5-Hydroxymethylfurfural was conveniently synthesized by dehydration of D-fructose in a good yield. To further build bioactive derivatives from 5-hydroxymethylfurfural, 2,3-unsaturated glycosides were directly obtained through the Ferrier-rearrange-ment reaction of various glycals. Noticeably, a solid acid catalyst was successfully applied in the preparation of 5-hydroxyme-thylfurfural and then recycled to promote the Ferrier-rearrang-ement reaction, making it possible to achieve two steps of reaction in an eco-friendly manner through the simple process.  相似文献

Irreversible bindings of cis-diamminedichloroplatinum(II) (cis-DDP) to human serum albumin (HSA) were investigated in a pH 7.4 buffer containing 0.1 M NaCl at various molar ratios (cis-DDP/HSA) up to 60 over a 14 d period (37 degrees C). The metal binding seemed to reach a plateau when incubated at less than 10 times excess of cis-DDP. As the molar ratio increased, the reaction rate was relatively fast within the first day, followed by a moderate increase in the metal binding. When incubated at 60 times excess of cis-DDP, the metal bound as much as 20 mol per mol of HSA in 14 d. Fluorescence quenching of the metal-bound protein suggested that the tryptophan residue was gradually exposed to a hydrophilic environment as the metal binding increased. Furthermore, cis-DDP cleaved disulfide bonds at the ratio of 1 mol of disulfide bond per 5.3 mol of the metal binding. It was therefore suggested that the metal binding also occurred at several sites other than the disulfide bond. Warfarin binding to the metal-bound protein, examined by fluorescence changes, also decreased with increasing metal binding or cleavage of the disulfide bonds. Thus, cis-DDP bound to multiple sites in addition to the lone sulfhydryl group (Cys-34), suggesting that massive conformational changes of the protein took place.  相似文献

Binding of the cationic dye auramine O (AuO) to the polysoap-type polymers poly(vinylbenzo-18-crown-6) (P18C6) and polyvinylbenzoglyme (PVBG) in water were studied by fluorimetry and dialysis. The quantum yield of P18C6-bound AuO was found to be 0.028, the value being 0.018 for AuO bound to PVBG. The intrinsic binding constants were found to be 2.2 × 104M?1 (P18C6) and 1.2 × 104M?1 (PVBG), the respective first binding constants being 317 and 63M?1. Addition of crown-ether-complexable cations such as K+, Tl+, or Cs+ converts the neutral poly(crown ether) into a polycation, causing repulsion of the cationic dye and a strong decrease in the AuO fluorescence. AuO fluorescence was also studied in the absence of polymer in ether solvents, giving θ values of 0.011 and 0.018 in THF and dioxane. Traces of water rapidly form a nonfluorescent species. Solutions of AuO in water without polymer present exhibit very strong fluorescence on addition of BPh4 anions owing to formation of AuO+, BPh4? ion pairs and higher aggregates.  相似文献

The synthesis, fluxionality and reactivity of the heterobimetallic complex [FeRu(CO)2(mu-CO)2(eta-C5H5)(eta-C5Me5)] are described. Complex exhibits enhanced photolytic reactivity towards alkynes compared to its homometallic analogues, forming the dimetallacyclopentenone complexes [FeRu(CO)(mu-CO){mu-eta]1:eta3-C(O)CR"CR'}eta]-C5H5)(eta-C5Me5)]( R'= R"= H; R'= R"= CO2Me; R'= H, R"= CMe2OH). Prolonged photolysis with diphenylethyne gives the dimetallatetrahedrane complex [FeRu(mu-CO)(mu-eta2:eta2-CPhCPh)(eta-C5H5)(eta-C5Me5)], which contains the first iron-ruthenium double bond. Complexes containing a number of organic fragments can be synthesised using , and . Heating a solution of gave the alkenylidene complex [FeRu(CO)2(mu-CO){mu-eta]1:eta2-C=C(CO2Me)2}(eta-C5H5)(eta-C5Me5)] through an unusual methylcarboxylate migration. Protonation and then addition of hydride to gives the ethylidene complex [FeRu(CO)2(mu-CO)(mu-CHCH3)(eta-C5H5)(eta-C5Me5)] via the ionic vinyl species [FeRu(CO)2(mu-CO)(mu-eta]1:eta2-CH=CH2)(eta-C5H5)(eta-C5Me5)][BF4]. Compound exhibits cis/trans isomerisation at room temperature. Protonation of dimetallacyclopentenone complexes gives the allenyl species [FeRu(CO)2(mu-CO)(mu-eta1:eta2-CH=C=CMe2)(eta-C5H5)(eta-C5Me5)][BF4]. Compound exist as three isomers, two cis and one trans. The two cis isomers are shown to be interconverting by sigma-pi isomerisation. The solid state structures of these compounds were established by X-ray crystallography and are discussed.  相似文献

