An improved method of synthesis of neplanocin and related cyclopentenyl-containing nucleosides

A facile one-step condensation of the cyclopentenyl α,-tosylate $\text{2c}$ with the alkali metal salts of 6-chloropurine and uracil, afforded the protected carbocyclic nucleosides $\text{5a}$ and $\text{8}$, respectively, which were converted to neplanocin a ($\text{6}$) and cyclopentenyl uracil ($\text{9}$) by established methods.     

Reaction between nucleoside base residues and the phosphorylating agent derived from 1-hydroxybenzotriazole and 2-chloroprenyl phosphorodichloridate

In the presence of 1-methylimidazole, 2-$\text{N}$-acyl guanine (as in $\text{4a}$), thymine (as in $\text{5a}$) and uracil (as in $\text{5b}$) residues react readily with the phosphorylating agent derived from 2-chlorophenyl phosphorodichloridate ($\text{1a}$) and 1-hydroxybenzotriazole.     

Novel nucleoside analogues via direct attack of carbon : Nucleophiles on nucleosides containing epoxy-sugars

Direct ring-opening of the epoxide ring in 1-(5′-$\text{O}$-trityl-2′,3′-anhydro-β-D-lyxo-furanosyl) uracil (1) by lithium acetylide or vinylmagnesium bromide/cuprous iodide affords the corresponding 5′-$\text{O}$-trity]-3′-$\text{C}$-substituted-3′-deoxy-$\text{ara}$-uridine species.     

The protection of uracil and guanine residues in oligonucleotide synthesis

The protection of uracil and 2-$\text{N}$-acyl guanine residues with 4-$\text{O}$-phenyl [or 4-$\text{O}$-(2,4-dimethylphenyl)] and 6-$\text{O}$-(2-nitrophenyl) groups as in $\text{7a}$ [or $\text{7b}$] and $\text{9}$, respectively, is described. These $\text{O}$-aryl protecting groups, which appear to withstand the usual conditions of oligonucleotide synthesis, may readily be removed by treatment with 2-nitrobenzaldoximate ions.     

Infrared spectra of hydrogen bonded crystals: Uracil

A theory of the infrared spectra of hydrogen bonds in molecular crystals of C$\text{5}\text{2}$h symmetry, containing, like uracil, four molecules and eight hydrogen bonds in the unit cell, is presented. The theoretical spectra are in good agreement in both the frequency and intensity distribution with experiment. The changes in the fine structure introduced by deuteration are quantitatively reproduced.     

[2+2] Cycloaddition of benzyne to cis,trans- and cis,cis-1,5-cyclooctadiene

The relative rates of formation of $\text{cis}$ and $\text{trans}$ [2+2]cycloadducts of benzyne to $\text{cis}$, $\text{trans}$- and $\text{cis}$, $\text{cis}$-1,5-cyclooctadiene, $\text{trans}$- and $\text{cis}$-cyclooctene are discussed and properties of the adducts (e.g., their unusual hydrogenation) are described.     

N-Acylated α-aminonitriles and their conversion into 5-aminooxazole, 5(4h)-oxazolone and 4(5h)-imidazolone derivatives

In contrast with the reaction of α-aminonitriles $\text{1}$a, the corresponding $\text{N}$-acylated α-aminonitriles $\text{1}$b-f and oxalyl chloride do not yield pyrazinone derivatives, but 5-aminooxazoles $\text{9}$-$\text{11}$ or 4(5$\text{H}$)-imidazolones $\text{12}$, the latter being converted in some cases into imidazo [2,1-$\text{a}$]isoquinoline-2,5,6(3$\text{H}$)-triones. Reactions of compounds $\text{1}$b-f and ethyl chlorooxoacetate provide evidence for a 5(4$\text{H}$)-iminooxazole intermediate $\text{7}$, which aromatizes to yield 5-aminooxazoles $\text{9}$-$\text{11}$; however, unaromatizable īntermediates of type $\text{7}$ - isolable as 5(4$\text{H}$)y-oxazolones $\text{13}$ after hydrolysis - undergo a catalyzed Dimroth-type rearrangement to give imidazolone derivatives $\text{12}$.     

Steroidal antifeedants from the dorid nudibranch Aldisasanguineacooperi

Two steroids, 3-oxo-chol-4-ene-24-oic acid ($\text{2}$) andi ts unsaturated analog $\text{4}$ have been isolated from the dorid nudibranch $\text{Aldisa}$$\text{sanguinea}$$\text{cooperi}$, and the acid $\text{2}$ has been shown to have antifeedant properties.     

Chemical synthesis of the 5-́half molecule of E.coli tRNA2GLY

A tritriacontanucleotide which has the sequence of the 5$\text{-}\text{?}$half molecule of E.coli glycine tRNA₂, was synthesized by the phosphotriester method involving p-anisidate protection for the 3$\text{-}\text{?}$phosphate ends. Di- and trinucleotide units were prepared from 5$\text{-}\text{?}$dimethoxytrityl-2$\text{-}\text{?}$O-tetrahydrofuranyl-3?-O-(o-chlorophenyl)phosphoryl derivatives of uridine, N-benzoylcytidine, N-benzolyadenosine and N-iso-butyrylguanosine by condensation with 3$\text{,}\text{?}$5$\text{-}\text{?}$unprotected nucleosides followed by phosphorylation to give 3$\text{-}\text{?}$phosphodiester blocks. The 3$\text{-}\text{?}$terminal dimers and trimers were synthesized by using 3$\text{-}\text{?}$(o- chlorophenyl)phosphoro-p-anisidates instead of 3?,5?-unprotected nucleosides. The 3?-phosphodiesters of oligonucleotides with a chain length of larger than 5 were obtained by removal of the 3?-phosphoro-p-anisidate with isoamyl nitrite. The 5$\text{-}\text{?}$dimethoxytrityl group was removed by treatment with zinc bromide under anhydrous conditions. Fragments were designed to use common dimer blocks and to reduce the step for 5$\text{-}\text{?}$deblocking of larger fragments. Finally a 3$\text{-}\text{?}$phosphodiester block with a chain length of 20 was condensed with a 5$\text{-}\text{?}$OH component (tridecanucleotide). The fully protected 33 mer was deblocked and purified by chromatography. The structural integrity of the product was confirmed by mobility shift analysis and complete digestion with RNase T2.     

