Yi is the seventh-largest ethnic group in China and features mountainous regional characteristics. The Liangshan Yi Autonomous Prefecture is the largest Yi agglomeration with isolated geographical conditions, profoundly impeding genetic communication. Here, we investigated 427 unrelated males of Liangshan from 193 Y-chromosome single nucleotide polymorphisms (Y-SNPs) and 27 Y-chromosome short tandem repeats (Y-STRs) to reveal the genetic structure and paternal phylogeny of the group. The haplogroup diversity reached 0.9169 with 46 different subhaplogroups by 193 Y-SNPs, and the haplotype diversity reached 0.9999 by 27 Y-STR loci. Multidimensional scaling (MDS), N-J tree, and Network were constructed to decipher and visualize the genetic relations between Yi and worldwide groups. Our results revealed: (1) the Network by Y-STRs and Y-SNPs showed the haplogroup D1a1a-M15 in the Liangshan Yi population was a ramification of Tibetan groups’ expansion from west to east on the plateau; (2) the haplogroup distribution and the mismatch mutation analysis indicated the haplogroup O2a2b1a1a1a4a2-Z25929 of Liangshan Yi derived from manifold Southeast Asian immigrants; (3) a high-resolution Y-SNPs panel is vital to depict accurate paternal derivations and build an integrated and refining genetic structure of ethnic groups.  相似文献

The Y-chromosome short tandem repeats (Y-STRs) loci with different mutation rates existing in the Y chromosome non-recombination region (NRY) allow to be applied in human forensics, genealogical researches, historical investigations and evolutionary studies. Currently, there is a high demand for pedigree search to narrow the scope of crime investigations. However, the commonly used Y-STRs kits generally contain Y-STRs with high mutation rates that could cause individuals from the same pedigree to display different haplotypes. Herein, we put forward a new strategy of Slowly Mutating (SM) Y-STRs plus Y-SNPs typing, which could not only improve the resolution and accuracy of pedigree search, but also be applicable to evolutionary research. First, we developed a nine SM Y-STRs assay by evaluating their mutation rates in 210 pedigrees. Then the gene diversity and efficiency of the SM Y-STRs and 172 Y-SNPs sets were investigated by 2304 unrelated males from 24 populations. Furthermore, network and time estimation analyses were performed to evaluate the new strategy's capability to reconstruct phylogenetic tree and reliability to infer the time to the most recent common ancestor (TMRCA). The nine SM Y-STRs assay even had a higher resolution and a comparable capacity of revealing population genetic differentiation compared to 172 Y-SNPs system. This new strategy could optimize the phylogenetic tree generated by commonly used Y-STR panels and obtain a quite consistent time estimations with the published dating.  相似文献

Y chromosome kits are successfully applied in cases where human biological material exists. With the development of genotyping ability, more Y chromosomal markers are needed for finer identification of male individuals and lineages. In this study, a developmental validation of a newly emerged Y chromosome kit that combines two different kinds of markers: 38 Y-STRs and 3 Y-indels are conducted. The results show that this kit has high sensitivity when there is a small amount of DNA (125 pg), more than one male (minor:major = 1:7), or a mixture of males and females (male:female = 125pg:1875pg), inhibited substances (800 μM hematin and more than 1600 ng/μL humic acid). The kit exhibits high precision level with a standard deviation of allele size no more than 0.14 nt. Locus DYS481 shows the largest stutter rate, with three stutters per true allele. Population samples are well identified (MP of 0.001106), and mutations can be observed in father–son pairs (46 mutations in 70 pairs, 10 in locus DYS627). Out of all the population samples, 13.2% belong to haplogroup M117-O2a2b1a1, with their ethnic group being Han Chinese. The results show that this kit can improve the performance of identifying male individuals, obtaining more unique haplotypes (increasing from 894 to 918 of 1000 male samples) and higher discrimination capacity (increasing from 0.942 to 0.955) in this study compared to previous widely used Yfiler Plus kit. Besides, it gives information about their paternal lineages in forensic genetic casework and genealogical database construction.  相似文献

