Summary Capillary electrophoresis with an electrolyte containing cyclodextrin was investigated for the simultaneous separation of the diastereoisomers of 6R,S-leucovorin and its active metabolite 6R,S-5-methyl-tetrahydrofolate. , and -cyclodextrin separated the diastereoisomers of 5-methyl-tetrahydrofolate, while only -cyclodextrin was found to be effective for the chiral separation of leucovorin. The effect of -cyclodextrin concentration was investigated, and subsequently a curve-fitting analysis for the quantitative estimation of the binding constants was attempted. The binding constants were found to be very small, in the range 2–4 M–1. Although the interaction between -cyclodextrin and the tetrahydrofolates is weak, the high efficiency of capillary electrophoresis and the use of high concentrations of -cyclodextrin allow baseline chiral separation of the diastereoisomers of leucovorin and 5-methyl-tetrahydrofolate. Changes in temperature exert differing effects on the separations of leucovorin and 5-methyl-tetrahydrofolate; higher temperatures improved the separation of leucovorin diastereoisomers but reduced the resolution of 5-methyl-tetrahydrofolate diastereoisomers. The effects of urea and buffer salt concentrations and of buffer pH were also investigated. Capillary electrophoresis with -cyclodextrin was used to analyse plasma samples spiked with clinically-relevant levels of leucovorin and 5-methyl-tetrahydrofolate. Resolution of these compounds in ultrafiltered plasma was demonstrated, but detection sensitivity was not adequate for the routine use of this method for the determination of leucovorin and 5-methyl-tetrahydrofolate in plasma. In addition, a simple technique to reverse the elution order of ionic stereoisomers was demonstrated. By adding a cationic surfactant into the buffer and reversing the separation potential, the elution order of the diastereoisomers of leucovorin and 5-methyl-tetrahydrofolate was reversed.  相似文献

The enantiomeric separation of 12 chiral dihydrobenzofurans is reported on derivatized -cyclodextrin stationary phases using high performance liquid chromatography. The hydroxypropyl -cyclodextrin chiral stationary phase (CSP) with acetonitrile/water mobile phases was the most effective combination as it baseline separated 9 of the 12 compounds. The acetyl -cyclodextrin and 2,3-dimethyl -cyclodextrin CSPs were also effective in the reverse phase mode. The native -cyclodextrin was far less effective than the non-aromatic derivatized CSPs. The aromatic functionalized CSPs showed no selectivity in the normal phase mode. Structural characteristics, such as substituent polarity and ring location, were important in the observed enantioselectivity.  相似文献

Summary The interaction between cyclodextrin and the drug (1R,2S,3S,4S)-(5Z)-7-(3-((phenylsulfonyl)amino)bicyclo[2.2.1]hept-2-yl)hept-5-enoic acid ((+)-S-145), was studied using -, -, and -cyclodextrin bonded-phase columns. Retention behavior of (+)-S-145 on these columns revealed that the strength of inclusion was -cyclodextrin. Interaction between -cyclodextrin and (+)-S-145 was found to increase as the proportion of carboxylic ion in the (+)-S-145 molecule increased. Comparison of binding capacities of these bondedsilica gels obtained by frontal analysis and surface coverage indicated that availability of the immobilized - and -cyclodextrin was 20–25%. The synthesized -cyclodextrin bonded-phase column was superior to that of commercial columns in terms of chiral separation of (±)-S-145. A typical usage of the -cyclodextrin column is discussed for separation of (±)-S-145 in plasma samples.  相似文献

Summary Capillary zone electrophoresis with -cyclodextrin as electrolyte additive has been investigated for the separation of the diastereomers of a 1,3-disubstituted tetrahydro--carboline. The effects of pH, buffer concentration, -cyclodextrin concentration and applied voltage on resolution were investigated. This study shows that optimizing the assay conditions leads to maximum chiral separation or resolution. The method enables further research on the quantitative analysis of these compounds.  相似文献