The synthesis and characterization of the Fe(III) complex of a novel crown ether-porphyrin conjugate, 52-N-(4-aza-18-crown-6)methyl-54,104,154,204-tetra-tert-butyl-56-methyl-5,10,15,20-tetraphenylporphyrin (H2Porph), as well as the corresponding hydroxo, dimeric, Fe(II), and peroxo species are reported. The crystal structure of [FeIII(Porph)Cl].H3O+.FeCl4-.C6H6.EtOH is also reported. [FeIII(Porph)(DMSO)2]+ and K[FeIII(Porph)(O22-)] are high-spin species (M?ssbauer data: delta = 0.38 mm s(-1), DeltaEq = 0.83 mm s(-1) and delta = 0.41 mm s(-1), DeltaEq = 0.51 mm s(-1), respectively), whereas in a solution of reduced [FeIII(Porph)(DMSO)2]+ complex the low-spin [FeII(Porph)(DMSO)2] (delta = 0.44 mm s(-1), DeltaEq = 1.32 mm s(-1)) and high-spin [FeII(Porph)(DMSO)] (delta = 1.27 mm s(-1), DeltaEq = 3.13 mm s(-1)) iron(II) species are observed. The reaction of [FeIII(Porph)(DMSO)2]+ with KO2 in DMSO has been investigated. The first reaction step, involving reduction to [FeII(Porph)(DMSO)2], was not investigated in detail because of parallel formation of an Fe(III)-hydroxo species. The kinetics and thermodynamics of the second reaction step, reversible binding of superoxide to the Fe(II) complex and formation of an Fe(III)-peroxo species, were studied in detail (by stopped-flow time-resolved UV/vis measurements in DMSO at 25 degrees C), resulting in kon = 36 500 +/- 500 M(-1) s(-1), koff = 0.21 +/- 0.01 s(-1) (direct measurements using an acid as a superoxide scavenger), and KO2- = (1.7 +/- 0.2) x 10(5) (superoxide binding constant kinetically obtained as kon/koff), (1.4 +/- 0.1) x 10(5), and (9.0 +/- 0.1) x 10(4) M(-1) (thermodynamically obtained in the absence and in the presence of 0.1 M NBu4PF6, respectively). Temperature-dependent kinetic measurements for kon (-40 to 25 degrees C in 3:7 DMSO/CH3CN mixture) yielded the activation parameters DeltaH = 61.2 +/- 0.9 kJ mol(-1) and DeltaS = +48 +/- 3 J K(-1) mol(-1). The observed reversible binding of superoxide to the metal center and the obtained kinetic and thermodynamic parameters are unique. The finding that fine-tuning of the proton concentration can cause the Fe(III)-peroxo species to release O2- and form an Fe(II) species is of biological interest, since this process might occur under very specific physiological conditions.  相似文献

The Mn(II) high-magnetic-field electron paramagnetic resonance (HFEPR) spectra of five different superoxide dismutases (SODs) were measured at 190 and 285 GHz. The native E. coli manganese SOD was found to be distinct from the other SODs by virtue of its large zero-field E-value. The two wild-type cambialistic proteins from Porphyromonas gingivalis and Rhodobacter capsulatus were also distinct. However, the Gly155Thr mutant of the P. gingivalis SOD changed the Mn(II) spectrum so that it closely resembled the spectrum of manganese reconstituted E. coli iron SOD. This observation paralleled enzyme activity measurements that show that this mutation causes the loss of activity with manganese and enhanced activity with iron indicating a conversion from a cambialistic to an iron-specific protein. The Mn(II) magnetic parameters were determined by simultaneously fitting the multifrequency data. Simulations were carried out by numerically diagonalizing the spin Hamiltonian and explicitly calculating all possible transition probabilities. The relationship between the Mn(II) zero-field interaction and structure of the metal binding site is also discussed.  相似文献