Y-chromosome SNP haplogroups exhibit geographic structuring in many populations around the world. Therefore, Y-chromosome haplogroups have been widely used to infer paternal biogeographical ancestry and high-resolution paternal lineage classification. In the present study, we designed a customized panel containing 183 Y-SNPs based on previous studies and evaluated the genotyping performance and repeatability, concordance, sensitivity, and ability of analysing case-type samples using a MALDI-TOF MS platform. The average call rate for duplicate typing of any one SNP in the panel was 97.0%. In the concordance and accuracy study, the results of haplogroup designation obtained from MALDI-TOF MS platform were fully consistent with those obtained from the next-generation sequencing (NGS) platform. The optimal amount of template DNA in the PCR seemed to be 10 ng. However, if less DNA (≥156.25 pg) was available, it was still possible to obtain meaningful haplogroup information. For the application part, this panel could be applied for the detection of blood, semen, and buccal swabs samples. Particularly, blood stain on FTA card samples could be dissected by direct PCR amplification on the MALDI-TOF MS platform. Besides, 371 unrelated male individuals from four Chinese ethnic groups (Han, Hui, Mongolian, and Kazak) were detected using this panel. Total 78 terminal haplogroups were found and the haplogroup diversity was 0.933576. The results demonstrate that this panel could be an accurate, fast, and cost-effective method for database construction where the amount of sample material is less of a concern and when the cost of the assay is taken into consideration.  相似文献

Acidity constants of six substituted 1-benzyl-1-methoxy-2-nitroethylenes (2-Z with Z = m-NO(2), m-CF(3), m-Cl, H, p-Me, p-MeO) and their respective nitronic acids were determined in 50% DMSO-50% water (v/v) at 20 degrees C. Kinetic data were obtained on the reversible deprotonation of all six 2-Z by OH(-) and piperidine and on the reversible deprotonation of 2-NO(2)() by piperazine, 1-(2-hydroxyethyl)piperazine, and morpholine in the same solvent. These data allowed a determination of the Br?nsted coefficients alpha (dependence on acidity of 2-Z) and beta (dependence on amine basicity). The fact that alpha > beta indicates the presence of a transition state imbalance which, however, is much smaller than that for the deprotonation of arylnitromethanes. The reasons for this reduction in the imbalance and their relevance to a recent study of the deprotonation of Fischer carbene complexes are discussed.  相似文献

Five new diterpene glycosides, virescenosides Z4-Z8 (1-5) have been isolated from a marine strain of Acremonium striatisporum KMM 4401 associated with the holothurian Eupentacta fraudatrix. Their structures were determined by HRESI-MS and 1D and 2D NMR. Virescenosides Z4-Z8 contain rare altruronic acid 4C1 and 1C4 conformations as their carbohydrate components. Virescenosides Z5 and Z7 exhibit an unusual 16-chloro, 15-hydroxyethylgroup as their side chains.  相似文献