An enantioselective silica rod type chiral stationary phase (CSP) is presented; a novel combination of the well known enantiomer separation properties of -cyclodextrin and the unique properties concerning the flow behavior of silica monoliths. Two different synthesis routes are described, and it was found that the in situ modification of a plain silica rod column turned out to be the best. The chromatographic behaviour of the -cyclodextrin silica rod was studied and compared with a very similar commercially available -cyclodextrin bonded particulate material (ChiraDex®). Even if the amount of -cyclodextrin bound to the silica rod was only about half of the amount of -cyclodextrin bound to ChiraDex® particles, good resolutions were achieved for a set of chiral test components like Chromakalin, Prominal, Oxazepam, Methadone and some other drugs. By taking advantage of the unique features of the silica rods relating to their flat H/u (Van Deemter) curves, fast enantiomer separations could be demonstrated.  相似文献

Aim of the present work was to investigate the effect of somecyclodextrins (CDs) on the solubility and ocular bioavailability of rufloxacin base (RUF), with theultimate goal of developing an ophthalmic formulation. Phase solubility studies of RUF inpH 7.4 buffer were carried out in the presence of -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD) and -cyclodextrin(-CD). The effect of hydroxypropyl methylcellulose (HPMC) on RUF solubility was evaluated after heating the solutionscontaining HP--CD at 120 °C.A significant enhancement of RUF solubility was achieved by associatingthe drug with CDs, particularly HP--CD. This CD formed with RUF a less stablecomplex than that formed by -CD, but did not suffer the solubility limitations ofthe parent CD, and showed a higher solubilizing capacity than -CD. Addition of 0.25%(w/v) HPMC to solutions containing HP--CD increased the solubilizing effect of this CD,thus allowing reduction of the amount necessary for solubilization of 0.3% (w/v) RUF.Preliminary pharmacokinetic data in rabbits indicated that theocular bioavailability of 0.3% (w/v) RUF solubilized by HP--CD was higher when compared witha 0.3% (w/v) RUF suspension used as reference.  相似文献

The binding of psychopharmaceutical drugs by -cyclodextrin (-CD) changed their effective electrophoretic mobilities. Based on this phenomenon the interactions between some psychopharmaceutical drugs and -cyclodextrin were studied and the binding constants of -CD with tiapride, imipramine, clomipramine, amitriptyline, trifluoperazine, perphenazine and carbamazepine were determined by affinity capillary electrophoresis. The presence of -CD improved the separation of these psychopharmaceutical drugs. Organic modifier was used as another kind of additive to be added in the running buffer to improve further the separation. 70 mM pH 7.2 phosphate buffer solution containing 12 mM -cyclodextrin and 15% (v/v) acetone was selected as optimal running buffer for separation by capillary electrophoresis.  相似文献

Inclusion complexation of all-trans-retinol, retinal and retinoic acid with -cyclodextrin (-CD) and heptakis(2,6-O-dimethyl)--cyclodextrin (DM--CD) were investigated by means of UV-vis spectroscopy. The association constants (K a) obtained for vitamin A with DM--CD is greater than with -CD. The influence of cyclodextrins (CDs) on the chemical stability of the contraceptive steroid progestin, ST1435, in aqueous solution has been studied using reversed phase high performance liquid chromatography. The effects of CD structure, temperature, and CD concentration on the rate of degradation were investigated. It was found that the drug degraded to different extents following a pseudo-first order reaction mechanism. The presence of the host molecules affected the degradation rate as a result of complexation which might result in protection of the labile moiety of the drug molecule against degradation. Hydroxypropyl--cyclodextrin (HP--CD) and hydroxyethyl--cyclodextrin (HE--CD) retarded the degradation in contrast to -cyclodextrin (-CD) which accelerated the steroid degradation. The stabilizing action of HP--CD is larger than that of HE--CD. The degradation rate increased upon increasing temperature and the Arrhenius equation is valid. Lineweaver-Burk equation analysis indicated that the steroid included inside the CD cavity degraded three times more slowly than did the free ST1435 in solution. This equation further supported the formation of a 1 : 1 inclusion complex between ST1435 and HP--CD with a stability constant of 934.5 M-1 at 65°C.  相似文献

