A polychromatic action spectrum for the induction of an ultraviolet-absorbing/screening mycosporine-like amino acid (MAA) has been determined in a filamentous and heterocystous nitrogen-fixing rice-field cyanobacterium, Anabaena sp. High-performance liquid chromatographic (HPLC) studies revealed the presence of only one type of MAA, which was identified as shinorine, a bisubstituted MAA containing both glycine and serine groups having a retention time at 2.8 min and an absorption maximum at 334 nm. Exposure of cultures to simulated solar radiation in combination with various cut-off filters (WG 280, 295, 305, 320, 335, 345, GG 400, 420, 455, 475, OG 515, 530, 570, RG 645, 665 and a broad-band filter, UG 11) clearly revealed that the induction of the MAA takes place only in the UV range. Photosynthetic active radiation (PAR) had no significant impact on MAA induction. The ratio of the absorption at 334 nm (shinorine) to 665 nm (chlorophyll a) and the action spectrum also showed the induction of MAA to be UV dependent peaking in the UV-B range at around 290 nm. The results indicate that the studied cyanobacterium, Anabaena sp. may protect itself from deleterious short wavelength solar radiation by its ability to synthesize a mycosporine-like amino acid in response to UV-B radiation and thereby screen the negative effects of UV-B.  相似文献

In the present investigation we show that the cyanobacterium Anabaena variabilis PCC 7937 produces a single mycosporine-like amino acid (MAA), shinorine (retention time = 2.3 min and absorption maximum at 334 nm) when isolated and purified by HPLC. Although there was significant induction of MAA synthesis from its initial value under 395 or 320 nm cutoff filters, MAA induction was significantly more pronounced in samples covered with 295 nm cutoff filters after 72 h of exposure. Heat as a stress factor had no effect on MAA induction with or without UV radiation. In contrast, salt and ammonium treatment had synergistic effects with UV stress. MAA synthesis was also induced by salt and ammonium in a concentration-dependent manner without UV stress in samples covered with 395 nm cutoff filters. The results indicate that MAAs may have other functions in addition to photoprotection in this organism.  相似文献

Using a high-resolution reverse-phase liquid chromatography method we found that the tissues of the hermatypic coral Pocillopora capitata (collected in Santiago Bay, Mexico) contain a high diversity of primary and secondary mycosporine-like amino acids (MAAs) typical of some reef-building coral species: mycosporine–glycine, shinorine, porphyra-334, mycosporine–methylamine–serine, mycosporine–methylamine–threonine, palythine–serine, palythine and one additional novel predominant MAA, with an absorbance maximum of 320 nm. Here we document the isolation and characterization of this novel MAA from the coral P. capitata. Using low multi-stage mass analyses of deuterated and non deuterated compounds, high-resolution mass analyses (Time of Flight, TOF) and other techniques, this novel compound was characterized as palythine–threonine. Palythine–threonine was also present in high concentrations in the corals Pocillopora eydouxi and Stylophora pistillata indicating a wider distribution of this MAA among reef-building corals. From structural considerations we suggest that palythine–threonine is formed by decarboxylation of porphyra-334 followed by demethylation of mycosporine–methylamine–threonine.  相似文献

Laser flash photolysis of ketone 1 in argon‐saturated methanol yields triplet biradical 1BR (τ = 63 ns) that intersystem crosses to form photoenols Z‐1P (λmax = 350 nm, τ ~ 10 μs) and E‐1P (λmax = 350 nm, τ > 6 ms). The activation barrier for Z‐1P re‐forming ketone 1 through a 1,5‐H shift was determined as 7.7 ± 0.3 kcal mol?1. In contrast, for ketone 2, which has a less sterically hindered carbonyl moiety, laser flash photolysis in argon‐saturated methanol revealed the formation of biradical 2BR (λmax = 330 nm, τ ~ 303 ns) that intersystem crosses to form photoenol E‐2P (λmax = 350 nm, τ > 42 μs), but photoenol Z‐2P was not detected. However, in more viscous basic H‐bond acceptor (BHA) solvent, such as hexamethylphosphoramide, triplet 2BR intersystem crosses to form both Z‐2P (λmax = 370 nm, τ ~ 1.5 μs) and E‐2P. Thus, laser flash photolysis of ketone 2 in methanol reveals that intersystem crossing from 2BR to form Z‐2P is slower than the 1,5‐H shift of Z‐2P, whereas in viscous BHA solvents, the 1,5‐H shift becomes slower than the intersystem crossing from 2BR to Z‐2P. Density functional theory and coupled cluster calculations were performed to support the reaction mechanisms for photoenolization of ketones 1 and 2 .  相似文献

