The alkylation of imidazole-4(5)-carboxylic acid anilide and methyl ester with bromoacetic acid and its methyl ester in neutral and alkaline media yields, along with monoalkylation products, 1,3-bis(2-methoxycarbonylmethyl)-4-R-imidazolium bromide. The monoalkylation of methyl imidazole-4(5)-carboxylate yields a 1:1 mixture of the 1,4- and 1,5-isomers, and the monoalkylation of imidazole-4(5)-carboxylic anilide selectively provides the 1,4-isomer.  相似文献

We have used 1H NMR to study the amine-imine tautomerism of some esters and amides of 2-N-labeled 4-methylthiazole-5-carboxylic acids and 5-carbamic acids. Derivatives of the anilide or ethyl ester of 5-cyano-6-oxo-2-styrylnicotinic acid are formed in the reaction of the anilide or ethyl ester of 5-cyano-2-methyl-6-oxonicotinic acid with aromatic aldehydes. On interaction with hydrazine hydrate the products are converted into hydrazides of 5-cyano-6-oxo-2-styrylnicotinic acid derivatives.  相似文献

Derivatives of 8-methylquinoline-5-carboxylic acid were synthesized. Alcoholysis of 8-methyl-5-cyanoquinoline gave methyl and ethyl 8-methylquinoline-5-carboxylates, which were converted to the corresponding 8-bromomethyl derivatives. The latter were used for the introduction of a methylamino group and the synthesis of N-acyl derivatives. We have carried out solvolysis of the previously described ethyl esters of 3-methyl(aryl)-4-methyl-2-thioxothiazoline-5-carboxylic acids, leading to the corresponding acids without breaking down the heterocycle. We synthesized a series of novel esters from the latter by treatment with dimethyl sulfate or reactive halides. Of these, only in the case of the ethyl ester of 3,4-dimethyl-2-thioxothiazoline-5-carboxylic acid did we obtain the hydrazinolysis product (the hydrazide), from which we synthesized novel hydrazones by treatment with aldehydes and ketones.  相似文献

A number of 8-hydroxy-6-methyl-1,6-naphthyridin-5(6H)-one-7-carboxylic acid alkyl esters 3 and the isomeric 5-hydroxy-7-methyl-1,7-naphthyridin-8(7H)-one-6-carboxylic acid alkyl esters 4 were synthesized from acyclic precursors obtained starting from quinolinic anhydride 5. Thus, methanolysis of 5 afforded the hemiester 6 which treated with oxalyl chloride and sarcosine ethyl ester gave 3-(N-ethoxycarbonylmethyl-N-methylcarbamoyl)pyridine-2-carboxylic acid methyl ester 8. Compound 8 was cyclized to naphthyridines 3a-e with sodium alkoxides. The isomeric naphthyridines 4a-c were obtained by cyclization of the open intermediary 2-(N-ethoxycarbonylmethyl-N-methylcarbamoyl)pyridine-3-carboxylic acid methyl ester 9 obtained by a route that involves treatment of 5 with sarcosine ethyl ester and esterification with diazomethane. Spectroscopic properties (1H nmr, uv, ir) of compounds 3 and 4 are discussed and confirmed the proposed structures.  相似文献

Diazotization of 2-amino-1,3,4-thiadiazoles gave 1,3,4-thiadiazole-2-diazonium sulfates which were converted to 2-azido-1,3,4-thiadiazoles. The latter reacted with ethyl acetoacetate in the presence of sodium methoxide in methanol to produce 1-(5-R1-1,3,4-thiadiazol-2-yl)-5-R2-1H-1,2,3-triazole-4-carboxylic acid derivatives. The reactions of 2-azido-5-methyl-1,3,4-thiadiazole and 2-azido-1,3-thiazole with ethyl 3-(1,3-benzodioxol-5-yl)-3-oxopropanoate led to the formation of 1,2,3-triazole ring under milder conditions (K2CO3, DMSO). Various 1,2,3-triazole-4-carboxylic acid derivatives were synthesized.  相似文献

Treatment of aminopyrimidines with a mixture of PhCOCl and NH4SCN and subsequent debenzoylation of the N-benzoyl-N′-(4,6-substituted pyrimidin-2-yl)thioureas obtained gave pyrimidinyl-2-thioureas. The reaction of ethyl esters of 4-methyl-2-oxo-2H-l-benzo(naphtho) pyran.3-carboxylic acids (1) with aromatic aldehy-des in the presence of piperidine yielded 4-styryl-3-carboxami-dopiperidyl connmrin derivatives 4. The reaction of hydrazlne hydrate with 1 gave acetophenone hydrozone derivatives 5 and acetophenone azine derivatives 6. The reaction of 1 with prima-ry amines afforded compomlds 7—9. Abstract

Two novel methods for synthesis of nicotinic acid derivatives with unsymmetrical substitution patterns were presented via ketene dithioacetals. The ketene N,S-acetals 2 reacted with β-ketoesters or β-cyanoesters to give 4-amino-5-cyano-2-alkyl-6-methylthio-nicotinic acid derivatives 3 or 2,4-diamino-5-cyano-6-methylthio-nicotinic acid ethyl ester 4. However, 6-amino-5-cyano-2-alkyl-4-methylthio-nicotinic acid esters 6 were obtained by the reaction of the ketene dithioacetals 1 and β-amino-crotonates.  相似文献

