An optical fiber assembly developed in our laboratory, which is based on detecting backward light scattering (BLS) signals, is now applied to detect the lead content in environmental samples. Due to effectively eliminating the interference of reflected light, this BLS signals based detection assembly can be used to determine analyte directly. In HAc-NaAc buffer medium (pH 4.8), the interaction of lead and sodium tetraphenylboron (TPB) in the presence of polyethylene glycol (PEG) yields large particles of ternary complex, resulting in strong enhanced backward light scattering (BLS) signals characterized at 371 nm. By measuring the BLS signals with the homemade optical fiber assembly coupled with a common spectrofluorometer, we found that the enhanced BLS intensity is proportional to lead content over the range of 0.03-1.0 μg ml−1 with the limit of determination (LOD) of 2.6 ng ml−1. Three artificial water samples containing various coexistent substances were detected with the recovery of 90.1-107.5%. Standard addition method was used to detect the lead content in drink tap water, and found that the lead is hardly to detect due to too low content. Prior enrichment should be made in order to detect river water samples, and it was found that the content of lead in Jialing River at Bebei Dock is about 14 ng ml−1, identical to the results using inductively coupled plasma atomic emission spectrometry (ICP-AES).  相似文献

Elucidation of physicochemical mechanisms of enzymatic processes is one of the main tasks of modern biology. High efficiency and selectivity of enzymatic catalysis are mostly ensured by conformational dynamics of enzymes and substrates. Here, we applied a stopped-flow kinetic analysis based on fluorescent spectroscopy to investigate mechanisms of conformational transformations during the removal of alkylated bases from DNA by ALKBH2, a human homolog of Escherichia coli AlkB dioxygenase. This enzyme protects genomic DNA against various alkyl lesions through a sophisticated catalytic mechanism supported by a cofactor (Fe(II)), a cosubstrate (2-oxoglutarate), and O2. We present here a comparative study of conformational dynamics in complexes of the ALKBH2 protein with double-stranded DNA substrates containing N1-methyladenine, N3-methylcytosine, or 1,N6-ethenoadenine. By means of fluorescent labels of different types, simultaneous detection of conformational transitions in the protein globule and DNA substrate molecule was performed. Fitting of the kinetic curves by a nonlinear-regression method yielded a molecular mechanism and rate constants of its individual steps. The results shed light on overall conformational dynamics of ALKBH2 and damaged DNA during the catalytic cycle.  相似文献

In this study, a method of dispersive liquid phase microextraction combined with the flame atomic absorption spectrometry was proposed for the determination of trace Hg using diphenylthiocarbazone as chelating reagent. Several factors which have effect on the microextraction efficiency of Hg, such as pH, extraction and dispersive solvent type and their volume, concentration of the chelating agent, extraction time were investigated, and the optimized experimental conditions were established. After extraction, the enrichment factor was 68. The detection limit of the method was 45 ng mL?1, and the relative standard deviation for eight determinations of 2 μg mL?1 Hg was 1.7%. The results for the determination of Hg in environmental water samples (tap water, well water, mineral water and Caspian sea water) have demonstrated the applicability of the proposed method.  相似文献

Summary: Polyurethane nano- and microparticles were synthesized by suspension-polyaddition technique, using aqueous polymerization medium. Castor oil, a vegetable triglyceride possessing hydroxyl groups was used as natural polyol and methylene diphenyl diisocyanate (MDI) as isocyanate. The levofloxacin, an antibacterial drug was used as model drug to measure the particles encapsulation efficiency. The effect of the addition of a second polyol, the poly(ethylene glycol) (PEG), and the stirring rate on the mean diameter and morphology of particles was also investigated. The poly(ethylene glycol) has an important effect in the reduction of particles size and their porosity. On the other hand, the poly(ethylene glycol) reduced the yield of encapsulation from 70% for the formulation without PEG to 20% for formulations with PEG. FTIR analysis confirmed the polyurethane formation. Dynamic light scattering study, transmission electron microscopy (TEM) and scanning electron microscopy (SEM) were used to determine the nanoparticles size and shape. 根据地质样品中钴、镍硫酸盐、镍黄铁矿、橄榄石、磁铁矿、菱铁矿5种相态不同的化学性质,选择了浸取剂,确定了最佳浸取条件,获取了各相态稳定的浸取率常数,并将遗传算法与钴、镍的化学物相分析相结合,建立了地质样品中钴、镍的五种相态同时测定的方法。方法简便、快速。其钴、镍的RSD(%)分别为:硫酸盐1.2、0.5,镍黄铁矿3.0、0.7,橄榄石1.0、0.25,磁铁矿2.9、2.4,菱铁矿5.5、2.4。平均相对误差小于5%。  相似文献

