We have developed a new immunosensor based on self-assembly chemistry for highly sensitive and label-free detection of 2,4,6-trinitrotoluene (TNT) using surface plasmon resonance (SPR). A monolayer of amine terminated poly(ethylene glycol) hydrazinehydrochloride (PEG-NH2) thiolate was constructed on an activated gold surface and immobilized with trinitrophenyl-β-alanine (TNPh-β-alanine) by amide coupling method. The binding interaction of a monoclonal anti-TNT Ab (M-TNT Ab) with TNPh-β-alanine immobilized thiolate monolayer surface was monitored and evaluated for detection of TNT based on the principle of indirect competitive immunoreaction. Here, the competition between the self-assembled TNT derivative and the TNT in solution for binding with antibody yields in the response signal that is inversely proportional to the concentration of TNT in the linear detection range. With the present immunoassay format, TNT could be detected in the concentration range from 0.008 ng/ml (8 ppt) to 30 ng/ml (30 ppb). The response time for an immunoreaction was 2 min and one immunocycle could be done with in 4 min including surface regeneration. Bound antibodies could be easily eluted from the self-assembled immunosurface at high recoveries (more than 100 cycles) using pepsin solution without any damage to the TNT derivatives immobilized on the surface. The compact self-assembled monolayer was highly stable and prevented the non-specific adsorption of proteins on the surface favoring error free measurement.  相似文献

Designing a surface recognition layer with high anti-fouling ability, high affinity, and high specificity is an important issue to produce high sensitivity biosensing transducers. In this study, a self-assembled monolayer (SAM) consisting of a homogeneous mixture of oligo(ethylene glycol) (OEG)-terminated alkanethiolate and mercaptohexadecanoic acid (MHDA) on Au was employed for immobilizing troponin T antibody and applied in detecting cardiac troponin T by using surface plasmon resonance (SPR). The mixed SAM showed no phase segregation and exhibited human serum albumin resistance, particularly with an antibody-immobilized surface. X-ray photoemission spectra revealed that the chemical composition ratio of OEG to the mixed SAM was 69% and the OEG packing density was 82%. The specific binding of troponin T on the designed surface indicated a good linear correlation (R = 0.991, P < 0.0009) at concentrations lower than 50 μg mL−1 with the limit of detection of 100 ng mL−1 using a SPR measuring instrument. It is concluded that the mixed SAM functions as designed since it has high detection capability, high accuracy and reproducibility, as well as shows strong potential to be applied in rapid clinical diagnosis for label-free detection within 2 min.  相似文献

A polyclonal antibody against trinitrophenyl (TNP) derivatives was raised in rabbit, and the antibody was applied to detection of trinitrotoluene (TNT) using a surface plasmon resonance (SPR) biosensor. TNP-keyhole limpet hemocyanine (TNP-KLH) conjugate was injected into a rabbit, and a polyclonal anti-TNP antibody was realized after purification of the sera using protein G. Aspects of the anti-TNP antibody against various nitroaromatic compounds, such as cross-reactivities and affinities, were characterized. The temperature dependence of the affinity between the anti-TNP antibody and TNT was also evaluated. The quantification of TNT was based on the principle of indirect competitive immunoassay, in which the immunoreaction between the TNP-β-alanine-ovalbumin (TNP-β-ala-OVA) and anti-TNP antibody was inhibited in the presence of free TNT in solution. TNP-β-ala-OVA was immobilized to the dextran matrix on the Au surface by amine coupling. The addition of a mixture of free TNT to the anti-TNP antibody was found to decrease the incidence angle shift due to the inhibitory effect of TNT. The immunoassay exhibited excellent sensitivity for the detection of TNT in the concentration range of 3 × 10−11 to 3 × 10−7 g/ml. To increase the sensitivity of the sensor, anti-rabbit IgG antibody was used. After flowing the mixture of free TNT and anti-TNP antibody, anti-rabbit IgG antibody was injected, and the incidence angle shift was measured. Amplification of the signal was observed and the detection limit was improved to 1 × 10−11 g/ml.  相似文献

