不同产地白芷药材高效液相色谱指纹图谱研究 总被引:3,自引:0,他引:3
本文采用高效液相色谱-二极管阵列检测器(HPLC-DAD)法建立中药白芷的指纹图谱.应用化学计量学中两种不同的模式识别方法(主成分分析法和系统聚类分析法)对实验数据进行处理,以找出来自三个不同产地30个中药白芷样品间的相似性及差异性.两种模式识别方法均能成功地按样品的来源将不同产地的样品正确分类.建立了不同产地中药白芷的识别方法,该方法能有效地控制中药白芷的质量,并能为其它中药产品的化学模式识别提供参考. 相似文献

高效液相色谱法研究当归指纹图谱 总被引:5,自引:0,他引:5
目的:研究当归药材的指纹图谱.方法:高效液相色谱法,Hypersil ODS柱(4.6 mm×250 mm,5μm),Kromasil ODS保护柱(4.6 mm×10 mm,5μm).甲醇-1%醋酸梯度流动相,流速1.0 mL/min,柱温25℃.以当归对照药材为对照品,色谱峰光谱采集范围:190~400 nm;以阿魏酸为参照物并测定了阿魏酸的含量,检测波长:323nm,流动相:甲醇-1%醋酸(40∶60,V/V),流速0.7 mL/min.结果:找出了22个共有峰,其中5号峰为阿魏酸,11个样品与当归对照药材之间的相似度均在90%以上,平均相似度为96.77%.结论:样品处理方法简单,研究所得的当归指纹图谱稳定性、重复性好,可以作为当归极性部分的特征性指纹图谱. 相似文献

高效液相色谱指纹图谱在中药芦根上的应用 总被引:16,自引:0,他引:16
利用现代分离分析技术如高效液相色谱(HPLC)与气相色谱(GC),能够获得中草药的色谱指纹图谱。本实验报道了高效液相指纹图谱应用于快速检测中草药提取物以及应用于研究、开发与生产中的质量控制体系中,利用HPLC建立芦根的色谱指纹图谱。实验条件为反相C18柱,流速为1.0mL/min,紫外检测器(λ=230nm)。通过分析比较色谱指纹图谱中的相对保留值α以及相对面积St对不同产地的芦根进行对比研究。这种基于HPLC的分析策略为中草药的精确鉴定和质量控制提供了有效的信息,并为HPLC在复杂组分样品中的应用开拓了新的领域。 相似文献

应用高效液相色谱(HPLC)法建立五加生化胶囊的指纹图谱。采用ANGLAVenusil XBP-C18色谱柱(250×4.6mm,5μm),以甲醇-0.1%磷酸溶液为流动相,梯度洗脱,检测波长为327nm,五加生化胶囊多数峰达到基线分离。采用2004A中药色谱指纹图谱相似度评价系统对11批样品的指纹图谱进行峰匹配,确定12个共有峰,11批五加生化胶囊指纹图谱的相似度均在0.90以上。结果表明:五加生化胶囊的HPLC指纹图谱特征性和专属性强,可较系统地用于五加生化胶囊的质量控制。 相似文献

应用高效液相色谱(HPLC)法建立桂枝茯苓丸的指纹图谱。采用ANGLA Promosil C18色谱柱(250×4.6mm,5μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,230nm检测,优化条件下,桂枝茯苓丸各成分得到较好的分离。经方法学验证,方法具良好的精密度、重复性和稳定性。采用中药色谱指纹图谱相似度评价系统软件(2004A版)进行指纹图谱分析,11批桂枝茯苓丸样品相似度均在0.90以上。实验表明:桂枝茯苓丸的HPLC指纹图谱特征性和专属性强,可较系统地用于桂枝茯苓丸的质量控制。 相似文献

建立了适用于王老吉广东凉茶颗粒质量控制的超高效液相色谱指纹图谱分析方法。样品采用甲醇超声萃取30 min,萃取液用超高效液相色谱法进行指纹图谱分析。色谱柱采用Waters ACQUITY HSS T3 C18(3.0mm×150 mm,1.8μm),以0.5%甲酸-乙腈为流动相进行梯度洗脱,流速为0.8 mL/min,柱温35℃。28个共有峰在18 min内得到良好分离,通过对照品对其中4个峰进行了确证。对王老吉广东凉茶颗粒样品进行了相似度分析,23个批次样品的相似度均达到0.95以上;以28个共有峰的相对峰面积进行主成分分析,23批次样品均落在同一区域。相似度评价和主成分分析结果均表明,王老吉广东凉茶颗粒的产品质量稳定性好。该方法快速、高效、稳定,可用于王老吉广东凉茶颗粒的质量控制。 相似文献