Phenolic compounds are a sort of common pollutants in water. Phenol index becomes an expedite indicator for the evaluation of the contamination level of water samples, in spite of the knowledge of the individual phenol and its derivatives are also important.In this work, an environmental friendly method for the determination of phenols, using a segmented flow system based on the conventional method's reactions without the liquid-liquid extraction step, was validated.Three linear dynamic ranges using C6H5OH: 1-10 μg l−1, 10-200 μg l−1 and 0.2-2.5 mg l−1, with a coefficient of variation lower than 2%, were obtained. Several method's performance parameters were determined: limits of detection, limits of quantification, precision through duplicate analysis and trueness using the reference materials purchased from LGC Promochem, RTC no. QCI-043-2 Lot:P1. Measurement uncertainty was evaluated using an interlaboratory approach based on proficiency testing data. Relative combined uncertainty for phenols in water samples, , of 0.054 were obtained, in according to those imposed by the Portuguese Legislation: target for 1 μg l−1 of phenol (surface waters) and target for 500 μg l−1 of phenol (wastewaters).A high efficiency reduction and elimination of reagents and wastes, reduction of analysis time and exposition of the analyst were also obtained.  相似文献

A new sample-treatment procedure has been developed for determination of total linear alkylbenzene sulfonate (LAS), i.e. homologues and isomers, in agricultural soil. The procedure involves two steps, ultrasound-assisted extraction of LAS from the samples with methanol then clean-up of the methanolic extracts and preconcentration of the LAS by solid-phase extraction on two adsorbent cartridges (SAX and C18). The ultrasound-assisted procedure reduces extraction time (10 min in contrast with 6–12 h for conventional Soxhlet extraction) and requires only small volumes of organic solvent. The effect of different variables interacting in the ultrasound-assisted extraction process was studied. Finally, separation and quantification of the homologues and isomers of LAS was performed by liquid chromatography with fluorescence detection (LC?FD). 2-Octylbenzenesulfonic acid sodium salt (Na-2ØC8-LAS) was used as internal standard. The proposed method was satisfactorily used for determination of LAS in agricultural soil samples from the fertile plain of Granada (Spain).

The analysis of samples contaminated by organic compounds is an important aspect of environmental monitoring. Because of the complex nature of these samples, isolating target organic compounds from their matrices is a major challenge. A new isolation technique, solid phase microextraction, or SPME, has recently been developed in our laboratory. This technique combines the extraction and concentration processes into one step; a fused silica fiber coated with a polymer is used to extract analytes and transfer them into a GC injector for thermal desorption and analysis. It is simple, rapid, inexpensive, completely solvent-free, and easily automated. To minimize matrix interferences in environmental samples, SPME can be used to extract analytes from the headspace above the sample. The combination of headspace sampling with SPME separates volatile and semi-volatile analytes from non-volatile compounds, thus greatly reducing the interferences from non-target compounds. This paper reports the use of headspace SPME to isolate volatile organic compounds from various matrices such as water, sand, clay, and sludge. By use of the technique, benzene, toluene, ethyl-benzene, and xylene isomers (commonly known as BTEX), and volatile chlorinated compounds can be efficiently isolated from various matrices with good precision and low limits of detection. This study has found that the sensitivity of the method can be greatly improved by the addition of salt to water samples, water to soil samples, or by heating. Headspace SPME can also be used to sample semi-volatile compounds, such as PAHs, from complex matrices.  相似文献

