The feasibility of a microcolumn electrophoresis technique was investigated with a 100 mm length, 2 mm I.D. fused-silica microcolumn packed with uniform quartz microncrystals prepared by hydrothermal synthesis. To evaluate the separation technique, tryptophan, phenylalanine and tyrosine were primarily separated by the microcolumn electrophoresis and detected at 216 nm without derivatization by an ordinary spectrophotometer. The separation conditions of the amino acids were optimized. With 1.5 mmol/L disodium phosphate buffer solution (pH 11.5) containing 25% (v/v) methanol and 10% (v/v) acetonitrile, the three amino acids were separated and the separation efficiency of tryptophan was 4.5 × 104 plates/m. The limits of detection were 0.035, 0.22 and 0.20 μmol/L, respectively. The sample capacity of the electrophoretic microcolumn achieved 35 μL. The proposed method was used to determine these amino acids in compound amino acid injection samples without derivatization. For the simplicity and portability of the microcolumn electrophoresis, it is studied as one of the high-performance separation techniques for an in situ and real-time electrokinetic flow analysis system. For its high detection sensitivity and large sample capacity, it can be developed for preparative electrophoresis.  相似文献

A liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based method for the detection of pesticides in tap and treated wastewater was developed and validated according to the ISO/IEC 17025:1999. Key features of this method include direct injection of 100 μL of sample, an 11 min separation by means of a rapid resolution liquid chromatography system with a 4.6 mm × 50 mm, 1.8 μm particle size reverse phase column and detection by electrospray ionization (ESI) MS-MS. The limits of detection were below 15 ng L−1 and correlation coefficients for the calibration curves in the range of 30-2000 ng L−1 were higher than 0.99. Precision was always below 20% and accuracy was confirmed by external evaluation. The main advantages of this method are direct injection of sample without preparative procedures and low limits of detection that fulfill the requirements established by the current European regulations governing pesticide detection.  相似文献

This work reports a sequential-injection analysis (SIA) method for the enzymatic assay of glucose with soluble glucose oxidase (GOD) and on-line sample dilution with chemiluminescence (CL) detection. A zone of sample was aspirated in the holding coil of the SIA manifold and, if necessary, was diluted on-line by means of an auxiliary dilution conduit. Then, a zone of GOD was aspirated adjacent to the sample zone and a stopped-flow period was applied to allow the enzymatic reaction to proceed with production of hydrogen peroxide. Then, zones of a catalyst (Co(II) solution) and alkaline luminol were aspirated into the holding coil. Finally, the flow was reversed and the stacked zones were sent to a flow-cell located in front of a photomultiplier tube (PMT) that monitored the CL intensity. The linear dynamic range was 1 × 10−5-1 × 10−3 mol L−1 glucose, the coefficient of variation at 8 × 10−5 mol L−1 of glucose was sr = 3.1% (n = 8), the limit of detection at the 3σ level was cL = 1 × 10−6 mol L−1 and the sampling frequency was 28 h−1. With on-line dilution by a factor of 1/200, the linear range could be extended up to 0.2 mol L−1 glucose. The advantages of the proposed method are the simple manifold and instrumentation used, the scope for automated on-line dilution, the low consumption of sample and reagents and the elimination of enzyme immobilisation procedures. The method was applied to the analysis of commercial drinks and honey with percent relative errors in glucose determination in the range 100 ± 6.1%.  相似文献

The voltammetric behaviour of Sildenafil citrate (SC) and its main metabolite UK-103,320 (UK) was studied by square wave techniques, leading to two methods for its total determination in biological samples (urine and human serum). The application of the square wave (SW), without the adsorptive accumulation, shows a maximum response at −0.950 V. The effect of experimental parameters that affect this determination are discussed. The stripping technique, proved to be more sensitive, yielding signals three times larger than those obtained by applying a square wave scan without the previous accumulation. Two calibration graphs to determine total SC and UK concentration were established. Calibration graph in urine sample was linear in the range 4.4×10−8 to 4.8×10−7 M by the stripping mode with an accumulation time of 10 s. In the same conditions but in serum sample regression line was linear in the range 3.4×10−8 to 9.7×10−7 M. The two proposed methods (SW and square-wave adsorptive stripping voltammetry (SWAdSV)) were applied to the total concentration analysis in eight different biological samples by the standard addition method with satisfactory results in the four different serum samples by the SW and in the four urines by SWAdSV.  相似文献

