An efficient methodology has been developed for the synthesis of quinoxalin‐2(1H)‐one derivatives of 2‐phenylimidazo[1,2‐a]pyridines by microwave‐irradiated Hinsberg heterocyclization between 2‐phenylimidazo[1,2‐a]pyridine‐3‐glyoxalates and o‐phenylenediamine using either montmorillonite K‐10 or Yb(OTf)3 as catalysts. Montmorillonite K‐10 was proven to be an efficient catalyst for the heterocyclization reaction between sterically hindered glyoxalate and o‐phenylenediamine only under microwave conditions. The use of Yb(OTf)3/tetrahydrofuran was also found to be an effective catalyst for the above chemical transformation among a series of Lewis acids screened under microwave conditions; however, comparatively lesser yields were obtained as compared with the use of montmorillonite K‐10.  相似文献

5‐(2‐Aminothiazol‐4‐yl)‐8‐hydroxyquinoline 2 has been synthesized by treating thiourea with 5‐chloroacetyl‐8‐hydroxyquinoline 1 . The amine 2 was treated with aromatic aldehydes to furnish schiff bases 6a‐c which on treatment with phenyl isothiocyanate gave the corresponding thiazolo‐s‐triazines 7a‐c . Reaction of 2 with phenyl isothiocyanate gave the corresponding aminocarbothiamide derivative 8 which on reaction with malonic acid in acetyl chloride afforded thiobarbituric acid derivative 9 . Coupling of 9 with diazonium salt gave the phenyl hydrazono derivative 10 . However, reaction of 2 with carbon disulphide and methyl iodide afforded dithiocarbamidate 12 which on treatment with ethylenediamine, o‐aminophenol and/or phenylenediamine gave the aminoazolo derivatives 13–15 , respectively. Other substituted fused thiazolopyrimidines 16–20 have been also prepared by the reaction of 2 with some selected dicarbonyl reagents. The characterisation of synthesized compounds has been done on the basis of elemental analysis, IR, 1H‐NMR and mass spectral data. All the newly synthesized compounds have been screened for their antimicrobial activities.  相似文献

In an endeavor to find a new class of antimicrobial agents, a series of novel substituted benzimidazole, benzoxazole, and benzothiazole derivatives 6 containing pyrazole moiety have been synthesized by reaction of 3‐aryl‐4‐formyl pyrazole 4 with substituted phenylenediamine or o‐aminophenol or o‐aminothiophenol 5 . Reaction of phenyl hydrazine or 2‐hydrazinopyridine 1 with substituted acetophenones 2 gave the corresponding hydrazones 3 , which on Vilsmeier–Haack reaction with POCl3–DMF gave substituted 3‐aryl‐4‐formyl pyrazoles 4 . All final compounds 6a , 6b , 6c , 6d , 6e , 6f , 6g , 6h , 6i , 6j , 6k were evaluated for in vitro antibacterial activities against Escherichia coli and Staphylococcus aureus strains and in vitro antifungal activity against Candida albicans and Aspergillus niger strains by using serial dilution method. The antimicrobial activities were expressed as the minimum inhibitory concentration in µg/mL. The compound containing benzimidazole and benzoxazole moiety gave better antibacterial and antifungal activities than benzothiazole compounds.  相似文献

Reaction of pyrone 1a or tetronic acid 1b with o‐phenylenediamine derivatives gave enaminone compounds 2 . When reacting with different electrophiles, these intermediates allowed versatile access to different heterocyclic structures: when DMA derivatives or triphosgene was used, cyclization occurred through nitrogen, leading to benzimidazoles 3 and benzimidazolones 4 , respectively, while reaction with benzaldehyde yielded benzodiazepines 5 .  相似文献

Highly efficient catalytic asymmetric Claisen rearrangements of O‐propargyl β‐ketoesters and O‐allyl β‐ketoesters have been accomplished under mild reaction conditions. In the presence of the chiral N,N′‐dioxide/NiII complex, a wide range of allenyl/allyl‐substituted all‐carbon quaternary β‐ketoesters was obtained in generally good yield (up to 99 %) and high diastereoselectivity (up to 99:1 d.r.) with excellent enantioselectivity (up to 99 % ee).  相似文献

