通过金硫键将腺苷适配体互补链(S1)和末端带羧基的DNA链(S2)修饰在金纳米粒子(GNPs)表面,以及甲苯胺蓝(TB)与S2的酰胺反应将TB标记在金纳米粒子表面形成甲苯胺蓝标记的DNA探针分子TB-S2-GNPs-S1,然后在玻碳电极表面电沉积一层金纳米粒子,以其为载体将末端带有巯基的腺苷适配体(Apt)固定在电极表面,以牛血清蛋白为封闭剂消除非特异性吸附,再通过TB-S2-GNPs-S1中的S1与Apt杂交将TB-S2-GNPs-S1负载到电极表面,成功建立了一种以甲苯胺蓝为电化学探针检测腺苷的适配体生物传感器。采用紫外可见光谱和扫描电镜对合成的金纳米粒子和TB-S2-GNPs-S1复合物进行表征。对电极的组装过程采用循环伏安法和电化学阻抗法(EIS)进行表征,对传感器的性能采用差分脉冲法(DPV)和电化学阻抗进行研究。该传感器在1.0×10-4~100.0 ng/m L范围内对腺苷具有良好的信号响应,相关系数(r)为0.994,检出限(S/N=3)为64.7 fg/m L。  相似文献

研制了一种简单和灵敏地检测环境激素双酚A(BPA)的电化学适配体传感器。首先在玻碳电极(GCE)表面采用电沉积法沉积一层多孔纳米金(NP-Au),再采用电聚合法将硫堇(TH)和适配体(APT)一步聚合到电极表面,以聚硫堇(PTH)作为电化学探针和APT的固定化载体,以牛血清白蛋白(BSA)抑制非特异性吸附,构筑GCE/NP-Au/PTH+APT/BSA传感器。采用差分脉冲伏安法对该传感器的电化学性能进行探究,发现双酚A在10.0 fg/mL~1.0 ng/mL浓度范围内有较好的信号响应,检出限为5.3 fg/mL。以GCE/PTH+APT/BSA传感器作为对照,其对双酚A在10.0 fg/mL~1.0 ng/mL浓度范围内呈线性关系,检出限为9.0 fg/mL。结果表明多孔纳米金的引入可有效提高传感器的灵敏度。GCE/NP-Au/PTH+APT/BSA传感器具有选择性高和检出限低等优点。  相似文献

采用电沉积法制备了铁氰化镍(NiHCF)氧化还原电化学探针, 以金纳米粒子(GNPs)为固定核酸适配体的载体构建了非标记型测定凝血酶(TB)和腺苷(AD)的核酸适配体传感器. 采用循环伏安法(CV)和SEM对NiHCF膜进行了表征; 利用电化学阻抗(EIS)对传感器的组装过程进行了监测; 用CV和差分脉冲伏安法(DPV)对该传感器的电化学行为进行了研究. 该传感器对凝血酶的检测在1.0 fg·mL-1~1.0 μg·mL-1范围内成良好的线性关系, 相关系数为0.997, 检测限为0.27 fg·mL-1; 对腺苷的检测在1.0 fg·mL-1~1.0 ng·mL-1范围内成良好的线性关系, 相关系数为0.997, 检测限为0.36 fg/mL. 该传感器制备简单, 灵敏度高, 抗干扰能力强.  相似文献

构建了一种可再生型三磷酸腺苷(ATP)适配体计时库仑电化学传感器.将一条短链DNA通过AuS键自组装固定在电极表面, ATP的核酸适配体与该短链DNA杂交而结合在电极表面.带负电的DNA通过静电吸引结合电解液中的六氨合钌(RuHex)阳离子.当传感器和靶分子ATP孵育后,ATP与核酸适配体结合,使适配体链从电极表面解离,电极表面吸附的DNA量减少,结合RuHex的量随之降低.通过计时库仑技术检测RuHex响应信号降低的量 ,可以对ATP进行定量测定.此传感器的电化学响应信号与ATP浓度对数值呈线性关系,线性检测范围为0.001~100 μmol/L,检出限(S/N=3)为0.5 nmol/L.此传感器检测靶分子ATP后,可以通过简单的操作步骤再生,再生5次后的响应信号为初始信号的90%以上.采用此传感器检测大鼠脑透析液中ATP的含量为(19.2±3.7) nmol/L (n=3).  相似文献