This paper presents a simple, rapid and reproducible method of analysis of salbutamol in pharmaceutical products, utilizing batch injection analysis (BIA) associated with amperometric detection. A study of salbutamol oxidation demonstrated a strong dependence between electrode fouling and pH. All determinations were done utilizing a glassy carbon electrode in presence of 3.0 mol l−1 NaOH. A large linear dynamic range from 8×10−7 to 2×10−4 mol l−1 was obtained by using an injected volume of 100 μl with a detection limit of 2.5×10−7 mol l−1. R.S.D. of 0.92% for 50 successive injections of 4×10−6 mol l−1 of salbutamol and a sample throughput of 60 samples per hour were achieved. The method was applied for salbutamol quantification in syrups.  相似文献

Simvastatin (SS) is an effective cholesterol-lowering medicine, and is hydrolyzed to simvastatin acid (SSA) after oral administration. Due to SS and SSA inter-conversion and its pH and temperature dependence, SS and SSA quantitation is analytically challenging. Here we report a high-throughput salting-out assisted liquid/liquid extraction (SALLE) method with acetonitrile and mass spectrometry compatible salts for simultaneous LC-MS/MS analysis of SS and SSA. The sample preparation of a 96-well plate using SALLE was completed within 20 min, and the SALLE extract was diluted and injected into an LC-MS/MS system with a cycle time of 2.0 min/sample. The seamless interface of SALLE and LC-MS eliminated drying down step and thus potential sample exposure to room or higher temperature. The stability of SS and SSA in various concentration ratios in plasma was evaluated at room and low (4 °C) temperature and the low temperature (4 °C) was found necessary to maintain sample integrity. The short sample preparation time along with controlled temperature (2-4 °C) and acidity (pH 4.5) throughout sample preparation minimized the conversion of SS → SSA to ≤0.10% and the conversion of SSA → SS to 0.00% The method was validated with a lower limit of quantitation (LLOQ) of 0.094 ng mL−1 for both SS and SSA and a sample volume of 100 μL. The method was used for a bioequivalence study with 4048 samples. Incurred sample reproducibility (ISR) analysis of 362 samples from the study exceeded ISR requirement with 99% re-analysis results within 100 ± 20% of the original analysis results.  相似文献

Dynamic gas sampling using solid phase microextraction (SPME) was evaluated for recovery of reactive terpenes and terpenoids in the presence of ozone. For limonene, α-terpineol and dihydromyrcenol in the 20-60 ppb range, this method achieves >80% recovery for ozone mixing ratios up to 100 ppb. Both the experimental results and a model analysis indicate that higher ozone concentrations and longer sampling times result in lower percent recovery. Typically greater than 90% recovery and ppb level method detection limits were achieved with a 5 min sample time. Increasing the flow rate from 100 to 400 sccm flow (5-20 cm s−1) through the active sampler did not significantly affect sensitivity or recovery in most cases, probably due to negligible mass-transfer improvements. The recovery for each compound improves when sampling from a mixture of different species than that from a single compound sample. This may be due to competition for ozone amongst adsorbed species. Dynamic SPME sampling can improve detection and quantification of terpenes in reactive environments, especially for low vapor pressure (<5 mm Hg at 25 °C) compounds that can be adsorbed to ozone scrubbers used in other methods.  相似文献

