A very simple and facile one‐pot method for the synthesis of pyrimidine derivatives is presented. In this method, 2‐aryl‐pyrimidine derivatives were prepared by the reaction of ketene N,S‐acetal 1 with commercially available aryl aldehydes by heating under solvent and catalyst‐free conditions. The synthesized pyrimidine derivatives showed blue‐green fluorescence in solid state.  相似文献

A new and easy method for the diastereoselective synthesis of 3-functionalized 2,3-dihydrobenzofuran derivatives from allyl 2-bromoaryl ethers is described. The key step of this transformation involves an intramolecular carbolithiation reaction of allyl 2-lithioaryl ethers. The substituents in both the allyl and the aryl moieties play an important and decisive role in stopping the reaction at the benzofuran thus avoiding a gamma-elimination reaction. Finally, this process is amenable to the synthesis of enantiomerically enriched compounds by using (-)-sparteine as a chiral inductor.  相似文献

A series of furo[2,3-d]pyrimidine derivatives were synthesized via the [3+2] cyclization of pyrimidine-4,6-diol and a variety of nitroolefins at catalyst-free conditions. The reaction is easy to perform simply mixing inexpensive starting materials in water under conventional heating at 90 °C. The reaction proceeds at a fast speed within 1.5–2 h and gives the high biological and pharmacological active substituent furo[2,3-d]pyrimidine derivatives with good to high yields. Mechanism of formation of these furo[2,3-d]pyrimidine derivatives are also proposed.  相似文献

A convenient method to obtain unknown chiral C2- and C3-functionalized aldono-1,4-lactone derivatives starting from l-ascorbic acid, which would be valuable in the synthesis of derivatives of various pharmacologically active agents for structure-activity studies, is described. The practicality of this approach is demonstrated by the synthesis of a series of 5,6-O-isopropylidene-2-allyl-3-keto-l-galactono-gamma-lactone and 5,6-O-isopropylidene-3-allyl-2-keto-l-galactono-gamma-lactone derivatives using the thermal Claisen rearrangement of the corresponding 3-O- and 2-O-allyl derivatives of 5,6-O-isopropylidene-l-ascorbic acid, respectively, followed by stereospecific reduction to the corresponding alcohols. The synthetic steps are shown to be efficient, and enantiospecific, and they proceed with high yields.  相似文献

Several kinds of cycloaddition reactions were applied to C3-ethynylated pyropheophorbide-a methyl ester to develop C3-functionalized chlorophyll derivatives. Its [2+2] cycloaddition with tetracyanoethylene, [2+3] cycloaddition with diazomethane, [2+3] Huisgen reaction with trimethylsilyl azide, and [2+4] Diels–Alder reaction with tetraphenylcyclopentadienone gave the expected products in moderate to high yields. Zinc complex of the resulting 3-pyrazolyl-chlorin was found to show self-aggregation in a less polar solvent, in an aqueous micelle, and in the solid thin films more readily than the corresponding zinc 3-triazolyl-chlorin.  相似文献

An efficient and high-yielding synthesis of N~2-arylaminopyrimidine-5-carbonitrile derivatives starting from arylamines and 2-methylthio-pyrimidine-5-carbonitriIe derivatives has been developed in the presence of cesium carbonate as basic reagent.This new protocol showed high chemical tolerance for a range of functional groups,and only the methylthio substituent on C2 of the pyrimidine ring was replaced with arylamine derivatives under the reaction conditions.  相似文献

o‐Aminothiophene dicarbonitrile 1 on neat reaction with cyclic ketones in anhydrous ZnCl2 yielded mixture of fused aminopyridine 3 and iminospirooxazine 4 derivatives. Similarly, pyrimidine derivatives 5 and 8 were obtained by the reaction of this intermediate 1 with formic acid and DMF‐DMA followed by hydrazine hydrate, respectively. The reaction of o‐amino‐thiophene dicarboxamide 2 at ambient temperature with cyclic ketones yielded spiropyrimidine 10 as a sole product in quantitative yield. The regioselective anellated pyrimidine 9 , 11 , and dihydropyrimidine 12 derivatives were also obtained by the reaction with aromatic aldehydes in presence of piperidine and iodine respectively. J. Pyrano[4,3‐d]pyrimidine derivative 3 was prepared by reaction of chlorocarbonyl isocyanate 1 with enaminonitrile 2 . Compound 3 reacted with nitrogen nucleophiles 4a‐f to afford 2‐substituted pyrido[4,3d]pyrimidine 5–8 , pyrimido[i]1,5a‐diaza‐9‐oxafluorene 9 and pyrimido[i]5a‐aza‐9‐thiafluorene 10 derivatives. Also, compound 3 reacted with active methylene compounds 4j to yield pyrimidine derivatives 14–16 which on reaction with EtONa 4k afforded 1,5,7‐triaza‐10‐oxaphenanthrene derivatives 17–19 .  相似文献