建立了微波辅助-微固相在线萃取/气相色谱-质谱联用(GC-MS)测定污泥中19种多氯联苯(PCBs)含量的分析方法。对微波辅助-微固相在线萃取条件进行优化,得出最优萃取条件为:萃取温度60℃,萃取时间25 min,解吸溶剂为乙酸乙酯,解吸剂用量150μL,解吸时间25 min。在优化条件下,方法的检出限为0.2~2.5 ng/g,相对标准偏差(RSD)小于14%,回收率为81.4%~102.1%。与传统的微波萃取、微固相萃取、超声萃取等方法相比,该方法集萃取、净化和浓缩于一体,极大地缩短了分析时间,适合于复杂环境样品体系中痕量PCBs的分析检测。  相似文献

在生产和贮存低分子量肝素的过程中,糖链上的硫酸酯基团会被水解而损失活性,因此肝素样品中常可以检测到游离的硫酸根离子,此外在生产过程中还会引入其他阴离子。为了检测低分子量肝素的质量稳定性,常用离子色谱检测低分子量肝素中游离的阴离子。但是相对分子质量较大的低分子量肝素会污染离子色谱柱和抑制器。为此发展了一种灵敏的毛细管电泳方法用于测定低分子量肝素中游离的SO~(2-)_4、Cl~-、F~-、PO~(3-)_4和OAc~-。不同于常用的背景吸收离子铬酸根,采用邻苯二甲酸根作为背景吸收电解质。与铬酸根相比,邻苯二甲酸根与所有待测阴离子电泳淌度匹配得更好,因此可以获得较窄的峰形。而且邻苯二甲酸根在230 nm检测波长下的摩尔吸光系数(4 754 L/(mol·cm))比铬酸根(254 nm,2 400 L/(mol·cm))高。因此,可以将毛细管电泳检测阴离子的灵敏度提高到与离子色谱法相当的水平。通过验证,该方法在0.002~1 mmol/L的浓度范围内具有较好的线性关系,日内(n=6)和日间(n=3)迁移时间和峰面积的相对标准偏差均小于3%。所测阴离子的检出限(S/N=3)和定量限(S/N=10)分别为0.4~0.8μmol/L和2~4μmol/L。该方法可用于监测低分子量肝素的稳定性。  相似文献

Determination of perfluorinated compounds (PFCs) is very important because of their potential hazards to the environment and human health. In present work, 1,8-bis (tetramethylguanidino)-naphthalene (TMGN), a superbasic proton sponge, was firstly employed as the matrix for quantitative detection of acidic PFCs in environmental water samples by Matrix-assisted Laser Desorption/Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS). Several acidic PFCs, such as perfluorooctanesulfonate (PFOS) and perfluorooctanoic acid (PFOA), were selected as model analytes for demonstrating the feasibility of the detection method. The results showed that deprotonated ions of these PFCs were detected without any other matrix ions interference. The achieved sensitivity with TMGN for PFOS detection was ten-fold higher than that with 1,8-bis (dimethyl-amino)-naphthalene (DMAN) which was used for the detection of fatty acid by MALDI-TOF-MS. The high sensitivity of this method made it feasible to monitor and quantify acidic PFCs in complicated environmental water samples. Furthermore, a novel combined strategy of solid phase extraction (SPE) followed by MALDI-TOF-MS detection was developed for quantifying PFCs in environmental water samples. The calibration curves with a wide linear dynamic range (0.1-10 ng L−1 for PFOS, PFHxS, and PFBS, and 0.5-50 ng L−1 for PFOA, PFNA, and PFDA.) were obtained. The limit of detection (LOD) for PFOS of this method was 0.015 ng L−1 (a signal-to-noise ratio of 3), which was lower than the LOD (0.036 ng L−1) obtained by high-pressure liquid chromatography/tandem mass spectrometry (LC-MS/MS) method. Moreover, the strategy was used to detect the selected PFCs in water samples collected from Xiaoqinghe river and Gaobeidian wastewater. The achieved concentrations of PFCs were closed to those obtained by LC-MS/MS method. It is indicated that the proposed MALDI-TOF-MS method with TMGN as the matrix is much reliable and can be used as an alternative method to detect trace PFCs in environmental water samples.  相似文献