The known aryne complex (PEt3)2Ni(eta2-C6H2-4,5-F2) (1a) reacts with a catalytic amount of Br2Ni(PEt3)2 over 1% Na/Hg to afford the dinuclear Ni(I) biarylyl complex [(PEt3)2Ni]2(mu-eta1:eta1-3,4-F2C6H2-3',4'-F2C6H2) (2a), which results from a combination of C-C bond formation and C-H bond rearrangement. The dinuclear benzyne [(PEt3)2Ni]2(mu-eta2:eta2-C6H2-4,5-F2) (3) was obtained by the reaction of 1a with a stoichiometric amount of Br2Ni(PEt3)2 over excess 1% Na/Hg, and 3 was found to catalyze the conversion of 1a to 2a. The reaction of 1a with B(C6F5)3 produced the trinuclear complex (PEt3)3Ni3(mu3:eta1:eta1:eta2-4,5-F2C6H2)(mu3:eta1:eta1:eta2-4,5-F2C6H2-4',5'-F2C6H2) (6). The addition of PEt3 to 6 produced 1 equiv of 1a and 1 equiv of [(PEt3)2Ni]2(mu-eta1:eta1-4,5-F2C6H2-4',5'-F2C6H2) (7a). Both 6 and 7a were identified as intermediates in the conversion of 1a to 2a. The analogue [(PEt3)(PMe3)Ni]2(mu-eta1:eta1-4,5-F2C6H2-4',5'-F2C6H2) (7b) was prepared by the addition of PMe3 to 6 and was structurally characterized. NMR spectroscopic evidence identified the additional asymmetric biarylyl [(PEt3)2Ni]2(mu-eta1:eta1-4,5-F2C6H2-3',4'-F2C6H2) (8a) during the conversion of 1a to 2a. The initial observation of 2 equiv of 8a for every equivalent of 2a produced from solutions of 7a suggests that 8a and 2a are formed from a common intermediate. A crossover labeling experiment shows that the C-H bond rearrangement steps in the conversion of 1a to 2a occur with the intermolecular scrambling of hydrogen and deuterium labels. The evidence collected suggests that Ni(I) complexes are capable of activating aromatic C-H bonds.  相似文献

Simmitecan (L-P) is an anticancer ester prodrug, which involves activation to chimmitecan (L-2-Z). In the current study, a liquid chromatography/tandem mass spectrometry-based method was developed for simultaneous determination of L-P and L-2-Z in various plasma samples. Because L-P is rapidly converted to L-2-Z by blood carboxylesterase during and after sampling, which hampers accurate determination of L-P and L-2-Z in the biological samples, different carboxylesterase inhibitors were tested. As a result, bis(4-nitrophenyl)phosphate gave the best results with respect to inhibitory capability, hemolysis, and matrix effects and was used to deactivate blood carboxylesterases when sampling. The plasma samples were precipitated with acetonitrile and the resulting supernatants were separated using a pulse gradient method on a C18 column. Irinotecan and camptothecin were used as internal standards for quantification of L-P and L-2-Z, respectively. Protonated L-P, L-2-Z and their internal standards were generated by electrospray ionization and their precursor-product ion pairs (m/z 599→124, 405→361, 587→195, and 349→305, respectively) were used for measurement. The newly developed bioanalytical assay processed favorable accuracy and precision with lower limits of quantification of 2.1 nM for L-P and 3.4 nM for L-2-Z, and was successfully applied to pharmacokinetic studies in tumor-bearing nude mice, rats, and dogs. There are substantial species differences in the ester activity. The experimental strategies illustrated in our report may be adopted for measurement of other prodrugs (including irinotecan) or drugs subject to ester hydrolysis, as well as their metabolites, in biological matrices.  相似文献

Lithium fluoroarylamidinates [(Ar(F)C(NSiMe(3))(2)Li)(n).xD] (Ar(F) = 4-CF(3)C(6)H(4), n = 2, D = OEt(2), x = 1 (2a); n = 1, D = TMEDA, x = 1 (4a); Ar(F) = 2-FC(6)H(4), n = 2, D = OEt(2), x = 1 (2b); Ar(F) = 4-FC(6)H(4), n = 2, D = OEt(2), x = 2 (2c); Ar(F) = 2,6-F(2)C(6)H(3), n = 2, D = OEt(2), x = 1 (2d); n = 2, D = 2,6-F(2)C(6)H(3)CN, x = 2 (3d); Ar(F) = C(6)F(5), n= 2, D = OEt(2), x = 1 (2e), n = 1, D = TMEDA, x = 1 (4e); n = 1, x = 2, D = OEt(2) (5e); D = THF (6e)) were prepared by the well-known method from LiN(SiMe(3))(2) and the corresponding nitrile in diethyl ether or by addition of the appropriate donor D to the respective diethyl ether complexes. Depending on the substituents at the aryl group and on the donors D, three different types of structures were confirmed by X-ray crystallography. Hydrolysis of 2e gave C(6)F(5)C(NSiMe(3))N(H)SiMe(3) (7e) and C(6)F(5)C(NH)N(H)SiMe(3) (8e). The lithium fluoroarylamidinates 2a-2d react with Me(3)SiCl to give the corresponding tris(trimethylsilyl)fluoroarylamidines Ar(F)C(NSiMe(3))N(SiMe(3))(2) (9a-9d). Attempts to prepare C(6)F(5)C(NSiMe(3))N(SiMe(3))(2) from 2e and Me(3)SiCl failed; however, the unprecedented cage [[C(6)F(5)C(NSiMe(3))(2)Li](4)LiF] (10e) in which a fluoride center is surrounded by a distorted trigonal bipyramid of five Li atoms was obtained from this reaction.  相似文献