A simple, inexpensive and reliable method for the routine analysis of riboflavin in beer by capillary electrophoresis-light emitting diode (CE-LED) induced fluorescence detection is described. A simple and straightforward sample preparation is involved and the method is based on an inexpensive blue LED as the light source combined with an on-line sample concentration technique. For this detection system, using a normal micellar electrokinetic chromatography (MEKC), stacking-MEKC and dynamic pH junction techniques, the detection limits were found to be 480, 20 and 1 ng mL−1, respectively. In addition, the number of theoretical plates for riboflavin was determined to be 3.8×104 by means of a dynamic pH junction and this was improved to 3.2×106 when the dynamic pH junction-sweeping mode was applied. The concentrations of riboflavin in 12 samples of different types of commercial beer were found to be in the range of 130-280 ng mL−1.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are common environmental pollutants in both living and working environments. The aim of this study was the development of a headspace solid-phase microextraction gas chromatography-isotope dilution mass spectrometry (HS-SPME/GC-IDMS) method for the simultaneous quantification of 13 PAHs in urine samples. Different parameters affecting PAHs extraction by HS-SPME were considered and optimized: type/thickness of fiber coatings, extraction temperature/time, desorption temperature/time, ionic strength and sample agitation. The stability of spiked PAHs solutions and of real urine samples stored up to 90 days in containers of different materials was evaluated. In the optimized method, analytes were absorbed for 60 min at 80 °C in the sample headspace with a 100 μm polydimethylsiloxane fiber. The method is very specific, with linear range from the limit of quantification to 8.67 × 103 ng L−1, a within-run precision of <20% and a between-run precision of <20% for 2-, 3- and 4-ring compounds and of <30% for 5-ring compounds, trueness within 20% of the spiked concentration, and limit of quantification in the 2.28-2.28 × 101 ng L−1 range. An application of the proposed method using 15 urine samples from subjects exposed to PAHs at different environmental levels is shown.  相似文献

The separation and determination of four parabens (methyl, ethyl, propyl, and butyl p-hydroxybenzoate) which are commonly used as preservatives in cosmetic products, by micellar electrokinetic capillary chromatography (MEKC) with and without large-volume sample stacking (LVSS) technique were compared. As an effective on-line concentration technique, LVSS was successfully combined with MEKC to determine neutral parabens in an acidic media. The effects of some typical parameters such as sample volume, buffer pH, temperature, and concentration of surfactant were examined. The detection limits for this LVSS-MEKC method were found to be 3.0 × 10−7 M for each of the parabens based on the signal-to-noise ratio of 3, which were around 300 times lower than normal MEKC technique. The curves of peak response versus concentration were linear from 1.0 × 10−6 to 5.0 × 10−5 M with regression coefficients of 0.9987, 0.9960, 0.9925 and 0.9864, respectively. A simple and easy-manipulative sample preparation method was developed and validated by analyzing commercially available cosmetic samples. It was found that with current sample preparation process and instrumentation system, 0.5 g of sample is enough for the analysis of parabens preservatives in cosmetic product with satisfactory results.  相似文献

A poly (acrylamide-co-methylenebisacrylamide) (poly (AAm-co-MBA)) monolith was prepared by thermal polymerization in the 100 or 250 μm i.d. capillary. The monolithic support was activated by ethylenediamine followed by glutaraldehyde. Trypsin was then introduced to form an immobilized enzyme reactor (IMER). The prepared IMER showed a reliable mechanical stability and permeability (permeability constant K = 2.65 × 10−13 m2). With BSA as the model protein, efficient digestion was completed within 20 s, yielding the sequence coverage of 57%, better than that obtained from the traditional in-solution digestion (42%), which took about 12 h. Moreover, BSA down to femtomole was efficiently digested by the IMER and positively identified by matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). To test the applicability of IMER for complex sample profiling, proteins extracted from Escherichia coli were digested by the IMER and further analyzed by nanoreversed phase liquid chromatography-electrospray ionization-mass spectrometry (nanoRPLC-ESI-MS/MS). In comparison to in-solution digestion, despite slightly fewer proteins were positively identified at a false discovery rate (FDR) of ∼1% (333 vs 411), the digestion time used was largely shortened (20 s vs 24 h), implying superior digestion performance for the high throughput analysis of complex samples.  相似文献