Plaunol A and B,new anti-ulcer diterpenelactones from Crotonsublyratus

Two new diterpenelactones named plaunol A ($\text{2}$) and B ($\text{3}$) exhibiting anti-Shay ulcer activity were isolated from Thai medicinal plant, $\text{Croton}$$\text{sublyratus}$. Structure of $\text{2}$ was determined by X-ray analysis of its $\text{p}$-bromobenzoate and structure of $\text{3}$ was deduced from the results of chemical correlation and the spectral data.     

12-S-CIS conformationally locked retinoids: Thermal and photochemical interconversions leading to new geometric isomers

For 12-$\text{s}$-$\text{cis}$ locked retinal analogues, thermal equilibria are established between 11-$\text{cis}$, 13-$\text{cis}$-1c and 13-$\text{cis}$-2c and between 9-$\text{cis}$, 11-$\text{cis}$, 13-$\text{cis}$-1d and 9-$\text{cis}$, 13-$\text{cis}$-2d; a photoisomerization occurs to transform 2c to $\text{all}$-$\text{trans}$-2a and 2d to 9-$\text{cis}$-2b

Synthesis of β-d-mannopyranosides and regioselective o-alkylation of dibutylstannylene complexes

Alkylation of dibutylstannylene complexes of 3,4,6-tri-$\text{O}$-benzyl-$\text{D}$-mannopyranose, methyl 6-$\text{O}$-trityl-α-$\text{D}$-mannopyranoside and methyl α-$\text{D}$-mannopyranoside gives respectively β-$\text{D}$-mannopyranoside and 3-$\text{O}$-alkyl derivatives with high selectivity.     

Boat-chair conformational preference of exo, exo-9-oxobicyclo[3.3.1]nonane-2,4-dicarboxylic acid

The ¹H and ¹³C nmr spectra of $\text{exo}$, $\text{exo}$-9-oxobicyclo[3.3.1]nonane-2,4-dicarboxylic acid, 3, in DMSO-d₆ or alkaline D₂O, clearly show that it exists in a $\text{boat}$-$\text{chair}$-conformation with $\text{equatorial}$ carboxyl groups, thus being the first case of $\text{boat}$-$\text{chair}$ preference of a bicyclo[3.3.1] nonan-9-one due to the presence of $\text{exo}$,$\text{exo}$-2,4- substituents.     

Synthesis and absolute configuration of +-lineatin,the pheromone of trypodendronlineatum

A new synthesis of (±), (+)- and (-)-lineatin (3,3,7-trimethyl-2, 9-dioxatricyclo [3.3.1.0^4,7] nonane, $\text{1}$) was achieved. The stereochemistry of (+)-lineatin was established as 1$\text{R}$, 4$\text{S}$, 5$\text{R}$, 7$\text{R}$ by an X-ray crystallographic analysis of an intermediate $\text{1}$$\text{5}$.     

Synthesis of N-benzoyl L- and D-2,3,6-trideoxy-3-aminoxylo-hexose from non-carbohydrate precursors

The enantiomeric alcohols ($\text{2}$) and ($\text{11}$), obtained from (2$\text{R}$,3$\text{R}$) tartaric acid and, respectively, $\text{L}$-threonine, have been used to construct the C₆, enantiomeric deoxy amino sugar derivatives ($\text{7}$) and ($\text{12}$)     

Conversion of 6-protoilludene into illudin-M and -S by omphalotusolearius

Deuterium labeled $\text{dl}$-6-protoilludene ($\text{7}$) was synthesized and fed to the illudin-producing fungus $\text{Omphalotus}$$\text{olearius}$ (ATCC 11719). The hydrocarbon ($\text{7}$) was incorporated into illudin-M ($\text{2}$) and -S ($\text{3}$).     

Isolation and structures of trans-laurencenyne,a possible precursor of the C15 halogenated cyclic ethers,and trans-neolaurencenyne from laurencia okamurai

Isolation and structural elucidation of two new acetylenic polyenes, $\text{trans}$-laurencenyne $\text{1}$ and $\text{trans}$-neolaurencenyne $\text{2}$ from the marine red alga $\text{Laurencia okamurai}$ were carried out, the former compound $\text{1}$ very recently being proposed to be a biosynthetic precursor of the C₁₅ halogenated cyclic ethers, $\text{7}$ and $\text{8}$.     

(10R̃, 11R̃)-(+)-squalene-10, 11-epoxide: Isolation from Laurencia okamurai and the asymmetric synthesis

From a red alga $\text{Laurencia okamurai}$ (10$\text{R}\text{?}$, 11$\text{R}\text{?}$)-(+)-squalene-10, 11-epoxide $\text{1}\text{?}$ was isolated and its asymmetric synthesis has been achieved starting from $\text{trans, trans}$-farnesol.
