  相似文献

A 6-alkyl-4,5-epoxy-6-hydroxy-2-cyclohexen-1-one derivative, a model compound for the hydrophilic moiety of scyphostatin, was stereoselectively synthesized from the Diels-Alder adduct. The key steps were the reductive cleavage of the 4,5-epoxide ring of the epoxidated adduct, the 1,3-carbonyl transposition of the 3-carbonyl group to the C1 position by a Wharton reaction and stereoselective bromination to provide a trans bromohydrin derivative, a precursor to the desired compound. Desilylation of the bromohydrin derivative with TBAF directly gave the target compound.  相似文献

The existence of a correlation has been established for pyrimidine but not for purine, nucleosides. It is suggested that the change in the chemical shift of the anomeric carbon is a consequence of 1,2-eclipsing interaction between O-2′ and N-1′ in the S type of conformation of the ribose ring. Possible reason for the absence of a correlation in the case of purine nucleosides are discussed. It is shown that the chemical shift of the anomeric carbon can be used in the conformational analysis of the ribose rings of pyrimidine nucleosides.  相似文献

The precise estimation of the degree of derivatization of functional groups in polymers is important for determining their macroscopic properties. In this work, the quantitative estimation of the extent of esterification of novolac copolymers with di-tert-butyl dicarbonate was studied. Although the extent of esterification has been calculated previously by quantifying the signals from FT-IR and UV-Vis spectroscopy, these were restricted to monitoring the progress of the derivatization process. The 13C NMR signal intensities from the inverse-gated 1H-decoupled NMR spectrum have been used recently for the quantitative estimation of the degree of esterification of polymers. An alternative methodology has been suggested by us based on the fully relaxed 1H chemical shift intensities. However, since the proton signals of novolac resins are generally broad and overlapping, the proton decoupled 13C NMR spectrum was used to identify the 1H NMR signals using the 2-D HSQC technique. A TOCSY experiment was also performed to confirm further the 1H NMR signal assignments and, finally, the deconvoluted 1H NMR spectrum was used for the calculation of the extent of derivatization.  相似文献

The minimal energy conformations of o-benzosemiquinone anion radical were calculated for several cases of assignments by using the INDO method and the molecular geometry adjusting method. In order to know the effect of lithium ion in the solvent, the minimal energy conformations of the system of Li? O? H and o-benzosemiquinone anion radical were calculated. The calculations of the minimal energy conformations of this radical in t-butyl alcohol, alkaline aqueous ethanol, alkaline water, neutral methanol, and acetonitrile were carried out. The total energies of the minimal energy conformations in the assignment |A3| > |A4| were lower than those in the assignment |A3| < |A4|.  相似文献

The proton spectra of the compounds propene (1) and 3-methylbutene-1 (2) have been measured in various solvents and, as neat liquids, at variable temperature. The effects of solvent and temperature on 1 are found to be small, but not negligible. The effect of temperature on the coupling constants of 2 are more pronounced, which is explained in terms of an anti-gauche equilibrium. To obtain a consistent ΔE° value from each temperature dependent coupling constant it is necessary to correct for the non-conformational temperature effects (obtained from 1) as well as to correct for changes in the geometry of the entire molecule, which are concurrent with a conformational transition. Such geometry changes have been calculated by a consistent force field technique, which, when coupled with INDO-MO calculated dependencies of coupling constants on valence (and torsional) angles, provided the changes in coupling constants. The observed temperature dependence of the coupling constants of the H2C?CH— moiety of 2 could be satisfactorily explained in this fashion. The difference in energy between the anti and gauche forms of 2 were determined as ΔE° = 127 ± 14 cal mol?1.  相似文献