Hexylester of 5-aminolevulinic acid (hALA) has been considered as an alternative to 5-aminolevulinic acid (ALA) for the treatment of malignancies of different origin. The present study addresses the ALA and hALA-induced PpIX pharmacokinetic profile using rat bladder carcinoma cells (AY27). The total PpIX content measured spectrofluorimetrically after extraction procedure at the end of 2 h incubation was at least 1.5-fold greater with hALA compared to ALA despite the difference in concentration of several orders between the two compounds (1 or 5 mM ALA and 5 or 10 x 10(-3) mM hALA). Considerable PpIX efflux was detected in the extracellular medium at the end of the incubation. With 5 mM ALA and 10 x 10(-3) mM hALA, PpIX build-up was continued beyond the incubation period pointing out to enzyme saturation in the biosynthetic pathway or/and the constitution of ALA reserve. Red laser light (lambda=630 nm) irradiation of AY27 cells after 2 h incubation with increasing ALA or hALA concentrations resulted in a nearly equal photocytotoxicity.  相似文献

Sialyloligosaccharides are synthesised by various glycosyltransferases and sugar nucleotides. All of these nucleotides are diphosphate compounds except for cytidine-5'-monophosphosialic acid (CMP-Neu5Ac). To obtain an insight into why cytidine-5'-diphosphosialic acid (CDP-Neu5Ac) has not been used for the sialyltransferase reaction and why it is not found in biological organisms, the compound was synthesised. This synthesis provided the interesting finding that the carboxylic acid moiety of the sialic acid attacks the attached phosphate group. This interaction yields an activated anhydride between carboxylic acid and the phosphate group and leads to hydrolysis of the pyrophosphate linkage. The mechanism was demonstrated by stable isotope-labelling experiments. This finding suggested that CMP-Neu5Ac might also form the corresponding anhydride structure between carboxylic acid and phosphate, and this seems to be the reason why CMP-Neu5Ac is acid labile in relation to other sugar nucleotides. To confirm the role of the carboxylic acid, CMP-Neu5Ac derivatives in which the carboxylic acid moiety in the sialic acid was substituted with amide or ester groups were synthesised. These analogues clearly exhibited resistance to acid hydrolysis. This result indicated that the carboxylic acid of Neu5Ac is associated with its stability in solution. This finding also enabled the development of a novel chemical synthetic method for CMP-Neu5Ac and CMP-sialic acid derivatives.  相似文献

The water-soluble monomers, 1-methyl-4-vinylimidazole, 1-methyl-5-vinylimidazole, 1-ethyl-5-vinylimidazole, and 1-propyl-5-vinylimidazole have been synthesized, polymerized, and copolymerized with 4(5)-vinylimidazole. The copolymers were characterized by 14C-labeling, NMR, pKa determination and viscosity measurements. The monomer reactivity ratios determined by 14C counting are r1 = 1.04; r2 = 0.94 [M1 = 4(5)-vinylimidazole, M2 = 1-methyl-4-vinylimidazole] and r1 = 1.01; r2 = 0.86 [M1 = 4(5)-vinylimidazole, M2 = 1-methyl-5-vinylimidazole]. The esterolytic activity of the copolymers for the hydrolysis of p-nitrophenyl acetate (PNPA) at pH 7–8 in 28.5% ethanol–water was higher than that of the mixtures of homopolymers. At pH 5–6 the esterolytic activities of the copolymers and the mixtures were similar. The most efficient esterolytic activity for PNPA hydrolysis at pH 7.11 in 28.5% ethanol–water occurred for copolymers containing 75 mole % 4(5)-vinylimidazole and for copolymers containing 1-methyl-4-vinylimidazole rather than 1-methyl-5-vinylimidazole.  相似文献