Asymmetric allylic alkylation of β‐ketoesters with allylic alcohols catalyzed by [Ni(cod)2]/(S)‐H8‐BINAP was found to be a superior synthetic protocol for constructing quaternary chiral centers at the α‐position of β‐ketoesters. The reaction proceeded in high yield and with high enantioselectivity using various β‐ketoesters and allylic alcohols, without any additional activators. The versatility of this methodology for accessing useful and enantioenriched products was demonstrated.  相似文献

A series of N‐substituted 1,4‐dihydro‐4‐oxo‐1,8‐naphthyridine‐3‐carboxylate esters has been prepared in two steps from ethyl 2‐(2‐chloronicotinoyl)acetate. Treatment of the β‐ketoester with N,N‐dimethylformamide dimethyl acetal in N,N‐dimethylformamide (DMF) gave a 95% yield of the 2‐dimethylaminomethylene derivative. Subsequent reaction of this β‐enaminone with primary amines in DMF at 120oC for 24 h then afforded the target compounds in 47–82% yields by a tandem SNAr‐addition‐elimination reaction. Synthetic and procedural details as well as a mechanistic rationale are presented.  相似文献

A simple, fast and efficient method for the preparation of several 2‐substituted benzimidazole derivatives is reported. Compounds were synthesized through a rapid one‐pot synthesis via microwave irradiation, starting from aldehydes and o‐phenylenediamine, in the presence of H2O2/HCl system in acetonitrile. The significant features of this method are short reaction times, high yields, easy and quick isolation of the products.  相似文献

The enantioselective tandem reaction of β,γ‐unsaturated α‐ketoesters with β‐alkynyl ketones was realized by a bimetallic catalytic system of achiral AuΙΙΙ salt and chiral N,N′‐dioxide‐MgΙΙ complex. The cycloisomerization of β‐alkynyl ketone and asymmetric intermolecular [4+2] cycloaddition with β,γ‐unsaturated α‐ketoesters subsequently occurred, providing an efficient and straightforward access to chiral multifunctional 6,6‐spiroketals in up to 97 % yield, 94 % ee and >19/1 d.r. Besides, a catalytic cycle was proposed based on the results of control experiments.  相似文献

3,3′,4,4′‐Diphenylsulfonetetracarboxylic dianhydride (1) was reacted with L ‐isoleucine (2) in acetic acid and the resulting imide‐acid (3) was obtained in high yield. The diacid chloride (4) was prepared from the diacid derivative (3) by reaction with thionyl chloride. The polycondensation reaction of diacid chloride (4) with several aromatic diamines such as 4,4′‐sulfonyldianiline (5a), 4,4′‐diaminodiphenyl methane (5b), 4,4′‐diaminodiphenylether (5c), p‐phenylenediamine (5d),m‐phenylenediamine (5e), 2,4‐diaminotoluene (5f) and 4,4′‐diaminobiphenyl (5g) was performed by two conventional methods: low temperature solution polycondensation and short period reflux conditions. In order to compare conventional solution polycondensation reaction methods with microwave‐assisted polycondensation, the reactions were also carried out under microwave conditions with a small amount of o‐cresol that acts as a primary microwave absorber. The reaction mixture was irradiated for 6 min with 100% of radiation power. Several new optically active poly(amide‐imide)s with inherent viscosity ranging from 0.23 to 0.41 dl/g were obtained with high yield. All of the earlier polymers were fully characterized by IR, elemental analyses and specific rotation techniques. Several β‐ketoesters were dilithiated with an excess of lithium diisopropylamide, followed by condensation with methyl 2‐(aminosulfonyl)benzoate to give intermediates that were not isolated but cyclized to 3‐substituted 1,2‐benzisothiazole‐1,1‐dioxides. In most instances involving the ester‐sulfonamide, a single β‐ketoester tautomer is usually formed after recrystallization from ethanol. The same dilithiated β‐ketoesters generally condense less well with 1,2‐benzisothiazol‐3(2H)‐one‐1,1‐dioxide (saccharin) under the same conditions to afford the same products usually in the same or lower yields. The use of N,N,N',N'‐tetramethylethylenediamine during these syntheses has sometimes resulted in improved yields of products.  相似文献