Nine 2-(pyrazol-1-yl)-4-methylquinolines bearing substituents on the pyrazole 3- or 5-positions (H, Me, Et, i-Pr, t-Bu) were regioselectively synthesized either using the direct condensation of 2-chloro-4-methylquinoline and sodium salt of 3(5)-substituted pyrazoles or by treatment of 2-hydrazino-4-methylquinoline with an appropriate β-ketoaldehyde. The 1H and 13C chemical shifts were discussed taking into account the preferred conformation about the C-2-N-1′ bond as calculated by the AM1 Hamiltonian. It appears that 5-ethyl and 5-isopropyl substituted derivatives present short C-H-N-1 interactions. Ortho steric effects appear to be responsible for these conformations.  相似文献

[reaction: see text] The coupling of 2-(1-alkynyl)-2-alken-1-ones with nucleophiles, either catalyzed by AuCl3 or induced by an electrophile, provides highly substituted furans in good to excellent yields under very mild reaction conditions. Various nucleophiles, including functionally substituted alcohols, H2O, carboxylic acids, 1,3-diketones, and electron-rich arenes, and a range of cyclic and acyclic 2-(1-alkynyl)-2-alken-1-ones readily participate in these cyclizations. Iodine, NIS, and PhSeCl have proven successful as electrophiles in this process. The resulting iodine-containing furans can be readily elaborated to more complex products using known organopalladium chemistry.  相似文献

A new synthetic pathway to the parent and substituted ABCD ring cores of the camptothecin family of alkaloids was developed. The N-alkylation of 1,6-dihydro-6-oxo-2-pyridinecarbonitrile (2) with 3-bromo-1-phenylpropyne provided 3a using Curran's protocol. Treatment of 3a with a catalytic amount of DBU (5 mol %) at 110 degrees C for 12 h produced indolizino[1,2-b]quinolin-9(11H)-one (6a), the parent ABCD ring core of camptothecin, in essentially quantitative yield.  相似文献

Three coordination polymers, {[Co(C10H5N3O5)(H2O)2]·H2O}n (1), {[Mn3(C10H5N3O5)2Cl2(H2O)6]·2H2O}n (2), and {[Cu3(C10H4N3O5)2(H2O)3]·4H2O}n (3), based on a T-shaped tripodal ligand 4-(4,5-dicarboxy-1H-imidazol-2-yl)pyridine 1-oxide (H3DCImPyO), were synthesized under hydrothermal conditions. The polymers showed diverse coordination modes, being characterized by elemental analysis, infrared spectroscopy, and single-crystal X-ray structure analysis. In 1, the HDCImPyO2? generated a 1-D chain by adopting a μ2-kN, O : kN′, O′ coordination mode to bridge two Co(II) ions in two bis-N,O-chelating modes. In 2, the HDCImPyO2? adopted a μ3-kN, O : kO′, O′′ : O′′′ coordination mode to bridge two crystallographically independent Mn(II) ions, forming a 2-D hcb network with {63} topology. In 3, by adopting μ4-kN, O : kO′, O′′ : kN′′, O′′′ : O′′′′ coordination, DCImPyO3? bridged three crystallographically independent Cu(II) ions to form a 3-D framework having the stb topology.  相似文献

A sensitive and selective determination of glycyrrhizin (GC) based on surface plasmon resonance (SPR) was performed by using an anti-GC monoclonal antibody (GC-MAb) and GC-bovine serum albumin (GC-BSA) conjugate (antigen). GC-BSA was immobilized on an Au thin film of the SPR sensor chip by physical adsorption, and GC determinations were performed by an indirect competitive method. The addition of GC into the GC-MAb solution (5 microg/ml) was found to decrease the incident-angle shift sharply because of an inhibition effect of GC. The RSDs (n = 3) of each point were less than 4%. The lowest detection limit for GC by SPR was almost the same as that by ELISA, 60-75 ng/ml. An evaluation of the affinity constant between GC-MAb and GC using the data from ELISA and those from SPR measurements was performed. The values of the association constant (KA) from three different analyses of ELISA data and from SPR measurements are discussed in detail. As a whole, the affinity constant (KA) between GC-MAb and GC was on the order of 10(7) M(-1).  相似文献

The ionization constants of several 4- and 5-substituted-2-pyrrolcarboxylic acids were determined in water at 25·0°. A good fit in the Hammett equation was found using σm, and σP constants.