Molecular imprinting technology is applied in surface plasmon resonance spectroscopy for highly sensitive and selective detection of morphine(MO).As SPR-based sensor of MO,the preparation of molecular imprinted polymer is as follows: methacryhc acids(MAA),ethylene glycol dimethacrylate(EGDMA),azodiisobutyronitrile(AIBN) were used as functional monomer,cross-linker and initiator,respectively.The experiment results showed that morphine imprinted polymer had the performance of high sensitivity and specificity,i.e.the relative signal of SPR response was proportional to the concentration of morphine in acetonitrile in the range of 10-9mol/L to 10-6mol/L(1 ppb-1 ppm) with LOD of 10-10mol/L,and MO was distinguished from its analogs,such as codeine.  相似文献

This paper describes the fabrication and sensing characteristics of a self-assembled monolayer (SAM)-based surface plasmon resonance (SPR) immunosensor for detection of benzaldehyde (BZ). The functional sensing surface was fabricated by the immobilization of a benzaldehyde–ovalbumin conjugate (BZ–OVA) on Au-thiolate SAMs containing carboxyl end groups. Covalent binding of BZ–OVA on SAM was found to be dependent on the composition of the base SAM, and it is improved very much with the use of a mixed monolayer strategy. Based on SPR angle measurements, the functional sensor surface is established as a compact monolayer of BZ–OVA bound on the mixed SAM. The BZ–OVA-bound sensor surface undergoes immunoaffinity binding with anti-benzaldehyde antibody (BZ-Ab) selectively. An indirect inhibition immunoassay principle has been applied, in which analyte benzaldehyde solution was incubated with an optimal concentration of BZ-Ab for 5 min and injected over the sensor chip. Analyte benzaldehyde undergoes immunoreaction with BZ-Ab and makes it inactive for binding to BZ–OVA on the sensor chip. As a result, the SPR angle response decreases with an increase in the concentration of benzaldehyde. The fabricated immunosensor demonstrates a low detection limit (LDL) of 50 ppt (pg mL−1) with a response time of 5 min. Antibodies bound to the sensor chip during an immunoassay could be detached by a brief exposure to acidic pepsin. With this surface regeneration, reusability of the same sensor chip for as many as 30 determination cycles has been established. Sensitivity has been enhanced further with the application of an additional single-step multi-sandwich immunoassay step, in which the BZ-Ab bound to the sensor chip was treated with a mixture of biotin-labeled secondary antibody, streptavidin and biotin–bovine serum albumin (Bio–BSA) conjugate. With this approach, the SPR sensor signal increased by ca. 12 times and the low detection limit improved to 5 ppt with a total response time of no more than ca. 10 min. The thermo-sensitive swelling behaviour of hydrogels based on 2-(2-methoxyethoxy)ethyl methacrylate (MEO2MA) and synthesized by free radical polymerization has been investigated. The homopolymer hydrogel presents a low critical solution temperature (LCST) close to room temperature, which can be modulated by copolymerization with longer oligo(ethylene glycol) side chain methacrylates (OEGxMA). Then, three series of copolymeric hydrogels synthesized with MEO2MA and several low ratios of OEGxMA with Mn = 475 g mol−1 (OEG8MA), Mn = 1100 g mol−1 (OEG23MA) and Mn = 2080 g mol−1 (OEG45MA) were studied. In addition to conventional tetra(ethylene glycol) dimethacrylate (TEGDMA) crosslinker, the use of biodegradable oligo(caprolactone) dimethacrylate (OCLDMA) was also tested. The hydrophilic/hydrophobic balance, function of the short and the long OEG side chains, establishes a swelling behaviour depending on monomer composition, side chain length and temperature. The swelling at equilibrium increases with increasing the amount of OEGxMA in the copolymer and, at the same time, the collapsing moves progressively to higher temperature. The temperature dependent volumetric response of some of these hydrogels can be compare with the most extended thermo-sensitive hydrogel, which is based on poly(N-isopropylacrylamide) (P(N-iPAAm)). Therefore, they are potential candidates to replace it in applications where biocompatibility is required.  相似文献