Chloroformylation of 5,5‐dimethyl‐1,2‐ oxathiolan‐4‐one 2,2‐dioxide 4 with Vilsmeier reagent (DMF/POCl3) led to the formation of cyclic β‐chloro‐vinylaldehyde (4‐chloro‐5,5‐dimethyl‐3‐formyl‐1,2‐oxathiolene 2,2‐dioxide 5 ). A novel series of imidazoles, pyrimidines, and thiazoles were synthesized using microwave irradiation and conventional method from commercial available p‐aminobenzoic acid. Thus, one‐pot condensation of p‐aminobenzoic acid, urea, and chloroacetic acid have been provided two types of imidazole derivatives as separated mixture 2a and 2b . [3 + 2 + 1] Cyclocondensation of 2a , benzaldehyde, and urea/thiourea in acidic medium afforded imidazolo oxazine derivative 3 and Imidazolothiazine 4 . Coupling of 2a with benzene diazonium salt gave the phenyldiazenyl imidazolidine 5 . While the reaction of 2a , thiourea, and benzaldehyde in sodium ethoxide afforded imidazolothiazine 6 . Oxidative cyclization of thiourea derivative 7 resulted a mixture of benzithiazole derivative 7a and oxathiazole derivative 7b . Cyclocondensation of 7a with phenylenediamine and 4‐methyl phenylenediamine furnished imidazole 8 and 9 , respectively. Reaction of P‐aminobenzioc acid with potassium cyanate followed by Biginelli reaction with (acetyl acetone, ethyl acetoacetate, and diethyl malonate) and salicyladehyde in HCl tolerated pyrimidine derivatives 10a–c , respectively. In the same manner, the reactions and short reaction time make microwave technique one of the greenest methodology for synthesis of this class of heterocyclic system.  相似文献

The condensation of 4‐amino‐3‐aryl‐5‐mercapto‐1, 2, 4‐triazoles (1a‐f) with 6‐/8‐substituted 1,4‐dihydro‐4‐oxo‐quinoline‐3‐carboxylic adds (2a‐d) in the presence of phosphorus oxychloride on refluxng or under microwave irradiation gave twenty four novel 3‐aryl‐6‐ (6‐/8‐substituted 4‐chloroquinoline‐3‐yl)‐s‐triazolo[3,4‐b]‐1, 3,4‐thiadiazoles (4a‐x), Considerable increase in the reaction rate has been observed with improved yields under microwave irradiation. The structures of the compounds synthesized were determined by elemental analyses, IR, 1H NMR and MS spectra. Their spectral properties and the reaction mechanism were also discussed. The preliminary biological test showed that some of compounds bad moderate antibacterial activities.  相似文献

Reaction of 6‐mercapto‐7H‐1,2,4‐triazolo[1,5‐d]tetrazole ( 1 ) wtih 1,2‐phenylenediamine afforded N‐{7H‐1,2,4‐triazolo[1,5‐d]tetrazol‐6‐yl}‐1,2‐phenylenediamine which was cyclized to benzimidazolyl‐1,2,4‐triazolo[1,5‐d]tetrazoles using various one‐carbon cyclizing agents. Also, the treatment of 1 with maleic anhydride or phthalic anhydride gave the corresponding thio derivatives followed by hydrazinolysis to afford the thio heterobicyclic systems. Former structures of the products have been established upon elemental and spectral analyses.  相似文献

N,N′‐Dioxide/nickel(II) complexes have been developed to catalyze the inverse‐electron‐demand hetero‐Diels–Alder reaction of β,γ‐unsaturated α‐ketoesters with acyclic enecarbamates. After detailed screening of the reaction parameters, mild optimized reaction conditions were established, affording 3,4‐dihydro‐2H‐pyranamines in up to 99 % yield, 99 % ee and more than 95:5 d.r. The catalytic system was also efficient for β‐substituted acyclic enecarbamates, affording more challenging 2,3,4‐trisubstituted 3,4‐dihydro‐2H‐pyranamine with three contiguous stereogenic centers in excellent yields, diastereoselectivities, and enantioselectivities. The reaction could be scaled up to a gram scale with no deterioration of either enantioselectivity or yield. Based on these experiments and on previous reports, a possible transition state was proposed.  相似文献

4‐Bromo‐o‐phenylenediamine and ethylimido‐p‐bromophenylacetate, 1 , were subjected to microwave irradiation to synthesize benzimidazole derivative, compound 2 . Ester derivative, 3 , and hydrazide derivative, 4 , of compound 2 were also synthesized, respectively. Finally, compound 4 was treated with 11 different aromatic aldehydes to obtain benzimidazole derivatives containing imine function. All reactions were carried out with microwave irradiation and conventional heating, and results were compared. Some of the newly synthesized compounds showed moderate antimicrobial activity against some tested organisms.  相似文献
