Noncatalysed anionic coupling of an arylsodium with haloarenes readily gives access to valuable biaryl structures. Compared to aryllithiums, the reaction with arylsodiums is faster and can be performed at lower temperature. The coupling with substituted haloarenes proceeds often with a high degree of regioselectivity. Since only cheap and readily available substrates are involved, this coupling is particularly suited for the large scale synthesis of basic biaryl building blocks.  相似文献

A highly regioselective allylic substitution of β‐silyl allylic alcohols has been achieved that provides the branched isomer as a single product. This high level of regiocontrol is achieved through the use of a vinyl silane group that can perform a Hiyama coupling providing 1,3‐disubstituted allylic amines. An unusual oxidative fragmentation product was also observed at elevated temperature that appears to proceed by a Fleming–Tamao‐type oxidation–elimination pathway.  相似文献

A survey of substitution reactions conducted in a phosphonium bistriflimide ionic liquid is presented. The results demonstrate high selectivity favoring substitution over typically competitive elimination and solvolytic processes even when challenging secondary and tertiary electrophiles are employed. The first reports of Kornblum substitution reactions in an ionic liquid are described that proceed with very high chemoselectivity in favor of nitro over nitroso products and elimination side products. The structure-reactivity study indicates that these reactions proceed through a narrow spectrum of pathways ranging from straight S(N)2 to a preassociation pathway along a saddle point that approaches the S(N)1 limit. The barrier to the formation of dissociated carbocations is attributed to the structural features of this ionic liquid that favor intervention of the associated nucleophile over dissociation, also preventing cross over to E1 processes. The lack of any basic entity in the phosphonium bistriflimide ionic liquid appears to prevent any potential base-mediated elimination reactions, which makes this a highly selective medium for use in general substitution reactions.  相似文献

Does the dehydrogenative coupling of aromatic compounds mediated by AlCl3 at high temperatures and also by FeCl3, MoCl5, PIFA, or K3[Fe(CN)6] at room temperature proceed by the same mechanism in all cases? With the growing importance of the synthesis of aromatic compounds by double C? H activation to give various biaryl structures, this question becomes pressing. Since some of these reactions proceed only in the presence of non‐oxidizing Lewis acids and some only in the presence of certain oxidants, the authors venture the hypothesis that, depending on the electronic structure of the substrates and the nature of the “catalyst”, two different mechanisms can operate. One involves the intermediacy of a radical cation and the other the formation of a sigma complex between the acid and the substrate. The goal of this Review is to encourage further mechanistic studies hopefully leading to an in‐depth understanding of this phenomenon.  相似文献

A series of novel P-chiral monophosphorus ligands exhibit efficiency in asymmetric Suzuki-Miyaura coupling reactions, enabling the construction of an array of chiral biaryl products in high yields and excellent enantioselectivities (up to 96% ee) under mild conditions. The carbonyl-benzooxazolidinone moiety in these chiral biaryl products allows facile derivatization for further synthetic applications. A computational study has revealed that a π-π interaction between the two coupling partners can enhance the enantioselectivity of the coupling reaction.  相似文献

The preparation and utilization of specific biaryl systems, particularly those which suffer hindered rotation, is a demanding goal not only in the synthesis of natural products and Pharmaceuticals, but also for example in the discovery of new reagents for asymmetric synthesis. This article will attempt to provide a topical review of modern, efficient processes for the specific preparation of biaryls. This appears to be of particular urgency, since, under the pressure of continually increasing demand, a wealth of new or modified synthetic approaches to the ever more important biaryl systems has been realized in recent years. The high standard which has been reached in this field is impressively demonstrated by regio- and stereoselective syntheses of important biaryl natural products such as steganone, ancistrocladine, ancistrocladisine, and dioncophylline A. Besides the utilization of asymmetric induction in the actual coupling step, the thermodynamically or kinetically controlled torsion of biaryl axes belongs to the most important concepts discussed.  相似文献