A miniaturized capillary electrophoresis system coupled to a surface plasmon resonance (SPR) sensor on a microfluidic platform fabricated from PDMS is detailed. A previously described split-flow injection technique is first utilized to manipulate sample into the microfluidic chip, followed by separation within the fused-silica capillary and final off-capillary detection of analytes via SPR. Instead of using commercial SPR flow cells requiring relatively large detection volumes, samples of less than 1 nL volume are utilized. The interface between the CE system and SPR sensor made it possible to detect minute volumes of sample with minimal dispersion. The flow cell has the potential to be applicable to miniaturized flow-injection (FI) systems where submicroliter volumes of sample are frequently only available for analysis. The components present in solution, but not bound to the sensor surface, were also investigated. The sensitivity of the CE-SPR system was similar to that found in UV-spectrometric instruments and nonchromophoric components could also be measured.  相似文献

Biotin was attached via a spacer group to thiomethyl and thiosulfate derivatives of carboxymethylcellulose (CMC) by DCC coupling. A degree of substitution (DS) 0.3 per modified anhydroglucose unit was reached for the biotin moieties. These biotinylated CMCs are able to recognize the receptor protein streptavidin in aqueous solution as demonstrated by gel electrophoresis. Furthermore, they spontaneously form stable monolayers of 1–2 nm thickness on planar gold surfaces as evidenced by surface plasmon resonance (SPR). These monolayers irreversibly bind streptavidin. The observed increase of layer thickness of 6 nm resembles the size of a single streptavidin molecule. Consequently, dense streptavidin monolayers must have been formed. The resulting functional monolayers of CMC derivatives on gold are interesting platforms for biosensors, because they are very stable, hydrophilic, and formed in a reproducible way.  相似文献

The primary role of lung surfactant is to reduce surface tension at the air–liquid interface of alveoli during respiration. Axisymmetric drop shape analysis (ADSA) was used to study the effect of poly(ethylene glycol) (PEG) on the rate of surface film formation of a bovine lipid extract surfactant (BLES), a therapeutic lung surfactant preparation. PEG of molecular weights 3350; 8000; 10,000; 35,000; and 300,000 in combination with a BLES mixture of 0.5 mg/mL was studied. The adsorption rate of BLES alone at 0.5 mg/mL was much slower than that of a natural lung surfactant at the same concentration; more than 200 s are required to reach the equilibrium surface tension of 25 mJ/m2. PEG, while not surface active itself, enhances the adsorption of BLES to an extent depending on its concentration and molecular weight. These findings suggest that depletion attraction induced by higher molecular weight PEG (in the range of 8000 to 35,000) may be responsible for increasing the adsorption rate of BLES at low concentration. The results provide a basis for using PEG as an additive to BLES to reduce its required concentration in clinical treatment, thus reducing the cost for surfactant replacement therapy.  相似文献

Thermoresponsive polymeric colloids attract great attention in several biotechnological applications owing to their ability to manipulate drug release characteristics in a controlled manner. Majority of these applications utilized N‐isopropylacrylamide (NIPAM)‐based particles for controlled drug release. Despite its advantages, such as easy chemical modification and well‐documented literature, a potentially important bottleneck for NIPAM in biological applications is its tendency for nonspecific protein adsorption. Herein, we report a simple way to prepare novel thermoresponsive colloids composed of oligo(ethylene glycol) side chains via precipitation polymerization technique. In addition to displaying highly reversible thermal response, these particles also have considerably low nonspecific protein adsorption when compared with NIPAM counterparts. These crosslinked poly(ethylene glycol) ethyl ether methacrylate particles were characterized using dynamic light scattering and transmission electron microscopy. The effects of co‐monomer, crosslinker and initiator on particle characteristics were investigated. A novel cell-impermeable zinc sensor was synthesized by incorporating poly(ethylene glycol)(PEG) to N-(8-quinolyl)-p-aminobenzenesulfonamide (HQAS) group.The polymeric zinc sensor combines both valuable features of HQAS and PEG.The HQAS of the sensor has the similar functions to TSQ,and exhibits a good fluorescence response to Zn2+ but poor fluorescence responses to other metal ions.The PEG chain can prevent the sensor to permeate healthy cell membrane.The stained experiments with the yeast cells as model showed that the sensor cannot stain the healthy yeast cells,but only the damaged or died yeast cells. These results indicated the novel zinc probe was a typical cell-impermeable zinc sensor.  相似文献