The formation of complexes at pH 4.7 of the Hg(II) with five monothiosemicarbazone and two dithiosemicarbazone has been studied. The mercury(II) reacts with monothiosemicarbazones of salicylaldehyde (λmax = 363 nm, E = 1.69 × 104liters · mol?1cm?1), pi-colinadehyde (λmax = 363 nm, E = 2.38 × 104liters · mol?1cm?1), 6-methyl-picolinaldehyde (λmax = 363 nm, E = 2.28 × 104liters · mol?1cm?1), di-2-pyridylketone (λmax = 380 nm, E = 2.08 × 104liters · mol?1cm?1), and o-naphthoquinone (λmax = 540 nm, E = 1.03 × 104liters · mol?1cm?1) and with dithiosemicarbazones of 1,4-dihydroxyphthalimide (λmax = 430 nm, E = 2.56 × 104liters · mol?1cm?1) and dipyridylglyoxal (λmax = 363 nm, E = 2.37 × 104liters · mol?1cm?1). A critical comparison of the stoichiometry and apparent stability constant of complexes with mono- and dithiosemicarbazones is given.  相似文献

The photoirradiation of trans‐ and cis‐poly(dimethylsilylenephenylenevinylene)s gave cis‐rich mixtures at equilibrium states. The degree of the photoisomerization could be exactly evaluated by comparing the UV spectra of the photoirradiated solutions with those of the trans and cis polymers. The geometric configuration of the trans and cis polymers was thermally stable and hardly changed even though they were heated. The trans and cis polymers exhibited different emission properties; e.g., trans polymer: λmax = 400 nm, quantum yield = 3.4×10–3; cis polymer: λmax = 380 nm, quantum yield = 1.5×10–3.  相似文献

To test the molecular exciton theory for heterodimeric chromophores, various heterodimers and clusters, in which two different dyes were stacked alternately, were prepared by hybridizing two oligodeoxyribonucleotides (ODNs), each of which tethered a different dye on D ‐threoninol at the center of the strand. NMR analyses revealed that two different dyes from each strand were stacked antiparallel to each other in the duplex, and were located adjacent to the 5′‐side of a natural nucleobase. The spectroscopic behavior of these heterodimers was systematically examined as a function of the difference in the wavelength of the dye absorption maxima (Δλmax). We found that the absorption spectrum of the heterodimer was significantly different from that of the simple sum of each monomeric dye in the single strand. When azobenzene and Methyl Red, which have λmax at 336 and 480 nm, respectively, in the single strand (Δλmax=144 nm), were assembled on ODNs, the band derived from azobenzene exhibited a small hyperchromism, whereas the band from Methyl Red showed hypochromism and both bands shifted to a longer wavelength (bathochromism). These hyper‐ and hypochromisms were further enhanced in a heterodimer derived from 4′‐methylthioazobenzene and Methyl Red, which had a much smaller Δλmax (82 nm; λmax=398 and 480 nm in the single‐strand, respectively). With a combination of 4′‐dimethylamino‐2‐nitroazobenzene and Methyl Red, which had an even smaller Δλmax (33 nm), a single sharp absorption band that was apparently different from the sum of the single‐stranded spectra was observed. These changes in the intensity of the absorption band could be explained by the molecular exciton theory, which has been mainly applied to the spectral behavior of H‐ and/or J‐aggregates composed of homo dyes. However, the bathochromic band shifts observed at shorter wavelengths did not agree with the hypsochromism predicted by the theory. Thus, these data experimentally verify the molecular exciton theory of heterodimerization. This coherent coupling among the heterodimers could also partly explain the bathochromicity and hypochromicity that were observed when the dyes were intercalated into the duplex.  相似文献