The separation of enantiomers of amino acid derivatives with modifiers of different structures on -cyclodextrin and aminated -cyclodextrin by reversed-phase high-performance liquid chromatography was studied. The dependence of the capacity factors of adsorbates on the concentration of the organic component in the mobile phase was examined. It was found that the retention of amino acid derivatives on unmodified -cyclodextrin and aminated -cyclodextrin increases with the hydrophobicity of the modifying agent of amino acid. In addition, for aminated -cyclodextrin, electrostatic interactions of ionized adsorbates and protonated surface amino groups substantially contribute to the retention. It was demonstrated that the recognition ability of aminated -cyclodextrin is affected by the structure of amino acid and its degree of dissociation.  相似文献

The interactions of nabumetone (NAB) with -cyclodextrin (-CD) and-cyclodextrin (-CD) were studied in aqueous solution by meansof phase-solubility analysis. Solid dispersions of NAB with -cyclodextrin(-CD), -cyclodextrin (-CD), methyl- (M-CD),hydroxypropyl-cyclodextrin (HP-CD) were prepared by coevaporationand kneading and also by coprecipitation in the case of -CD. X-ray diffractometry, thermal analysis and infrared spectroscopy (FTIR) were used to study the possibility of complexation of the drug with the different cyclodextrins. Solid dispersions of nabumetone with -CD showed a remarkable improvement in the dissolution rate of nabumetone.  相似文献

Stoichiometric amounts of triethylamine (TEA) were found to enhance the chiral induction by -cyclodextrin (-CD) in the reduction of acetophenone (ACPH) by aqueous NaBH4. The enantioselectivity obtained depends upon the molar ratio -CD:ACPH:TEA. Evidence for the formation of a three-component inclusion compound was obtained from detailled1H and2H NAM studies. The restriction of the molecular motion of the prochiral center probably accounts for the strong enhancement of the chiral induction observed.  相似文献

The inclusion complex of the anti-inflammatory drug, loxoprofen, with -cyclodextrin-(CD), sulfated -CD, and glycerol ether -CD was studied by UV-VIS absorption and 1H-NMR spectroscopy in solution. The inclusion complex of loxoprofen with -CDs was prepared by freeze-drying, and then characterized in the solid state by thermal analysis, X-ray diffraction, FT-IR and FT-Raman spectroscopy, and scanning electron microscopy (SEM). Furthermore, a physical mixture of loxoprofen/-CD (1/1, mol-%) in the solid state was also characterized. The solubility of the loxoprofen increased on addition of -CDs. The solubility enhancement of the loxoprofen with -CDs is in the following order: glycerol ether -CD > sulfated -CD > -CD.  相似文献

-cyclodextrin-coated sensors chips were obtained by grafting amino--cyclodextrin or amino-polymers of -cyclodextrin (poly--CD-NH2) to functionalized gold surfaces. An additional carboxymethylated dextran bearing adamantyl groups (Ad-Dex-COOH) was then immobilized onto the surface by formation of inclusion complexes between -cyclodextrin cavities and adamantyl groups. Such multilayered structures were stable in aqueous media. However, the initial -cyclodextrin-coated surface could be recovered by using sodium dodecylsulfate solutions (SDS). After activation withN-hydoxysuccinimide, dextran-coated sensor chips were used to bind antibodies. The immunoreactivity of the resulting biosensors was examined. Moreover, conditions leading to the complete regeneration of the initial surface were investigated. Throughout this study, interfacial adsorption and desorption phenomena were followed in real time by an optical technique, Surface Plasmon Resonance (SPR).  相似文献