A modified Rheodyne 7520 microsample injector was used as a new solid phase microextraction (SPME)–liquid chromatography (LC) interface. The modification was focused on the construction of a new sample rotor, which was built by gluing two sample rotors together. The new sample rotor was further reinforced with 3 pieces of stainless steel tubing. The enlarged central flow passage in the new sample rotor was used as a desorption chamber. SPME fiber desorption occurred in static mode. But all desorption solvent in the desorption chamber was injected into LC system with the interface. The analytical performance of the interface was evaluated by SPME–LC analysis of PAHs in water. At least 90% polycyclic aromatic hydrocarbons (PAHs) were desorbed from a polyacrylonitrile (PAN)/C18 bonded fuse silica fiber in 30 s. And injection was completed in 20 s. About 10–20% total carryovers were found on the fiber and in the interface. The carryover in the interface was eliminated by flushing the desorption chamber with acetonitrile at 1 mL min−1 for 2 min. The repeatability of the method was from 2% to 8%. The limit of detection (LOD) was in the mid pg mL−1 range. The linear ranges were from 0.1 to 100 ng mL−1. The new SPME–LC interface was reliable for coupling SPME with LC for both qualitative and quantitative analysis.  相似文献

In this paper, a rapid and effective method based on capillary zone electrophoresis (CZE) coupled with electrospray ionization mass spectrometry (ESI-MS) was established for the trace analysis of microcystin (MC) isomers in crude algae sample. The experimental conditions including the composition, acidity and concentration of buffer, separation voltage, injection time, and MS detection parameters were investigated in detail. A capillary separation system was as follows: a uncoated fused-silica capillary tube (50 μm i.d. × 90 cm), 40 mmol L−1 ammonium acetate solution (pH 9.86) as running buffer, 25 kV as separation voltage, 20 kV × 3 s water first and 20 kV × 20 s for sample injection. Mass analysis was performed in ESI source, with sheath gas temperature 150 °C, sheath gas pressure 10 psi, and sheath gas flow 6 L min−1. And sheath liquid was 7.5 mmol L−1 acetic acid in 50% isopropanol-water (3 μL min−1). Protonation and ammonium adduct molecular ions m/z 506.9 (MC-LR) and 532.0 (MC-YR) were used for the quantification of MCs. Under these conditions, two MCs were baseline separated within 9 min, the calibration curves were obtained in the range of 0.11-10.0 μg mL−1 and 0.16-10.5 μg mL−1 for MC-LR and MC-YR, respectively. Meanwhile, limits of detection were 0.05 and 0.08 μg mL−1 for MC-LR and MC-YR, respectively. The recoveries for the two MCs were in the range of 95.8-108%. The developed approach had been successfully applied to the analysis of MCs in crude algae samples.  相似文献

The voltammetric behavior of cefixime was studied using cyclic, linear sweep, differential pulse and square wave voltammetric techniques. The oxidation of cefixime was irreversible and exhibited diffusion controlled process depending on pH. The oxidation mechanism was proposed and discussed. Different parameters were tested to optimize the conditions for the determination of cefixime. The dependence of current intensities and potentials on pH, concentration, scan rate, nature of the buffer was investigated. According to the linear relationship between the peak current and the concentration, differential pulse (DPV) and square wave (SWV) voltammetric methods for cefixime assay in pharmaceutical dosage forms and biological fluids were developed. For the determination of cefixime were proposed in acetate buffer at pH 4.5, which allows quantitation over the 6 × 10−6-2 × 10−4 M range in supporting electrolyte and spiked serum sample; 8 × 10−6-2 × 10−4 M range in urine sample; 6 × 10−6-1 × 10−4 M range in breast milk samples for both techniques. The repeatability, reproducibility, precision and accuracy of the methods in all media were investigated. No electroactive interferences from the excipients and endogenous substances were found in the pharmaceutical dosage forms and in the biological samples, respectively.  相似文献