The detailed mechanism for arylation of styrene or its α-CF3 substituted analog using aryliron complex [CpFe(CO)2Ar] was studied using density functional theory calculations. Results of calculations show that the arylation mechanism mainly involves three steps: (1) a ligand exchange process between a CO and styrene or its derivative; (2) migration of Ar group from Fe to β-C of styrene; (3) β-H (or β-F) elimination and dissociation of the stilbene derivative from the CpFeHCO (or CpFeFCO) moiety. Both of Steps (2) and (3) experience a similar four-memberred cyclic transition state. The dπ-pπ interaction stabilizes the CC π coordinated complexes and the agostic interaction plays an important role in stabilizing intermediates and promoting elimination of the β-H (or β-F if available). For arylation of the α-CF3 substituted styrene, our calculations clarified that the dissociation of ethylene derivatives to give PF (product for β-F elimination) is kinetically and thermodynamically more favorable than to give PH (product for β-H elimination), which is the determined step for the selectivity of the final products.  相似文献

[RuCl2(PPh3)3] reacts with thallium(I) fluoride to give either [Tl(mu-F)3Ru(PPh3)3] (1) or [Tl(mu3-F)(mu2-Cl)2Ru2(mu2-Cl)(mu2-F)(PPh3)4] (2) depending on the excess of TlF used. Both 1 and 2 were fully characterized, including X-ray structure determinations. Complex 1 reacts with dihydrogen to form the known ruthenium hydride complex [Ru(H)2(H2)(PPh3)3] upon hydrogenolysis of the Ru-F bond. The reaction of 1 with activated alkyl bromides (R-Br) gives the corresponding alkyl fluorides and the trinuclear complex [Tl(mu3-F)(mu2-F)(mu2-X)Ru2(mu2-Br)(mu2-F)(PPh3)4] (X=Br, F) (3), whose structure closely resembles that of 2. However, 1 is not active as catalyst for the nucleophilic fluorination of R-Br in the presence of thallium fluoride. The effect of the bridging coordination mode of fluoride on the Ru-F bond is discussed in terms of the HSAB principle, which suggests a more general model for predicting the stability of d6 and d8 complexes containing hard ligands (such as fluoro, oxo, and amido).  相似文献