Acetylcholinesterase inhibitors are the most frequently prescribed anti-Alzheimer's drugs. A series of 5H-thiazolo[3,2-a]pyrimidine-6-carboxylic acid ethyl ester derivatives as the novel acetylcholinesterase inhibitors was designed based on virtual screening methods. Several new 3-arylsulfonylmethyl-1,2,4-oxadiazole-5-carboxylic acid derivatives have been synthesized. A typical example, 3-[(4-chlorophenylsulfonyl)methyl]-1,2,4-oxadiazole-5-carboxylic acid ethyl ester ( 2c ), was prepared from the reaction of 2-(4-chlorophenylsulfonyl)acetamide oxime ( 1c ) with ethyl oxalyl chloride. The hydrazide derivative ( 3f ) showed antihypertensive activity in rats. Various structural modifications to improve activity are discussed.  相似文献

Cycloaddition of intermediates formed from the nucleophilic addition of aryl isothiocyanate to acidic cyanomethylenes and α-halocarbonyl compounds gave only 4-thiazolidinones. 5-Thiazolidinones were not observed. Cyano-(4-oxo-3-phenylthiazolidin-2-ylidene)-acetic acid ethyl ester (1) and cyano-[5-(4-methoxybenzylidene)-4-oxo-3-phenylthiazolidin-2-ylidene]-acetic acid ethyl ester (2a) also were shown to exhibit moderate antiviral activity.

以甘草次酸(1)为原料,将其11位羰基还原、30位羧基酯化得11-脱氧甘草次酸-30-乙酯(3)。再以四氢呋喃为溶剂,N,N’-二环己基碳二亚胺(DCC)/4-二甲基氨基吡啶(DMAP)为偶合剂,选用Fmoc保护氨基酸对11-脱氧甘草次酸-30-乙酯的3位羟基进行酯化,得到11-脱氧甘草次酸-30-乙酯-3位氨基酸酯衍生物(4a~4d)。化合物4a~4d在V(CHCl2)∶V(Et2NH)=1∶1溶液中脱去Fmoc保护基得到最终产物(5a~5d),产率80%~87%。化合物5a~5d用1H NMR、EI-MS进行了表征。活性实验结果表明,化合物5a~5d对在高浓度二甲基甲酰胺下生长的枯草芽孢杆菌、大肠杆菌和酵母菌具有保护作用。  相似文献

A simple synthesis of furo[2,3-c]pyridine and its 2- and 3-methyl derivatives from ethyl 3-hydroxyisonicotinate ( 2 ) is described. The hydroxy ester 2 was O-alkylated with ethyl bromoacetate or ethyl 2-bromopropionate to give the diester 3a or 3b . Cyclization of compound 3a afforded ethyl 3-hydroxyfuro [2,3-c]pyridine-2-carboxylate ( 4 ) which was hydrolyzed and decarboxylated to give furo[2,3-c]pyridin-3(2H)-one ( 5a ). Cyclization of 3b gave the 2-methyl derivative 5b . Reduction of 5a and 5b with sodium borohydride yielded the corresponding hydroxy derivative 6a and 6b , respectively, which were dehydrated with phosphoric acid to give furo[2,3-c]pyridine ( 7a ) and its 2-methyl derivative 7b . 4-Acetylpyridin-3-ol ( 8 ) was O-alkylated with ethyl bromoacetate to give ethyl 2-(4-acetyl-3-pyridyloxy) acetate ( 9 ). Saponification of compound 9 , and the subsequent intramolecular Perkin reaction gave 3-methylfuro[2,3-c]pyridine ( 10 ). Cyclization of 9 with sodium ethoxide gave 3-methylfuro[2,3-c]pyridine-2-carboxylic acid, which in turn was decarboxylated to give compound 10 .  相似文献

We have obtained 4-aryl-2-carbamoylmethylthio-5-ethoxycarbonyl-1,4-dihydropyridine-3-carboxylic acid nitriles by S-alkylation of the corresponding 2-thioxo-1,2,3,4-tetrahydropyridine-3-carboxylic acid nitrile by iodoacetamide or one-pot multicomponent synthesis methods: condensation of 2-arylidene-acetoacetic acid ethyl ester, 2-cyanothioacetamide, piperidine, and iodoacetamide; acetoacetic acid ethyl ester, 3-aryl-2-cyanothioacrylamide, piperidine, and iodoacetamide; acetoacetic acid ethyl ester, an aromatic aldehyde, 2-cyanothioacetamide, piperidine, and iodoacetamide. We have carried out a comparative analysis of the capability of 2-alkylthio-4-aryl-5-ethoxycarbonyl-1,4-dihydropyridine-3-carboxylic acid nitriles for electrochemical oxidation as a function of the electronic properties of the aryl substituent in the 4 position of the heterocycle and the 2-alkylthio substituent. X-ray diffraction data indicate the existence of a hydrogen bond between the C=O of the 2-carbamoylmethylthio substituent and the NH of the hydrogenated heterocycle, which explains the more facile oxidation of the studied compounds compared with 2-methylthio-substituted 1,4-dihydropyridines.Dedicated to Academician V. 以取代吡啶为原料,在羟胺-O-磺酸的作用下,得到取代的N-氨基吡啶的硫酸盐,再通过1,3-偶极环加成反应,与丙炔酸乙酯生成吡唑[1,5-a]并吡啶-3-羧酸乙酯衍生物,然后在质量分数30%的NaOH水溶液作用下水解成酸。 该方法将取代的N-氨基吡啶的硫酸盐直接投入到下步反应,省去传统方法中将硫酸盐转化为碘盐的步骤,解决了碘盐不易析出的问题,并将取代的N-氨基吡啶硫酸盐和丙炔酸乙酯分别用水和二甲基甲酰胺溶解后再混合,增加了原料和K2CO3在体系中的溶解性,提高了产率。 本文成功合成了6种化合物(4a~4f),产率为88%~93%,该方法条件温和,后处理简单,成本低,是适合大规模生产的新工艺。  相似文献