A simple sample preparation method was developed by using a centrifugal ultrafiltration (CF-UF) device with hollow fiber (HF) for the determination of cefaclor in plasma by HPLC. Samples were placed into a homemade device, which was consisted of a glass tube and a U-shaped hollow fiber. The filtrate was withdrawn from the hollow fiber into a syringe after centrifugation and 20 μL was directly injected into the HPLC for analysis. The HPLC method had a linear calibration curve in the concentration range of 6.00 × 10−2–30.7 μg mL−1(r = 0.9996). The limit of detection (LOD) and limit of quantitation (LOQ) were 0.02 and 0.06 μg mL−1, respectively. The intra and inter-day precisions (RSD) were 1.7%, 1.2%, 1.0% and 3.6%, 2.5%, 1.9%, respectively, for three concentrations. Assay accuracy was higher than 99.2% and the absolute recovery was 86.8–92.5%. It is feasible to use this novel and low cost device for sample pretreatment for the analysis of cefaclor in plasma.  相似文献

In this work, two kinds of chelating resin, bis(2-aminoethylthio)methylated resin (BAETM) and γ-aminobutyrohydroxamate resin (γ-ABHX) were synthesized. Of these, the former has a hydrophobic skeleton, and the latter a hydrophilic skeleton. The functionalities of each were 0.91 and 2.21 mmol g−1, respectively. The chelating behavior of these resins towards vanadium, molybdenum and tungsten as a function of pH was studied. To perform trace metals analysis in complex matrices, a hyphenated method-chelation ion chromatography (CIC) coupled on-line detection with inductively coupled plasma mass spectrometry (ICP-MS) was developed. With a BAETM resin column (5×0.4 cm i.d.) as the separator, a sample volume of 20 μl, nitric acid (pH 1.5) as the eluent and a flow rate of 1 ml min−1, the detection limits for the determination of vanadium, molybdenum and tungsten were lower than 0.05 ng ml−1and the linear ranges were up to 100 ng ml−1 for each element. By increasing the injected sample volume to 250 μl, the resin concentrator improved the detection limit to 0.01 ng ml−1. For the determination of these elements (5 ng ml−1 for each) spiked in artificial sea water samples, γ-ABHX resin column (3×0.6 cm i.d.) demonstrated well resolved peak separation between the analytes and the matrix elements—calcium and magnesium, by using sodium nitrate (10 ml, 10−4 M) as the eliminator.  相似文献

Conventional immunoassays on microtiterplates are very useful analytical tools in environmental analysis, but the long assay times, usually in the range of hours, are a drawback. To overcome this disadvantage, the development of fast (express) assay formats is described, which use polyelectrolytes as carriers. Two semi-homogeneous immunochemical methods, namely the polyelectrolyte-ELISA (enzyme-linked immunosorbent assay) and the express-FIIAA (flow injection immunoaffinity analysis) for the analysis of the herbicide atrazine were set-up. Using polyclonal antisera for atrazine, the following results were obtained. Standard curves for atrazine showed a linear range from 3 to 100 μg l−1 in polyelectrolyte-ELISA and 0.3-100 μg l−1 in express-FIIAA. The test midpoints in polyelectrolyte-ELISA and express-FIIAA were 12 and 5 μg l−1, respectively. The duration time of the immunochemical reaction was in both assays 15 min, but the total assay time differed (30 min (polyelectrolyte-ELISA) and 18 min (express-FIIAA)). A significant difference between the formats could be observed in the number of samples that can be determined per day. The polyelectrolyte-ELISA can handle samples in parallel on a microtiterplate (usually 20/plate), whereas in the express-FIIAA the samples are automatically analysed one after another. This first demonstration of these techniques shows the potential of these methods, but also their limitations.  相似文献