The utility of the Suzuki cross-coupling to synthesize biaryl compounds is expanded herein to include reactions of resin-supported chloropyrimidines with boronic acids. In particular, an efficient method is described for the synthesis of a library of biaryl compounds from solid-supported chloropyrimidines. The Suzuki reaction was performed in an inert atmosphere using Pd(2)(dba)(3)/P(t-Bu)(3) as catalyst, spray-dried KF as base, and THF as solvent. The reaction was allowed to proceed overnight at 50 degrees C. Upon cleavage with acid, a library of 4-(substituted amino)-6-arylpyrimidines was obtained in moderate yield and high purity.  相似文献

A palladium-assisted coupling reaction of aryl triflate with arene was investigated, and a novel Pd reagent prepared from equimolar Pd(OAc)2, 1,3-Bis[diphenylphosphino]propane (DPPP), and Bu3P was developed. This method is useful for intramolecular biaryl coupling reactions, not only between aryl triflate and arene (triflate-amide), but also between aryl halide and arene (halo-amide).  相似文献

Described herein is the Pd(0)-catalyzed coupling cyclization reaction of 1,2-allenyl ketones with organic halides leading efficiently and conveniently to not only 2,3,4- and 2,3,5-trisubstituted furans but also 2,3,4,5-tetrasubstituted furans. Furthermore, this method showed high substituent-loading capability and tolerance of various substituents. The reactions of 1,2-allenyl ketones 1 e, 1 p, 1 q, and deuterated [D]1 c were performed for a mechanistic study, which demonstrated that instead of an enolization pathway, the reaction may proceed via the intermediacy of dienolate palladium and intramolecular nucleophilic attack on the pi-allyl palladium intermediate by the carbonyl oxygen.  相似文献

Alkynes have two sets of mutually orthogonal π‐bonds that are different from the π‐bonds of alkenes. These π‐bonds are able to bond with transition metal compounds. Alkynes easily bond with the various kinds of compounds having a π‐bond such as carbon monoxide, alkenes, other alkynes and nitriles in the presence of the transition metal compounds. The most representative reaction of alkynes is called the Pauson–Khand reaction. The Pauson–Khand reactions include the cyclization of alkynes with alkenes and carbon monoxide in the presence of cobalt carbonyls. Similar Pauson–Khand reactions also proceed in the presence of other transition metal compounds. These reactions are the first type of characteristic reaction of alkynes. Other various kinds of cyclizations with alkynes also proceed in the presence of the transition metal compounds. These reactions are the second type of characteristic reaction of alkynes. These include cyclooligomerizations and cycloadditions. The cyclooligomerizations include mainly cyclotrimerizations and cyclotetramerizations, and the cycloadditions are [2 + 2], [2 + 2 + 1], [2 + 2 + 2], [3 + 2], [4 + 2], etc., type cycloadditions. Alkynes are fairly reactive because of the high s character of their σ‐bonds. Therefore, simple coupling reactions with alkynes also proceed besides the cyclizations. The coupling reactions are the third type of characteristic reactions of alkynes in the presence of, mainly, the transition metal compounds. These reactions include carbonylations, dioxycarbonylations, Sonogashira reactions, coupling reactions with aldehydes, ketones, alkynes, alkenes and allyl compounds. The photoinduced regio‐ and enantioselective coupling of naphthols and derivatives thereof is achieved in the confined chiral coordination space of a RuII metalloligand based cage. The racemic or enantiopure cages encapsulate naphthol guests, which then undergo a regiospecific 1,4‐coupling, rather than the normal 1,1‐coupling, to form 4‐(2‐hydroxy‐1‐naphthyl)‐1,2‐napthoquinones; moderate stereochemical control is achieved with homochiral cages. The photoreactions proceed under both aerobic and anaerobic conditions but through distinct pathways that nevertheless involve the same radical intermediates. This unusual dimerization constitutes a very rare example of asymmetric induction in biaryl coupling by making use of coordination cages with dual functionality—photoredox reactivity and stereoselectivity.  相似文献