The interaction of calf thymus DNA with [CuL(ClO(4))]ClO(4)·H(2)O (1) and [ZnLBr]Br·H(2)O (2) (L = 9,12,15,18,27,28-hexaaza-29-oxatetracyclo[24.2.1.0(2,7).0(20,25)]enneicosa-2,4,6,20,22,24,26,28(1)-octaene) dicationic complexes in aqueous solution at neutral pH, was investigated by variable-temperature UV-vis absorption, circular dichroism and fluorescence spectroscopy. The values of the DNA-binding constants of these complexes, determined by competitive binding spectrofluorimetric titrations of ethidium bromide (EB)-DNA solutions, are (6.7 ± 0.5) × 10(6) M(-1) for CuL(2+) and (4.7 ± 0.5) × 10(5) M(-1) for ZnL(2+). These data together with a through analysis of the spectroscopic behaviour consistently suggest that both compounds are effective DNA binders. Interestingly, the DNA-binding strength of these complexes has been found to be correlated to their in vitro cytotoxic activity toward human breast carcinoma cells, although the complex with lower DNA-binding affinity is more active. In fact, biological studies showed that when the compounds are delivered through the cell membrane by a lipidic carrier, the cell survival is sensibly reduced, up to 58% with 1 and to 31% with 2.  相似文献

The crystal structure of the title compound, [nBu2Sn(5-Cl-2-OH-C6H3CO2)2], shows that in the monomeric species the six-coordinate Sn atom exists in a skew trapezoidal bipyramidal geometry in which the four O donor atoms, derived from two asymmetrically chelating carboxylate ligands, define the basal plane and the n-butyl substituents lie over the weaker Sn? O interactions defining a C? Sn? C angle of 147.6(2)°. An unusual feature of the structure is the presence of both intra- and inter-molecular hydrogen bonding contacts which serve to stabilize the structure. The in vitro antitumour activity of this compound against mammary and colon carcinoma cell lines is also reported.  相似文献

It may be hypothesized that the lipoxygenase (LOX) metabolic pathway plays an important role in photodynamic therapy (PDT) of malignant tumours, and modification of this pathway may result in administration of lower doses of photodynamic active agents accompanied by reduced side effects. In this study, we examine in more detail the cytokinetic parameters of human colon adenocarcinoma HT-29 cells pre-treated for 48 or 24h with LOX inhibitor MK-886, followed by PDT induced by hypericin. Based on MTT assay the concentrations of both agents (MK-886 and hypericin) with relatively slight (non-significant) cytotoxic effects were selected. These concentrations were used for combined treatment, where MTT response, total cell number, floating cells quantification, viability, cell cycle progression and DNA synthesis were detected. Hoechst/PI staining, PARP fragmentation and mitochondrial membrane potential (MMP) were evaluated to determine the extent of apoptosis. While MK-886 alone caused mainly necrosis, 48h pre-treatment of cells with MK-886 followed by PDT with hypericin clearly shifted the type of cell death to apoptosis. PDT with hypericin alone caused apoptosis in 19% of the cell population. Some combined modalities significantly potentiated the apoptotic effect (31% of apoptotic cells; 2.5microM MK-886/0.1microM hypericin), i.e., by 60% more than after single treatment with hypericin. Increased apoptosis was confirmed by PARP (116kDa) cleavage to characteristic 89kDa fragments and changes in MMP. Increasing concentration of MK-886 was accompanied by massive changes in the cell cycle progression. Combined treatment with lower concentrations of MK-886 and hypericin increased accumulation of cells in the S phase, accompanied by inhibition of DNA synthesis. Increasing concentration of MK-886 in this combination caused the opposite effect, manifesting significant accumulation of cells in the G0/G1 phase. More pronounced effects were observed after the 48h pre-treatment schedule. This anti-proliferative effect was confirmed by BrdU incorporation. These results indicate that combined treatment involving PDT and LOX inhibitor MK-886 may improve the therapeutic effectiveness of PDT.  相似文献

A new analytical reagent 5-(p-aminobenzylidene)-rhodanine (ABR) was synthesized. The acidic dissociation constant of ABR has been determined. The properties, the acid-base behavior of ABR and the reactions of ABR with metallic ions have been studied. The color reactions of the reagent with Pd(II), Au(III), Ag(I), Ru(III), Hg(II) and Cu(II) are studied in detail. [structure: see text]. Flexible, convergent access to 2,3-disubstituted benzo[b]thiophenes has been developed. The most concise approach involves sequential coupling of o-bromoiodobenzenes with benzylmercaptan and zinc acetylides to give benzyl o-ethynylphenyl sulfides which react with iodine to give 3-iodobenzo[b]thiophenes in a 5-endo-dig iodocyclization. These iodides can be further elaborated using palladium-mediated coupling and/or metalation techniques. This method has been applied to the synthesis of some novel tubulin binding agents.  相似文献