In this work, a large-volume on-column injection method for the analysis of 21 organochlorine pesticides, including HCH isomers, DDT derivatives and cyclodiene derivatives, was optimized. The solvent selected to carry out the study was ethyl acetate and the injection volume was 100 μl. Some factors were introduced in a Plackett-Burman design to determine their influence in the vaporization efficiency. The significant factors were then studied by a univariate procedure and sorted according to their importance on the response. The effect of the injection conditions on the peak resolution was also noted. The conditions selected according to sensitivity and resolution were: initial oven temperature 75 °C, injection speed 20 μl s−1, solvent vapor exit (SVE) valve closure time 60 s, initial pressure 100 kPa and isothermal oven time 1 min. Analytical characteristics expressed in terms of precision, linear range, and limit of detection have been determined and compared with those obtained by splitless injection. The degradation of endrin and p,p′-DDT thermolabile pesticides was evaluated for both injection techniques. Finally the developed method was successfully applied to the ultratrace analysis of pesticides in natural waters. With this purpose a micro liquid-liquid extraction method using 2 ml of ethyl acetate to extract 10 ml of water sample was proposed. Recoveries between 69 and 107% were obtained with a very good precision (0.2-1.3%) for the studied pesticides, except for p,p′-DDD. Detection limits between 0.3 and 25 ng l−1, which fulfill the limits established by the new water directive 2008/105/EC, were achieved. The MLLE method was compared with the SPE method by the analysis of some water samples using both procedures, and good concordance was obtained.  相似文献

In this study a method for the determination of cadmium in fuel alcohol using solid-phase extraction with a flow injection analysis system and detection by flame atomic absorption spectrometry was developed. The sorbent material used was a vermicompost commonly used as a garden fertilizer. The chemical and flow variables of the on-line preconcentration system were optimized by means of a full factorial design. The selected factors were: sorbent mass, sample pH, buffer concentration and sample flow rate. The optimum extraction conditions were obtained using sample pH in the range of 7.3-8.3 buffered with tris(hydroxymethyl)aminomethane at 50 mmol L−1, a sample flow rate of 4.5 mL min−1 and 160 mg of sorbent mass. With the optimized conditions, the preconcentration factor, limit of detection and sample throughput were estimated as 32 (for preconcentration of 10 mL sample), 1.7 μg L−1 and 20 samples per hour, respectively. The analytical curve was linear from 5 up to at least 50 μg L−1, with a correlation coefficient of 0.998 and a relative standard deviation of 2.4% (35 μg L−1, n = 7). The developed method was successfully applied to spiked fuel alcohol, and accuracy was assessed through recovery tests, with recovery ranging from 94% to 100%.  相似文献

A boron-doped diamond (BDD) electrode was used for the electroanalytical determination of estriol hormone in a pharmaceutical product and a urine sample taken during pregnancy by square-wave voltammetry. The optimized experimental conditions were: (1) a supporting electrolyte solution of NaOH at a pH of 12.0, and (2) a frequency of 20 Hz, a pulse height of 30 mV and a scan increment of 2 mV (for the square-wave parameters). The analytical curve was linear in the concentration range of 2.0 × 10−7 to 2.0 × 10−5 mol L−1 (r = 0.9994), with a detection limit of 1.7 × 10−7 mol L−1 and quantification limit of 8.5 × 10−7 mol L−1. Recoveries of estriol were in the range of 98.6-101.0%, for the pharmaceutical sample, and 100.2-103.4% for the urine sample, indicating no significant matrix interference effects on the analytical results. The accuracy of the electroanalytical methodology proposed was compared to that of the radioimmunoassay method. The values for the relative error between the proposed and standard methods were −7.29% for the determination of estriol in the commercial product and −4.98% in a urine sample taken during pregnancy. The results obtained suggest a reliable and interesting alternative method for electroanalytical determination of estriol in pharmaceutical products and urine samples taken during pregnancy using a boron-doped diamond electrode.  相似文献