The catalytic and selective construction of carbon–carbon bonds for the generation of complex molecules is one of the most important tasks in organic chemistry. This was clearly highlighted by the 2010 Nobel Prize in Chemistry, which was awarded for the development of Pd‐catalyzed cross‐coupling reactions. The underlying concept of cross‐linking building blocks to generate molecular complexity can also be widely found in natural product biosynthesis. Impressive examples for such natural cross‐coupling reactions are biosynthetic processes for the assembly of biaryl moieties in natural products—highly efficient enzymatic reactions that often achieve synthetically yet unmatched selectivities. A library of phosphite‐pyridine ligands L1 – L12 a – g has been successfully applied for the first time in the Pd‐catalyzed allylic substitution reactions of several di‐ and trisubstituted substrates by using a wide range of C, N and O nucleophiles, among which are the little studied α‐substituted malonates, β‐diketones, and alkyl alcohols. The highly modular nature of this ligand library enables the substituents/configuration at the ligand backbone, and the substituents/configurations at the biaryl phosphite moiety to be easily and systematically varied. We found that the introduction of an enantiopure biaryl phosphite moiety played an essential role in increasing the versatility of the Pd‐catalytic systems. Enantioselectivities were therefore high for several hindered and unhindered di‐ and trisubstituted substrates by using a wide range of C, N and O nucleophiles. Of particular note were the high enantioselectivities (up to>99 % ee) and high activities obtained for the trisubstituted substrates S6 and S7 , which compare favorably with the best that have been reported in the literature. We have also extended the use of these new catalytic systems in alternative environmentally friendly solvents such as propylene carbonate and ionic liquids. Studies on the Pd‐π‐allyl intermediates provide a deeper understanding of the effect of ligand parameters on the origin of enantioselectivity.  相似文献

An efficient and rapid solution-phase combinatorial synthesis of the SAG library was developed. The salient features for this library synthesis is the application of carbothioamide-derived palladacycle-catalyzed Suzuki coupling reactions for the parallel synthesis of a series of pyridine-based biaryl aldehydes under aerobic conditions and a direct N-alkylation of carbamates using NaH as base in DMF in the presence of catalytic amount of water. The resultant library has been submitted to biological screening to evaluate their potential role in the regulation of Hedgehog pathway.  相似文献

The first enantioselective construction of a new class of axially chiral naphthyl‐indole skeletons has been established by organocatalytic asymmetric coupling reactions of 2‐naphthols with 2‐indolylmethanols (up to 99 % yield, 97:3 e.r.). This approach not only affords a new type of axially chiral heterobiaryl backbone, but also provides a new catalytic enantioselective strategy for constructing axially chiral biaryl scaffolds by making use of the C3‐electrophilicity of 2‐indolylmethanols.  相似文献

We have developed a modification of the Miyaura arylboronate synthesis(1a) by substituting a ligandless palladium catalyst for PdCl(2)(dppf). Palladium acetate, free of ligand, was found highly effective for such coupling reactions. This modified procedure is advantageous over the original Miyaura synthesis in ease of workup, catalyst removal, and low catalyst cost. Furthermore, the boronates formed in this manner can be used directly for Suzuki coupling reactions in a one-pot fashion. The biaryl products have improved impurity profiles and reduced heavy metal contamination.  相似文献

Aromatization of 4‐cyano‐3‐oxotetrahydrothiophene by sulfuryl chloride gives the new building block 4‐cyano‐3‐pyrrolidylthiophene, which forms unsymmetrical regioregular oligothiophenes with a strict alternation of the donor and acceptor groups along the conjugated system. The self‐coupling reactions that form the oligomers are shown to proceed by a regioselective electrophilic aromatic substitution mechanism involving a stabilized Wheland intermediate.  相似文献

Copper-catalyzed reactions of vinyldiazoacetates with imines occur via a pathway in which the activated imine undergoes electrophilic addition to the vinyldiazo compound, whereas reactions catalyzed by rhodium(II) proceed through a metal carbene to an intermediate iminiumylide.