Asymmetric halogenation and hydrohalogenation of styrene in microcrystalline cyclodextrin complexes were studied in the gas-solid state, and compared with the homogenous reactions in aqueous or dimethyl sulfoxide solutions. The gas-solid brominations in the - and -cyclodextrin complexes produced predominantly (–)-1,2-dibromo-l-phenylethane. The chiral induction for the reaction of the -cyclodextrin complex rose to 9 times that of the -cyclodextrin complex. Brominations in the homogenous solutions containing the - or -cyclodextrin complexes gave no dibromide but racemic bromohydrin. In the gas-solid chlorination, the -cyclodextrin complex gave (–)-dichloride,S-(+)-2-chloro-l-phenylethanol (14% ee) and (+)-1,2,2-trichloro-l-phenylethane, and the -cyclodextrin complex produced (+)-dichloride,S-(+)-chlorohydrin (8% ee) and (+)-trichloride. The chiral induction of the gas-solid halogenation using the solid cyclodextrin complexes is attributed to the ability to hold rigidly a chiral conformation of the crystalline state. The chiral recognition by cyclodextrins and permethylated cyclodextrins have been investigated on the basis of the X-ray data of crystalline inclusion complexes. The macrocyclic ring of - and -cyclodextrin shows a round and symmetrical structure. -Cyclodextrin includes racemic 1-phenylethanol with the statistical disorder of the hydroxyl group. A pair of the R- and S-isomers of flurbiprofen are included within the cylindrical cavity formed by dimeric -cyclodextrin molecules with a head-to-head arrangement. The macrocyclic ring of permethylated cyclodextrins is remarkably distorted from the regular polygonal symmetry and has more flexibility in the conformational change than cyclodextrins. Owing to the distorted conformation and steric hindrance involving methyl groups, permethylated cyclodextrins do not equally include both isomers, as demonstrated by the permethylated -cyclodextrin complexes with mandelic acids. Permethylated -cyclodextrin binds D-mandelic acid more tightlyvia a host-guest hydrogen bond and induced-fit conformational change. Permethylated -cyclodextrin forms a hydrated crystalline complex with R-flurbiprofen, but S-flurbiprofen forms a non-hydrated crystalline complex. Significant differences between the two complexes are found in the orientation of the phenyl group and hydrogen-bond formation involving the carboxyl group.  相似文献

A series of sulfoalkyl ether -cyclodextrin derivatives, including sulfoethyl, sulfopropyl and sulfobutyl ethyl -cyclodextrins, have been synthesized and characterized. Each sulfoalkyl ether -cyclodextrin is a mixture of various degrees of substitution and different positional isotherms. Elemental analysis, 1H NMR, MS, and Differential Scanning Calorimetry analysis were used to determine the average degree of substitution for each -cyclodextrin derivative. The average degrees of substitution are 3.4, 1.6 and 2.5 for sulfoethyl, sulfopropyl and sulfobutyl ether -cyclodextrin, respectively. The water solubility of these derivatives is substantially higher than that of -cyclodextrin. 1H NMR indicates thatsulfoethyl ether -cyclodextrin may have major substitution on the secondaryhydroxyl group while the major substitution in sulfopropyl and sulfobutyl ether-cyclodextrin could be on the primary hydroxyl group. MS spectra showthat no more than one substitution occurred on a single glucose unit.  相似文献

The synthesis of the following neutral plasmalogens has been effected: -O-(octadec-1-enyl)-, distearoylglycerol, -O-(octadec-1-enyl)-, -dipalmitoylglycerol, -O-(hexadec-1-enyl)-, -distearoylglycerol, and -O-(pentadec-1-enyl)-, -dipalmitoylglycerol.2. The crystal structures of two forms of -cyclodextrin barbital complexes (Form I and Form II) were investigated by X-ray analysis. In Form I crystal, two -cyclodextrin(-CyD) molecules including two barbital molecules form a dimeric structure by the hydrogen bonds among their secondary hydroxyl groups. The unit cell volume of Form II is about twice as large as that of Form I and there exist two -CyD dimers in Form II. Although the (Host)/(Guest) ratio is 1/1, only one barbital molecule can be found in Form II at the present stage. The sinilarities and differences between the two crystal structures are mainly discussed here.  相似文献