为了检测三磷酸腺苷(ATP)的浓度,利用微系统(MEMS)技术小批量加工薄膜金电极,采用自组装法将巯基修饰的三磷酸腺苷适体固定到金电极表面,以三磷酸腺苷适体作为识别元件,构建了一种基于共面薄膜金电极的三磷酸腺苷适体传感器。依据核酸磷酸骨架荷负电特性静电排斥[Fe(CN)6]3!/4!所引起的阻抗变化实现对ATP浓度的检测。首先采用电化学阻抗谱法研究了裸金电极及ATP加入前后、6-巯基己醇封闭电极前后以及不同自组装时间(3,8,15,24和30 h)条件下,电极在电化学阻抗溶液中阻抗值变化。然后研究了不同浓度ATP适体传感器的电化学阻抗谱以及适体传感器的线性度和重复性。结果表明,在自组装时间为24 h,使用6-巯基己醇封闭金电极的条件下,此传感器线性测量范围可达到1~500 nmol/L,检出限为1 nmol/L,线性相关系数为0.9842。此传感器制作简单,检出限低且重复性好。  相似文献

构建了一种新型的基于二硫化钼/纳米金和硫堇/纳米金信号放大的检测17β-雌二醇的电化学适配体传感器. 利用巯基自组装技术将17β-雌二醇的适配体探针DNA固定在二硫化钼/纳米金修饰玻碳电极表面, 与末端带巯基的部分互补DNA链杂交, 将硫堇/纳米金电化学指示剂自组装在杂交后的双链DNA上, 制备了17β-雌二醇电化学适配体传感器. 二硫化钼/纳米金复合材料增加了电极的有效表面积和DNA探针的固定量. 纳米金作为信号物质载体负载硫堇, 实现了电化学指示剂的信号放大. 加入目标物17β-雌二醇后, 目标物与适配体DNA特异性结合, 导致互补DNA链脱落, 双链上结合的硫堇/纳米金电化学指示剂数量减少, 电化学信号降低. 实验结果表明, 在1.0×10 -14~5.0×10 -12 mol/L范围内17β-雌二醇浓度与峰电流的线性关系良好, 检出限为4.2×10 -15 mol/L(S/N=3). 该传感器可望用于其它环境激素类物质的检测.  相似文献

本文基于双酚A(BPA)与其适配体互补链(cDNA)对适配体的竞争结合作用构建了检测BPA的电化学传感器。制备了金纳米粒子与二硫化钼的纳米复合物(Au-MoS_2),并将其修饰到玻碳电极(GCE)表面,制得修饰电极Au-MoS_2/GCE。通过巯基修饰的cDNA与适配体的杂交反应,生成双链DNA(dsDNA),利用其中cDNA的巯基在金纳米粒子表面生成Au-S键的化学吸附作用,将dsDNA修饰到电极表面。利用具有卟啉平面结构的氯化血红素(hemin)在dsDNA沟槽中的嵌插作用,制得电化学传感器Hemin-dsDNA/Au-MoS_2/GCE。基于hemin对于H_2O_2和对苯二酚(HQ)的化学反应的电催化作用,建立了差分脉冲伏安法(DPV)测定BPA的分析方法,由于BPA与适配体结合的亲和性较强,在一定实验条件下,溶液中BPA浓度越高,导致更多的hemin从电极表面脱落,结果表明,在优化的实验条件下,DPV峰电流值与BPA浓度在1.0 nmol·L~(-1)至10.0μmol·L~(-1)的范围内呈线性关系,检测限为0.8 nmol·L~(-1)。本文还考察了传感器的稳定性与选择性,并将其用于实际样品中BPA的检测,结果满意。  相似文献