The reaction of (C5Me5)2U(CH3)2 with 2 equiv of N[triple bond]C-ArF gives the fluorinated uranium(IV) bis(ketimide) complexes (C5Me5)2U[-N=C(CH3)(ArF)]2 [where ArF=2-F-C6H4 (4), 3-F-C6H4 (5), 4-F-C6H4 (6), 2,6-F2-C6H3 (7), 3,5-F2-C6H3 (8), 2,4,6-F3-C6H2 (9), 3,4,5-F3-C6H2 (10), and C6F5 (11)]. These have been characterized by single-crystal X-ray diffraction, 1H and 19F NMR, cyclic voltammetry, UV-visible-near-IR absorption spectroscopy, and variable-temperature magnetic susceptibility. Density functional theory (DFT) results are reported for complexes 6 and 11 for comparison with experimental data. The most significant structural perturbation imparted by the F substitution in these complexes is a rotation of the fluorinated aryl (ArF) group out of the plane defined by the N=C(CMe)(Cipso) fragment in complexes 7, 9, and 11 when the ArF group possesses two o-fluorine atoms. Excellent agreement is obtained between the DFT-calculated and experimental crystal structures for 11, which displays the distortion, as well as for 6, which does not. In 7, 9, and 11, the out-of-plane rotation results in large angles (phi=53.7-89.4 degrees) between the planes formed by ketimide atoms N=C(CMe)(Cipso) and the ketimide aryl groups. Complexes 6 and 10 do not contain o-fluorine atoms and display interplanar angles in the range of phi=7-26.8 degrees. Complex 4 with a single o-fluorine substituent has intermediate values of phi=20.4 and 49.5 degrees. The distortions in 7, 9, and 11 result from an unfavorable steric interaction between one of the two o-fluorine atoms and the methyl group [-N=C(CH3)] on the ketimide ligand. All complexes exhibit UV/UIV and UIV/UIII redox couples, although the distortion in 7, 9, and 11 appears to be a factor in rendering the UIV/UIII couple irreversible. The potential separation between these couples remains constant at 2.15+/-0.03 V. The electronic spectra are dominated by unusually intense f-f transitions in the near-IR that retain nearly identical band energies but vary in intensity as a function of the fluorinated ketimide ligand, and visible and near-UV bands assigned to metal (5f)-to-ligand (pi*) charge-transfer and interconfiguration (5f2-->5f16d1) transitions, respectively. Variable-temperature magnetic susceptibility data for these complexes indicate a temperature-independent paramagnetism (TIP) below approximately 50 K that results from admixing of low-lying crystal-field excited states derived from the symmetry-split 3H4 5f2 manifold into the ground state. The magnitude of the TIP is smaller for the complexes possessing two o-fluorine atoms (7, 9, and 11), indicating that the energy separation between ground and TIP-admixed excited states is larger as a consequence of the greater basicity of these ligands.  相似文献

Starting material KN(H)C(6)H(3)-2,6-F(2) was prepared via a transamination reaction from KNH(2) and 2,6-F(2)C(6)H(3)NH(2) in THF and crystallized from 1,4-dioxane (diox) as the three-dimensional polymer [(diox)(1.5)K{N(H)-2,6-F(2)C(6)H(3)}.diox(0.5)](infinity) (1). The metathesis reaction of (THF)(4)CaI(2) with KN(Me)Ph in THF yields monomeric (THF)(4)Ca[N(Me)Ph](2) (2) with a nearly linear N-Ca-N moiety of 179.84(8) degrees . The metathesis reaction of (THF)(4)CaI(2) with KN(H)Mes yields trinuclear (THF)(6)Ca(3)[N(H)Mes](6) (3) with a linear Ca(3) fragment and bridging 2,4,6-trimethylphenylamido groups. The reaction of 1 with (THF)(4)CaI(2) gives dinuclear (THF)(5)Ca(2)[N(H)-2,6-F(2)C(6)H(3)](4).2THF (4) with three bridging and one terminally bound 2,6-difluorophenylamide. A similar reaction of (THF)(5)SrI(2) with KN(H)-2,6-F(2)C(6)H(3) yields dinuclear (THF)(6)Sr(2)[N(H)-2,6-F(2)C(6)H(3)](3)I.THF (5) in which the iodide anion binds terminally. This iodide ligand cannot be substituted as easily by excess KN(H)-2,6-F(2)C(6)H(3). The metathesis reaction of (THF)(5)BaI(2) with KN(H)-2,6-F(2)C(6)H(3) leads to the formation of [(THF)(2)Ba{N(H)-2,6-F(2)C(6)H(3)}(2)](infinity) (6) which crystallizes as a one-dimensional polymer with bridging 2,6-difluorophenylamide anions and additional Ba-F-bonds.  相似文献