We herein report the preparation of thermo- and redox-responsive branched polymers by the condensation reaction of three-armed oligo(ethylene glycol) (trisOEG) and cystamine (CA). The prepared branched polymers exhibited a soluble–insoluble transition at a lower critical solution temperature (LCST) and formed coacervate droplets through a liquid–liquid phase separation process. We then demonstrated control of the LCSTs of the branched polymers by varying the feed ratio of CA and the surrounding salt concentration close to body temperature. In addition, the trisOEG-cys x polymer formed coacervate droplets above the LCST, in which hydrophobic molecules were condensed. The redox response of the branched polymers was also investigated. Interestingly, the branched polymers degraded to low-molecular-weight materials (i.e., trisOEG) in the presence of dithiothereitol as a reducing agent through cleavage of the disulfide bond of CA. This facile preparation of branched polymers is expected to be valuable in the context of functional biomedical materials and modifiers for materials surfaces, such as the bases for drug delivery carriers and separation materials. © 2018 Wiley Periodicals, Inc. The immobilisation of biological recognition elements onto a sensor chip surface is a crucial step for the construction of biosensors. While some of the optical biosensors utilise silicon dioxide as the sensor surface, most of the biosensor surfaces are coated with metals for transduction of the signal. Biological recognition elements such as proteins can be adsorbed spontaneously on metal or silicon dioxide substrates but this may denature the molecule and can result in either activity reduction or loss. Self assembled monolayers (SAMs) provide an effective method to protect the biological recognition elements from the sensor surface, thereby providing ligand immobilisation that enables the repeated binding and regeneration cycles to be performed without losing the immobilised ligand, as well as additionally helping to minimise non-specific adsorption. Therefore, in this study different surface chemistries were constructed on SPR sensor chips to investigate protein and DNA immobilisation on Au surfaces. A cysteamine surface and 1%, 10% and 100% mercaptoundecanoic acid (MUDA) coatings with or without dendrimer modification were utilised to construct the various sensor surfaces used in this investigation. A higher response was obtained for NeutrAvidin immobilisation on dendrimer modified surfaces compared to MUDA and cysteamine layers, however, protein or DNA capture responses on the immobilised NeutrAvidin did not show a similar higher response when dendrimer modified surfaces were used.  相似文献

Surface plasmon resonance (SPR) spectroscopy is utilized to study in real-time and, by label-free means, the reversible and quasi-irreversible adsorption of small ionic or neutral molecules, pharmaceuticals, and proteins on poly(dimethylsiloxane) (PDMS) surfaces. The SPR sensor is covered with 0.2% (w/v) PDMS in octane. During the timescale of a typical lab-on-a-chip analysis or an electrophoretic separation, it was found that small neutral components containing a hydrophobic part do not adsorb or absorb onto PDMS, while larger, water-soluble polymer-like materials like proteins generally irreversibly adsorb to PDMS. The technique can be used to monitor the kinetics of adsorption and desorption of the molecules. For the non-specific adsorption of teicoplanin to PDMS, a Langmuir-like adsorption isotherm was obtained (Kd = 32 ± 2 μmol L−1).  相似文献

Phage contamination is a very serious and unavoidable problem in modern fermentation industry.It is necessary to develop sensitive and rapid phage detection methods for the early detection of phage contamination.In the present work,a real-time,rapid,specific and quantitative phage T4 detection method based on surface plasmon resonance(SPR) technique has been introduced.Escherichia coli was immobilized onto the preformed MPA self-assembled monolayer(SAM) through the widely used EDC/NHS cross-linking reaction as the recognition element.The bacteria immobilization was verified efficiently through the electrochemical measurements and fluorescence microscopy observations.The specific adsorption was much stronger than the non-specific adsorption of phage T4 binding to the biosensor surface modified by E.coli,and the latter could be neglected.The detection sensitivity reached 1×10 7 PFU/mL within 10 min.Within the experimental phage concentrations,the linear correlation between the SPR response and the phage concentration was good.The results suggest that the SPR technique is a potentially powerful tool for the phage or other virus detections,as a label-free,real-time,and rapid method.  相似文献