该文以DNA四面体纳米结构探针(TSP)为捕获探针,将辣根过氧化物酶标记的IgG抗体结合在纳米金颗粒表面(AuNPs-IgG-HRP)作为信号分子,构建了一种新型DNA甲基化电化学传感器。利用一步热变性法组装成TSP后,通过Au—S键固定在修饰纳米金颗粒的金电极表面,经过靶标DNA杂交、5-甲基胞嘧啶(5-mc)抗体及AuNPs-IgG-HRP结合后,用差分脉冲伏安法(DPV)进行检测。采用循环伏安法(CV)和电化学阻抗谱(EIS)对修饰电极的构建过程进行电化学表征。探究了杂交时间、5-mc抗体浓度、IgG-HRP加入体积、氢醌(HQ)和过氧化氢(H2O2)浓度对传感器的影响。在最佳条件下,该传感器对甲基化DNA的线性响应范围为1.0×10-15~1.0×10-10 mol/L,检出限(S/N=3)为4.4×10-16 mol/L。该传感器具有良好的选择性和稳定性,为DNA甲基化检测提供了新方法。  相似文献

用溶胶-凝胶法以磷钼酸(MPA)的镍盐溶液水解钛酸四丁酯制备了NiPMo/TiO2催化剂.使用ICP、 XRD、 TG-DTA、 IR、 TPD-MS和微反应技术研究了催化剂的化学组成、热稳定性、化学吸附性质和催化反应性能.杂多钼酸盐与TiO2通过O2-在TiO2表面发生了键合.在623 K下,杂多阴离子仍保持原有的Keggin结构.CO2在Lewis酸位Ni(Ⅱ)和Lewis碱位Ni-O-Mo的桥氧协同作用下生成CO2卧式吸附态Ni(Ⅱ)←O-(CO)←(O--Ni).丙烯有多种吸附态在催化剂上吸附.在563 K、 1 MPa和空速1500 h-1的反应条件下,丙烯的摩尔转化率为3.2%,产物MAA选择性为95%.  相似文献

A new and simple synthesis of novel N-protected methyl 5-substituted-4-hydroxypyrrole-3-carboxylates, which exist in equilibrium with their 4-oxo tautomers, has been developed in two steps starting from N-protected α-amino acids. The key intermediates are enaminones, which can also be isolated, characterized, and used for the construction of other functionalized heterocycles, before they spontaneously decompose to pyrrole products. 4-Hydroxypyrroles are prone to partial aerial oxidation but can be efficiently alkylated or reduced to stable polysubstituted pyrrolidine derivatives.  相似文献

The chemoselectivity in the intramolecular CH insertion of various diazosulfonamides has been experimentally studied. The results reveal that the aliphatic 1,4-, 1,5-, or 1,6-C(sp3)?H insertions of diazosulfonamides are not accessible, while the aromatic 1,5-C(sp2)?H insertion can be realized specifically by adjusting the diazo-adjacent group. In addition, the general chemoselectivities in the intramolecular CH insertions of diazosulfonyl compounds are summarized. Generally, diazosulfones undergo both aromatic 1,5-C(sp2)?H and aliphatic 1,5- and 1,6-C(sp3)?H insertions, while diazosulfonates undergo aliphatic 1,5- and 1,6-C(sp3)?H insertions. However, diazosulfonamides only undergo aromatic 1,5-C(sp2)?H insertion.  相似文献

The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula.  相似文献

KMnO4-mediated oxidative CN bond cleavage of tertiary amines producing secondary amine was introduced, which was trapped by electrophiles (acyl chloride and sulfonyl chloride) to form amides and sulfonamides. The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines.  相似文献

N-Heterocyclic carbene-palladacyclic complexes 3 were successfully achieved in a one-pot procedure under mild conditions. An efficient iodine-mediated oxidative Pictet-Spengler reaction in dimethyl sulphoxide (DMSO) using terminal alkynes as the 2-oxoaldehyde surrogate for the synthesis of aryl (9H-pyrido[3,4-b]indol-1-yl)methanones is described. The scope of the protocol includes the total synthesis of Fascaplysin, Eudistomins Y1 and Y2. The methodology is extended for preparing pyrrolo[1,2-a]-quinoxaline and indolo[1,5-a]quinoxaline derivatives. The utility of 1-aroyl-β-carbolines was demonstrated by performing palladium-catalyzed β-carboline directed ortho-C(sp2)-H functionalization of the phenyl ring with thiomethyl (SMe) group using DMSO as source and for accessing 4-aryl-canthin-6-ones.  相似文献