SPECTRAL AND PHOTOCHEMICAL PROPERTIES OF CURCUMIN   总被引:2,自引:0,他引:2  
Curcumin, bis(4-hydroxy-3-methoxyphenyl)-l,6-heptadiene-3,5-dione, is a natural yellow-orange dye derived from the rhizome of Curcuma longa, an East Indian plant. In order to understand the photobiology of curcumin better we have studied the spectral and photochemical properties of both curcumin and 4-(4-hydroxy-3-methoxy-phenyl)-3-buten-2-one (hC, half curcumin) in different solvents. In toluene, the absorption spectrum of curcumin contains some structure, which disappears in more polar solvents, e.g. ethanol, acetonitrile. Curcumin fluorescence is a broad band in acetonitrile (λmax= 524 nm), ethanol (λmax= 549 nm) or micellar solution (λmax= 557 nm) but has some structure in toluene (λmax= 460, 488 nm). The fluorescence quantum yield of curcumin is low in sodium dodecyl sulfate (SDS) solution (φ= 0.011) but higher in acetonitrile (φ= 0.104). Curcumin produced singlet oxygen upon irradiation (φ > 400 nm) in toluene or acetonitrile (Φ= 0.11 for 50 μM curcumin); in acetonitrile curcumin also quenched 1O2 (kq, = 7 × 106 M?1 s?1). Singlet oxygen production was about 10 times lower in alcohols and was hardly detectable when curcumin was solubilized in a D2O micellar solution of Triton X-100. In SDS micelles containing curcumin no singlet oxygen phosphorescence could be observed. Curcumin photogenerates superoxide in toluene and ethanol, which was detected using the electron paramagnetic resonance/spin-trapping technique with 5,5-dimethyl-pyrroline-.N-oxide as a trapping agent. Unidentified carbon-centered radicals were also detected. These findings indicate that the spectral and photochemical properties of curcumin are strongly influenced by solvent. In biological systems, singlet oxygen, superoxide and products of photodegradation may all participate in curcumin phototoxicity depending on the environment of the dye.  相似文献

2,2′-Dihydroxybenzophenone thiosemicarbazone forms complexes with Cu(II) (λmax = 385 nm, ? = 8.60 × 103 liter · mol?1 · cm?1); Ni(II) (λmax = 380 nm, ? = 15.4 × 103 liter · mol?1 · cm?1); Co(II) (λmax = 380 nm, ? = 12.3 × 103 liter · mol? · cm?1); and Fe(III) (λmax = 365 nm, ? = 7.9 × 103 liter · mol?1 · cm?1) and have been applied to the analysis of these metal ions in binary, ternary, and quaternary mixtures. The determination procedures are based exclusively on the different pH values of the formation complexes, hence the extraction step is not necessary.  相似文献

A phycocyanin (PC) with γmax= 632 nm (PC632) was isolated from extracts of the cyanobacterium Mastigocladus laminosus. The complex contained three polypeptides migrating in SDS-PAGE around 22 kDa. The N-termini of the three polypeptides are identical. They are homologous to rod-core linker polypeptides from this and another cyanobacterium and are identical to one of them. Reconstitution of PC (γmax=614–620 nm) with the 22 kDa polypeptide produced again the red-shifted PC632. Phycobilisomes isolated at pH 6 or 7 contain, if any, only traces of 22 kDa polypeptides. Origin and functions of the polypeptides are discussed, they are most likely proteolytic fragments of allophycocyanin-PC rod-core linkers that contain the PC-binding domain.  相似文献

A spectral-luminescent study of the hydrogel formed by the crosslinking of chitosan succinate and hyaluronic acid dialdehyde (HAD) macromolecules, which is a dosage form of the Lucentis drug (active ingredient, ranibizumab), has been conducted. The individual fluorescence (FL) spectra of Lucentis (λmax = 336 nm), HAD (λmax = 328 and 438 nm), and the hydrogel (λmax = 396 nm) have been obtained. An analysis of these spectra suggests the participation of carbonyl groups and proteins contained in HAD in its gelation with chitosan succinate. The diffusion of Lucentis from the hydrogel into a physiological solution was studied by a FL method to demonstrate the high prolonged-release properties of the new dosage form.  相似文献

We develop the chemistry of boron difluoride hydrazone dyes (BODIHYs) bearing two aryl substituents and explore their properties. The low-energy absorption bands (λmax=427–464 nm) of these dyes depend on the nature of the N-aryl groups appended to the BODIHY framework. Electron-donating and extended π-conjugated groups cause a redshift, whereas electron-withdrawing groups result in a blueshift. The title compounds were weakly photoluminescent in solution and strongly photoluminescent as thin films (λPL=525–578 nm) with quantum yields of up to 18 % and lifetimes of 1.1–1.7 ns, consistent with the dominant radiative decay through fluorescence. Addition of water to THF solutions of the BODIHYs studied causes molecular aggregation which restricts intramolecular motion and thereby enhances photoluminescence. The observed photoluminescence of BODIHY thin films is likely facilitated by a similar molecular packing effect. Finally, cyclic voltammetry studies confirmed that BODIHY derivatives bearing para-substituted N-aryl groups could be reversibly oxidized (Eox1=0.62–1.02 V vs. Fc/Fc+) to their radical cation forms. Chemical oxidation studies confirmed that para-substituents at the N-aryl groups are required to circumvent radical decomposition pathways. Our findings provide new opportunities and guiding principles for the design of sought-after multifunctional boron difluoride complexes that are photoluminescent in the solid state.  相似文献