An ultra-sensitive and highly selective parathion methyl(PM) detection method by surface plasmon resonance(SPR) combined with molecularly imprinted films(MIF) was developed. The PM-imprinted film was prepared by thermo initiated polymerization on the bare Au surface of an SPR sensor chip.Template PM molecules were quickly removed by an organic solution of acetonitrile/acetic acid(9:1,v/v), causing a shift of 0.58 in SPR angle. In the concentrations range of 10à13–10à10mol/L, the refractive index showed a gradual increase with higher concentrations of template PM and the changes of SPR angles were linear with the negative logarithm of PM concentrations. In the experiment, the minimum detectable concentration was 10à13mol/L. The selectivity of the thin PM-imprinted film against diuron,tetrachlorvinphose and fenitrothion was examined, but no observable binding was detected. The results in the experiment suggested that the MIF had the advantages of high sensitivity and selectivity.  相似文献

The poly-o-phenylenediamine (PoPD) nonconducting film and gold nanoparticles (AuNPs) were combined to fabricate AuNPs/PoPD film, which is used as a novel biocompatible interface for the immobilization of antibody and develop a simple and sensitive label-free immunoassay for the detection of the related antigen (human immunoglobulin G (IgG)). Surface plasmon resonance (SPR) and electrochemical methods were used to provide the real-time information about the polymer film growth, assembling of various sizes of gold nanoparticles, anti-human IgG antibody (anti-hIgG) immobilization and the antigen–antibody interaction. The microstructures of the PoPD and AuNPs/PoPD films were characterized by atomic force microscopy (AFM). These results demonstrated that AuNPs were uniformly dispersed on the porous surface of PoPD film, which formed a nano-structure biocompatible AuNPs/PoPD interface. The use of gold nanoparticles and PoPD film could enhance the immunoassay sensitivity and anti-nonspecific property of the resulting immunoassay electrode. Additionally, the reproducibility and preliminary application of anti-hIgG/AuNPs/PoPD/Au electrode for SPR detection of hIgG was also evaluated.  相似文献

We report in this study the presence of Janus particles, which are candidates for use with electronic color papers. We used negatively charged polystyrene particles (370 nm) as the core particles, and gold was then sputtered onto their packed monolayer under several conditions. The sputtered particles were next redispersed into the aqueous medium by gentle sonication. Gold nanoparticles localized on one side of the cores could also serve as seeds for subsequent shell growth by electroless gold plating. Through these treatments, a series of well-dispersed Janus particles were obtained with gold nanostructures of different size and shape only on one side. Their dispersions showed different colors originating from the surface plasmon resonance absorption of gold nanoparticles localized on the hemisphere. The particles obtained by this approach have potential applications such as in sensors and electronic color paper.  相似文献

A highly sensitive reverse sandwich immunoassay for the detection of human cardiac myoglobin (cMb) in serum was designed utilizing a gold nanoparticle (AuNP)-enhanced surface plasmon resonance (SPR) biosensor. First, a monoclonal anti-cMb antibody (Mab1) was covalently immobilized on the sensor surface. AuNPs were covalently conjugated to the second monoclonal anti-cMb antibody (Mab2) to form an immuno-gold reagent (Mab2-AuNP). The reverse sandwich immunoassay consists of two steps: (1) mixing the serum sample with Mab2-AuNP and incubation for the formation of cMb/Mab2-AuNP complexes and (2) sample injection over the sensor surface and evaluation of the Mab1/cMb/Mab2-AuNP complex formation, with the subsequent calculation of the cMb concentration in the serum. The biosensor signal was amplified approximately 30-fold compared with the direct reaction of cMb with Mab1 on the sensor surface. The limit of detection of cMb in a human blood serum sample was found to be as low as 10 pM (approx. 0.18 ng mL−1), and the inter-assay coefficient of variation was less than 3%. Thus, the developed SPR-based reverse sandwich immunoassay has a sensitivity that is sufficient to measure cMb across a wide range of normal and pathological concentrations, allowing an adequate estimation of the disease severity and the monitoring of treatment.  相似文献