Copolymerization of fluorine ring-substituted 2-phenyl-1,1-dicyanoethenes, RC6H3CH?C(CN)2 (R is 2,3-F,F, 2,4-F,F, 2,5-F,F, 2,6-F,F, and 4-CF3) with 4-fluorostyrene were prepared in the presence of a radical initiator (ABCN) at 70°C. The composition of the copolymers was calculated from nitrogen analysis, and the copolymers were characterized by IR, 1H and 13C-NMR, GPC, DSC, and TGA. The monomer reactivity ratios for 4-fluorostyrene (M1), r1 = 0.6 and 2-(2,4-difluorophenyl)-1,1-dicyanoethene (M2), r2 = 0 were determined from Fineman-Ross plot. The order of relative reactivity (1/r1) for difluoro-substituted monomers is 2,4-F,F (0.31) > 2,3-F,F (0.25) > 2,5-F,F (0.22) > 2,6-F,F (0.10). DSC curves showed that the copolymers were amorphous with high T g in comparison with that poly(4-fluorostyrene) indicating a substantial decrease in chain mobility of the copolymer due to the high dipolar character of the trisubstituted ethylene monomer units. From the thermogravimetric analysis, the copolymers began to degrade in the range 214–260°C. The copolymer of 4-fluorostyrene and 2-(2,4-difluorophenyl)-1,1-dicyanoethene and poly(4-fluorostyrene) were dielectrically characterized in the range 25–200°C. The dominating relaxation process detected in both materials was the α-relaxation, associated with the dynamic glass transition. The relationship polarity-permittivity was discussed.  相似文献

The effects of different terphenyl ligand substituents on the quintuple Cr-Cr bonding in arylchromium(I) dimers stabilized by bulky terphenyl ligands (Ar) were investigated. A series of complexes, ArCrCrAr (1-4; Ar = C6H2-2,6-(C6H3-2,6-iPr2)2-4-X, where X = H, SiMe3, OMe, and F), was synthesized and structurally characterized. Their X-ray crystal structures display similar trans-bent C(ipso)CrCrC(ipso) cores with short Cr-Cr distances that range from 1.8077(7) to 1.8351(4) A. There also weaker Cr-C interactions [2.294(1)-2.322(2) A] involving an C(ipso) of one of the flanking aryl rings. The data show that the changes induced in the Cr-Cr bond length by the different substituents X in the para positions of the central aryl ring of the terphenyl ligand are probably a result of packing rather than electronic effects. This is in agreement with density functional theory (DFT) calculations, which predict that the model compounds (4-XC6H4)CrCr(C6H4-4-X) (X = H, SiMe3, OMe, and F) have similar geometries in the gas phase. Magnetic measurements in the temperature range of 2-300 K revealed temperature-independent paramagnetism in 1-4. UV-visible and NMR spectroscopic data indicated that the metal-metal-bonded solid-state structures of 1-4 are retained in solution. Reduction of (4-F3CAr')CrCl (4-F3CAr' = C6H2-2,6-(C6H3-2,6-iPr2)2-4-CF3) with KC8 gave non-Cr-Cr-bonded fluorine-bridged dimer {(4-F3CAr')Cr(mu-F)(THF)}2 (5) as a result of activation of the CF3 moiety. The monomeric, two-coordinate complexes [(3,5-iPr2Ar*)Cr(L)] (6, L = THF; 7, L = PMe3; 3,5-iPr2Ar* = C6H1-2,6-(C6H-2,4,6-iPr3)2-3,5-iPr2) were obtained with use of the larger 3,5-Pri2-Ar* ligand, which prevents Cr-Cr bond formation. Their structures contain almost linearly coordinated CrI atoms, with high-spin 3d5 configurations. The addition of toluene to a mixture of (3,5-iPr2Ar*)CrCl and KC8 gave the unusual dinuclear benzyl complex [(3,5-iPr2Ar*)Cr(eta3:eta6-CH2Ph)Cr(Ar*-1-H-3,5-iPr2)] (8), in which a C-H bond from a toluene methyl group was activated. The electronic structures of 5-8 have been analyzed with the aid of DFT calculations.  相似文献