Cyclic tetraaryl[5]cumulenes ( 1 a–f ) have been synthesized and studied as a function of increasing ring strain. The magnitude of ring strain is approximated by the extent of bending of the cumulenic core as assessed by a combination of X-ray crystallographic analysis and DFT calculations. Trends are observed in 13C NMR, UV-vis, and Raman spectra associated with ring strain, but the effects are small. In particular, the experimental HOMO-LUMO gap is not appreciably affected by bending of the [5]cumulene framework from ca. 174° (λmax=504 nm) in 1 a to ca. 178° (λmax=494 nm) in 1 f .  相似文献

π‐Conjugated polymers (Poly1–Poly3) containing a 2,2′‐bipyridine (bpy) unit were subjected to coordination to nickel and copper dithiolate for the purpose of manipulating the photophysical properties. The absorption maximum peak of Poly1 [maximum wavelength (λmax) = 446 nm] redshifted by 36 nm upon the coordination of bpy to NiCl2, which produced Poly1–NiCl2. A further bathochromic shift was observed in the spectrum of Poly1–mntNi [mntNi = (maleonitrile dithiolate)nickel; λmax = 499 nm] bearing the dithiolate ligand, which stemmed from the extension of the conjugated system over the nickel dithiolate moiety through the bpy unit. An increase in the [Ni]/[bpy] ratio in Poly1–mntNi rendered the original maximum peak at 446 nm smaller and the lower energy charge‐transfer peak at 499 nm larger; the isosbestic points remained at 380 and 475 nm. The green fluorescence (λmax = 504 nm) emitted from Poly1 markedly diminished upon the coordination of nickel dithiolate because of the effective energy transfer. The absorption maximum peak of Poly1–mntNi in chloroform at 499 nm blueshifted to 471 nm when the volume ratio of the chloroform/N,N‐dimethylformamide solvent reached 10:90. The coordination of nickel dithiolate to Poly2 and Poly3 also brought about redshifts of the absorption maximum peaks of as much as 55 and 61 nm, respectively. The absorption maximum peak of Poly1–(phenyldithiolate)nickel(pdtNi) (λmax = 474 nm) redshifted by 28 nm in comparison with that of Poly1, whereas the magnitude of the shift of Poly1–bis(thiophenoxide)nickel(btpNi) bearing two thiophenoxide ligands was 20 nm. Photoreactive and thermoresponsive N‐isopropylacrylamide (NIPAM)–surfmer copolymer hydrogels containing 4,4′‐di(6‐sulfato‐hexyloxy)azobenzene (DSHA) dianions are described. The functional hydrogels are obtained in a two steps. First a micellar aqueous solution of (11‐(acryloyloxy)undecyl)trimethylammonium bromide (AUTMAB) and NIPAM is exposed to 60Co‐gamma irradiation, and a thermoresponsive copolymer gel is obtained. Second, DSHA is included by shrinking the gel at 50 °C and subsequent reswelling in an aqueous solution of DSHA disodium salt at 20 °C. Reswelling is accompanied by electrostatic adsorption of DSHA dianions at the positively charged AUTMAB headgroups replacing the bromide ions. Gels containing trans‐DSHA are transparent yellow at room temperature (λmax = 370 nm), while gels containing cis‐rich DSHA are orange (λmax = 460 and 330 nm). Energy dispersive X‐ray measurements indicate that 41% of the bromide ions are exchanged if trans‐DSHA is used for adsorption, and only 7.5% if cis‐DSHA is used. The incorporation of DSHA lowers the lower critical solution temperature (LCST) from 34 to 32 °C. Below the LCST, DSHA can be switched from the trans‐ to the cis‐rich state and vice versa upon irradiation with UV (λ = 366 nm) or visible light (λ ≥ 450 nm). New through‐space cyano‐substituted poly(p‐arylenevinylene)s containing a [2.2]paracyclophane unit were synthesized by the Knoevenagel reaction. Polymers 5 and 7 have cyano groups at α‐positions and β‐positions from the dialkoxyphenylene unit, respectively. Their optical and electrochemical behaviors were investigated in detail in comparison with their model compounds. Polymers 5 and 7 exhibited through‐space conjugation via the cyclophane units. Polymer 5 showed greenish blue emission (λmax = 477 nm) in diluted solution with fluorescence quantum efficiency (?F) of only 0.007, whereas polymer 7 emitted in the bluish green region (λmax = 510 nm) with ?F of 0.32. © 2009 Wiley Periodicals, Inc. The effect of intermolecular proton transfer on the spectral properties of levofloxacin in the ground and excited electronic states was studied. The preferred direction of possible protolytic reactions induced by UV irradiation in this compound was studied. It was found that the proton transfer processes have a considerable effect on the capability of the compound to emit light and occur on the nanosecond timescale. The photochemical reactions of the tree forms of levofloxacin (pH: 4.0, 7.0, 10.0) were studied by laser flash photolysis and product studies. Irradiation at pH 4 yielded a pulse and transient (λmax = 395, 515, 575 nm) assigned to the protonated triplet. Irradiation at pH 7 yielded a transient species (λmax = 525, 610 nm) assigned to the neutral form. Protonation of the anionic singlet excited state was also observed (λmax = 440, 570, 680 nm).  相似文献