An efficient synthetic method for 4-functionalized quinoline derivatives, 4-((1,3-dithian-2-ylidene)methyl)quinolines, has been developed. Mediated by trifluoromethanesulfonic acid, ethynyl ketene-S,S-acetals can react in a one-pot procedure with various arylamines and aldehydes under mild conditions to give the corresponding quinoline derivatives in good to high yields via a consecutive arylimine formation, regiospecific aza-Diels-Alder (Povarov) reaction, and reductive amination.  相似文献

The reaction of butyrolactone acetal with CH acids was used to synthesize 2-methylenetetrahydrofuran derivatives. The latter react with dimethylformimide acetal to give a dieneamine that is capable of undergoing cyclization to furo [3,2-c]pyrimidine derivatives. This two-ring system was also synthesized by the reaction of cyanacetamide with 3-dimethylaminomethylenebutyrolactone acetal. Continuing investigations in the field of the synthesis of condensed heterocyclic systems containing pyrrole and indole fragments ¦1–3¦, we have condensed of 2-aminopyrrole derivatives with various 1, 3-dicarbonyl compounds. The reaction took place at the boil in solutions in pyridine or acetic acid, and also without a solvent at 150–160° C, forming in a single stage derivatives of pyrrolo[1, 2-a]pyrimidine (I–III) and pyrrolo[2, 1-b]-tetrahydroquinazoline (IV–VI). The condensation of 2-aminopyrrole with acetoacetic and substituted acetoacetic esters led to 4-oxo derivatives of pyrrolo[1, 2-β]pyrimidine (VII and VIII).  相似文献

Condensation of 3-amino-4-cyanopyrazole (1) with ethylacetoacetate, ethyl cyanoacetate, diethyl malonate and acetylacetone afforded pyrazolo[1,5-a]pyrimidine derivatives (2—8a). Other compounds (8b—h) of this ring system were obtained by treatment of 1 with arylidenemalononitrile and ethylarylidenecyanoacetate. A new synthetic route for the access to [1,2,3]triazolo[1,5-a][1,4]benzodiazepines and other derivatives is described. This strategy is based on the cycloaddition of 2-oxoalkylidenephosphoranes to o-functionalized aryl azides followed by the reaction of the corresponding triazole intermediate with amines. An electrooxidative [3 + 2] annulation of phenols and electron-deficient alkenes for the synthesis of C3-functionalized 2-aryl-2,3-dihydrobenzofuran derivatives was achieved. The ring construction starts by a unique α-addition of carbon radicals derived from anodic oxidation of phenols to electron-deficient alkenes. The subsequent anodic oxidation of the resulting alkyl radical intermediates followed by trapping with the phenolic hydroxy group assembles the 2,3-dihydrobenzofuran core. Such a pathway enables the installation of various electrophilic functionalities including alkoxycarbonyl, alkylaminocarbonyl, trifluoromethyl, and cyano groups at the C-3 of the 2,3-dihydrobenzofuran framework, which is unattainable by other intermolecular reactions. The application of this method for a rapid synthesis of a bioactive natural product is demonstrated.

7-Chloro-2-(propylthio)thiazolo[5,4-d]pyrimidine 4 was prepared and used as a precursor for synthesizing new derivatives of thiazolo[5,4-d]pyrimidine through the nucleophilic displacement of the chlorine atom by different types of amino derivatives. Reaction of compound 4 with primary and secondary amines produced the corresponding substituted amino derivatives. Reaction of 4 with hydrazine hydrate then followed by condensation with aromatic aldehydes afforded hydrazone derivatives. Reaction of 4 with hydrazine hydrate then followed by reaction with isothiocyanates gave the disubstituted thiosemicarbazides. Finally, treatment of 4 with hydrazine derivatives afforded the novel disubstituted hydrazine derivatives. Preliminary antiviral screening on the bovine viral diarrhea virus (BVDV) was carried out, which was classified as a member of the same family of Hepatitis C virus. The results indicated that some of the tested compounds exhibited anti-BVDV activity which may be suitable for a new lead molecule to design more potent anti-BVDV agents.  相似文献

[reaction: see text] The synthesis of 2'-deoxycytidine nucleosides bearing amino and thiol groups appended to the 5-position of the nucleobase via a butynyl linker is described. The corresponding triphosphates were then synthesized from the nucleoside and incorporated into oligonucleotides by Vent (exo(-)) DNA polymerase. The ability of Vent (exo(-)) polymerase to amplify oligonucleotides containing these functionalized cytidine derivatives in a polymerase chain reaction (PCR) was demonstrated for the amino-functionalized derivative.  相似文献