The aim of this work was to characterize biogenic amines (BAs) in different parts of Lycium barbarum L. using HPLC with dansyl chloride derivatization, and jointly, to provide referential data for further exploration and utilization of Lycium barbarum L. The linear correlation coefficients for all BAs were above 0.9989. The limits of detection and quantification were 0.015–0.075 and 0.05–0.25 μg/mL, respectively. The relative standard deviations for the intra-day and inter-day precision were 0.66–2.69% and 0.91–4.38%. The described method has good repeatability and intermediate precision for the quantitative determination of BAs in different parts of Lycium barbarum L. Satisfactory recovery for all amines was obtained (79.3–110.3%). The result showed that there were four kinds of BAs. The highest putrescine content (20.9 ± 3.2 mg/kg) was found in the flower. The highest histamine content (102.7 ± 5.8 mg/kg) was detected in the bark, and the highest spermidine (13.3 ± 1.6 mg/kg) and spermine (23.7 ± 2.0 mg/kg) contents were detected in the young leaves. The high histamine (HIS) content in the bark may be one of the reasons why all of the parts of Lycium barbarum L., except the bark, are used for medicine or food in China. A glycoconjugate designated as LbGp5B was isolated from the fruit of Lycium barbarian L. and purified to homogeneity by gel filtration. LbGpSB is composed of rhamnose (Una), arabinose (Ara), galactose (Gal), glucose (Glc), galacturonic add (GalA) and seventeen amino adds. The molecular weight of LbGpSB was determined by sodium dodecyl sulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) and by matrix‐assisted laser desorption/ionization (MALDI) time of fight (TOF) mass spectrometry (MS). A system for transformation and regeneration of Lycium barbarum L. , an important Chinese medical plant, has been established. Young stem segments from Lycium barbarum L. were infected with Agrobacterium tumefaciens C58cl(pGV3850: :ineo1130), and the transformed calli selected from the callus induction medium containing 50 fig/ml kanamycin could regenerate buds on differentiation medium containing 25 μg/ml kanamycin. 30% of the regenerated buds were normal in morphology. The normal buds could develop into whole plantlets after they were transferred to the rooting medium to induce roots. Nopa-line detection, NPT-Ⅱ enzyme activity assay and Southern blotting hybridization indicated that the foreign genes had been integrated into the genome of Lycium barbarum L. and expressed in the plant. Lycium barbarum L. (LB) fruits have high nutritive values and therapeutic effects. The aim of this study was to comprehensively evaluate the differences in phenolic composition of LB fruits from different geographical regions. Different methods of characterization and statistical analysis of data showed that different geographic sources of China could be significantly separated from each other. The highest total phenolic compound (TPC) content was observed in LB fruits from Ningxia (LBN), followed by those from Gansu (LBG) and Qinghai (LBQ). The Fourier transform infrared (FTIR) spectra of LB fruits revealed that LBQ had a peak at 2972 cm−1 whereas there was no similar peak in LBG and LBQ. A new HPLC method was established for the simultaneous determination of 8 phenolic compounds by quantitative analysis of multiple components by a single marker (QAMS), including 4 phenolic acids (chlorogenic acid, caffeic acid, 4-hydroxycinnamic acid, and ferulic acid), 1 coumarin (scopoletin), and 3 flavonoids (kaempferol-3-O-rutinoside, rutin, and narcissoside). It was showed that rutin was the most dominant phenolic compound in LBQ, although the average content of 4 phenolic acids was also high in LBQ, and scopoletin was the richest in LBG. UHPLC-Q-TOF-MS was used to qualitatively analyze the phenolics, which showed LBN was abundant in phenolic acids, LBQ was rich in flavonoids, and coumarins were the most plentiful in LBG. In conclusion, this study can provide references for the quality control and evaluation of phenolics in LB fruits and their by-products. 相似文献

Water-distilled essential oils from the fruits of Lycium barbarum and L. ruthenicum were analyzed by GCMS. The main components in the oil of L. barbarum were found to be hexadecanoic acid (47.5%), linoleic
acid (9.1%), β-elemene (5.4%), myristic acid (4.2%), and ethyl hexadecanoate (4.0%). The essential oil of L. ruthenicum has heptacosane (14.3%), ethyl linoleate (10.0%), hexacosane (7.0%), nonacosane (6.2%), and ethyl hexadecanoate (5.8%) as

Novel food, super food, wolfberry, the Himalayan longevity fruit. Several different and, even more, colourful names define the fruits of Lycium barbarum L., one of the most popular food supplements and anti-aging remedies of the recent years. Goji derivatives find application at the interface between food and medicine thanks to the claimed antioxidant and immunomodulatory properties. Aiming at identifying the most effective extraction protocol for processing different goji berries samples (fresh and dried, commercial and from an Italian cultivar), combinations of extractive and analytical techniques were attempted. The concentration of bioactive compounds and of total phenols were measured and the antioxidant activity of the obtained extracts was studied. The results highlight the most suitable combination of source and process to obtain an extract that can be suitable for nutraceutical applications. 相似文献

The detailed structure of an O‐glycan derived from the fruit of Lycium barbarum L. was elucidated based on glycosidic linkage analysis, complete and partial acid hydrolysis, 1H‐NMR and 13C NMR spectroscopy. According to the experiments, the carbohydrate was in the form of polysacchride (arabinogalactan) chains with highly branched 3, 4‐galactans and terminal arabinofuranosyl substituents. The immuno‐modulating mechanism of glycoconjugate and its glycan were investigated using tritium thymidine incorporation assay, flow cytometry assay and electrophoretical mobility shift assay (EMSA). The results suggested that the immunoactive components of the fruit of Lycium barbarum L. could enhance the splenocyte proliferation in normal mice and the effects of glycan chain were stronger than those of glycoconjugate. The target cell was most likely to be B‐lymphocyte, on which existed receptor binding site acting with the glycan. In addition, the immuno‐stimulatory effect of glycoconjugate (LbGp4) and its glycan (LbGp4‐OL) was associated with activating the expression of nuclear factor KB (NF‐KB) and activator protein 1 (AP‐1). 相似文献

干枸杞经粉碎、匀浆、离心后,通过阳离子交换柱脱去样品中其它氨基酸,再通过Zorbax-C8柱进行柱前衍生分离。衍生剂:A.4%OPA甲醇溶液;B.尿素∶磷酸钠盐缓冲液(pH6.8)=1∶3(W/V)。流动相:甲醇∶0.01mol/L乙酸钠溶液(pH6.8)=35∶65(V/V)。紫外检测波长330nm。牛磺酸浓度在0.1~1.0mmol/L范围内可被定量测定。回收率可达100.31%±1.98%,变异系数(CV)为1.94%。 相似文献