The complexation of Cu2+ by N-isopropyl-2-methyl-1,2-propanediamine (L) has been studied by potentiometric and spectrophotometric titration. The dominant complexes formed in this system are [CuL]2+, [CuL2]2+, [Cu2L2(OH)2]2+, and [CuL(OH)2]. The data were thoroughly tested for different models with [CuL(OH)]+, [CuL(OH)]+, [Cu(OH)]+, and [Cu2(OH)2]2+ as additional species. The importance of steric factors is indicated by the d-d* spectra: for [CuL2]2+, (λmax = 499 nm) the absorption maximum is shifted by 50 nm to high energies relative to [Cu(en)2]2+, (λmax = 549 nm), whereas the opposite is true for the 1:1 complexes ([CuL]2+ : λmax = 712 nm,s [Cu(en)]2+ : λmax = 660 nm).  相似文献

A new regioregular head‐to‐tail (HT)‐type polypyridine with methoxyethoxyethoxy (MEEO) side chains, HT‐PMEEOPy, was synthesized by means of Kumada‐Tamao coupling polymerization of a Grignard monomer with a Ni catalyst. Although the polymer was precipitated in THF during polymerization, multiangle laser light scattering (MALLS) analysis indicated that the weight‐average molecular weight (Mw) was about 25,000. The HT content in the polymer was 95%. A solution of HT‐PMEEOPy in CHCl3 was found to emit a strong blue light when the solution was irradiated with UV light; the UV‐vis absorption maximum (λmax) and photoluminescence maximum (λmax em) were at 392 and 460 nm, respectively. To clarify the effect of regioregularity of PMEEOPy on the photoluminescence, head‐to‐head (HH) PMEEOPy was synthesized by means of Yamamoto coupling polymerization. Evaluation of three subclasses of boron difluoride formazanate complexes bearing o‐, m‐, and p‐anisole N‐aryl substituents (Ar) as readily accessible alternatives to boron dipyrromethene (BODIPY) dyes for cell imaging applications is described. While the wavelengths of maximum absorption (λmax) and emission (λem) observed for each subclass of complexes, which differed by their carbon‐bound substituents (R), were similar, the emission quantum yields for 7 a – c (R=cyano) were enhanced relative to 8 a – c (R=nitro) and 9 a – c (R=phenyl). Complexes 7 a – c and 8 a – c were also significantly easier to reduce electrochemically to their radical anion and dianion forms compared to 9 a – c . Within each subclass, the o‐substituted derivatives were more difficult to reduce, had shorter λmax and λem, and lower emission quantum yields than the p‐substituted analogues as a result of sterically driven twisting of the N‐aryl substituents and a decrease in the degree of π‐conjugation. The m‐substituted complexes were the least difficult to reduce and possessed intermediate λmax, λem, and quantum yields. The complexes studied also exhibited large Stokes shifts (82–152 nm, 2143–5483 cm?1). Finally, the utility of complex 7 c (Ar=p‐anisole, R=cyano), which can be prepared for just a few dollars per gram, for fluorescence cell imaging was demonstrated. The use of 7 c and 4′,6‐diamino‐2‐phenylindole (DAPI) allowed for simultaneous imaging of the cytoplasm and nucleus of mouse fibroblast cells.  相似文献