The isomeric S-methyldihydropyrrins 9-Z and 9-E exhibit markedly different behavior in Pd(0)-catalyzed cross-coupling reactions. Thioimidates 9-Z are readily converted to imines 10-Z employing Pd(0)/AlkZnI. Under identical conditions 9-E are inert. Oxidative addition to Pd(0) requires activation by Zn or other Lewis acids, which is sterically unfavorable with 9-E. Analogous results were obtained with the related thioimidates 11-E,Z as well as with methylthiopyridines 19alpha-gamma. In the case of both 11 and 19 oxidative addition to Pd(0) was greatly facilitated in the presence of BF(3) x Et(2)O. The importance of Lewis acid activation to Pd(0) oxidative addition in such substrates appears to be a general phenomenon not previously recognized.  相似文献

The visual pigment rhodopsin is a seven-transmembrane (7-TM) G protein-coupled receptor (GPCR). Activation of rhodopsin involves two pH-dependent steps: proton uptake at a conserved cytoplasmic motif between TM helices 3 and 6, and disruption of a salt bridge between a protonated Schiff base (PSB) and its carboxylate counterion in the transmembrane core of the receptor. Formation of an artificial pigment with a retinal chromophore fluorinated at C14 decreases the intrinsic pKa of the PSB and thereby destabilizes this salt bridge. Using Fourier transform infrared difference and UV-visible spectroscopy, we characterized the pH-dependent equilibrium between the active photoproduct Meta II and its inactive precursor, Meta I, in the 14-fluoro (14-F) analogue pigment. The 14-F chromophore decreases the enthalpy change of the Meta I-to-Meta II transition and shifts the Meta I/Meta II equilibrium toward Meta II. Combining C14 fluorination with deletion of the retinal beta-ionone ring to form a 14-F acyclic artificial pigment uncouples disruption of the Schiff base salt bridge from transition to Meta II and in particular from the cytoplasmic proton uptake reaction, as confirmed by combining the 14-F acyclic chromophore with the E134Q mutant. The 14-F acyclic analogue formed a stable Meta I state with a deprotonated Schiff base and an at least partially protonated protein counterion. The combination of retinal modification and site-directed mutagenesis reveals that disruption of the protonated Schiff base salt bridge is the most important step thermodynamically in the transition from Meta I to Meta II. This finding is particularly important since deprotonation of the retinal PSB is known to precede the transition to the active state in rhodopsin activation and is consistent with models of agonist-dependent activation of other GPCRs.  相似文献

Novel trisubstituted ethylenes, halogen ring-substituted butyl 2-cyano-3-phenyl-2-propenoates, RPhCH ? C(CN)CO2C4H9 (where R 2-Br, 3-Br, 4-Br, 2-Cl, 3-Cl, 4-Cl, 2-F, 3-F, 4-F) were prepared and copolymerized with styrene. The monomers were synthesized by the piperidine catalyzed Knoevenagel condensation of ring-substituted benzaldehydes and butyl cyanoacetate, and characterized by CHN analysis, IR, 1H and 13C-NMR. All the ethylenes were copolymerized with styrene (M1) in solution with radical initiation (ABCN) at 70°C. The compositions of the copolymers were calculated from nitrogen analysis and the structures were analyzed by IR, 1H and 13C?NMR. The order of relative reactivity (1/r1) for the monomers is 3-F (22.97) > 3-Br (6.05) > 2-Cl (4.32) > 4-F (3.19) > 4-Br (2.47) > 4-Cl (1.92) > 2-F (1.74) > 3-Cl (1.10) > 2-Br (1.07). Decomposition of the copolymers in nitrogen occurred in two steps, first in the 200–500°C range with residue (2.3–5.9% wt.), which then decomposed in the 500–800°C range.  相似文献