A flow injection analysis (FIA) technique for the determination of Fe(II) and total-Fe in environmental water samples has been developed with a high sensitivity. The resin used for preconcentration of iron was the macroporous resin, Amberlite XAD-4 functionalized by N-hydroxyethylethylenediamine (HEED) groups. The technique employed was FIA by combination of on-line chelate resin preconcentration and chemiluminescence detection (CL), using brilliant sulfoflavine and hydrogen peroxide reagent solutions. The interference by coexisting Fe(III) could be eliminated by addition of 1×10−6 mol of deferrioxamine B solution. The detection limits of Fe(II) and total-Fe were 0.80 and 0.36 nmol l−1 for 5.6-ml seawater samples with a concentration of 2 nmol l−1. The relative standard deviations for both samples were less than ±4%. A typical analysis for Fe(II) can be performed in 7.5 min. The technique was ascertained by comparing the analytical value of total-Fe with the certified value of Fe in the reference standard seawater CASS-3.  相似文献

Increased blood glucose in diabetic individuals results in the formation of advanced glycation end products (AGEs), causing various adverse effects on kidney cells, thereby leading to diabetic nephropathy (DN). In this study, the antiglycative potential of Swertiamarin (SM) isolated from the methanolic extract of E. littorale was explored. The effect of SM on protein glycation was studied by incubating bovine serum albumin with fructose at 60 °C in the presence and absence of different concentrations of swertiamarin for 24 h. For comparative analysis, metformin was also used at similar concentrations as SM. Further, to understand the role of SM in preventing DN, in vitro studies using NRK-52E cells were done by treating cells with methylglyoxal (MG) in the presence and absence of SM. SM showed better antiglycative potential as compared to metformin. In addition, SM could prevent the MG mediated pathogenesis in DN by reducing levels of argpyrimidine, oxidative stress and epithelial mesenchymal transition in kidney cells. SM also downregulated the expression of interleukin-6, tumor necrosis factor-α and interleukin-1β. This study, for the first time, reports the antiglycative potential of SM and also provides novel insights into the molecular mechanisms by which SM prevents toxicity of MG on rat kidney cells.  相似文献

The voltammetric behavior of isoniazid and hydrazine at an overoxidized polypyrrole modified glassy carbon electrode has been investigated. The obtained cyclic voltammograms showed that their oxidation peaks were overlapped and it is difficult to determine them individually from a mixture without separation. To overcome this limitation, a procedure was proposed for resolution of overlapped voltammetric signals from mixtures of isoniazid and hydrazine. In this procedure, genetic algorithm was used for the selection of potentials for partial least squares. A feed forward artificial neural network with back propagation error algorithm was used to process the nonlinear relationship between currents and concentrations of hydrazine and isoniazid. The proposed method was suitable for determination of isoniazid in pharmaceutical tablets and detection of hydrazine impurities in the same samples.  相似文献

Along with histone modifications, RNA interference and delayed replication timing, DNA methylation belongs to the key processes in epigenetic regulation of gene expression. Therefore, reliable information about the methylation level of particular DNA fragments is of major interest. Herein the methylation level at two positions of the promoter region of the gene methylguanine‐O6‐DNA‐Methyltransferase (MGMT) was investigated. Previously, it was demonstrated that the epigenetic status of this DNA region correlates with response to alkylating anticancer agents. An automated CGE method with LIF detection was established to separate the six DNA fragments resulting from combined bisulfite restriction analysis of the methylated and non‐methylated MGMT promoter. In COBRA, the DNA was treated with bisulfite converting cytosine into uracil. During PCR uracil pairs with adenine, which changes the original recognition site of the restriction enzyme Taql. Artificial probes generated by mixing appropriate amounts of DNA after bisulfite treatment and PCR amplification were used for validation of the method. The methylation levels of these samples could be determined with high accuracy and precision. DNA samples prepared by mixing the corresponding clones first and then performing PCR amplification led to non‐linear correlation between the corrected peak areas and the methylation levels. This effect is explained by slightly different PCR amplification of DNA with different sequences present in the mixture. The superiority of CGE over PAGE was clearly demonstrated. Finally, the established method was used to analyze the methylation levels of human brain tumor tissue samples.  相似文献
